Cover Story: Our research indicates a defect in proinsulin (PI) chaperone GRP94 leads to an accumulation of mishandled proinsulin in β-cells. This results in ER stress, inflammatory pathway activation, int-proteasome assembly, and GRP94-PI complex secretion. Simultaneously, increased MHC-I expression on β-cells enhances their visibility to immune cells. The secreted GRP94-PI complexes are taken up by antigen-presenting cells and cross-presented to CD8+ T-cells to trigger an immune attack against β-cells. View this paper
