Fruit flies that belong to the genus Zeugodacus (Diptera: Tephritidae) pose significant threats as invasive pests of agricultural crops in Asia and sub-Saharan Africa. The intensification of transboundary trade in fresh horticultural produce has increased the risk of introducing invasive species such as fruit flies, more so through the inadvertent transport of their immature developmental stages. Such immature stages of fruit flies belonging to the Tephritidae family are frequently intercepted at the international borders worldwide and are unable to be identified to the species level using morphological characteristics. Molecular identification using mitochondrial Cytochrome Oxidase I (COI) gene has proven to be quite useful, as they are not constrained by developmental stages, sex, or colour morphs of the pest species in question. Also, real-time PCR-based species-specific assays offer quicker turnaround time since they do not require any post-PCR procedures. This study evaluated the utility of a real-time PCR assay based on the COI gene region to identify Zeugodacus cucurbitae from other Tephritid species. The developed real-time PCR assay provides a swift and precise way of discriminating between these highly invasive pest species during an interception event for rapid decision making. High specificity, having no cross-reactions with closely related Tephritids, and sensitivity of the developed assay will be extremely useful in discriminating Z. cucurbitae from other closely related fruit fly species. Z. cucurbitae-specific real-time PCR developed in this study is appropriate for organizations that carry out routine diagnostics to facilitate fresh produce imports and exports. Our assay is fully optimized for rapid deployment at international borders, offering reliable detection of the target species regardless of developmental stage, sex, or geographic origins. Physicochemical and Microbiological Properties of Hazelnuts from Three Varieties Cultivated in Portugal

Hazelnut is an important crop worldwide, and the characteristics of the fruits are quite variable according to a number of factors, including variety and cultivation conditions, which in turn can vary according to harvest year. This study aimed to investigate some physical and chemical characteristics of three hazelnut varieties grown in Portugal (Grada de Viseu, Tonda di Giffoni and Butler) along two different harvesting years (2021 and 2022). Also, the microbial quality was investigated for its relevance to the conservation of the fruits. The physical properties evaluated were biometric characteristics and colour, the chemical components analysed were moisture, lipids, protein, ash and fibre, and the microbial properties investigated were the microorganisms, moulds and yeasts. The results showed that, generically, statistically significant differences were found between the three varieties under study on several properties investigated. The kernel was confirmed as the lighter part of all hazelnuts, and when comparing between varieties, Tonda di Giffoni presented the lighter fruits in both harvesting years. With respect to weight, the Tonda di Giffoni variety was the lightest in both harvest years. Moisture content was observed to be higher than the recommended limits for two of the samples (Grada de Viseu in 2021: 6.01 ± 0.26 g/100 g and Butler in 2022: 6.02 ± 0.37 g/100 g), although the difference was marginal given that the recommended limit is 6%. Not surprisingly, lipids were the major chemical component, ranging from 66.46 ± 1.67 to 70.14 ± 1.75 g/100 g in 2021 and from 64.38 ± 1.67 to 77.77 g/100 g in 2022. It was further observed that the three varieties presented a satisfactory microbiological quality. Finally, applying factor analysis with principal components and Varimax rotation, a solution that explains 92.8% of the variance was obtained. This study provided information that is relevant for the characterisation and evaluation of variability according to the year of hazelnuts of three varieties cultivated in Portugal. Full article

Kiwifruit (Actinidia spp.) is a globally important economic fruit with high nutritional value. Fruit peelability, defined as the mechanical ease of separating the peel from the fruit flesh, is a critical quality trait influencing consumer experience and market competitiveness and has emerged as a critical breeding target in fruit crop improvement programs. The present review systematically synthesized existing studies on kiwifruit peelability, and focused on its evolutionary trajectory, genotypic divergence, quantitative evaluation, possible underlying mechanisms, and artificial manipulation strategies. Kiwifruit peelability research has advanced from early exploratory studies in New Zealand (2010s) to systematic investigations in China (2020s), with milestones including the development of evaluation metrics and the identification of genetic resources. Genotypic variation exists among kiwifruit genera. Several Actinidia eriantha accessions and the novel Actinidia longicarpa cultivar ‘Guifei’ exhibit superior peelability, whereas most commercial Actinidia chinensis and Actinidia deliciosa cultivars exhibit poor peelability. Quantitative evaluation highlights the need for standardized metrics, with “skin-flesh adhesion force” and “peel toughness” proposed as robust, instrument-quantifiable indicators to minimize operational variability. Mechanistically, peelability is speculated to be governed by cell wall polysaccharide metabolism and phytohormone signaling networks. Pectin degradation and differential distribution during fruit development form critical “peeling zones”, whereas ethylene, abscisic acid, and indoleacetic acid may regulate cell wall remodeling and softening, collectively influencing skin-flesh adhesion. Owing to the scarcity of easy-to-peel kiwifruit cultivars, artificial manipulation methods, including manual peeling benchmarking, lye treatment, and thermal peeling, can be employed to further optimize kiwifruit peelability. Currently, shortcomings include incomplete genotype-phenotype characterization, limited availability of easy-peeling germplasms, and a fragmented understanding of the underlying mechanisms. Future research should focus on methodological innovation, germplasm development, and the elucidation of relevant mechanisms. Full article

In an increasingly challenging agricultural environment, the identification of novel tools for protecting crops from stress agents while securing marketable production is a key objective. Here we investigated the effects of three previously characterized Prosystemin-derived functional peptide fragments as protective agents against salt stress and as biostimulants modulating tomato yield and quality traits. The treatments of tomato plants with femtomolar amounts of the peptides alleviated salt stress symptoms, likely due to an increase in root biomass up to 18% and the upregulation of key antioxidant genes such as APX2 and HSP90. In addition, the peptides exhibited biostimulant activity, significantly improving root area (up to 10%) and shoot growth (up to 9%). We validated such activities through two-year field trials carried out on industrial tomato crops. Peptide treatments confirmed their biostimulant effects, leading to a nearly 50% increase in marketable production compared to a commonly used commercial product and consistently enhancing fruit °Brix values. Produce supply chains play a critical role in ensuring fruits and vegetables reach consumers efficiently, affordably, and at optimal freshness. In recent decades, hub-and-spoke network models have emerged as valuable tools for optimizing sustainable cold food supply chains. Traditional optimization efforts typically focus on removing inefficiencies, minimizing lead times, refining inventory management, strengthening supplier relationships, and leveraging technological advancements for better visibility and control. However, the majority of models rely on deterministic approaches that overlook the inherent uncertainties of crop yields, which are further intensified by climate variability. Rising atmospheric CO2 concentrations, along with shifting temperature patterns and extreme weather events, have a substantial effect on crop productivity and availability. Such uncertainties can prompt distributors to seek alternative sources, increasing costs due to supply chain reconfiguration. This research introduces a stochastic hub-and-spoke network optimization model specifically designed to minimize transportation expenses by determining optimal distribution routes that explicitly account for climate variability effects on crop yields. A use case involving a cold food supply chain (CFSC) was carried out using several weather scenarios based on climate models and real soil data for California. Strawberries were selected as a representative crop, given California’s leading role in strawberry production. Simulation results show that scenarios characterized by increased rainfall during growing seasons result in increased yields, allowing distributors to reduce transportation costs by sourcing from nearby farms. Conversely, scenarios with reduced rainfall and lower yields require sourcing from more distant locations, thereby increasing transportation costs. Nonetheless, supply chain configurations may vary depending on the choice of climate models or weather prediction sources, highlighting the importance of regularly updating scenario inputs to ensure robust planning. This tool aids decision-making by planning climate-resilient supply chains, enhancing preparedness and responsiveness to future climate-related disruptions. Full article

Enhancing the yield and qualitative traits of horticultural crops without further hampering the environment constitutes an urgent challenge that could be addressed by implementing innovative agronomic tools, such as plant biostimulants. This study investigated the effects of three commercial biostimulants—BIO1 (fulvic/humic acids), BIO2 (leonardite-humic acids), and BIO3 (plant-based extracts)—on leaf ecophysiology, yield, and fruit quality in two raspberry cultivars, ‘Autumn Bliss’ (AB) and ‘Zeva’ (Z), grown in an open-field context, to assess their effectiveness in raspberry cultivation. Experimental activities involved two Research Years (RYs), namely, year 2023 (RY 1) and 2024 (RY 2). Leaf parameters such as chlorophyll, flavonols, anthocyanins, and the Nitrogen Balance Index (NBI) were predominantly influenced by the interaction between Treatment, Year and Cultivar factors, indicating context-dependent responses rather than direct biostimulant effects. BIO2 showed a tendency to increase yield (g plant−1) and berry number plant−1, particularly in RY 2 (417.50 g plant−1, +33.93% vs. control). Fruit quality responses were cultivar and time-specific: BIO3 improved soluble solid content in AB (12.8 °Brix, RY 2, Intermediate Harvest) and Z (11.43 °Brix, +13.91% vs. BIO2). BIO2 reduced titratable acidity in AB (3.12 g L−1) and increased pH in Z (3.02, RY 2) but also decreased °Brix in Z. These findings highlight the potential of biostimulants to modulate raspberry physiology and productivity but underscore the critical role of cultivar, environmental conditions, and specific biostimulant composition in determining the outcomes, which were found to critically depend on tailored application strategies. The extensive use of conventional pesticides has been a fundamental strategy in modern agriculture for controlling pests and increasing crop productivity; however, their improper application poses significant risks to human health and environmental sustainability. This review compiles scientific evidence linking pesticide exposure to oxidative stress and genotoxic damage, particularly affecting rural populations and commonly consumed foods, even at levels exceeding the maximum permissible limits in fruits, vegetables, and animal products. Additionally, excessive pesticide use has been shown to alter soil microbiota, negatively compromising long-term agricultural fertility. In response to these challenges, recent advances in nanotechnology offer promising alternatives. This review highlights the development of nanopesticides designed for controlled release, improved stability, and targeted delivery of active ingredients, thereby reducing environmental contamination and increasing efficacy. Moreover, emerging nanobiosensor technologies, such as e-nose and e-tongue systems, have shown potential for real-time monitoring of pesticide residues and soil health. Although pesticides are still necessary, it is crucial to implement stricter laws and promote sustainable solutions that ensure safe and responsible agricultural practices. The need for evidence-based public policy is emphasized to regulate pesticide use and protect both human health and agricultural resources. Full article

Postharvest losses amongst small-scale farmers in developing countries are high due to inadequate resources and infrastructure. Among the various affected crops, tomatoes are particularly vulnerable; however, studies on postharvest losses of most fruits and vegetables are limited. Therefore, this study aimed to assess postharvest tomato losses under different production systems within the small-scale supply chain using the indirect assessment (questionnaires and interviews) and direct quantification of losses. Farmers reported tomato losses due to insects (82.35%), cracks, bruises, and deformities (70.58%), and diseases (64.71%). Chemical sprays were the main form of pest and disease control reported by all farmers. The direct quantification sampling data revealed that 73.07% of the tomatoes were substandard at the farm level, with 47.92% and 25.15% categorized as medium-quality and poor-quality, respectively. The primary contributors to the losses were decay (39.92%), mechanical damage (31.32%), and blotchiness (27.99%). Postharvest losses were significantly higher under open-field production systems compared to closed tunnels. The fungi associated with decay were mainly Geotrichum, Fusarium spp., and Alternaria spp. These findings demonstrate the main drivers behind postharvest losses, which in turn highlight the critical need for intervention through training and support, including the use of postharvest loss reduction technologies to enhance food security. Indoor horticulture requires a substantial quantity of electricity to meet crops extended photoperiodic requirements for optimal photosynthetic rate. Simultaneously, global electricity costs have grown dramatically in recent years, endangering the sustainability and profitability of indoor vertical farms and/or modern greenhouses that use artificial lighting systems to accelerate crop development and growth. This study investigates the growth rate and physiological development of cherry tomato plants cultivated in a pilot indoor vertical farm at the Agricultural University of Athens’ Laboratory of Farm Structures (AUA) under continuous and disruptive lighting. The leaf physiological traits from multiple photoperiodic stress treatments were analyzed and utilized to estimate the plant’s tolerance rate under varied illumination conditions. Four different photoperiodic treatments were examined and compared, firstly plants grew under 14 h of continuous light (C-14L10D/control), secondly plants grew under a normalized photoperiod of 14 h with intermittent light intervals of 10 min of light followed by 50 min of dark (NI-14L10D/stress), the third treatment where plants grew under 14 h of a load-shifted energy demand response intermittent lighting schedule (LSI-14L10D/stress) and finally plants grew under 13 h photoperiod following of a load-shifted energy demand response intermittent lighting schedule (LSI-13L11D/stress). Plants were subjected also under two different light spectra for all the treatments, specifically WHITE and Blue/Red/Far-red light composition. The aim was to develop flexible, energy-efficient lighting protocols that maintain crop productivity while reducing electricity consumption in indoor settings. Results indicated that short periods of disruptive light did not negatively impact physiological responses, and plants exhibited tolerance to abiotic stress induced by intermittent lighting. Post-harvest data indicated that intermittent lighting regimes maintained or enhanced growth compared to continuous lighting, with spectral composition further influencing productivity. Plants under LSI-14L10D and B/R/FR spectra produced up to 93 g fresh fruit per plant and 30.4 g dry mass, while consuming up to 16 kWh less energy than continuous lighting—highlighting the potential of flexible lighting strategies for improved energy-use efficiency. Drosophila suzukii is an invasive pest of many fruit crops worldwide. Employing the Sterile Insect Technique (SIT) could mitigate D. suzukii population growth and crop damage. This study evaluated the efficacy of SIT on commercial fruit, by (1) validating the quality of irradiated sterile males (male mating competitiveness, courtship, and flight performance) in the laboratory, and (2) assessing population suppression and fruit damage reduction in commercial raspberry fields. Treatment with SIT was compared to the grower’s standard chemical insecticide program throughout the season. The principal metrics of efficacy were trap counts of wild adult female D. suzukii in crops and larvae per fruit during harvesting. These metrics together with monitoring of border areas allowed targeting of high-pressure areas with higher releases of sterile males, to maximise efficacy for a given release number. The sterile male D. suzukii were as competitive as their fertile non-irradiated counterparts in laboratory mating competitiveness and flight performance studies while fertility egg-to-pupae recovery was reduced by 99%. In commercial raspberry crops, season-long releases of sterile males significantly suppressed the wild D. suzukii population, compared to the grower standard control strategy; with up to 89% reduction in wild female D. suzukii and 80% decrease in numbers of larvae per harvested fruit. Additionally, relative fruit waste (i.e., percentage of harvested fruits rejected for sale) at harvest was reduced for early, mid and late harvest crops, by up to 58% compared to the grower standard control. SIT has the potential to provide an effective and sustainable strategy for managing D. suzukii in raspberries, increasing marketable yield by reducing adult populations, fruit damage and waste fruit. SIT could therefore serve as a valuable tool for integrated pest management practices in berry production systems. Full article

Currently, most crop yield estimation studies based on the YOLO model are only conducted during a single stage of maturity. Combining multi-growth period data for crop analysis is of great significance for crop growth detection and early yield estimation. In this study, a new network model, YOLOv8-RL, was proposed using citrus multigrowth period characteristics as a data source. A citrus yield estimation model was constructed and validated by combining network identification counts with manual field counts. Compared with YOLOv8, the number of parameters of the improved network is reduced by 50.7%, the number of floating-point operations is decreased by 49.4%, and the size of the model is only 3.2 MB. In the test set, the average recognition rate of citrus flowers, green fruits, and orange fruits was 95.6%, the mAP@.5 was 94.6%, the FPS value was 123.1, and the inference time was only 2.3 milliseconds. This provides a reference for the design of lightweight networks and offers the possibility of deployment on embedded devices with limited computational resources. The two estimation models constructed on the basis of the new network had coefficients of determination R2 values of 0.91992 and 0.95639, respectively, with a prediction error rate of 6.96% for citrus green fruits and an average error rate of 3.71% for orange fruits. Compared with network counting, the yield estimation model had a low error rate and high accuracy, which provided a theoretical basis and technical support for the early prediction of fruit yield in complex environments. Genetic transformation is an essential tool for investigating gene function and editing genomes. Kiwifruit, recognized as a significant global fresh fruit crop, holds considerable economic and nutritional importance. However, current genetic transformation techniques for kiwifruit are impeded by low efficiency, lengthy culture durations (a minimum of six months), and substantial labor requirements. In this research, we established an efficient system for shoot regeneration and the stable genetic transformation of the ‘Hayward’ cultivar, utilizing leaf explants in conjunction with two strains of Agrobacterium that harbor the expression vector pBI121-35S::GFP, which contains the green fluorescent protein (GFP) gene as a visible marker within the T-DNA region. Our results show that 93.3% of leaf explants responded positively to the regeneration medium, producing multiple independent adventitious shoots around the explants within a six-week period. Furthermore, over 71% of kanamycin-resistant plantlets exhibited robust GFP expression, and the entire transformation process was completed within four months of culture. Southern blot analysis confirmed the stable integration of GFP into the genome, while RT-PCR and fluorescence microscopy validated the sustained expression of GFP in mature plants. This efficient protocol for regeneration and transformation provides a solid foundation for micropropagation and the enhancement of desirable traits in kiwifruit through overexpression and gene silencing techniques. Anchote (Coccinia abyssinica) is a neglected high-potential food and nutrition security tuber crop in Ethiopia. Phenotyping core germplasm collections using agro-morphological and physiological markers is essential for effective crop improvement and utilization. A total of 282 anchote germplasms were profiled using six qualitative and twenty-six quantitative agro-morphological and physiological traits. Augmented Block Design was used for the experiment at the Debre Zeit Agricultural Research Center. The chi-square test and Shannon diversity index indicated the presence of substantial phenotypic variation and diversity among the accessions based on the predominant qualitative traits studied. The quantitative agro-morphological and physiological traits showed wider variability and ranges for the accessions. The broad-sense heritability and genetic advance as a percentage of the mean were notably high for quantitative traits such as root yield, vine length, and leaf area index. A significantly positive correlation was observed among agronomically important traits such as root yield and root diameter as well as root yield and leaf area. The principal component analysis for qualitative and quantitative traits found that ten components explained 72.2% of the variation for qualitative traits, whereas nine components accounted for 69.96% of the variation in quantitative traits. The primary contributors to the variations are traits such as root (shape, flesh color, and yield), leaf (color, length, diameter, area) and fruit (length, diameter, and weight). Further, the accessions were grouped into two and three clusters based on qualitative and quantitative traits, respectively, indicating that quantitative characters better differentiated among the accessions. Similarly, the tanglegram showed little similarity between the qualitative and quantitative agro-morphological and physiological traits in clustering the accessions. These findings indicate the presence of sizable trait variation among the accessions that can be exploited as a selection marker to design and facilitate conservation and breeding strategies of anchote. Tomato crops are treated with high concentrations of synthetic fertilizers and insecticides to increase yields, but the careless use of these chemicals harms the environment and human health and affects plant pathogen resistance. The effect of foliar spray of three concentrations of chitosan (500, 1000, and 2000 mg L−1) on plant growth, yield, fruit quality, and physiological performance in two tomato varieties (Floradade and Candela F1) was studied. Physiological traits such as photosynthesis, chlorophyll content, and leaf area index of the plants were positively affected by chitosan, an effective compound that biostimulates growth, with increases in biomass of organs with respect to the control treatment. Chitosan also improved tomato quality, such as increases in polyphenols, antioxidant capacity, flavonoids, carotenoids, vitamin C, and total soluble solids in both tomato varieties. Finally, yield increased by 76.4% and 65.4% in Floradade and Candela F1, respectively. The responses of tomato plants to chitosan application were different depending on the variety evaluated, indicating a differential response to the biostimulant. The use of chitosan in agriculture is a tool that has no negative effects on plants and the environment and can increase the productive capacity of tomato plants. Full article

Effects of Foliar and Root Application of Different Amino Acids on Mini-Watermelon

Biostimulants, particularly single amino acids, can increase plant growth and crop quality, gaining significant attention. This study investigates the effects of 10 amino acids via root/foliar application on the growth, quality, taste, and volatile flavor of mini-watermelons and compares the differences between the application methods. Here, we employed electronic noses, electronic tongues, and gas chromatography–ion mobility spectrometry to investigate these effects. Root application excels in fruit growth and pectin accumulation, while foliar application boosts soluble protein and specific nutrients. Specifically, root application (except for Val) significantly increases fruit weight, with Gly being most effective for longitudinal diameter, while most amino acids (except Val/Lys) promote transverse diameter. Pectin content shows bidirectional regulation: root application of Glu/Gly/Lys/Pro/Trp/Val enhances pectin, whereas foliar application inhibits it. For taste indices, most treatments improve soluble solids (except Glu root/Arg-Leu foliar), and Ala/Asp/Glu/Gly reduce titratable acids, optimizing the sugar–acid ratio. Foliar application is more efficient for soluble protein accumulation (Ala/Glu/Gly/Pro/Leu). For nutritional quality, except for Lys, all treatments increase vitamin C and widely promote total phenolics and lycopene, with only minor exceptions, and only Arg foliar application enhances ORAC. Additionally, the results revealed that root-applied lysine and valine greatly raised the levels of hexanal and 2-nonenal, whereas foliar-applied valine significantly increased n-nonanal and (Z)-6-nonenal. Overall, we found that amino acids can considerably improve mini-watermelon production, quality, taste, and antioxidant capacity, providing theoretical and practical references for their widespread use in agriculture.
