Reliable, non-destructive grading of fresh fruit requires simultaneous assessment of external morphology and hidden internal defects. Camera-based grading of fresh fruit using colorimetric (RGB) and near-infrared (NIR) imaging often misses subsurface bruising and cannot capture the fruit’s true shape, leading to inconsistent quality assessment and increased waste. To address this, we developed a 4D-grading pipeline that fuses visible and near-infrared (VNIR) and short-wave infrared (SWIR) hyperspectral imaging with structured-light 3D scanning to non-destructively evaluate both internal defects and external form. Our contributions are (1) flagging the defects in fruits based on the reflectance information, (2) accurate shape and defect measurement based on the 3D data of fruits, and (3) an interpretable, decision-tree framework that assigns USDA-style quality (Premium, Grade 1/2, Reject) and size (Small–Extra Large) labels. We demonstrate this approach through preliminary results, suggesting that 4D hyperspectral imaging may offer advantages over single-modality methods by providing clear, interpretable decision rules and the potential for adaptation to other produce types. Background: The implementation of advanced agronomical strategies, including the use of antitranspirant, in order to mitigate the negative effects of environmental stress, particularly heat stress on plants, has become a focal area of research in the Mediterranean basin. This region is characterized by hot and dry summer that affects plant physiology. Methods: The experiment was carried out in Sicily (South Italy) on 12-year-old avocado cv. Hass grafted onto Walter Hole rootstock. Two subplots each of forty homogenous trees were selected and treated (1) with calcium carbonate (DECCO Shield®) and (2) with water (control) at the following phenological phases: 711, 712 and 715 BBCH. The climatic parameters were recorded throughout the year. Physiological measurements (leaf transpiration, net photosynthesis, stomatal conductance, leaf water potential) were measured at 105, 131 and 168 days after full bloom. Fruit growth was monitored, and physico-chemical analyses were carried out at harvest. Results: The antitranspirant increased photosynthesis and stomatal conductance and reduced leaf transpiration (−26.1%). Fruit growth rate increased during summer, although no morphological and qualitative difference was observed at harvest. PCA highlighted the positive effect of the calcium carbonate on overall plant physiology. Conclusions: Antitranspirant foliar application reduced heat stress effects by improving physiological responses of avocado trees. Full article

To improve the postharvest preservation of cherry tomatoes and combat pathogenic, both bacterial and fungal contamination (particularly Alternaria alternata), a novel biodegradable coating was developed based on a water-in-water (W/W) Pickering emulsion system. The emulsion was stabilized by L. plantarum (Lactobacillus plantarum), with maltodextrin (MD) as the dispersed phase and hydroxypropyl methylcellulose (HPMC) as the continuous phase. Characterization of emulsions at varying concentrations revealed that the optimized W/W-PL^8 film exhibited superior stability, smooth morphology, and low water vapor permeability (WVP = 220.437 g/(m2·24 h)), making it a promising candidate for fruit and vegetable preservation. Furthermore, the coating demonstrated strong antioxidant activity, with scavenging rates of 58.99% (ABTS) and 94.23% (DPPH), along with potent antimicrobial effects, showing inhibition rates of 12.8% against Escherichia coli and 23.7% against Staphylococcus aureus. Applied to cherry tomatoes, the W/W-PL^8 coating significantly reduced respiration rates, minimized decay incidence, and maintained nutritional quality during storage. Remarkably, the coating successfully controlled Alternaria alternata contamination, enhancing the storage duration of cherry tomatoes. These findings highlight the potential of W/W-PL^8 as an eco-friendly and functional packaging material for fresh produce preservation. Full article

This study aimed to develop a functional powder using whey and milk matrices, leveraging the protective capacity of chia–alginate hydrogels and the advantages of electrohydrodynamic spraying (EHDA), a non-thermal technique suitable for encapsulating probiotic cells under stress conditions commonly encountered in food processing. A hydrogel matrix composed of chia seed mucilage and sodium alginate was used to form a biopolymeric network that protected probiotic cells during processing. The encapsulation efficiency reached 99.0 ± 0.01%, and bacterial viability remained above 9.9 log10 CFU/mL after lyophilization, demonstrating the excellent protective capacity of the hydrogel matrix. Microstructural analysis using confocal laser scanning microscopy (CLSM) revealed well-retained cell morphology and homogeneous distribution within the hydrogel matrix while, in contrast, scanning electron microscopy (SEM) showed spherical, porous microcapsules with distinct surface characteristics influenced by the encapsulation method. Encapsulates were incorporated into beverages flavored with red fruits and pear and subsequently freeze-dried. The resulting powders were analyzed for moisture, protein, lipids, carbohydrates, fiber, and color determinations. The results were statistically analyzed using ANOVA and response surface methodology, highlighting the impact of ingredient ratios on nutritional composition. Raman spectroscopy identified molecular features associated with casein, lactose, pectins, anthocyanins, and other functional compounds, confirming the contribution of both matrix and encapsulants maintaining the structural characteristics of the product. The presence of antioxidant bands supported the functional potential of the powder formulations. Chia–alginate hydrogels effectively encapsulated L. reuteri, maintaining cell viability and enabling their incorporation into freeze-dried beverage powders. This approach offers a promising strategy for the development of next-generation functional food gels with enhanced probiotic stability, nutritional properties, and potential application in health-promoting dairy systems. Mushrooms are eukaryotic organisms with absorptive heterotrophic nutrition, capable of feeding on organic matter rich in cellulose and lignocellulose. Since ancient times, they have been considered allies and, in certain cultures, they were seen as magical beings or food of the gods. Of the great variety of edible mushrooms identified worldwide, less than 2% are traded on the market. Although mushrooms have been valued for their multiple nutritional and healing benefits, some cultures perceive them as toxic and do not accept them in their culinary practices. Despite the existing skepticism, several researchers are promoting the potential of edible mushrooms. There are two main methods of mushroom cultivation: solid-state fermentation and submerged fermentation. The former is the most widely used and simplest, since the fungus grows in its natural environment; in the latter, the fungus grows suspended without developing a fruiting body. In addition, submerged fermentation is easily monitored and scalable. Both systems are important and have their limitations. This article discusses the main methods used to increase the performance of submerged fermentation with emphasis on the modes of operation used, types of bioreactors and application of morphological bioengineering of filamentous fungi, and especially the use of intelligent automatic control technologies and the use of non-invasive monitoring in fermentation systems thanks to the development of machine learning (ML), neural networks, and the use of big data, which will allow more accurate decisions to be made in the fermentation of filamentous fungi in submerged environments with improvements in production yields. In Brazil, Plasmodium infections in non-human primates (NHPs) have been associated with P. simium and P. brasilianum, which are morphologically and genetically similar to the human-infecting species P. vivax and P. malariae, respectively. Surveillance and monitoring of wild NHPs are crucial for understanding the distribution of these parasites and assessing the risk of zoonotic transmission. This study aimed to detect the presence of Plasmodium spp. genetic material in Platyrrhini primates from 47 municipalities in the states of Bahia and Minas Gerais. The animals were captured using Tomahawk-type live traps baited with fruit or immobilized with tranquilizer darts. Free-ranging individuals were chemically restrained via inhalation anesthesia using VetBag® or intramuscular anesthesia injection. Blood samples were collected from the femoral vein. A total of 298 blood and tissue samples were collected from 10 primate species across five genera: Alouatta caraya (25), Alouatta guariba clamitans (1), Callicebus melanochir (1), Callithrix geoffroyi (28), Callithrix jacchus (4), Callithrix kuhlii (31), Callithrix penicillata (175), Callithrix spp. hybrids (15), Leontopithecus chrysomelas (16), Sapajus robustus (1), and Sapajus xanthosthernos (1). Molecular diagnosis was performed using a nested PCR targeting the 18S small subunit ribosomal RNA (18S SSU rRNA) gene, followed by sequencing. Of the 298 samples analyzed, only one (0.3%) from Bahia tested positive for Plasmodium brasilianum/P. malariae. This represents the first detection of this parasite in a free-living C. geoffroyi in Brazil. These findings highlight the importance of continued surveillance of Plasmodium infections in NHPs to identify regions at risk for zoonotic transmission. Full article

Effective postharvest cooling of olive fruit is increasingly critical under rising harvest temperatures driven by climate change. This study models passive cooling dynamics using a trait-based, mixed-effects statistical framework. Ten olive groups—representing seven cultivars and different ripening or size stages—were subjected to controlled cooling conditions. Surface temperature was recorded using infrared thermal imaging, and morphological and compositional traits were quantified. Temperature decay was modeled using Newton’s Law of Cooling, extended with a quadratic time term to capture nonlinear trajse thectories. A linear mixed-effects model was fitted to log-transformed, normalized temperature data, incorporating trait-by-time interactions and hierarchical random effects. The results confirmed that fruit weight, specific surface area (SSA), and specific heat capacity (SHC) are key drivers of cooling rate variability, consistent with theoretical expectations, but quantified here using a trait-based statistical model applied to olive fruit. The quadratic model consistently outperformed standard exponential models, revealing dynamic effects of traits on temperature decline. Residual variation at the group level pointed to additional unmeasured structural influences. This study demonstrates that olive fruit cooling behavior can be effectively predicted using interpretable, trait-dependent models. The findings offer a quantitative basis for optimizing postharvest cooling protocols and are particularly relevant for maintaining quality under high-temperature harvest conditions. Full article

With the objective of identifying superior Vernicia montana trees grounded in phenotypic and agronomic traits, this study sought to develop and implement a comprehensive evaluation method which would provide a practical foundation for future clonal breeding initiatives. Using the Vernicia montana propagated from seedling forests grown in the Suxian District of Chenzhou City in southern Hunan Province, we conducted pre-selection, primary selection, and re-selection of Vernicia montana forest stands and took the nine trait indices of single-plant fruiting quantity, single-plant fruit yield, disease and pest resistance, fruit ripening consistency, fruit aggregation, fresh fruit single-fruit weight, fresh fruit seed rate, dry seed kernel rate, and seed kernel oil content rate as the optimal evaluation indexes and carried out cluster analysis and a comprehensive evaluation in order to establish a comprehensive evaluation system for superior Vernicia montana trees. The results demonstrated that a three-stage selection process—consisting of pre-selection, primary selection, and re-selection—was conducted using a comprehensive analytical approach. The pre-selection phase relied primarily on sensory evaluation criteria, including fruit count per plant, tree size, tree morphology, and fruit clustering characteristics. Through this rigorous screening process, 60 elite plants were selected. The primary selection was based on phenotypic traits, including single-plant fruit yield, pest and disease resistance, and uniformity of fruit ripening. From this stage, 36 plants were selected. Twenty plants were then selected for re-selection based on key performance indicators, such as fresh fruit weight, fresh fruit seed yield, dry seed kernel yield, and oil content of the seed kernel. Then the re-selected optimal trees were clustered and analyzed into three classes, with 10 plants in class I, 7 plants in class II, and 3 plants in class III. In class I, the top three superior plants exhibited outstanding performance across key traits: their fresh fruit weight per fruit, fresh fruit seed yield, dry seed yield, and seed kernel oil content reached 41.61 g, 42.80%, 62.42%, and 57.72%, respectively. Compared with other groups, these figures showed significant advantages: 1.17, 1.09, 1.12, and 1.02 times the average values of the 20 reselected superior trees; 1.22, 1.19, 1.20, and 1.08 times those of the 36 primary-selected superior trees; and 1.24, 1.25, 1.26, and 1.19 times those of the 60 pre-selected trees. Fruits counts per plant and the number of fruits produced per plant of the best three plants in class I were 885 and 23.38 kg, respectively, which were 1.13 and 1.18 times higher than the average of 20 re-selected superior trees, 1.25 and 1.30 times higher than the average of 36 first-selected superior trees, and 1.51 and 1.58 times higher than the average of 60 pre-selected superior trees. Class I superior trees, especially the top three genotypes, are suitable for use as mother trees for scion collection in grafting. The findings of this study provide a crucial foundation for developing superior clonal varieties of Vernicia montana through selective breeding. Full article

Anchote (Coccinia abyssinica) is a neglected high-potential food and nutrition security tuber crop in Ethiopia. Phenotyping core germplasm collections using agro-morphological and physiological markers is essential for effective crop improvement and utilization. A total of 282 anchote germplasms were profiled using six qualitative and twenty-six quantitative agro-morphological and physiological traits. Augmented Block Design was used for the experiment at the Debre Zeit Agricultural Research Center. The chi-square test and Shannon diversity index indicated the presence of substantial phenotypic variation and diversity among the accessions based on the predominant qualitative traits studied. The quantitative agro-morphological and physiological traits showed wider variability and ranges for the accessions. The broad-sense heritability and genetic advance as a percentage of the mean were notably high for quantitative traits such as root yield, vine length, and leaf area index. A significantly positive correlation was observed among agronomically important traits such as root yield and root diameter as well as root yield and leaf area. The principal component analysis for qualitative and quantitative traits found that ten components explained 72.2% of the variation for qualitative traits, whereas nine components accounted for 69.96% of the variation in quantitative traits. The primary contributors to the variations are traits such as root (shape, flesh color, and yield), leaf (color, length, diameter, area) and fruit (length, diameter, and weight). Further, the accessions were grouped into two and three clusters based on qualitative and quantitative traits, respectively, indicating that quantitative characters better differentiated among the accessions. Similarly, the tanglegram showed little similarity between the qualitative and quantitative agro-morphological and physiological traits in clustering the accessions. These findings indicate the presence of sizable trait variation among the accessions that can be exploited as a selection marker to design and facilitate conservation and breeding strategies of anchote. Neuroinflammation, driven by activated microglia, contributes to the progression of neurodegenerative diseases. Extracellular vesicles mediate intercellular communication and influence immune responses. Chrysin, a natural flavone found in fruits and propolis, has demonstrated anti-inflammatory effects. This study explored the immunomodulatory potential of chrysin-loaded EVs (EVs-Chry) derived from BV2 microglial cells. BV2 cells were treated with chrysin for 24 h to assess cytotoxicity and proliferation. EVs were isolated from treated and untreated cells, characterized by nanoparticle tracking analysis, and applied to naïve BV2 cells prior to LPS stimulation. Effects on cell morphology, migration, cytokine expression (IL-1β, IL-6), inflammasome activity (caspase-1), and apoptosis-related protein Bcl-xL were investigated. Our results show that EVs-Chry significantly reduced LPS-induced cell proliferation, restored resting microglial morphology, and reduced migratory capacity. Furthermore, co-treatment with EVs-Chry and LPS reduced pro-inflammatory cytokines such as IL-1β, IL-6, and caspase-1 expression while enhancing anti-apoptotic Bcl-xL levels, indicating a shift toward an anti-inflammatory, neuroprotective micro-glial phenotype. Together, our results demonstrated that EVs-Chry have neuroprotective effects on LPS-induced microglial activation and modulate microglial responses to inflammatory stimuli, attenuating pro-inflammatory signaling and promoting cellular homeostasis. These findings support the therapeutic potential of EVs-Chry in the context of neuroinflammatory and neurodegenerative disorders. Full article

High Carbon Dioxide Concentration Inhibits Pileus Growth of Flammulina velutipes by Downregulating Cyclin Gene Expression

Flammulina velutipes is a widely cultivated edible mushroom in East Asia, recognized for its nutritional benefits and distinct morphology characterized by a long stipe and a compact, hemispherical pileus. The pileus not only plays a critical biological role in reproduction through spore formation but also serves as a key commercial trait influencing consumer preference and market value. Despite its economic importance, pileus development in F. velutipes is highly sensitive to environmental factors, among which carbon dioxide (CO2) concentration is particularly influential under indoor cultivation conditions. While previous studies have reported that elevated CO2 levels can inhibit pileus expansion in other mushroom species, the molecular mechanisms by which CO2 affects pileus growth in F. velutipes remain poorly understood. In this study, we investigated the impact of CO2 concentration on pileus morphology and gene expression in F. velutipes by cultivating fruiting bodies under two controlled atmospheric conditions: low (1000 ppm) and high (10,000 ppm) CO2. Morphometric analysis revealed that elevated CO2 levels significantly suppressed pileus expansion, reducing the average diameter by more than 50% compared to the low CO2 condition. To elucidate the underlying genetic response, we conducted RNA sequencing and identified 102 differentially expressed genes (DEGs), with 78 being downregulated under elevated CO2. Functional enrichment analysis highlighted the involvement of cyclin-dependent protein kinase regulatory pathways in this response. Two cyclin genes were found to be significantly downregulated under elevated CO2 conditions, and their suppression was validated through quantitative real-time PCR. These genes, possessing conserved cyclin_N domains, are implicated in the regulation of the eukaryotic cell cycle, particularly in mitotic growth. These results indicate that CO2-induced downregulation of cyclin genes may underlie cell cycle arrest, contributing to inhibited pileus development. This study is the first to provide transcriptomic evidence that elevated CO2 concentrations specifically repress PHO80-like cyclin genes in F. velutipes, revealing a molecular mechanism by which CO2 stress inhibits pileus development. These findings suggest that elevated CO2 triggers a morphogenetic checkpoint by repressing PHO80-like cyclins, thereby modulating cell cycle progression during fruiting body development. This study provides the first evidence of such a transcriptional response in edible mushrooms and offers promising molecular targets for breeding CO2-resilient strains and optimizing commercial cultivation conditions. Xylosalsola chiwensis (Popov) Akhani & Roalson is listed in the Red Data Book of Kazakhstan as a rare species with a limited distribution, occurring in small populations in Kazakhstan, Uzbekistan, and Turkmenistan. The aim of this study is to deepen the understanding of the ecological conditions of its habitats, the floristic composition of its associated plant communities, the species’ morphological and anatomical characteristics, and its molecular phylogeny, as well as to identify the main threats to its survival. The ecological conditions of the X. chiwensis habitats include coastal sandy plains and the slopes of chinks and denudation plains with gray–brown desert soils and bozyngens on the Mangyshlak Peninsula and the Ustyurt Plateau at altitudes ranging from −3 to 270 m above sea level. The species is capable of surviving in arid conditions (less than 100 mm of annual precipitation) and under extreme temperatures (air temperatures exceeding 45 °C and soil surface temperatures above 65 °C). In X. chiwensis communities, we recorded 53 species of vascular plants. Anthropogenic factors associated with livestock grazing, industrial disturbances, and off-road vehicle traffic along an unregulated network of dirt roads have been identified as contributing to population decline and the potential extinction of the species under conditions of unsustainable land use. The morphometric traits of X. chiwensis could be used for taxonomic analysis and for identifying diagnostic morphological characteristics to distinguish between species of Xylosalsola. The most taxonomically valuable characteristics include the fruit diameter (with wings) and the cone-shaped structure length, as they differ consistently between species and exhibit relatively low variability. Anatomical adaptations to arid conditions were observed, including a well-developed hypodermis, which is indicative of a water-conserving strategy. The moderate photosynthetic activity, reflected by a thinner palisade mesophyll layer, may be associated with reduced photosynthetic intensity, which is compensated for through structural mechanisms for water conservation. The flow cytometry analysis revealed a genome size of 2.483 ± 0.191 pg (2n/4x = 18), and the phylogenetic analysis confirmed the placement of X. chiwensis within the tribe Salsoleae of the subfamily Salsoloideae, supporting its taxonomic distinctness. To support the conservation of this rare species, measures are proposed to expand the area of the Ustyurt Nature Reserve through the establishment of cluster sites. Full article

Identification of Avocado Fruit Disease Caused by Diaporthe phaseolorum and Colletotrichum fructicola in China

Persea americana (avocado) is a healthy fruit, rich in unsaturated fatty acids, various minerals, and vitamins. As avocado cultivation continues to expand globally, its development is increasingly constrained by concomitant diseases, among which fruit rot and anthracnose have emerged as significant threats to fruit quality. Menglian in Yunnan Province is the largest avocado production area in China. In November 2024, fruit rot was observed on avocado fruits in Yunnan, China, characterized by reddish-brown discoloration, premature ripening, softening, and pericarp decay, with a field infection rate of 22%. Concurrently, anthracnose was detected in avocado fruits, presenting as small dark brown spots that developed into irregular rust-colored lesions, followed by dry rot depressions, ultimately leading to soft rot, peeling, or hardened dry rot, with a field infection rate of 15%. Infected fruit samples were collected, and fungal strains were isolated, purified, and inoculated via spore suspension, followed by re-isolation. The strains were conclusively identified as Diaporthe phaseolorum (SWFU20, SWFU21) and Colletotrichum fructicola (SWFU12, SWFU13) through an integrated approach combining DNA extraction, polymerase chain reaction (PCR), sequencing, phylogenetic reconstruction, and morphological characterization. This is the first report of D. phaseolorum causing fruit rot and C. fructicola causing anthracnose on avocado in China. In future research, we will test methods for the control of D. phaseolorum and C. fructicola. The identification of these pathogens provides a foundation for future disease management research, supporting the sustainable development of the avocado industry. Full article

In this study the objective was to morphologically characterize fruits, seeds and seedlings of Cereus fernambucensis Lem., as well as evaluate the seed germination and phenological dynamics of these columnar cacti, native to Brazil, which occur in restinga ecosystems. Biometric and morphological determinations were performed using 100 fruits, describing seed morphology in external and internal aspects and considering five stages of development for the characterization of seedlings. In the study of seed germination, two light conditions (12 h photoperiod and complete darkness) were tested under 25 °C, in a completely randomized design with four replicates of 50 seeds each. In the estimation of reproductive phenology, information was collected from herbarium specimens on the SpeciesLink online platform, and the exsiccatae were analyzed for the notes on their labels to evaluate reproductive aspects. Fruits showed an average mass of 21.11 g, length of 44.76 mm, diameter of 28.77 mm and about 336 seeds per fruit. Seeds behave as positive photoblastic, with a high percentage of germination under controlled conditions (94%). Germination is epigeal and phanerocotylar, with slow growth and, at 30 days after sowing, the seedling measures approximately 2 cm, which makes it possible to visualize the appearance of the epicotyl and the first spines. The species blooms and bears fruit throughout the year, with peaks of flowering and fruiting in January and March, respectively. The various characteristics make C. fernambucensis a key species for maintaining the biodiversity of restingas. Full article

As a unique woody oil crop in China, Camellia oleifera Abel. germplasm resources show significant genetic diversity in Ya’an City. This study measured 60 phenotypic traits (32 quantitative, 28 qualitative) of 302 accessions to analyze phenotypic variation, establish a classification system, and screen high-yield, high-oil germplasms. The phenotypic diversity index for fruit (H’ = 1.36–1.44) was significantly higher than for leaf (H’ = 1.31) and flower (H’ < 1), indicating genetic diversity concentrated in reproductive traits, suggesting potential genetic variability in these traits. Fruit quantitative traits (e.g., single fruit weight CV = 35.37%, fresh seed weight CV = 38.93%) showed high genetic dispersion. Principal component analysis confirmed the fruit factor and economic factor as main phenotypic differentiation drivers. Quantitative traits were classified morphologically, and correlation analysis integrated them into 13 key indicators classified using LSD and range methods. Finally, TOPSIS evaluation selected 10 excellent germplasms like TQ122 and TQ49, with fruit weight, fresh seed yield, and kernel oil content significantly exceeding the population average. This study provides data for C. oleifera DUS test guidelines and proposes a multi-trait breeding strategy, supporting high-yield variety selection and germplasm resource protection. Full article
