Cover Story: This study analyzed tumor-associated macrophages (TAMs) in intrahepatic cholangiocarcinoma (ICC) using integrated RNA sequencing data. TAMs exhibited significant heterogeneity, co-expressing features of both inflammatory (M1) and anti-inflammatory (M2) macrophages, with specific markers like TREM2 and CD9 identified and validated. Immune profiling revealed a reduced M1/M2 macrophage ratio and an altered adaptive immune landscape, consistent with an immunotolerant tumor microenvironment. Protein network analysis pinpointed the TREM2-LGALS3 axis as a potential therapeutic target. The findings highlight TREM2-positive TAMs as a promising target for immunotherapy in ICC via the TREM2-LGALS3 pathway. View this paper
