Humic Substances Promote the Activity of Enzymes Related to Plant Resistance

The extensive use of pesticides has significant implications for public health and the environment. Breeding crop plants is the most effective and environmentally friendly approach to improve the plants’ resistance. However, it is time-consuming and costly, and it is sometimes difficult to achieve satisfactory results. Plants induce defense responses to natural elicitors by interpreting multiple genes that encode proteins, including enzymes, secondary metabolites, and pathogenesis-related (PR) proteins. These responses characterize systemic acquired resistance. Humic substances trigger positive local and systemic physiological responses through a complex network of hormone-like signaling pathways and can be used to induce biotic and abiotic stress resistance. This study aimed to assess the effect of humic substances on the activity of phenylalanine ammonia-lyase (PAL), peroxidase (POX), and β-1,3-glucanase (GLU) used as a resistance marker in various plant species, including orange, coffee, sugarcane, soybeans, maize, and tomato. Seedlings were treated with a dilute aqueous suspension of humic substances (4 mM C L−1) as a foliar spray or left untreated (control). Leaf tissues were collected for enzyme assessment two days later. Humic substances significantly promoted the systemic acquired resistance marker activities compared to the control in all independent assays. Overall, all enzymes studied in this work, PAL, GLUC, and POX, showed an increase in activity by 133%, 181%, and 149%, respectively. Among the crops studied, citrus and coffee achieved the highest activity increase in all enzymes, except for POX in coffee, which showed a decrease of 29% compared to the control. GLUC exhibited the highest response to HS treatment, the enzyme most prominently involved in increasing enzymatic activity in all crops. Plants can improve their resistance to pathogens through the exogenous application of HSs as this promotes the activity of enzymes related to plant resistance. In the context of increasing demand for sustainable floriculture, this study evaluated the effects of salicylic acid (SA) on phenotypic traits of poinsettia (Euphorbia pulcherrima Willd.). A factorial experiment was conducted in a commercial glasshouse using rooted poinsettia cuttings treated with three SA concentrations (10−3, 10−4, 10−5 M) applied via foliar or root application. Morphological parameters, colorimetric traits (CIELAB), canopy development, and biomass accumulation were assessed throughout the cultivation cycle. SA had no significant influence on the plant height, leaf number, or biomass of stems, leaves, and roots. However, notable phenotypic changes were observed. Foliar applications, particularly at 10−5 M, induced visible changes in leaf and bract color, including reduced brightness, saturation, and red pigmentation, especially in newly developed tissues. Conversely, root applications had milder effects and were generally associated with a more stable bract color. The 10−4 M root treatment promoted greater bract surface and color saturation. Canopy expansion and dry matter accumulation were also influenced by SA in a dose- and method-dependent manner, with high-dose foliar treatments (10−3 M) exerting suppressive effects. These findings suggest that the application mode and concentration of SA are critical in modulating ornamental quality traits, with low-to-moderate doses—particularly via root application—offering promising strategies to enhance plant performance in sustainable poinsettia cultivation. Cucumbers (Cucumis sativus L.) are highly sensitive to cold, but grafting onto cold-tolerant rootstocks can enhance their low-temperature resilience. This study investigates the physiological and molecular mechanisms by which exogenous vitamin C (Vc) mitigates cold stress in grafted cucumber seedlings. Using cucumber ‘Chiyu 505’ as the scion and pumpkin ‘Chuangfan No.1’ as the rootstock, seedlings were grafted using the whip grafting method. In the third true leaf expansion stage, seedlings were foliar sprayed with Vc at concentrations of 50, 100, 150, and 200 mg L−1. Three days after initial spraying, seedlings were subjected to cold stress (8 °C) for 3 days, with continued spraying. After that, morphological and physiological parameters were assessed. Results showed that 150 mg L−1 Vc treatment was most impactive, significantly reducing the cold damage index while increasing the root-to-shoot ratio, root vitality, chlorophyll content, and activities of antioxidant enzymes (SOD, POD, CAT). Moreover, this treatment enhanced levels of soluble sugars, soluble proteins, and proline compared to control. However, 200 mg L−1 treatment elevated malondialdehyde (MDA) content, indicating potential oxidative stress. For transcriptomic analysis, leaves from the 150 mg L−1 Vc and CK treatments were sampled at 0, 1, 2, and 3 days of cold stress. Differential gene expression revealed that genes associated with photosynthesis (LHCA1), stress signal transduction (MYC2-1, MYC2-2, WRKY22, WRKY2), and antioxidant defense (SOD-1, SOD-2) were initially up-regulated and subsequently down-regulated, as validated by qRT-PCR. Overall, we found that the application of 150 mg L−1 Vc enhanced cold tolerance in grafted cucumber seedlings by modulating gene expression networks related to photosynthesis, stress response, and the antioxidant defense system. This study provides a way for developing Vc biostimulants to enhance cold tolerance in grafted cucumbers, improving sustainable cultivation in low-temperature regions. Full article

Enhancing the nutritional content of crops is crucial for safeguarding human health and mitigating global hunger. A viable method for attaining this goal is the planned implementation of various agronomic practices, including tillage and nutrient provision. A field experiment was executed at the Hungarian University of Agriculture and Life Sciences in Gödöllő in the 2023 and 2024 growing seasons. The study aimed to assess the effects of foliar nutrient supply and soil tillage methods on the grain nutritional composition and mineral content of winter barley. Employing a split-plot design with three replications, the experiment included four nutrient treatments (control, bio-cereal, bio-algae, and MgSMnZn blend) and two soil tillage types (i.e., plowing and cultivator). The results indicated that while protein content was not influenced by the main effects of nutrients and tillage, the levels of β-glucan, starch, crude ash, and moisture content were significantly (p < 0.05) affected by the nutrient treatments and by growing year, treated as a random factor. Notably, bio-algae and bio-cereal nutrients, combined with cultivator tillage, enhanced β-glucan content. All applied nutrient treatments increased the level of starch compared to the control. With regard to grain mineral content, the iron and zinc content responded to the nutrient supply, tillage, and growing year. However, applying a multiple-nutrient composition-based treatment did not increase iron and zinc levels, suggesting that individual applications may be more effective for increasing the content of these minerals in grains. Cultivator tillage improved iron and zinc levels. Moreover, manganese (Mn) and copper (Cu) were predominantly affected by nutrient availability and by growing seasons as a random factor. Therefore, to improve grain quality, this study emphasizes the significance of proper nutrient and tillage methods by focusing on the intricate relationships between agronomic techniques and environmental factors that shape barley’s nutritional profile. Weeds pose a significant challenge to agricultural productivity, requiring control strategies that are both effective and environmentally sustainable. Therefore, this study evaluated the inhibitory potential of aqueous extracts from Ricinus communis L. leaves to manage the weeds Oryza sativa L. (weedy rice) and Cyperus ferax. Extracts were obtained through pressurized liquid extraction using water as the solvent. Bioassays were conducted during pre- and post-emergence stages by foliar spraying 15 and 30 days after sowing (DAS). The effect of extraction time (1–30 min) on inhibitory efficacy was also assessed. Chemical profiles of the extracts were characterized using high-performance liquid chromatography. The extracts significantly inhibited seed germination, with suppression rates reaching 92.7%. Plant growth was also diminished, particularly with earlier treatments (at 15 DAS), resulting in reductions of up to 32% and 53% in shoot length, and 69% and 73% in total dry mass for O. sativa L. and C. ferax, respectively. Mortality rates of O. sativa L. and C. ferax reached 64% and 58%, respectively. Phenolic compounds were identified in the extracts, and higher concentrations were observed at shorter extraction times. These findings underscore the potential of R. communis L. leaf extracts as an ecologically sustainable alternative for weed management, providing an effective and natural approach that may reduce reliance on synthetic herbicides and mitigate their environmental impact. Tomato crops are treated with high concentrations of synthetic fertilizers and insecticides to increase yields, but the careless use of these chemicals harms the environment and human health and affects plant pathogen resistance. The effect of foliar spray of three concentrations of chitosan (500, 1000, and 2000 mg L−1) on plant growth, yield, fruit quality, and physiological performance in two tomato varieties (Floradade and Candela F1) was studied. Physiological traits such as photosynthesis, chlorophyll content, and leaf area index of the plants were positively affected by chitosan, an effective compound that biostimulates growth, with increases in biomass of organs with respect to the control treatment. Chitosan also improved tomato quality, such as increases in polyphenols, antioxidant capacity, flavonoids, carotenoids, vitamin C, and total soluble solids in both tomato varieties. Finally, yield increased by 76.4% and 65.4% in Floradade and Candela F1, respectively. The responses of tomato plants to chitosan application were different depending on the variety evaluated, indicating a differential response to the biostimulant. The use of chitosan in agriculture is a tool that has no negative effects on plants and the environment and can increase the productive capacity of tomato plants. Full article

Biostimulants, particularly single amino acids, can increase plant growth and crop quality, gaining significant attention. This study investigates the effects of 10 amino acids via root/foliar application on the growth, quality, taste, and volatile flavor of mini-watermelons and compares the differences between the application methods. Here, we employed electronic noses, electronic tongues, and gas chromatography–ion mobility spectrometry to investigate these effects. Root application excels in fruit growth and pectin accumulation, while foliar application boosts soluble protein and specific nutrients. Specifically, root application (except for Val) significantly increases fruit weight, with Gly being most effective for longitudinal diameter, while most amino acids (except Val/Lys) promote transverse diameter. Pectin content shows bidirectional regulation: root application of Glu/Gly/Lys/Pro/Trp/Val enhances pectin, whereas foliar application inhibits it. For taste indices, most treatments improve soluble solids (except Glu root/Arg-Leu foliar), and Ala/Asp/Glu/Gly reduce titratable acids, optimizing the sugar–acid ratio. Foliar application is more efficient for soluble protein accumulation (Ala/Glu/Gly/Pro/Leu). For nutritional quality, except for Lys, all treatments increase vitamin C and widely promote total phenolics and lycopene, with only minor exceptions, and only Arg foliar application enhances ORAC. Additionally, the results revealed that root-applied lysine and valine greatly raised the levels of hexanal and 2-nonenal, whereas foliar-applied valine significantly increased n-nonanal and (Z)-6-nonenal. Overall, we found that amino acids can considerably improve mini-watermelon production, quality, taste, and antioxidant capacity, providing theoretical and practical references for their widespread use in agriculture. Although favorable effects of Selenium nanoparticles (SeNPs or nSe) in tomato have been reported, research has concentrated on stress alleviation and disease management. From the above it is noticeable that the effect of NPs varies greatly depending on the model plant, nanoparticle (concentration, size, shape), and application (foliar or drenching). For this reason, the objective of this study was to investigate the impact of biostimulating tomato plants under no stressor conditions (Solanum lycopersicum cv. ‘Pomodoro’ L.) with SeNPs on morpho-physiological and fruit quality parameters. Three doses of Selenium nanoparticles (5, 15, and 30 mg L−1), and a control were applied via a foliar application after transplanting. The results indicate that a 5 mg L−1 SeNP treatment improved the growth and yield of the tomato, with the exception of the root length and leaf weight. Moreover, all doses modified the evaluated physiology, bioactive compounds, and fruit quality parameters. This research helped in understanding the SeNPs’ effect on tomato plants in greenhouses under a no stressor condition. Full article

This study evaluates the agronomic potential of two types of vermicompost—one produced solely from wine industry residues (WIR) and one incorporating sewage sludge (WIR + SS)—under rainfed and deficit irrigation conditions in Mediterranean vineyards. The vermicompost was obtained through a two-phase process involving initial thermophilic pre-composting, followed by vermicomposting using Eisenia fetida for 90 days. The conditions were optimized to ensure aerobic decomposition and maintain proper moisture levels (70–85%) and temperature control. This resulted in end products that met the legal standards required for agricultural use. However, population dynamics revealed significantly higher worm reproduction and biomass in the WIR treatment, suggesting superior substrate quality. When applied to grapevines, WIR vermicompost increased soil organic matter, nitrogen availability, and overall fertility. Under rainfed conditions, it improved vegetative growth, yield, and must quality, with increases in yeast assimilable nitrogen (YAN), sugar content, and amino acid levels comparable to those achieved using chemical fertilizers, as opposed to the no-fertilizer trial. Foliar analyses at veraison revealed stronger nutrient uptake, particularly of nitrogen and potassium, which was correlated with improved oenological parameters compared to the no-fertilizer trial. In contrast, WIR + SS compost was less favorable due to lower worm activity and elevated trace elements, despite remaining within legal limits. These results support the use of vermicompost derived solely from wine residues as a sustainable alternative to chemical fertilizers, in line with the goals of the circular economy in viticulture. The adoption of biological control strategies plays a crucial role in ensuring the sustainability of organic agricultural practices. A field experiment was conducted in 2023 and 2024 to evaluate the impact of biological treatments using lactic acid bacteria (LAB) Lactiplantibacillus plantarum and mycoparasitic fungus (MPF) Trichoderma virens applied through seed treatment and foliar application separately and in combination on agronomic characteristics and pea yield in organic cultivation. Seed treatment with LAB and MPF resulted in a notable improvement in shoot length and root dry weight, while an increase in root nodule number was observed exclusively with LAB. The combined application of MPF as a seed treatment and LAB as a foliar application at the flowering stage significantly enhanced pod weight per plant, seed number per pod and per plant, and seed weight compared to treatments with LAB applied as either a foliar or seed treatment separately, as well as the untreated control. However, the yield responses to individual and combined treatments under field conditions demonstrated variability and inconsistency. Protein content ranged from 21.24% to 21.61%, and no significant differences observed between treatments. This is the first field report directly comparing the effectiveness of treatments on organic pea production. The findings offer promising avenues for assessing the long-term impacts of these treatments on the sustainable intensification of pea cultivation. Full article

Foliar application of biostimulants can be a valid option to reach the goal of sustainable intensification in agriculture, especially in extensive crops such as durum wheat. However, due to the wide range of active ingredients and their mixtures available in the market, the need to select the most efficient product in a specific growing environment is of dramatic importance to achieve remarkable results in yield and grain quality. To analyze the potential of different active ingredients, a field trial was performed in two consecutive growing seasons (2023 and 2024) under Mediterranean climatic conditions. A randomized block design with three replicates was used. Durum wheat cultivar “Iride” was treated with the following five foliar biostimulants in comparison with the untreated control (T0): seaweed and plant extracts (T1); micronized vaterite (T2); culture broth of Pseudomonas protegens (T3); humic and fulvic acids (T4); organic nitrogen fertilizer (N 5%) containing glycine betaine (T5). Biostimulant treatment was applied at the end of tillering and at heading. Root length, chlorophyll content, grain yield, yield components and grain quality were measured and subjected to a one-way analysis of variance. As compared to the control, seaweed and plant extracts as well as micronized vaterite showed the best results in terms of grain yield (29% and 24% increase, respectively), root length (120% and 77% increase, respectively) and grain protein content (one percentage point increase, from approx. 12% to 13%). In arid regions, yields from safflower plants are appreciably lower than normal. Foliar application of zinc or glycine betaine has been reported to increase yields in other grown crops. A field experiment was conducted to compare the specific effects and mechanisms of foliar-applied zinc or glycine betaine on safflower yield in this study. Seven foliar spraying treatments were implemented, including a control (spraying water), three concentrations of zinc sulfate (Zn1: 0.6 g L−1, Zn2: 0.8 g L−1, Zn3: 1.0 g L−1), and three concentrations of glycine betaine (GB1: 0.23 g L−1, GB2: 0.47 g L−1, GB3: 0.70 g L−1). Results showed that Zn1 treatment had the highest grain yield at 2197 kg ha−1, which was 45.4% higher than the control. GB3 treatment resulted in a grain yield at 2127 kg ha−1, which was 40.8% higher than the control. The yield increase mechanism for the zinc treatment was primarily due to optimized plant morphology and improved photosynthetic performance, while glycine betaine improved yield mainly through antioxidant regulation. This study has important implications for water-saving and sustainable agriculture development in arid regions. Full article

To explore the effects of various zinc (Zn) fertilizer application methods and concentrations on foxtail millet quality and flavonoid biosynthesis, we used Zhangzagu 13 as the experimental material. The transcriptome and metabolome were used to examine variations in flavonoid biosynthesis and metabolism in foxtail millet under different Zn application methods. The results showed that different Zn application methods significantly increased the total polyphenol, carotenoid, total flavonoid, and Zn contents in the grains of foxtail millet. Under the basal soil application (S3) and foliar spray (F2) treatments, the total flavonoid content significantly increased by 45.87% and 64.40%, respectively, compared with that of CK. Basal soil Zn fertilization increased the flavonoid content of foxtail millet by up-regulating genes related to flavonoid metabolism and biosynthesis, including flavanone-3-hydroxylase, chalcone isomerase, and leucoanthocyanidin reductase. Foliar Zn application enhanced flavonoid content by up-regulating genes involved in flavonoid metabolic and biosynthetic processes and chalcone isomerase. In conclusion, using Zn fertilizer can improve the synthesis and metabolism of foxtail millet flavonoids, effectively increase the content of functional substances in grains, and realize the biofortification of foxtail millet grains. Sclerotinia sclerotiorum (S. sclerotiorum), a necrotrophic phytopathogen, causes sclerotinia stem rot (SSR) in many crops like oilseed rape, resulting in severe economic losses. Developing eco-friendly compound fungicides has become a critical research priority. This study explored the combination of sodium selenite and cuminic acid to screen for the optimal mixing ratio and investigate its inhibitory effects and mechanisms against S. sclerotiorum. The results demonstrated that synergistic effects were observed with a 1:3 combination ratio of sodium selenite to cuminic acid. After treatment with the selenium compound agent, ultrastructural observations revealed that the hyphae of S. sclerotiorum became severely shriveled, deformed, and twisted. The agent significantly reduced oxalic acid production and the activities of polymethylgalacturonide (PMG) and carboxymethylcellulose enzymes (Cx), while increasing the exocytosis of nucleic acids and proteins from the mycelium. Foliar application of the selenium compound agent significantly reduced lesion areas in rapeseed. Combined with the results of transcriptome sequencing, this study suggests that the compound agent effectively inhibits the growth of S. sclerotiorum by disrupting its membrane system, reducing the activity of cell wall-degrading enzymes, and suppressing protein synthesis, etc. This research provides a foundation for developing environmentally friendly and effective fungicides to control S. sclerotiorum. Plant growth regulators (PGRs) enhance crop stress resistance but their roles in microbial-mediated phosphorus cycling within intercropping systems are unclear. Thus, We conducted a two-year field study using corn (Zea mays L. cv. Denghai 605) and soybean (Glycine max L. cv. Hedou 22) in fluvisols and luvisols soil according to World Reference Base for Soil Resources (WRB) standard. Under a 4-row corn and 6-row soybean strip intercropping system, three treatments were applied: a water control (CK), and two plant growth regulators—T1 (EC: ethephon [300 mg/L] + cycocel [2 g/L]) and T2 (ED: ethephon [300 mg/L] + 2-Diethyl aminoethyl hexanoate [10 mg/L]). Foliar applications were administered at the V7 stage (seventh leaf) of intercropped corn plants to assess how foliar-applied PGRs (T1/T2) modulated the soil phosphorus availability, microbial communities, and functional genes in maize intercropping systems. PGRs increased the soil organic phosphorus and available phosphorus contents, and alkaline phosphatase activity, but not total phosphorus. PGRs declined the α-diversity in fluvisols soil but increased the α-diversity in luvisols soil. The major taxa changed from Actinobacteria (CK) to Proteobacteria (T1) and Saccharibacteria (T2) in fluvisols soil, and from Actinobacteria/Gemmatimonadetes (CK) to Saccharibacteria (T1) and Acidobacteria (T2) in luvisols soil. Functional gene dynamics indicated soil-specific regulation, where fluvisols soil harbored more phoD (organic phosphorus mineralization) and relA (polyphosphate degradation) genes, whereas phnP gene dominated in luvisols soil. T1 stimulated organic phosphorus mineralization and inorganic phosphorus solubilization in fluvisols soil, upregulating regulation genes, and T2 enhanced polyphosphate synthesis and transport gene expression in luvisols soil. Proteobacteria, Nitrospirae, and Chloroflexi were positively correlated with organic phosphorus mineralization and polyphosphate cycling genes, whereas Bacteroidetes and Verrucomicrobia correlated with available potassium (AP), total phosphorus (TP), and alkaline phosphatase (ALP) activity. Thus, PGRs activated soil phosphorus by restructuring soil type-dependent microbial functional networks, connecting PGRs-induced shifts with microbial phosphorus cycling mechanisms. These findings facilitate the targeted use of PGRs to optimize microbial-driven phosphorus efficiency in strategies for sustainable phosphorus management in diverse agricultural soils. Full article
