Salinization hinders the restoration of vegetation in salt-affected soils by negatively impacting plant growth and development. Halophytes play a key role in the restoration of saline and degraded lands due to unique features explaining their growth aptitude in such extreme ecosystems. Suaeda fruticosa is an euhalophyte well known for its medicinal properties and its potential for saline soil phytoremediation. However, excessive salt accumulation in soil limits the development of this species. Research findings increasingly advocate the use of extremophile rhizosphere bacteria as an effective approach to reclaim salinized soils, in conjunction with their salt-alleviating effect on plants. Here, a pot experiment was conducted to assess the role of a halotolerant plant growth-promoting actinobacterium, Glutamicibacter sp., on the growth, nutritional status, and shoot content of proline, total soluble carbohydrates, and phenolic compounds in the halophyte S. fruticosa grown for 60 d under high salinity (600 mM NaCl). Results showed that inoculation with Glutamicibacter sp. significantly promoted the growth of inoculated plants under stress conditions. More specifically, bacterial inoculation increased the shoot concentration of proline, total polyphenols, potassium (K+), nitrogen (N), and K+/Na+ ratio in shoots, while significantly decreasing Na+ concentrations. These mechanisms partly explain S. fruticosa tolerance to high saline concentrations. Our findings provide some mechanistic elements at the ecophysiological level, enabling a better understanding of the crucial role of plant growth-promoting rhizobacteria (PGPRs) in enhancing halophyte growth and highlight their potential for utilization in restoring vegetation in salt-affected soils. Soil salinization severely restricts crop growth and presents a major challenge to global agriculture. In this study, a plant-growth-promoting rhizobacterium (PGPR) was isolated and identified as Proteus sp. through 16S rDNA analysis and was subsequently named Proteus sp. JHY1. Under salt stress, exogenous dopamine (DA) significantly enhanced the production of indole-3-acetic acid and ammonia by strain JHY1. Pot experiments revealed that both DA and JHY1 treatments effectively alleviated the adverse effects of 225 mM NaCl on rice, promoting biomass, plant height, and root length. More importantly, the combined application of DA-JHY1 showed a significant synergistic effect in mitigating salt stress. The treatment increased the chlorophyll content, net photosynthetic rate, osmotic regulators (proline, soluble sugars, and protein), and reduced lipid peroxidation. The treatment also increased soil nutrients (ammoniacal nitrogen and available phosphorus), enhanced soil enzyme activities (sucrase and alkaline phosphatase), stabilized the ion balance (K+/Na+), and modulated the soil rhizosphere microbial community by increasing beneficial bacteria, such as Actinobacteria and Firmicutes. This study provides the first evidence that the synergistic effect of DA and PGPR contributes to enhanced salt tolerance in rice, offering a novel strategy for alleviating the adverse effects of salt stress on plant growth. Soil salinity adversely affects crop growth and development, leading to reduced soil fertility and agricultural productivity. The indigenous salt-tolerant plant growth-promoting rhizobacteria (PGPR), as a sustainable microbial resource, do not only promote growth and alleviate salt stress, but also improve the soil microecology of crops. The strain H5 isolated from saline-alkali soil in Bachu of Xinjiang was studied through whole-genome analysis, functional annotation, and plant growth-promoting, salt-tolerant trait gene analysis. Phylogenetic tree analysis and 16S rDNA sequencing confirmed its classification within the genus Halomonas. Functional annotation revealed that the H5 genome harbored multiple functional gene clusters associated with plant growth promotion and salt tolerance, which were critically involved in key biological processes such as bacterial survival, nutrient acquisition, environmental adaptation, and plant growth promotion. The pot experiment under moderate salt stress demonstrated that seed inoculation with Halomonas sp. H5 not only significantly improved the agronomic traits of tomato seedlings, but also increased plant antioxidant enzyme activities under salt stress. Additionally, soil analysis revealed H5 treatment significantly decreased the total salt (9.33%) and electrical conductivity (8.09%), while significantly improving organic matter content (11.19%) and total nitrogen content (10.81%), respectively (p < 0.05). Inoculation of strain H5 induced taxonomic and functional shifts in the rhizosphere microbial community, increasing the relative abundance of microorganisms associated with plant growth-promoting and carbon and nitrogen cycles, and reduced the relative abundance of the genera Alternaria (15.14%) and Fusarium (9.76%), which are closely related to tomato diseases (p < 0.05). Overall, this strain exhibits significant potential in alleviating abiotic stress, enhancing growth, improving disease resistance, and optimizing soil microecological conditions in tomato plants. These results provide a valuable microbial resource for saline soil remediation and utilization. Full article

With the increasing severity of global climate change and soil salinization, the development of microorganisms that enhance crop salt tolerance has become a critical focus of agricultural research. In this study, we explored the potential of a novel Streptomyces species Qhu-G9 as a plant growth-promoting rhizobacterium (PGPR) under salt stress conditions, employing whole-genome sequencing and functional annotation. The genomic analysis revealed that Qhu-G9 harbors various genes related to plant growth promotion, including those involved in phosphate solubilization, indole-3-acetic acid (IAA) biosynthesis, antioxidant activity, and nitrogen fixation. A total of 8528 coding genes were annotated in Qhu-G9, with a significant proportion related to cell metabolism, catalytic activity, and membrane transport, suggesting its broad growth-promoting potential. In vitro experiments demonstrated that Qhu-G9 exhibited strong iron siderophore production, IAA synthesis, phosphate solubilization, and 1-aminocyclopropane-1-carboxylate (ACC) deaminase activity, all of which correlate with its plant growth-promoting capacity. Further plant growth trials revealed that Qhu-G9 significantly enhances the growth of Avena sativa and Onobrychis viciifolia seedlings under salt stress conditions, improving key physiological parameters, such as chlorophyll content, relative water content, and photosynthetic efficiency. Under salt stress conditions, inoculation with Qhu-G9 resulted in notable increases in total biomass, root length, and plant height. Biochemical analyses further confirmed that Qhu-G9 alleviates the oxidative damage induced by salt stress by boosting antioxidant enzyme activities, reducing peroxide levels, and promoting the accumulation of osmotic regulators. These findings suggest that Qhu-G9 holds great promise as a PGPR that not only promotes plant growth, but also enhances plant tolerance to salt stress; thus, it has significant agricultural potential. Plant growth-promoting rhizobacteria (PGPR) are beneficial soil microorganisms that enhance plant growth and stress tolerance through various mechanisms, including phytohormone production, EPS production, phosphate solubilization, and extracellular enzyme production. These bacteria establish endosymbiotic relationships with plants, improving nutrient availability and overall crop productivity. Despite extensive research on PGPR isolation, their practical application in agricultural fields has faced challenges due to environmental stresses and limited survival during storage. To address these limitations, the present study aimed to isolate salt-tolerant bacterial strains and formulate them with organic carriers to enhance their stability and effectiveness under saline conditions. The isolated bacterial strains exhibited high salt tolerance, surviving NaCl concentrations of up to 850 millimolar. These strains demonstrated basic key plant growth-promoting traits, including phosphate solubilization, auxin production, and nitrogen fixation. The application of carrier-based formulations with both strains, Bacillus wiedmannii (RR2) and Bacillus paramobilis (RR3), improved physiological and biochemical parameters in wheat plants subjected to salinity stress. The treated plants, when subjected to salinity stress, showed notable increases in chlorophyll a (73.3% by Peat + RR3), chlorophyll b (41.1% by Compost + RR3), carotenoids (51.1% by Peat + RR3), relative water content (77.7% by Compost + RR2), proline (75.8% by compost + RR3), and total sugar content (12.4% by peat + RR2), as compared to the stressed control. Plant yield parameters such as stem length (35.1% by Peat + RR3), spike length (22.5% by Peat + RR2), number of spikes (67.6% by Peat + RR3), and grain weight (39.8% by Peat + RR3) were also enhanced and compared to the stressed control. These results demonstrate the potential of the selected salt-tolerant PGPR strains (ST-strains) to mitigate salinity stress and improve wheat yield under natural field conditions. The study highlights the significance of carrier-based PGPR applications as an effective and sustainable approach for enhancing crop productivity in saline-affected soils. Full article

Synergistic Effects of Salt-Tolerant PGPR and Foliar Silicon on Pak Choi Antioxidant Defense Under Salt Stress

Salinization severely impairs crop growth by inducing oxidative stress and disrupting cellular homeostasis. This study systematically investigates the synergistic effects of salt-tolerant plant-growth-promoting rhizobacteria (ST-PGPR) and foliar silicon fertilizer spraying (FSFS) on antioxidant responses in Pak choi under salt stress. Two-season pot experiments were carried out to evaluate key indicators, including antioxidant enzyme activities (superoxide dismutase: SOD; peroxidase: POD; catalase: CAT), oxidative stress (malondialdehyde: MDA), osmolyte accumulation (proline, soluble protein), and hormones (Jasmonic Acid: JA; Salicylic Acid: SA; Abscisic acid: ABA). The results demonstrate that combining ST-PGPR with FSFS significantly enhances SOD (6.18–2353.85%), POD (3.44–153.29%), and CAT (25.71–319.29%) activities while reducing MDA content (8.12–35.87%). Proline and soluble protein levels increased by 1.56–15.71% and 5.03–188.87%, respectively. Hormonal regulation increased JA, SA, and ABA levels by 1.05–31.81%, 2.09–34.29%, and 3.18–30.09%, respectively. Notably, ST-PGPR treatments at 104 and 106 cfu·mL−1, combined with foliar silicon application, consistently ranked highest in overall antioxidant performance across both seasons based on a principal component analysis. These findings provide novel insights into microbial–mineral interactions for sustainable saline agriculture. Full article

Salinity and heavy metal stress significantly reduce agricultural productivity in arable lands, particularly affecting crops like rice (Oryza sativa L.). This study aimed to evaluate the efficacy of heavy metal-tolerant plant growth-promoting rhizobacteria (HMT-PGPR) in mitigating the harmful effects of salt (NaCl), chromium (Cr), and combined NaCl + Cr stress on rice plants. Two pre-isolated and well-characterized heavy metal-tolerant epiphytic (Ochrobactrum pseudogrignonense strain P14) and endophytic (Arthrobacter woluwensis strain M1R2) PGPR were tested. The LSD test (p ≤ 0.05) was used to assess the statistical significance between treatment means. Stresses caused by NaCl, Cr, and their combination were found to impair plant growth and biomass accumulation through mechanisms, including osmotic stress, oxidative damage, ionic imbalance, reduced photosynthetic pigment, lowered relative water content, and compromised antioxidant defense systems. Conversely, inoculation with HMT-PGPR alleviated these adverse effects by reducing oxidative stress indicators, including malondialdehyde (MDA), hydrogen peroxide (H2O2) content and electrolyte leakage (EL) and enhancing plant growth, osmolyte synthesis, and enzymatic antioxidant activity under single- and dual-stress conditions. The application of HMT-PGPR notably restricted Na+ and Cr6+ uptake, with an endophytic A. woluwensis M1R2 demonstrating superior performance in reducing Cr6+ translocation (38%) and bioaccumulation (42%) in rice under dual stress. The findings suggest that A. woluwensis effectively mitigates combined salinity and chromium stress by maintaining ion homeostasis and improving the plant’s antioxidant defenses. Full article

Halotolerant, plant growth-promoting rhizobacteria (PGPR) are known to alleviate plant growth under abiotic stresses, especially those isolated from saline arid soils. In this study, 66 bacterial isolates, obtained from various habitats in Saudi Arabia, were characterized for their plant growth-promoting (PGP) traits, and screened for heat and salt stress resilience. Finally, selected halotolerant PGPR strains were assessed for their potential to improve maize (Zea mays L.) growth under salinity stress using in vitro assays. Our results indicated that many isolates possessed key PGP traits such ACC deaminase, N-fixation, and phytohormone production. Additionally, several isolates were able to tolerate high temperatures, and 20 bacterial isolates were classified as halotolerant. Furthermore, among the isolates, Pseudomonas soyae (R600), Bacillus haynesii (SFO145), Salinicola halophilus (SFO075), and Staphylococcus petrasii (SFO132) significantly enhanced various maize growth parameters under salt stress conditions when compared to uninoculated plants. These halotolerant PGPR are good candidates to be explored as bioinoculants for sustainable agriculture under saline arid soil conditions. Full article

Synergistic Effect of Serratia fonticola and Pseudomonas koreensis on Mitigating Salt Stress in Cucumis sativus L.

Beneficial microbes enhance plant growth and development, even under stressful conditions. Serratia fonticola (S1T1) and Pseudomonas koreensis (S4T10) are two multi-trait plant growth-promoting rhizobacteria (PGPRs) that are resistant to saline conditions. This study evaluated the synergistic effect of these PGPRs on mitigating salinity stress (200 mM) in Cucumis sativus. Presently, the synergistic effect of both strains enhances the plant growth-promoting attributes of cucumber, and the growth parameters were significantly higher than those of uninoculated plants. The PGPR-treated plants revealed a significantly higher biomass and improved chlorophyll content. The inoculation of S1T1 and S4T10 and the synergistic effect of both promoted 23, 24, and 28% increases, respectively, in the fresh biomass and 16, 19.8, and 24% increases, respectively, in the dry biomass. Similarly, S1T1 and S4T10 and their synergistic effects led to 16.5, 28.4, and 38% increases, respectively, in the water potential and 18, 22, and 28% decreases, respectively, in abscisic acid (ABA). A reduction in the electrolytic leakage (EL) was additional proof of successful PGPR activities. Similarly, a decrease in the antioxidant levels, including those of malondialdehyde (21–30%), hydrogen peroxide (19–38%), and superoxide anions (24–34%), was observed, alongside an increase in antioxidant enzymes such as catalase (22–29%) and superoxide dismutase (17–27%). Additionally, the synergistic inoculation of the PGPRs enhanced the NaCl stress tolerance by upregulating the expression of the ion transporter genes HKT1 (1–2-fold), NHX (1–3-fold), and SOS1 (2–4-fold). Conclusively, the synergistic effect of the multi-trait PGPRs significantly enhances C. sativus L. growth under salt stress. Full article

Comparison of Two Bacillus Strains Isolated from the Coastal Zone in Barley (Hordeum vulgare L.) Under Salt Stress

Salt stress is one of the most important abiotic stress factors that negatively affects sustainable crop production, agricultural productivity, and microbial life. Increasing salt stress negatively affects the growth and development of barley, posing a threat to global food security. It is now known that inoculation of plant growth-promoting rhizobacteria (PGPR) has significant potential in increasing stress tolerance and yield in agricultural products. This study focused on the effects of Bacillus cereus CUN6 and Bacillus thuringiensis SIRB2, isolated from the coastal zone and tested for their PGPR capacities, on physiological (root length, shoot length, biomass, dry weight) and biochemical (total chlorophyll, total protein, hydrogen peroxide, lipid peroxidation, peroxidase activity (POX), catalase activity (CAT)) analyses in Hordeum vulgare L. seedlings under salt stress. The results showed that the two bacterial inoculations alleviated the negative effects of salt stress by increasing the root-shoot length, biomass, dry weight, chlorophyll content, and total protein content in barley plants. However, B.thuringiensis increased growth and development especially in root length, biomass, and dry weight compared to B.cereus. On the other hand, B.cereus significantly increased root length, biomass, and chlorophyll content under salt stress; these increases were 17%, 5%, and 7%, respectively. B.thuringiensis chlorophyll content increased by 4% in 300 mM NaCl compared to the control. When compared in terms of the antioxidant defense system, B.thuringiensis inoculation was more effective on CAT activity, while B.cereus inoculation was more effective on POX activity. Under salt stress, B.cereus and B.thuringiensis inoculation significantly decreased H2O2 content in barley; these decreases were 16% and 10%, respectively. Additionally, TBARs content was significantly decreased by B.cereus and B.thuringiensis inoculation under salt stress; these decreases were determined as 8% and 9%, respectively, compared to the control. These results indicated that both bacterial inoculations can alleviate the salt tolerance of barley seedlings by regulating antioxidant metabolism. This research focused on the potential of B.cereus and B.thuringiensis as biofertilizers against salt stress in barley based on physiological and biochemical analysis. Full article

Using Klebsiella sp. and Pseudomonas sp. to Study the Mechanism of Improving Maize Seedling Growth Under Saline Stress

The increasing salinization of cultivated soil worldwide has led to a significant reduction in maize production. Using saline–alkaline-tolerant growth-promoting bacteria (PGPR) in the rhizosphere can significantly improve the saline tolerance of maize and ensure the stability of maize yields, which has become a global research hotspot. This study screened salt-tolerant microorganisms Klebsiella sp. (GF2) and Pseudomonas sp. (GF7) from saline soil to clarify the mechanism in improving the saline tolerance of maize. In this study, different application treatments (GF2, GF7, and GF2 + GF7) and no application (CK) were set up to explore the potential ecological relationships between the saline tolerance of maize seedlings, soil characteristics, and microorganisms. The results showed that co-occurrence network and Zi-Pi analysis identified Klebsiella and Pseudomonas as core microbial communities in the rhizosphere soil of maize seedlings grown in saline soil. The deterministic process of microbial assembly mainly controlled the bacterial community, whereas bacteria and fungi were governed by random processes. The application of saline–alkaline-resistant PGPR under saline stress significantly promoted maize seedling growth, increased the activity of soil growth-promoting enzymes, and enhanced total nitrogen, soil organic carbon, and microbial carbon and nitrogen contents. Additionally, it reduced soil salt and alkali ion concentrations [electrical conductivity (EC) and exchangeable Na+]. Among them, GF2 + GF7 treatment had the best effect, indicating that saline–alkaline-tolerant PGPR could directly or indirectly improve the saline tolerance of maize seedlings by improving the rhizosphere soil ecological environment. EC was the determining factor to promote maize seedling growth under saline–alkaline stress (5.56%; p < 0.01). The results provided an important theoretical reference that deciphers the role of soil factors and microecology in enhancing the saline tolerance of maize. Full article

Salinity stress poses a major obstacle to agricultural productivity. Employing plant growth-promoting rhizobacteria (PGPR) has attracted significant attention due to its potential to improve plant development in challenging conditions. Yet, additional investigation is essential to fully understand the potential of PGPR in mitigating salinity stress, especially in field applications. Hence, this study investigated the resistance mechanisms of soybean (Glycine max (L.) Merr.) under salt stress with PGPR application through a field experiment with four treatments: normal soybean planting (NN), normal planting + PGPR (NP), salt stress planting (SN), and salt stress planting + PGPR (SP). This research investigated how applying PGPR under salt stress influences soybean photosynthetic traits, osmotic regulation, rhizosphere microbial communities, and yield quality. The results demonstrated that salt stress enhanced leaf temperature and significantly reduced the leaf area index, SPAD value, stomatal conductance, photosynthetic rate, and transpiration rate of soybeans. Compared to SN treatment, SP treatment significantly improved the stomatal conductance, photosynthetic rate, and transpiration rate by 10.98%, 16.28%, and 35.59%, respectively. Salt stress substantially increased sodium (Na+) concentration and Na+/K+ ratio in leaves, roots, and grains while reducing potassium (K+) concentration in roots and leaves. Under salinity stress, PGPR application significantly minimized Na+ concentration in leaves and enhanced K⁺ concentration in leaves, roots, and grains by 47.05%, 25.72%, and 14.48%, respectively. PGPR application boosted carbon assimilation (starch synthesis) by enhancing the activities of sucrose synthase, fructokinase, and ADP-glucose pyrophosphorylase. It improved physiological parameters and increased soybean yield by 32.57% compared to SN treatment. Additionally, PGPR enhanced antioxidant enzyme activities, including glutathione reductase, peroxidase, ascorbate peroxidase, and monodehydroascorbate reductase, reducing oxidative damage from salt stress. Analysis of rhizosphere microbial communities revealed that PGPR application enriched beneficial bacterial phyla such as Bacteroidetes, Firmicutes, Nitrospirae, and Patescibacteria and fungal genera like Metarhizium. These microbial shifts likely contributed to improved nutrient cycling and plant–microbe interactions, further enhancing soybean resilience to salinity. This study demonstrates that PGPR enhances soybean growth, microbial diversity, and salt tolerance under salinity stress, while future efforts should optimize formulations, explore synergies, and scale up for sustainable productivity. Soil salinity, affecting 20–50% of irrigated farmland globally, poses a significant threat to agriculture and food security, worsened by climate change and increasing droughts. Traditional methods for managing saline soils—such as leaching, gypsum addition, and soil excavation—are costly and often unsustainable. An alternative approach using plant growth-promoting microorganisms (PGPMs) offers promise for improving crop productivity in saline conditions. This study tested twenty-three bacterial strains, one yeast, and one fungal strain, isolated from diverse sources including salicornia plants, sandy soils, tomato stems or seeds, tree leaves, stems, and flowers. They were initially submitted to in vitro selection tests to assess their ability to promote plant growth under salt stress. In vitro tests included auxin production, phosphate solubilization, and co-culture of microorganisms and tomato seedlings in salt-supplemented media. The Bacillus sp. strain 44 showed the highest auxin production, while Bacillus megaterium MJ had the strongest phosphate solubilization ability. Cryptococcus sp. STSD 4 and Gliomastix murorum (4)10-1(iso1) promoted germination and the growth of tomato seedlings in an in vitro co-culture test performed on a salt-enriched medium. This innovative test proved particularly effective in selecting relevant strains for in planta trials. The microorganisms that performed best in the various in vitro tests were then evaluated in vivo on tomato plants grown in greenhouses. The results showed significant improvements in growth, including increases in fresh and dry biomass and stem size. Among the strains tested, Gliomastix murorum (4)10-1(iso1) stood out, delivering an increase in fresh biomass of 94% in comparison to the negative control of the salt modality. These findings highlight the potential of specific PGPM strains to enhance crop resilience and productivity in saline soils, supporting sustainable agricultural practices. Full article

Microbes produce various bioactive metabolites that can influence plant growth and stress tolerance. In this study, a plant growth-promoting rhizobacterium (PGPR), strain S14, was identified as Micrococcus luteus (designated as MlS14) using de novo whole-genome assembly. The MlS14 genome revealed major gene clusters for the synthesis of indole-3-acetic acid (IAA), terpenoids, and carotenoids. MlS14 produced significant amounts of IAA, and its volatile organic compounds (VOCs), specifically terpenoids, exhibited antifungal activity, suppressing the growth of pathogenic fungi. The presence of yellow pigment in the bacterial colony indicated carotenoid production. Treatment with MlS14 activated the expression of β-glucuronidase (GUS) driven by a promoter containing auxin-responsive elements. The application of MlS14 reshaped the root architecture of Arabidopsis seedlings, causing shorter primary roots, increased lateral root growth, and longer, denser root hairs; these characteristics are typically controlled by elevated exogenous IAA levels. MlS14 positively regulated seedling growth by enhancing photosynthesis, activating antioxidant enzymes, and promoting the production of secondary metabolites with reactive oxygen species (ROS) scavenging activity. Pretreatment with MlS14 reduced H2O2 and malondialdehyde (MDA) levels in seedlings under drought and heat stress, resulting in greater fresh weight during the post-stress period. Additionally, exposure to MlS14 stabilized chlorophyll content and growth rate in seedlings under salt stress. MlS14 transcriptionally upregulated genes involved in antioxidant defense and photosynthesis. Furthermore, genes linked to various hormone signaling pathways, such as abscisic acid (ABA), auxin, jasmonic acid (JA), and salicylic acid (SA), displayed increased expression levels, with those involved in ABA synthesis, using carotenoids as precursors, being the most highly induced. Furthermore, MlS14 treatment increased the expression of several transcription factors associated with stress responses, with DREB2A showing the highest level of induction. In conclusion, MlS14 played significant roles in promoting plant growth and stress tolerance. Metabolites such as IAA and carotenoids may function as positive regulators of plant metabolism and hormone signaling pathways essential for growth and adaptation to abiotic stress. Full article

This study investigates arbuscular mycorrhizal fungi (AMF), plant growth-promoting rhizobacteria (PGPR), and their combined application under salt stress (200 mM NaCl), emphasizing their synergistic potential to enhance plant resilience. Conducted in a controlled climate chamber, key parameters such as plant height, biomass, SPAD values, ion leakage, relative water content (RWC), osmotic potential, and mineral uptake were assessed. Salt stress significantly reduced plant growth, chlorophyll content, and nutrient absorption. However, AMF and PGPR improved plant performance, with co-inoculation showing the highest efficacy in increasing RWC, nutrient uptake, and maintaining membrane stability. AMF and PGPR treatments enhanced potassium retention and reduced sodium and chloride accumulation, mitigating ionic imbalances. The improved chlorophyll content and water relations under co-inoculation demonstrate the potential of these biostimulants to boost photosynthesis and plant resilience. These findings highlight AMF and PGPR as eco-friendly solutions for sustainable agriculture, promoting crop productivity and stress tolerance under saline conditions. Full article
