Advanced and Personalized Drug Delivery Systems: From Formulation Science to Clinical Application

Dear Colleagues,
The development of advanced drug delivery systems has emerged as one of the most dynamic and clinically consequential areas in contemporary pharmaceutical sciences. Despite significant progress in pharmacology, a substantial proportion of therapeutic failures are not attributable to insufficient drug potency, but rather to suboptimal delivery: poor bioavailability, inadequate biodistribution, erratic release profiles, and low patient adherence. These persistent challenges define the core problems that innovative delivery technologies are called to solve.
Over the past two decades, research in this field has produced a growing arsenal of nanostructured carriers—including solid lipid nanoparticles, polymeric nanoparticles, liposomes, nanoemulsions, and lipid–polymer hybrids—alongside macroscopic platforms such as orodispersible films, transdermal patches, and hydrogels. Each of these systems addresses specific pharmacokinetic limitations, whether by protecting labile molecules from enzymatic degradation, enabling controlled or stimulus-responsive release, enhancing mucosal permeation, or facilitating non-invasive administration. Yet translating these technologies from the laboratory to clinical practice remains a formidable challenge. Questions of scalability, in vitro–in vivo correlation, and the design of patient-centered formulations—particularly in the context of compounding and personalized medicine—are not yet fully resolved.
This Special Issue aims to serve as a comprehensive platform for sharing the latest scientific advances and clinical insights in the field. This Special Issue will explore innovative carrier technologies, novel excipients and functional polymers, personalized compounding strategies, and the clinical outcomes associated with optimized delivery approaches across therapeutic areas including oncology, dermatology, pain management, neurology, and chronic disease.
We cordially invite researchers, pharmacists, and clinicians to submit original research articles and comprehensive reviews. Topics of interest include, but are not limited to, nanoparticulate drug delivery systems, transdermal and transmucosal delivery platforms, orodispersible and fast-dissolving formulations, sustained- and controlled-release systems, bioavailability enhancement strategies, in vitro–in vivo correlations, and the clinical application of personalized compounding.
