Background/Objectives: Traditional dietary screeners face significant limitations: they rely on subjective self-reporting, average intake estimates, and are influenced by a participant’s awareness of being observed—each of which can distort results. These factors reduce both accuracy and reproducibility. The Guide Against Age-Related Disease (GARD) addresses these issues by applying Assembly Theory to objectively quantify food and food behavior (FFB) complexity. This study aims to validate the GARD as a structured, bias-resistant tool for dietary assessment in clinical and research settings. Methods: The GARD survey was administered in an internal medicine clinic within a suburban hospital system in the southeastern U.S. The tool assessed six daily eating windows, scoring high-complexity FFBs (e.g., fresh plants, social eating, fasting) as +1 and low-complexity FFBs (e.g., ultra-processed foods, refined ingredients, distracted eating) as –1. To minimize bias, patients were unaware of scoring criteria and reported only what they ate the previous day, avoiding broad averages. A computer algorithm then scored responses based on complexity, independent of dietary guidelines. Internal (face, convergent, and discriminant) validity was assessed using Spearman rho correlations. Results: Face validation showed high inter-rater agreement using predefined Assembly Index (Ai) and Copy Number (Ni) thresholds. Positive correlations were found between high-complexity diets and behaviors (rho = 0.533–0.565, p < 0.001), while opposing constructs showed moderate negative correlations (rho = –0.363 to −0.425, p < 0.05). GARD scores aligned with established diet patterns: Mediterranean diets averaged +22; Standard American Diet averaged −10. Full article

Background/Objectives: Porcine circovirus type 2 (PCV2) infection causes porcine circovirus disease (PCVD), a global immunosuppressive disease in pigs. Its clinical manifestations include post-weaning multisystemic wasting syndrome (PMWS) and porcine dermatitis and nephropathy syndrome (PDNS), which cause significant economic losses to the swine industry. The Cap protein, which is the major protective antigen of PCV2, can self-assemble to form virus-like particles (VLPs) in the insect baculovirus expression system. Few studies have compared the expression of Cap proteins in different baculovirus expression systems. Methods: In this study, we compared two commonly commercialized baculovirus construction systems with the Cap protein expression in various insect cells. Results: The results demonstrate that the flashBAC system expressed the Cap protein at higher levels than the Bac-to-Bac system. Notably, when expressing four copies of the Cap protein, the flashBAC system achieved the highest protein yield in High Five cells, where it reached 432 μg/mL at 5 days post-infection (dpi) with 27 °C cultivation. Animal experiments confirmed that the purified Cap protein effectively induced specific antibody production in mice and swine. Conclusions: This study provides critical data for optimizing the production of the PCV2 Cap protein, which is of great significance for reducing the production cost of PCV2 vaccines and improving the industrial production efficiency. The human Tau protein stands for one of the most conspicuous and crucial hallmarks of Alzheimer’s disease (AD) diagnosis, along with other tauopathies. However, the assay for direct detection of tiny Tau protein concentrations in human samples continues to pose a significant challenge for the early diagnosis of AD. Thus, an amplification-based strategy is required. In this proposed work, we established an impedimetric immunosensor to detect human Tau-441 protein in PBS buffer using a sandwich approach, wherein we employed two distinct monoclonal antibodies (HT7 and BT2) that specifically recognize the amino acids 159–198 of the target protein. Through this strategy, we were able to detect as low as 0.08 pg/mL. These findings were attributed to the use of a biotinylated antibody (BT2)-streptavidin complex, which facilitated the amplification of the normalized signal, resulting in a lower limit of detection in comparison to the directly based immunosensors. Subsequently, we investigated the designed immunosensor to assess the assay’s selectivity in the presence of different off-targets, and no cross-interaction was recorded. The outcomes of our study provide valuable new insights into the application of sandwich-based assay as a highly sensitive and selective immunosensor for the detection of small protein. Full article

Background: During food processing and storage, traditional protein-based delivery systems encounter significant challenges in maintaining the structural and functional integrity of bioactive compounds, primarily due to their temporal instability. Methods: In this study, a nanocomposite hydrogel was prepared through the co-assembly of a self-assembling peptide, 9-Fluorenylmethoxycarbonyl-phenylalanine-arginine-glycine-aspartic acid-phenylalanine (Fmoc-FRGDF), and hyaluronic acid (HA). The stability of this hydrogel as a quercetin (Que) delivery carrier was systematically investigated. Furthermore, the impact of Que co-assembly on the microstructural evolution and physicochemical properties of the hydrogel was characterized. Concurrently, the encapsulation efficiency (EE%) and controlled release kinetics of Que were quantitatively evaluated. Results: The findings indicated that HA significantly reduced the storage modulus (G′) from 256.5 Pa for Fmoc-FRGDF to 21.1 Pa with the addition of 0.1 mg/mL HA. Despite this reduction, HA effectively slowed degradation rates; specifically, residue rates of 5.5% were observed for Fmoc-FRGDF alone compared to 14.1% with 0.5 mg/mL HA present. Notably, Que enhanced G′ within the ternary complex, increasing it from 256.5 Pa in Fmoc-FRGDF to an impressive 7527.0 Pa in the Que/HA/Fmoc-FRGDF hydrogel containing 0.1 mg/mL HA. The interactions among Que, HA, and Fmoc-FRGDF involved hydrogen bonding, electrostatic forces, and hydrophobic interactions; furthermore, the co-assembly process strengthened the β-sheet structure while significantly promoting supramolecular ordering. Interestingly, the release profile of Que adhered to the Korsmeyer–Peppas pharmacokinetic equations. Conclusions: Overall, this study examines the impact of polyphenol on the rheological properties, microstructural features, secondary structure conformation, and supramolecular ordering within peptide–polysaccharide–polyphenol ternary complexes, and the Fmoc-FRGDF/HA hydrogel system demonstrates a superior performance as a delivery vehicle for maintaining quercetin’s bioactivity, thereby establishing a multifunctional platform for bioactive agent encapsulation and controlled release. Recombinant protein hydrogels have emerged as transformative biomaterials that overcome the bioinertness and unpredictable degradation of traditional synthetic systems by leveraging genetically engineered backbones, such as elastin-like polypeptides, SF, and resilin-like polypeptides, to replicate extracellular matrix (ECM) dynamics and enable programmable functionality. Constructed through a hierarchical crosslinking strategy, these hydrogels integrate reversible physical interactions with covalent crosslinking approaches, collectively endowing the system with mechanical strength, environmental responsiveness, and controlled degradation behavior. Critically, molecular engineering strategies serve as the cornerstone for functional precision: domain-directed self-assembly exploits coiled-coil or β-sheet motifs to orchestrate hierarchical organization, while modular fusion of bioactive motifs through genetic encoding or site-specific conjugation enables dynamic control over cellular interactions and therapeutic release. Such engineered designs underpin advanced applications, including immunomodulatory scaffolds for diabetic wound regeneration, tumor-microenvironment-responsive drug depots, and shear-thinning bioinks for vascularized bioprinting, by synergizing material properties with biological cues. By uniting synthetic biology with materials science, recombinant hydrogels deliver unprecedented flexibility in tuning physical and biological properties. This review synthesizes emerging crosslinking paradigms and molecular strategies, offering a framework for engineering next-generation, adaptive biomaterials poised to address complex challenges in regenerative medicine and beyond. In this study, growth mechanisms are proposed to understand how banded dendritic crystal aggregates in poly(1,4-butylene adipate) (PBA) transform into straight dendrites upon dilution with a large quantity of poly(ethylene oxide) (PEO) (25–90 wt.%). In growth packing, crystal plates are deformed in numerous ways, such as bending, scrolling, and twisting in self-assembly, into final aggregated morphologies of periodic bands or straight dendrites. Diluting PBA with a significant amount of PEO uncovers intricate periodic banded assemblies, facilitating better structural analysis. Both circularly banded and straight dendritic PBA aggregates have similar basic lamellar patterns. In straight dendritic PBA spherulites, crystal plates can twist from edge-on to flat-on, similar to those in ring-banded spherulites. Therefore, twists—whether continuous or discontinuous—are not limited to the conventional models proposed for classical periodic-banded spherulites. Thus, it would not be universally accurate to claim that the periodic circular bands observed in polymers or small-molecule compounds are caused by continuous lamellar helix twists. Straight dendrites, which do not exhibit optical bands, may also involve alternate crystal twists or scrolls during growth. Iridescence tests are used to compare the differences in crystal assemblies of straight dendrites vs. circularly banded PBA crystals. Full article

Car interior design should evoke emotions, offer comfort, convey safety and at the same time project the brand identity of the car manufacturer. Lighting is used to address these functions. Modules required for automotive interior lighting often feature injection-moulded (IM) light guides, whereas woven fabrics with polymer optical fibres (POFs) offer certain technological advantages and show first-series applications in cars. In the future, car interior illumination will become even more important in the wake of megatrends such as autonomous driving. Since the increase in deployment of these technologies facilitates a need for an economical comparison, this paper aims to deliver a cost-driven approach to fulfil the aforementioned objective. Therefore, the cost structures of the supply chains for an IM-based and a POF-based illumination module are analysed. The employed research methodologies include an activity-based costing approach for which the data is collected via document analysis and guideline-based expert interviews. To account for data uncertainty, Monte Carlo simulations are conducted. POF-based lighting modules have lower initial costs due to continuous fibre production and weaving processes, but are associated with higher unit costs. This is caused by the discontinuous assembly of the rolled woven fabric which allows postponement strategies. The development costs of the mould generate high initial costs for IM light guides, which makes them beneficial only for high quantities of produced light guides. For the selected scenario, the POF-based module’s self-costs are 11.05 EUR/unit whereas the IM module’s self-costs are 14,19 EUR/unit. While the cost structures are relatively independent from the selected scenario, the actual self-costs are highly dependent on boundary conditions such as production volume. Full article

Poly(ε-caprolactone)-b-polysarcosine (PCL-b-PSar) block copolymers (BCPs) emerge as a promising alternative to conventional poly(ε-caprolactone)-b-poly(ethylene oxide) BCPs for biomedical applications, leveraging superior biocompatibility and biodegradability. In this study, we synthesized two series of PCL-b-PSar BCPs with controlled polymerization degrees (DP of PCL: 45/67; DP of PSar: 28–99) and low polydispersity indexes (Đ ≤ 1.1) and systematically investigated their crystallization-driven self-assembly (CDSA) in alcohol solvents (ethanol, n-butanol, and n-hexanol). It was found that the limited solubility of PSar in alcohols resulted in competition between micellization and crystallization during self-assembly of PCL-b-PSar, and thus coexistence of lamellae and spherical micelles. To overcome this morphological heterogeneity, we developed a modified self-seeding method by employing a two-step crystallization strategy (i.e., Tc1 = 33 °C and Tc2 = 8 °C), achieving conversion of micelles into crystals and yielding uniform self-assembled structures. PCL-b-PSar BCPs with short PSar blocks tended to form well-defined two-dimensional lamellar crystals, while those with long PSar blocks induced formation of hierarchical structures in the PCL45 series and polymer aggregation on crystal surfaces in the PCL67 series. Solvent quality notably influenced the self-assembly pathways of PCL45-b-PSar28. Lamellar crystals were formed in ethanol and n-butanol, but micrometer-scale dendritic aggregates were generated in n-hexanol, primarily due to a significant Hansen solubility parameter mismatch. This study elucidated the CDSA mechanism of PCL-b-PSar in alcohols, enabling precise structural control for biomedical applications. Full article

Against the backdrop of global energy transition and strict environmental regulations, supercritical carbon dioxide (scCO2) fracturing and oil displacement technologies have emerged as pivotal green approaches in shale gas exploitation, offering the dual advantages of zero water consumption and carbon sequestration. However, the inherent low viscosity of scCO2 severely restricts its sand-carrying capacity, fracture propagation efficiency, and oil recovery rate, necessitating the urgent development of high-performance thickeners. The current research on scCO2 thickeners faces a critical trade-off: traditional fluorinated polymers exhibit excellent philicity CO2, but suffer from high costs and environmental hazards, while non-fluorinated systems often struggle to balance solubility and thickening performance. The development of new thickeners primarily involves two directions. On one hand, efforts focus on modifying non-fluorinated polymers, driven by environmental protection needs—traditional fluorinated thickeners may cause environmental pollution, and improving non-fluorinated polymers can maintain good thickening performance while reducing environmental impacts. On the other hand, there is a commitment to developing non-noble metal-catalyzed siloxane modification and synthesis processes, aiming to enhance the technical and economic feasibility of scCO2 thickeners. Compared with noble metal catalysts like platinum, non-noble metal catalysts can reduce production costs, making the synthesis process more economically viable for large-scale industrial applications. These studies are crucial for promoting the practical application of scCO2 technology in unconventional oil and gas development, including improving fracturing efficiency and oil displacement efficiency, and providing new technical support for the sustainable development of the energy industry. This study innovatively designed an amphiphilic modified amino silicone oil polymer (MA-co-MPEGA-AS) by combining maleic anhydride (MA), methoxy polyethylene glycol acrylate (MPEGA), and amino silicone oil (AS) through a molecular bridge strategy. The synthesis process involved three key steps: radical polymerization of MA and MPEGA, amidation with AS, and in situ network formation. Fourier transform infrared spectroscopy (FT-IR) confirmed the successful introduction of ether-based CO2-philic groups. Rheological tests conducted under scCO2 conditions demonstrated a 114-fold increase in viscosity for MA-co-MPEGA-AS. Mechanistic studies revealed that the ether oxygen atoms (Lewis base) in MPEGA formed dipole–quadrupole interactions with CO2 (Lewis acid), enhancing solubility by 47%. Simultaneously, the self-assembly of siloxane chains into a three-dimensional network suppressed interlayer sliding in scCO2 and maintained over 90% viscosity retention at 80 °C. This fluorine-free design eliminates the need for platinum-based catalysts and reduces production costs compared to fluorinated polymers. The hierarchical interactions (coordination bonds and hydrogen bonds) within the system provide a novel synthetic paradigm for scCO2 thickeners. The synthesis and structural characterization of three new triple-stranded helical complexes ([Dy2(L2)3]2Cl∙15H2O (C1), [Y2(L2)3]3(NO3)Cl∙14H2O∙DMSO (C2), and [Eu2(L4)3]∙12H2O (C3), where L2 and L4 are ligands derived from pyridoxal hydrochloride and succinic or adipic acid dihydrazides, respectively, were described. The X-ray data, combined with spectroscopic measurements, indicated that L2 and L4 act as bis-tridentate ligands, presenting two tridentate chelating cavities O,N,O to obtain the dinuclear complexes C1C3. Their antiviral profile was predicted via in silico calculations in terms of interaction with the structural severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) spike glycoprotein in the down- and up-states and complexed with the cellular receptor angiotensin-converting enzyme 2 (ACE2). The best affinity energy values (−9.506, −9.348, and −9.170 kJ/mol for C1, C2, and C3, respectively) were obtained for the inorganic complexes docked in the model spike-ACE2, with C1 being suggested as the most promising candidate for a future in vitro validation. The obtained in silico antiviral trend was supported by the prediction of the electronic and physical–chemical properties of the inorganic complexes via the density functional theory (DFT) approach, representing an original and relevant contribution to the bioinorganic and medicinal chemistry fields. Systemic and Mucosal Humoral Immune Responses to Lumazine Synthase 60-mer Nanoparticle SARS-CoV-2 Vaccines

Background: Vaccines that stimulate systemic and mucosal immunity to a level required to prevent SARS-CoV-2 infection and transmission are an unmet need. Highly protective hepatitis B and human papillomavirus nanoparticle vaccines highlight the potential of multivalent nanoparticle vaccine platforms to provide enhanced immunity. Here, we report the construction and characterization of self-assembling 60-subunit icosahedral nanoparticle SARS-CoV-2 vaccines using the bacterial enzyme lumazine synthase (LuS). Methods and Results: Nanoparticles displaying prefusion-stabilized SARS-CoV-2 spike ectodomains fused to the surface-exposed amino terminus of LuS were designed using structure-guided approaches. Negative stain-electron microscopy studies of purified nanoparticles were consistent with self assembly into 60-mer nanoparticles displaying 20 spike trimers. After two intramuscular doses, these purified spike-LuS nanoparticles elicited significantly higher SARS-CoV-2 neutralizing activity than spike trimers in vaccinated mice. Furthermore, intramuscular DNA priming and intranasal boosting with a SARS-CoV-2 LuS nanoparticle vaccine stimulated mucosal IgA responses. Conclusion: These data identify LuS nanoparticles as highly immunogenic SARS-CoV-2 vaccine candidates and support the further development of this platform against SARS-CoV-2 and its emerging variants. The optical properties of a heterostructure containing GaSb/GaAs quantum dots (QDs) have been systematically investigated via photoluminescence (PL) measurements to gain insights into carrier dynamics. The QD and wetting layer (WL) emissions exhibit a complementary dependence on the excitation intensity and temperature, reflecting the interplay between carrier localization in the WL and carrier relaxation from the WL to the QDs. Carrier dynamics related to localization, injection, and recombination are further validated by time-resolved photoluminescence (TRPL). These findings highlight the necessity of carefully optimizing GaSb/GaAs QD structures to mitigate the impact of carrier localization, thereby enhancing the ultimate performance of devices utilizing these QDs as active region materials. Background/Objectives: SARS-CoV-2 vaccine candidates comprising the receptor binding domain (RBD) of the spike protein have been shown to confer protection against infection. Previous research evaluating vaccine candidates with SARS-CoV-2 RBD fused to ferritin (RBD-ferritin) and other scaffolds suggested that multimeric assemblies of RBD can enhance antigen presentation to improve the potency and breadth of immune responses. Though RBDs directly fused to a self-assembling scaffold can be delivered as messenger RNA (mRNA) formulated with lipid nanoparticles (LNPs), reports of SARS-CoV-2 vaccine candidates that combine these approaches remain scarce. Methods: Here, we designed RBD fused to AP205 or TIP60 self-assembling nanoparticles following a search of available structures focused on several scaffold properties. RBD-AP205 and RBD-TIP60 were tested for antigenicity following transfection and for immunogenicity and neutralization potency when delivered as mRNA in mice, with RBD-ferritin as a direct comparator. Results: All scaffolded RBD constructs were readily secreted to transfection supernatant and showed antigenicity in ELISA, though clear heterogeneity in assembly was observed. RBD-AP205 and RBD-TIP60 also exhibited robust antibody binding and neutralization titers in mice that were comparable to those elicited by RBD-ferritin or a full-length membrane-bound spike. Conclusions: These data suggest that AP205 and TIP60 can present RBD as effectively as ferritin and induce similar immune responses. By describing additional scaffolds for multimeric display that accommodate mRNA delivery platforms, this work can provide new tools for future vaccine design efforts. Full article

The scope of this paper is the development of a cost-effective wireless torque measurement system for vehicle drivetrain shafts. The prototype integrates strain gauges, an HX711 conditioner, a Wemos D1 Mini ESP8266, and a rechargeable battery directly on the rotating shaft, forming a self-contained sensor node. Calibration against a certified dynamometric wrench confirmed an operating span of ±5–50 N·m. Within this range, the device achieved a mean absolute error of 0.559 N·m. It also maintained precision better than ±2.5 N·m at 95% confidence, while real-time data were transmitted via Wi-Fi. The total component cost is below EUR 30 based on current prices. The novelty of this proof-of-concept implementation demonstrates that reliable, IoT-enabled torque sensing can be realized with low-cost, readily available parts. The paper details assembly, calibration, and deployment procedures, providing a transparent pathway for replication. By aligning with Industry 4.0 requirements for smart, connected equipment, the proposed torque measurement system offers an affordable solution for process monitoring and predictive maintenance in automotive and industrial settings. This capsule was obtained through a reaction involving 2,8,14,20-tetra-undecyl-resorcin[4]arene and water in chloroform. The formation of an inclusion complex was deduced from a combination of spectral measurements (UV-visible spectroscopy, 1H, 31P{1H} NMR and DOSY). The rhodium complex was evaluated in the [2+2+2] cycloaddition between N,N-dipropargyl-p-toluenesulfonamide and arylacetylene derivatives. In the presence of two equivalents of arylacetylenes in water-saturated chloroform at 60 °C for 24 h, the 4-methyl-N-(prop-2-yn-1-yl)-N-((2-tosylisoindolin-5-yl)methyl)benzenesulfonamide, the homocycloaddition product of 1,6-diyne is predominantly formed. In the presence of the supramolecular capsule, a selectivity inversion in favor of 5-aryl-2-tosylisoindoline is observed, with heterocycloaddition products formed in proportions between 53 and 69%.
