Comments and Suggestions for AuthorsIn the review “Roles of small noncoding vault RNAs in intestinal epithelium homeostasis and diseases”, the authors consider the biogenesis of vault RNA (vtRNA), the molecular mechanisms of regulation of gene expression at the post-transcriptional level using vtRNA. In addition, the significance and change of vtRNA in diseases (with inflammation or damage, dysfunction of the intestinal barrier) of the human intestinal mucosa are described.
The authors of the manuscript present complex and very significant material, describe the alleged mechanisms underlying vtRNA in intestinal epithelium homeostasis and diseases.
Despite the well-written manuscript, there are a number of comments:
- When listing keywords, the authors of the manuscript should indicate the full name of the abbreviated expression IBD and put IBD in parentheses.
- The authors need to provide more detailed information about the type of small RNAs with a unique composition that were detected using electron microscopy. The link is [14], line -76.
- In the caption to Figure 1, the full name of the vtRNA - NSUN2 fragmentation pathway must be indicated.
- On line 435, the authors presented a small section “1.9. Involvement of vtRNAs in Gut Mucosal Disorders”, which appeared after subsection 2.3. This section needs to be renumbered and, of course, information added.
- At the beginning of the section “3. Inflammatory Bowel Disease”, the authors describe two types of patients with inflammation of the intestinal mucosa and provide only one reference – [10]. It is necessary to supplement the information provided and add links.
- It is advisable to enlarge sections 3.2. and 3.3..

Comments and Suggestions for AuthorsThe author delivered a well organized review for vault RNAs in human. I have the following comments:
- Please include a paragraph describing the relationship of vault protein and vault RNA, especially the whether the vault RNA is inside the vault nanoparticle.
- Please include a table of graph summarizing the therapeutic relevance of vault RNA for section 3
