Chain topology offers a chemistry-preserving route to tune block copolymer (BCP) self-assembly by modifying intrachain correlations and relaxation pathways without changing monomer interactions. Here, we perform a unit-matched comparison of four lamella-forming AB architectures reconstructed from an identical constitutive diblock unit (N0): a linear diblock (DB), a linear multiblock (MB), a comb-like architecture (CB), and a star-like architecture (SB). Using dynamical density functional theory (DDFT), we quantify topology-dependent bulk ordering thresholds and show that architectural reconfiguration systematically stabilizes the ordered phase, reducing the order–disorder transition relative to DB (MB/CB/SB 0.793/0.762/0.752 of the diblock value), in semi-quantitative agreement with random phase approximation (RPA) spinodal trends. We also compare topology-dependent directed self-assembly in a common trench geometry under matched reduced quench depth Δ(χN0)=χN0(χN0)ODT, thereby isolating kinetic differences at comparable thermodynamic distance from bulk ordering. A Fourier-based alignment order parameter α(t) reveals sigmoidal alignment kinetics over decades in time and is well captured by a logistic form in lnt, enabling compact descriptors (t50, t90, and a steepness parameter k) that separate alignment onset from late-stage defect annihilation, while selective sidewalls robustly template sidewall-parallel lamellae across all topologies, the late-stage kinetics remain strongly connectivity dependent and can exhibit long-tailed completion associated with slow late-stage defect annihilation. These results demonstrate a dual role of topology in DSA: lowering the segregation strength required for bulk ordering while reshaping defect-mediated alignment pathways under confinement. Amphiphilic copolymers based on polystyrene and polyglycidol combine the chemical inertness of polystyrene with the biocompatibility of polyglycidol, making them attractive materials for polymeric micelles. While comb-like architectures have been explored to control micellization behavior and biological response, a direct comparison between comb-like and coil-comb topologies in polystyrene–polyglycidol copolymers at identical polyglycidol content remains insufficiently investigated. In this work, amphiphilic comb-like and coil-comb polystyrene–polyglycidol copolymers were synthesized via copper-catalyzed azide–alkyne click chemistry by grafting a monoalkyne-terminated polyglycidol precursor onto azide-functionalized random and block styrene copolymers. The copolymers were characterized by size exclusion chromatography and nuclear magnetic resonance. Polymeric micelles were prepared by nanoprecipitation, and their self-assembly in aqueous solution was investigated by critical micelle concentration determination, dynamic and electrophoretic light scattering, and atomic force microscopy. Both copolymers formed stable aqueous dispersions and exhibited comparable critical micelle concentrations. At identical polyglycidol content, the random copolymer formed a uniform, monomodal micellar population, whereas the block-based coil-comb architecture led to bimodal size distributions, indicating the coexistence of two distinct micellar populations. The investigated systems showed low cytotoxicity and did not induce significant oxidative stress within the studied concentration range. On isolated rat brain sub-cellular fractions (synaptosomes, mitochondria and microsomes), administered alone, the comb-like and coil-comb polystyrene-polyglycidol copolymers did not reveal statistically significant neurotoxic effects. The results demonstrate that macromolecular architecture plays a key role in governing micellar organization and in vitro biological response in polystyrene–polyglycidol copolymers, highlighting their potential as architecture-controlled polymer-based nanocarriers. Combining Brownian dynamics simulations and self-consistent field theory, we demonstrate that stable assembled structures, such as vesicles, toroidal micelles, bowl-like micelles, sheet-like micelles, non-spherical vesicles, and cylindrical micelles, are dependent on the molecular parameters of amphiphilic comb-like copolymers. Importantly, we find that vesicle formation involves two intermediate states, sheet-like and bowl-like micelles, and the difference in their free energies is minimal, which illustrates the coexisting phase between them. Moreover, the assembled vesicles can be modulated in the membrane thickness with overall size, unchanged only by adjusting the backbone length. We also demonstrate the coexistence of toroidal and cylindrical micelles because neither structure has a significant advantage over the other in free energy. Our work points out how to obtain different morphologies by adjusting the molecular parameters of amphiphilic comb-like copolymers, instilling confidence in their potential for stable drug encapsulation and enhanced targeted drug delivery. Amphiphilic block copolymers with complex topologies (e.g., star and brush topologies) have attracted significant attention in drug delivery owing to their superior performance over linear micelles. However, their precise synthesis and structure–property relationships require further investigation. In this study, hydroxylated polybutadiene with adjustable topology and hydroxyl group density was employed as a macroinitiator to synthesize well-defined amphiphilic poly (ethylene oxide)-b-poly(ε-caprolactone-ran-δ-valerolactone) (PEO-b-P(CL-ran-VL)) copolymers via ring-opening polymerization (ROP). A series of linear, star, linear–comb, and star–comb copolymers were prepared as curcumin-loaded micellar carriers for the study. The self-assembly behavior, drug encapsulation efficiency, and in vitro release profiles of these copolymers in aqueous environments were systematically investigated. The results demonstrated that increasing the branch length of star–comb copolymers effectively reduced micelle size from 143 to 96 nm and enhanced drug encapsulation efficiency from 27.3% to 39.8%. Notably, the star–comb architecture exhibited 1.2-fold higher curcumin encapsulation efficiency than the linear counterparts. Furthermore, the optimized star–comb nanoparticles displayed sustained release kinetics (73.38% release over 15 days), outperforming conventional linear micelles. This study establishes a quantitative structure–property relationship between copolymer topology and drug delivery performance, providing a molecular design platform for programmable nanocarriers tailored to diverse therapeutic requirements of various diseases. Brush-like graft copolymers (A-g-B), in which linear A-blocks are randomly grafted onto the backbone of a brush-like B-block, exhibit intense strain-stiffening and high mechanical strength on par with load-bearing biological tissues such as skin and blood vessels. To elucidate molecular mechanisms underlying this tissue-mimetic behavior, in situ synchrotron X-ray scattering was measured during uniaxial stretching of bottlebrush- and comb-like graft copolymers with varying densities of poly(dimethyl siloxane) and poly(isobutylene) side chains. In an undeformed state, these copolymers revealed a single interference peak corresponding to the average spacing between the domains of linear A-blocks arranged in a disordered, liquid-like configuration. Under uniaxial stretching, the emergence of a distinct four-spot pattern in the small-angle region indicated the development of long-range order within the material. According to the affine deformation of a cubic lattice, the four-spot pattern’s interference maxima correspond to 110 reflections upon stretching along the [111] axis of the body-centered unit cell. The experimental findings were corroborated by computer simulations of dissipative particle dynamics that confirmed the formation of a bcc domain structure. Full article

In this work, comb homopolymers as well as comb-type copolymers of thermo-responsive oligo(ethylene glycol methyl ether methacrylate)s, OEGMAs, with various chain lengths (DEGMA, PEGMA500, and PEGMA950 containing 2, 9, or 19 repeating ethylene glycol units, respectively) were synthesized through free radical (co)polymerization. For the copolymers, either the functional hydrophobic glycidyl methacrylate (GMA) or the inert hydrophilic N,N-dimethylacrylamide (DMAM) were selected as comonomers. The self-assembly and thermo-responsive behavior of the products was investigated through Nile Red fluorescence probing, turbidimetry, and dynamic light scattering (DLS). Interestingly, it was found that all OEGMA-based homopolymers exhibit a tendency to self-organize in aqueous media, in addition to thermo-responsiveness. The critical aggregation concentration (CAC) increases with the number of repeating ethylene oxide units in the OEGMA macromonomers (CAC was found to be 0.003, 0.01, and 0.03% w/v for the homopolymers PDEGMA, PPEGMA500, and PPEGMA950, respectively). Moreover, the CAC of the copolymers in aqueous media is highly affected by the incorporation of hydrophobic GMA or hydrophilic DMAM units, leading to lower or higher values, respectively. Thus, the CAC decreases down to 0.003% w/v for the GMA-richest copolymer of PEGMA950, whereas CAC increases up to 0.01% w/v for the DMAM-richest copolymer of DEGMA. Turbidimetry and DLS studies proved that the thermo-sensitivity of the polymers is governed by several parameters such as the number of repeating ethylene glycol groups in the side chains of the OEGMAs, the molar percentage of the hydrophobic or hydrophilic comonomers, along with the addition of salts in the aqueous polymer solutions. Thus, the cloud point of the homopolymer PDEGMA was found at 23 °C and it increases to 33.5 °C for the DMAM-richest copolymer of DEGMA. Lastly, the formation of a hydrogel upon heating aqueous mixtures of the GMA-comprising copolymers with silica nanoparticles overnight is strong evidence of the functional character of these polymers. A detailed understanding of the interactions between wax and asphaltenes with other components of crude oils and the effect of treatments with paraffin inhibitors (PIs) and asphaltene dispersants (ADs), with a focus on identifying specific structure-activity relationships, is necessary to develop effective flow assurance strategies. The morphological and rheological consequences of treating wax and asphaltenes in oils of differing composition with a series of ADs having structural features in common with an alpha olefin-maleic anhydride (AO-MA) comb-like copolymer PI were assessed alone and in combination with said PI. Of the four ADs studied, two were identified as being effective dispersants of asphaltenes in heptane-induced instability tests and in a West Texas (WT) crude. The degree to which a low concentration of asphaltenes stabilizes wax in the absence of treatment additives is lessened in oils having greater aromatic fractions. This is because these stabilizing interactions are replaced by more energetically favorable aromatic–asphaltene interactions, increasing oil viscosity. Treatment with AD alone also reduces the extent of wax–asphaltene interactions, increasing oil viscosity. In concert with the PI, treatment with the AD having greater structural similarity with the PI appears to improve wax solubility in both the presence and absence of asphaltenes. However, the viscosity of the treated oils is greater than that of the oil treated with PI alone, while treatment with AD having lesser structural similarity with the PI does not adversely affect oil viscosity. These data suggest that rather than treating both wax and asphaltenes, AD may poison the function of the PI. These data illuminate the pitfalls of designing flow assurance additives to interact with both wax and asphaltenes and developing treatment plans. Full article

Polymers with crystallizable side chains have numerous applications, and their properties depend on their crystal morphologies and phase separation. Structural analysis on a wide spatial scale plays an important role in controlling the thermal properties and higher-order structures of these polymers. In this study, we elucidated the melting and crystallization processes of copolymers with varying crystallizable side-chain fractions over a wide spatial range. Differential scanning calorimetry revealed that the enthalpies of melting and crystallization increased linearly with increasing crystallizable side-chain fraction. The results of wide-angle X-ray scattering indicated that the crystal lattice was hexagonal. Conversely, spherulite-like higher-order architectures with linear structures and radial spreading were observed in the highly crystallizable components, but no micrometer-scale structures were observed in the less crystallizable components. In situ small-angle X-ray scattering was used to elucidate the phase separation and mixing processes. Lamellar crystallites were observed at crystallizable side-chain fractions of >55 wt.%, whereas small crystallites were observed at fractions of <45 wt.%. At temperatures above the order-disorder transition temperature, density fluctuations caused by correlation holes were observed. These properties have a strong effect on the crystallizable side-chain fraction. Full article

Adenosine is a vital biological small molecule that regulates various physiological processes in the human body. A high expression of adenosine in cells can facilitate tumor growth. Therefore, detecting adenosine is crucial for early disease diagnosis. In this paper, we designed a fluorescent biosensor for the sensitive detection of adenosine based on the cationic comb-type copolymer PLL-g-Dex for assisted rapid hybridization of nucleic acids at room temperature. In this strategy, adenosine preferentially binds to the aptamer immobilized on the surface of magnetic nanobeads, releasing free aDNA in solution as the primer strand, which rapidly forms DNA nanowires with auxiliary probes of bDNA with the assistance of PLL-g-Dex. SYBR Green I is embedded in DNA duplexes to generate strong fluorescence. The experimental results showed that PLL-g-Dex promotes DNA hybridization reactions at room temperature to form ultra-long DNA nanowires, thus achieving signal amplification and shortening the detection time. In addition, magnetic nanobeads can reduce the background signal during the reaction. Compared with several previous studies on the fluorescence detection of adenosine, this strategy has a lower detection limit of 2.32 nM. Furthermore, this novel system exhibited a good detection performance even under complex environments, such as serum, providing some reference for the quantitative detection of adenosine in early disease diagnosis. A series of amphiphilic arborescent copolymers of generations G1 and G2 with an arborescent poly(γ-benzyl L-glutamate) (PBG) core and poly(ethylene oxide) (PEO) chain segments in the shell, PBG-g-PEO, were synthesized and evaluated as drug delivery nanocarriers. The PBG building blocks were generated by ring-opening polymerization of γ-benzyl L-glutamic acid N-carboxyanhydride (Glu-NCA) initiated with n-hexylamine. Partial or full deprotection of the benzyl ester groups followed by coupling with PBG chains yielded a comb-branched (arborescent polymer generation zero or G0) PBG structure. Additional cycles of deprotection and grafting provided G1 and G2 arborescent polypeptides. Side chains of poly(ethylene oxide) were then randomly grafted onto the arborescent PBG substrates to produce amphiphilic arborescent copolymers. Control over the branching density of G0PBG was investigated by varying the length and the deprotection level of the linear PBG substrates used in their synthesis. Three G0PBG cores with different branching densities, varying from a compact and dense to a loose and more porous structure, were thus synthesized. These amphiphilic copolymers behaved similar to unimolecular micelles in aqueous solutions, with a unimodal number- and volume-weighted size distributions in dynamic light scattering measurements. It was demonstrated that these biocompatible copolymers can encapsulate hydrophobic drugs such as doxorubicin (DOX) within their hydrophobic core with drug loading efficiencies of 42–65%. Sustained and pH-responsive DOX release was observed from the unimolecular micelles, which suggests that they could be useful as drug nanocarriers for cancer therapy. Full article

Thin-film composite mixed-matrix membranes (TFC-MMMs) have potential applications in practical gas separation processes because of their high permeance (gas flux) and gas selectivity. In this study, we fabricated a high-performance TFC-MMM based on a rubbery comb copolymer, i.e., poly(2-[3-(2H-benzotriazol-2-yl)-4-hydroxyphenyl] ethyl methacrylate)-co-poly(oxyethylene methacrylate) (PBE), and metal–organic framework MOF-808 nanoparticles. The rubbery copolymer penetrates through the pores of MOF-808, thereby tuning the pore size. In addition, the rubbery copolymer forms a defect-free interfacial morphology with polymer-infiltrated MOF-808 nanoparticles. Consequently, TFC-MMMs (thickness = 350 nm) can be successfully prepared even with a high loading of MOF-808. As polymer-infiltrated MOF is incorporated into the polymer matrix, the PBE/MOF-808 membrane exhibits a significantly higher CO2 permeance (1069 GPU) and CO2/N2 selectivity (52.7) than that of the pristine PBE membrane (CO2 permeance = 431 GPU and CO2/N2 selectivity = 36.2). Therefore, the approach considered in this study is suitable for fabricating high-performance thin-film composite membranes via polymer infiltration into MOF pores. Full article

An important problem for the oil industry is the deposition of paraffin on pipelines during the transit of crude oil and restart processes at low temperature. In this regard, the need for suitable methods of wax deposition has attracted substantial attention. Therefore, pour point depressants (PPDs) are considered a critical processing aid to modify the paraffin crystallization and improve the flow of waxy crude oil. The effect of pendants in comb-type copolymers on the ability of crude oil to flow in the cold is examined in the current study. Such PPDs were first created by the free radical polymerization of maleic anhydride with benzyl oleate to create the poly (benzyl oleate-co-maleic anhydride). The resultant copolymer was then aminated with alkyl amine (stearyl amine) (C18H39N) to form pendant alkyl amine chains. The esterified copolymers were structurally characterized by Fourier Transform Infrared, X-ray diffraction spectral analysis, and scanning electron microscopy. Moreover, the potential interactions between PPD and waxes were investigated by using differential scanning calorimetry, X-ray diffraction, and light microscopy. The obtained PPDs, which are effective at a dose of 2000 ppm, were able to reduce the pour point by up to 3 °C. The viscosity and yield stress of the petroleum waxy crude oil were revealed by rheometer. Full article

The amphiphilic block copolymer poly(N-isopropylacrylamide)–Ge(C6F5)2–poly(2,2,3,3-tetrafluoropropyl methacrylate) was prepared by the reaction of chain transfer to bis-(pentafluorophenyl)germane during the polymerization of N-isopropylacrylamide and the subsequent postpolymerization of isolated functional polymers in 2,2,3,3–tetrafluoropropyl methacrylate. The conversion of the block copolymer was 68% and the molecular weight of the sample was 490,000 g/mol. The colloidal chemical properties of Langmuir monolayers and Langmuir-Blodgett films of synthesized block copolymer have been studied. For comparison, a functional polymer, namely, poly-N-isopropylacrylamide with terminal –Ge(C6F5)2H group, was synthesized and studied. The concentrations of spreading solutions were selected and the effect of subphase acidity on the formation of monolayers of macromolecules of the block copolymer was studied. It was found that regardless of the acidity of the subphase, high pressure of fracture of films are characteristic of monolayers of collapse pressures πmax = (48–61) mN/m. The morphology of the Langmuir-Blodgett films of functional polymer exhibit isolated elongated micelles with high densities in the form of “octopus” on the periphery of which there are terminal hydrophobic groups. For the Langmuir-Blodgett film of block copolymer, a comb-like structure is observed with characteristic protrusions. Full article

Densely grafted comb-like macromolecules (bottlebrushes) with alternating solvophobic and solvophilic side chains were studied in a selective solvent and at the liquid interface using mesoscopic computer simulations. The effects of backbone length and copolymer composition were considered. While self-assembly in solution revealed only spherical aggregates for all ar-chitectures studied, adsorption onto the liquid interface in particular cases resulted in morpho-logical changes, with worm-like aggregates or a continuous monolayer observed. In turn, the compression of macromolecules at the interface also leads to morphological transitions, includ-ing the formation of a mesh-like percolated structure. The obtained results may be useful for the preparation of solid nanoparticles of anisotropic shape or nanostructured ultra-thin copolymer films. Full article

We computationally investigate the conformational behavior, “bridging” chain, between different the phase-separated domains vs “looping” chain on the same domain, for two chain architectures of ABA triblock copolymers, one with a linear architecture (L-TBC) and the other with comb architecture (C-TBC) at various segregation regimes using dissipative particle dynamics (DPD) simulations. The power-law relation between the bridge fraction (Φ) and the interaction parameter (χ) for C-TBC is found to be Φχ1.6 in the vicinity of the order-disorder transition (χODT), indicating a drastic conversion from the bridge to the loop conformation. When χ further increases, the bridge-loop conversions slow down to have the power law, Φχ0.18, approaching the theoretical power law Φχ1/9 predicted in the strong segregation limit. The conformational assessment conducted in the present study can provide a strategy of designing optimal material and processing conditions for triblock copolymer either with linear or comb architecture to be used for thermoplastic elastomer or molecular nanocomposites.
