Downy mildew caused by Plasmopara viticola is an important disease in grape production, particularly in the highly susceptible, widely cultivated Vitis vinifera L. Breeding for disease resistance is an effective solution, and V. vinifera intraspecific crosses can yield progeny with both disease resistance and high quality. To assess the potential of intraspecific recurrent selection in V. vinifera (IRSV) in improving grapevine resistance to downy mildew and to analyze the pattern of disease resistance inheritance, the disease-resistant variety Ecolly was selected as one of the parents and crossed with Cabernet Sauvignon, Marselan, and Dunkelfelder, respectively, creating three reciprocal combinations, resulting in 1657 hybrid F1 progenies. The primary results are as follows: (1) significant differences in disease resistance among grape varieties and, significant differences in disease resistance between different vintages of the same variety were found; (2) the leaf downy mildew resistance levels of F1 progeny of different hybrid combinations conformed to a skewed normal distribution and showed some maternal dominance; (3) the degree of leaf bulbous elevation was negatively correlated with the level of leaf downy mildew resistance, and the correlation coefficient with the level of field resistance was higher; (4) five progenies with higher levels of both field and in vitro disease resistance were obtained. Intraspecific hybridization can improve the disease resistance of offspring through super-parent genetic effects, and Ecolly can be used as breeding material for recurrent hybridization to obtain highly resistant varieties. Discovering, analyzing, and finding a key to understanding the physiological and biochemical responses that Vitis vinifera L. undertakes against drought stress is of fundamental importance for this profitable crop. Today’s considerable climatic fluctuations force researchers and farmers to focus on this issue with solutions inclined to respect the ecosystem. In this academic work, we focused on describing the drought stress consequences on several parameters of secondary metabolites on Vitis vinifera leaves (quercetins, kaempferol, resveratrol, proline, and xanthophylls) and on some ecophysiological characteristics (e.g., water potential, stomatal conductance, and leaf temperature) to compare the answers that diverse agronomic management techniques (i.e., irrigation with and without zeolite, pure zeolite and no application) could instaurate in the metabolic pathway of this important crop with the aim to find convincing and thought-provoking responses to use this captivating and versatile mineral, the zeolite known as the “magic rock”. Stressed grapevines reached 56.80 mmol/m2s gs at veraison and a more negative stem Ψ (+10.63%) compared to plants with zeolite. Resveratrol, in the hottest season, fluctuated from 0.18–0.19 mg/g in zeolite treatments to 0.37 mg/g in stressed vines. Quercetins were inclined to accumulate in response to drought stress too. In fact, we recorded a peak of quercetin (3-O-glucoside + 3-O-glucuronide) of 11.20 mg/g at veraison in stressed plants. It is interesting to note how the pool of metabolites was often unchanged for plants treated with zeolite and for plants treated with water only, thus elevating this mineral to a “stress reliever”. Climate change is usually recognized as the most significant challenge facing viticulture in the 21st century. As a result, experts are increasingly emphasizing the need to explore the biodiversity within the species Vitis vinifera L. In this context, the present study investigated the intra-varietal biodiversity of two widely cultivated grapevine varieties on the Canary Islands of Lanzarote and Fuerteventura (Spain). These islands, characterized by desert-like climates, strong winds, volcanic soils, and phylloxera-free conditions, have presented uninterrupted grapevine cultivation for the past three to five centuries. Intra-varietal variability was detected in 93.46% of the 107 accessions analyzed. The most divergent samples were a Malvasia Dubrovacka (LNZ-87) and a Listan prieto (FTV-8), each exhibiting five distinct variations. Another Listan prieto accession (FTV-13) showed four variations. A group of seven individuals displayed three variations including two Malvasia volcanica accessions (LNZ-12, LNZ-72) and five Listan prieto accessions (FTV-1, FTV-2, FTV-7, FTV-9, FTV-12). A set of 100 SSR markers was used to analyze this grapevine collection, of which 17 revealed variability. The most informative markers were VChr15b, VVIp34, VVMD32, VChr9b, VVMD5, VVMD28, and VMC4F3, while the least informative was VVNTM1, which detected no variation. The parentage of Malvasia volcanica (Malvasia Dubrovacka × Bermejuela) was supported by all SSR markers, assuming that three of them may involve a mutated parent. Full article

Entomopathogenic fungi (EPF) are substantial biocontrol agents reducing the populations of economically important pests in numerous crops. Recent findings indicate that their role in agroecosystems is more complex and extends to affecting plant physiology and growth. This study examined the effects of Beauveria bassiana and Isaria fumosorosea, as well as Salicylic acid (SA), on physiological parameters of grapevine (Vitis vinifera cv. Sauvignon Blanc). Additionally, the impact of SA on spore germination and pathogenicity of EPF against larvae of the European grapevine moth (Lobesia botrana) was tested. Foliar application of EPF was found to increase the electron transport rate (ETR) from PSII to PSI, indicating higher photosynthetic activity compared to control plants. EPF also elevated the transpiration rate (E) and stomatal conductance (gs). In contrast, SA treatments decreased E and gs, while the high dose (10 mM) exhibited reduced Fv/Fm value, accompanied by phytotoxic spots on leaves. Spore germination of both fungi was significantly reduced only by the SA concentration of 2 mM, while 0.5 and 1 mM did not affect germination. Combination EPF and SA treatments presented the highest larval mortality of L. botrana (87.5% at 28 °C and 77.5% at 24 °C for B. bassiana and I. fumosorosea, respectively). However, SA reduced larval mycosis in most cases. Overall, the results suggest that EPF and SA can be co-applied and included in vineyard integrated strategies to support grapevine health. Caffeic acid-O-methyltransferase (COMT) is a key enzyme in lignin synthesis and secondary metabolism in plants, and it participates in the regulation of plant growth and development as well as plants’ stress response. To further investigate the function of COMT in grapevine, a total of 124 COMT family genes were identified from three Vitis species in this study, namely Pinot noir (Vitis vinifera L.), Vitis amurensis, and Vitis riparia. The amino acid sequence encoded by these genes ranged from 55 to 1422 aa, and their molecular mass ranged from 6640.82 to 77,034.43 Da. Subcellular localization prediction inferred that they were mainly located in the plasma membrane and cytoplasm. The prediction of secondary structures showed that α-helix and irregular coiled-coil were primary structural elements. These genes were unevenly distributed across 10 different chromosomes, respectively. Phylogenetic tree analysis of the amino acid sequences of VvCOMT, VaCOMT, VrCOMT, and AtCOMT proteins showed that they were closely related and were divided into four subgroups. The motif distribution was similar among the cluster genes, and the gene sequence was notably conserved. The 124 members of the COMT gene family possessed a variable number of exons, ranging from 2 to 13. The promoter region of all of these COMTs genes contained multiple cis-acting elements related to hormones (e.g., ABA, IAA, MeJA, GA, and SA), growth and development (e.g., endosperm, circadian, meristem, light response), and various stress responses (e.g., drought, low temperature, wounding, anaerobic, defense, and stress). The intraspecies collinearity analysis suggested that there were one pair, three pairs, and six pairs of collinear genes in Va, Pinot noir, and Vr, respectively, and that tandem duplication contributed more to the expansion of these gene family members. In addition, interspecific collinearity revealed that the VvCOMTs had the strongest homology with the VaCOMTs, followed by the VrCOMTs, and the weakest homology with the AtCOMTs. The expression patterns of different tissues and organs at different developmental stages indicated that the VvCOMT genes had obvious tissue expression specificity. The majority of VvCOMT genes were only expressed at higher levels in certain tissues. Furthermore, we screened 13 VvCOMT genes to conduct qRT-PCR verification according to the transcriptome data of VvCOMTs under abiotic stresses (NaCl, PEG, and cold). The results confirmed that these genes were involved in the responses to NaCl, PEG, and cold stress. This study lays a foundation for the exploration of the function of the COMT genes, and is of great importance for the genetic improvement of abiotic stress resistance in grapes. Full article

Influence of Biochar Foliar Application on Malvazija Istarska Grapevine Physiology

Biochar has attracted interest in viticulture for its potential to enhance nutrient uptake and improve grapevine physiology under changing climatic conditions, particularly in Mediterranean regions. However, the widespread adoption of biochar has been limited due to economic and logistical constraints associated with its large-scale application. To address these barriers hindering the widespread adoption of biochar, this study investigates the effects of foliar-applied water suspensions of biochar at concentrations of 300 mg/L (B300), 600 mg/L (B600), and 1200 mg/L (B1200), compared to a water-only control (C), as a practical alternative application method. The research focused on Malvazija istarska (Vitis vinifera L.), an indigenous Croatian grapevine variety, conducted in an experimental vineyard in Poreč, Croatia. The key physiological parameters examined included photo-synthetic activity, leaf water potential, the elemental composition of the grapevine leaves, and grape yield. Foliar applications were administered three times during the growing season, with five replicates per treatment. The results indicated that biochar treatments had no significant impact on photosynthetic activity, suggesting that foliar application did not cause leaf shading. However, higher biochar concentrations (B600 and B1200) led to increased leaf concentrations of nitrogen (2.1–3.8%), potassium (10.1–18.4 g/kg), sulfur (2.2–2.5 g/kg), boron (65.1–83.6 mg/kg), and manganese (42.4–69.8 mg/kg) compared to B300 and C treatments. Conversely, magnesium content decreased (2.1–2.7 g/kg), likely due to potassium–magnesium antagonism. Furthermore, the B600 treatment produced the highest grape yield (2.67 kg/vine), representing up to a 37% increase compared to other treatments. These findings suggest that the foliar application of biochar can be an effective and sustainable strategy to enhance vineyard productivity. Moreover, it offers a circular economy approach by valorizing grapevine pruning waste as a biochar source. Full article

Grape (Vitis vinifera L.) is one of the most extensively cultivated crops in temperate climates, with its primary fate being wine production, which is paired with a great generation of grape pomace (GP). GP contains a plethora of antioxidant phenolic compounds, being well-known for its high content of various tannins, liable for the astringency of this fruit. Winemaking produces a great mass of by-products that are rich in tannins. Grape seed (GSd) and pulp waste, as well as leaves and stems (GSt), are rich in condensed tannins (CTs), while its skin (GSk) contains more flavonols and phenolic acids. CTs are polymers of flavan-3-ols, and their antioxidant and anti-inflammatory properties are well-accounted for, being the subject of extensive research for various applications. CTs from the diverse fractions of grapefruit and grapevine share similar structures given their composition but diverge in their degree of polymerization, which can modulate their chemical interactions and may be present at around 30 to 80 mg/g, depending on the grape fraction. Thus, this prominent agroindustrial by-product, which is usually managed as raw animal feed or further fermented for liquor production, can be valorized as a source of tannins with high added value. The present review addresses current knowledge on tannin diversity in grapefruit and grapevine by-products, assessing the differences in composition, quantity, and degree of polymerization. Current knowledge of their reported bioactivities will be discussed, linking them to their current and potential applications in food and feed. Full article

Among grapevine trunk diseases, Eutypa dieback, caused by the fungus Eutypa lata, is one of the most critical ones, due to its widespread infection in vineyards and the lack of effective treatments. This fungus is a vascular pathogen that enters grapevines through pruning wounds. The infection process is associated with phytotoxic metabolites produced by the fungus, and as such, the identification of new metabolites from different culture conditions and broths could provide valuable insights into the fungus’s enzymatic system and help its control. For the purposes of this study, the OSMAC (one strain, many compounds) approach was applied to investigate the secondary metabolism of E. lata strain 311 isolated from Vitis vinifera plants in Spain. A total of twenty metabolites were isolated, including five reported for the first time from E. lata and four that are newly identified compounds in the literature: eulatagalactoside A, (R)-2-(4′-hydroxy-3′-methylbut-1′-yn-1′-yl)-4-(hydroxymethyl)phenol, (S)-7-hydroxymethyl-3-methyl-2,3-dihydro-1-benzoxepin-3-ol, and (3aR,4S,5R,7aS)-4,5-dihydroxy-6-((R)-3′-methylbuta-1′,3′-dien-1′-ylidene)hexahydrobenzo[d][1,3]dioxol-2-one. These compounds were extracted from fermentation broths using silica gel column chromatography and high-performance liquid chromatography (HPLC). Their structures were elucidated through extensive 1D and 2D NMR spectroscopy, along with high-resolution electrospray ionization mass spectrometry (HRESIMS). Compounds were evaluated for phytotoxicity against Phaseolus vulgaris, with only eulatagalactoside A producing white spots after 48 h. Additionally, the antibacterial activity against Escherichia coli, Staphylococcus aureus, and Klebsiella pneumoniae of selected compounds was tested. The compounds (R)-2-(4′-hydroxy-3′-methylbut-1′-yn-1′-yl)-4-(hydroxymethyl)phenol and (S)-7-(hydroxymethyl)-3-methyl-2,3-dihydrobenzo[b]oxepin-3-ol showed the most significant antimicrobial activity against Gram-positive bacteria, inhibiting S. aureus by over 75%, with IC50 values of 511.4 µg/mL and 617.9 µg/mL, respectively. Excessive shoot vigor in grapevines negatively impacts plant growth and fruit quality, necessitating the use of plant growth regulators (PGRs) for canopy management. This study investigated the effects of mepiquat chloride (MC) and chlormequat chloride (CCC) on shoot growth (including new shoot length, relative chlorophyll content, leaf area, etc.) and fruit quality in Vitis vinifera cv. ‘Shine Muscat’. Different concentrations of MC (100, 300, 500, 700 mg/L) and CCC (100, 300, 500, 700 mg/L) were applied via foliar spraying at multiple stages before flowering. The results demonstrated that both PGRs effectively suppressed shoot elongation, with CCC exhibiting superior inhibitory efficacy compared to MC. However, high concentration of either compound also restricted leaf and cluster development. Optimal treatments MC (500 mg/L) and CCC (100 mg/L) significantly enhanced berry size, soluble solids content (SSC), and solid–acid ratio while maintaining effective shoot control. For practical application, we recommend spraying MC (500 mg/L) or CCC (100 mg/L) during the new shoot growth, flower-cluster separation, and flowering stages of ‘Shine Muscat’ grapevines to improve the new shoot control effect and fruit quality. The commercial production of grapevines (Vitis vinifera L.) relies heavily on rootstocks that are hybrids of non-vinifera parentage. The relatively newly released GRN rootstocks (GRN-1, GRN-2, GRN-3, GRN-4, and GRN-5) were bred from especially under-studied genetic backgrounds. This study aimed to evaluate ungrafted GRN-series grape rootstocks under moderate water-stress conditions and to characterize and compare their physiological performances. Each of the GRN rootstocks had specific physiological characteristics that would make them suitable for a wide range of growing conditions and vineyard management goals. GRN-1 had growth habits which were more vigorous and the highest carbohydrate storage levels, while GRN-2 had the highest level of nitrogen and the largest leaf area, but the lowest levels of carbohydrate storage. GRN-3 was less tolerant to high-salinity soils, and had the longest internodes, while GRN-4 had high boron levels, which supports flowering and fruit set, and short internodes. GRN-5 was consistently moderate across all measured areas, except internode thickness, for which it was the highest. These findings show the variations in physiological growth habits among the ungrafted GRN-series rootstocks and suggest that growth habits, carbohydrate storage, leaf canopy, fruit production, and nutrition vary based on rootstock parentage. Further investigation is needed to determine whether these characteristics persist when grafted onto Vitis vinifera L. scions. Full article

Abstract
Grapevines (Vitis vinifera L.) face significant challenges from abiotic stresses caused by climate change, including drought, salinity, and temperature extremes. Grapevines (Vitis vinifera L.) face significant challenges from abiotic stresses caused by climate change, including drought, salinity, and temperature extremes. This comprehensive review examined the role of beneficial microorganisms in enhancing grapevine tolerance to these stresses, focusing on arbuscular mycorrhizal fungi (AMF), plant growth-promoting rhizobacteria (PGPR), and endophytes. The study analyzes species-specific effects and their molecular mechanisms, highlighting how single and consortium inoculations improve plant resilience. AMF species, particularly Funneliformis mosseae and Rhizophagus irregularis, demonstrated significant enhancement in drought and salinity tolerance through improved nutrient uptake and stress response modulation. The PGPRs, Bacillus and Pseudomonas species, show remarkable abilities to mitigate various abiotic stresses through mechanisms including phytohormone production and antioxidant defense enhancement. Endophytic microorganisms such as Pseudomonas fluorescens RG11 and Serendipita indica play crucial roles in stress mitigation through melatonin production and improved water retention, respectively. The synergistic effects of combined AMF, PGPR, and PGPF applications led to a significant increase in grapevine drought and salinity tolerance, improving nutrient uptake, photosynthesis rates, and antioxidant defense mechanisms. Molecular analysis revealed that these microbial consortia regulate the expression of stress-responsive genes, particularly VvNCED and VvP5CS, enhancing grapevine resilience through improved osmotic adjustment, ROS scavenging, and hormonal regulation. These findings provide valuable insights into the molecular pathways underlying stress tolerance, offering promising strategies for sustainable viticulture under climate change. Full article

N-(2-chloro-4-pyridyl)-N′-phenylurea (CPPU) and N-phenyl-1,2,3-thidiazole-5ylurea (TDZ) are plant growth regulators used for seedless treatment in grape. In this study, the flowers of ‘Shine Muscat’ (Vitis labruscana Bailey × V. vinifera L.) were treated with 3, 5, and 10 mg/L CPPU and TDZ one week before flowering. The results showed that both CPPU and TDZ treatments reduced the pollen germination rate and caused abnormal stamen and pollen grain phenotypes, resembling the male sterility observed in ‘Y_14’ (a novel grapevine germplasm derived from the self-progeny of ‘Shine Muscat’). Using RNA-seq technology, the stamens of flowers treated with 10 mg/L CPPU (CPPU_10), 10 mg/L TDZ (TDZ_10), and the control (CK) were analyzed. A total of 520 and 722 differentially expressed genes (DEGs) were identified in CPPU and TDZ treatments, respectively. GO and KEGG analyses revealed that the common pathways leading to pollen abortion in both treatments were primarily associated with hydrolase activity (acting on glycosyl bonds), phenylpropanoid biosynthesis, pentose and glucuronate interconversions, and ABC transporters. By comparing the DEGs across the three groups (Y_14 vs. SM, CPPU_10 vs. CK, TDZ_10 vs. CK), 16 DEGs exhibited similar expression patterns. Further tissue-specific expression analysis identified nine genes that were highly expressed in stamens and shared the same expression pattern in sterile lines. These findings provide a foundation for further studies on the impact of CPPU and TDZ treatments on grape stamen fertility. Full article

Functional Characterization of Grapevine VviMYC4 in Regulating Drought Tolerance by Mediating Flavonol Biosynthesis

Drought ranks among the key abiotic stresses that limit the growth and yield of grapevines (Vitis vinifera L.). Flavonols, a class of antioxidants commonly found in grapevines, play a crucial role in combating drought stress. In this study, we characterized the function and regulatory mechanism of the grapevine VviMYC4 in mediating flavonol biosynthesis in response to drought stress. VviMYC4 encodes a protein of 468 amino acids with conserved bHLH-MYC_N and bHLH domains. Phylogenetic analysis confirmed its homology with the grapevine VviMYC2 and similarity in function. The expression of VviMYC4 in ‘Cabernet Sauvignon’ grapevine seedling leaves increased initially and then decreased during prolonged drought stress. The homologous and heterologous transformation of VviMYC4 in grape suspension cells, Arabidopsis plants, tobacco leaves, and grapevine leaves demonstrated its ability to positively regulate flavonol biosynthesis and accumulation by promoting the expression of flavonol-related genes, thereby enhancing the drought tolerance of transgenic plants. Furthermore, VviMYC4 could bind to specific E-box sites on the promoters of VviF3H and VviFLS to improve their activities. This study highlights VviMYC4 as a pivotal positive regulator of drought tolerance in grapevines and proposes that VviMYC4 enhances the antioxidant and reactive oxygen species (ROS) scavenging abilities of grapevines in challenging environments and improves their stress resilience by mediating flavonol biosynthesis. Our findings offer crucial candidate genes and valuable insights for the molecular breeding of grapevine drought resistance. Full article

The cultivation of table grapes in Brazil is economically significant, with production influenced by edaphoclimatic factors and rootstock selection. The cultivar ‘BRS Núbia’ (Vitis vinifera L. x Vitis labrusca L.) is a promising alternative; however, its phenological behavior, thermal requirements, and compatibility with different rootstocks under subtropical conditions require further evaluation. This study aimed to assess the duration of phenological stages, thermal requirement, and ripening dynamics of ‘BRS Núbia’ grapevines grafted onto the rootstocks ‘IAC 572 Jales’, ‘IAC 766 Campinas’, and ‘Paulsen 1103’. The experiment was conducted in São Manuel, São Paulo, Brazil during the 2021 and 2022 production cycles using a split-plot experimental design (3 × 2). Evaluations included the duration of phenological stages from pruning to budburst, flowering, fruit set, onset of ripening, and harvest, as well as the ripening curve and thermal accumulation from pruning to harvest. Rootstocks did not significantly affect (p > 0.05) the duration of phenological stages; however, differences were observed between production cycles. The 2022 cycle was longer (167.7 days) compared to 2021 (142.6 days), with greater thermal accumulation (1871.7 GDDs vs. 1743.4 GDDs). The analysis of phenological stages revealed that, across both production cycles evaluated, the ‘BRS Núbia’ cultivar required an average accumulation of 1807.5 growing degree days from pruning to harvest. Soluble solids content ranged from 17.43 to 18.50°Brix, and titratable acidity decreased throughout maturation. The maturation index was highest in vines grafted onto ‘Paulsen 1103’, indicating its positive influence on fruit quality. The ‘BRS Núbia’ grapevine exhibited a mean thermal requirement of 1807.5 growing degree days (GDDs) to complete its phenological cycle, which lasted approximately 150 days under subtropical conditions. Bunch compactness (BC) is a complex, multi-trait characteristic that has been studied mostly in the context of wine grapes, with table grapes being scarcely considered. As these groups have marked phenotypic and genetic differences, including BC, the study of this trait is reported here using a genetically diverse collection of 116 Vitis vinifera L. cultivars and lines enriched for table grapes over two seasons. For this, 3D scanning-based morphological data were combined with ground measurements of 14 BC-related traits, observing high correlations among both approaches (R2 > 0.90–0.97). The multivariate analysis suggests that the attributes ‘berries per bunch’, ‘berry weight and width’, and ‘bunch weight and length’ could be considered as the main descriptors for BC, optimizing evaluation times. Then, GWASs based on a set of 70,335 SNPs revealed that GBS analysis in this same population enabled the detection of several SNPs associated with different sub-traits, with a locus for ‘berries per bunch’ in chromosome (chr) 18 being the most prominent. Enrichment analysis of significant and frequent SNPs found simultaneously in several traits and seasons revealed the over-representation of discrete functions such as alpha-linolenic acid metabolism and glycan degradation. In summary, the utility of 3D automated phenotyping was validated for table grape backgrounds, and new SNPs and candidate genes associated with the BC trait were detected. The latter could eventually become a selection tool for grapevine breeding programs. Full article
