Special Issue Information

Dear Colleagues,

Over the last decade, there has been a significant progress in our understanding of the etiology of multiple sclerosis (MS); that is, on the crucial role played by Epstein–Barr virus (EBV) and its interaction with the host immune system, as well as on the pathophysiology of progression, through the characterization of chronic, compartimentalized inflammation mainly driven by microglia. Despite these advancements, regarding the former process, there is a need to better define the molecular mechanisms through which EBV induces central nervous system autoimmunity in predisposed individuals. Concerning progression, further investigations on persistent microglial activation and its impact on oligodendrocyte differentiation, myelin repair and irreversible neuro-axonal damage could open up major therapeutic avenues. This Special Issue, entitled “Multiple Sclerosis: Molecular Basis and New Therapy”, will focus on the molecular basis of MS etiology, with a specific interest on the role of EBV and gut microbiota, on the determinants of progression and the mechanisms underlying remyelination failure in affected individuals. Particular attention will be given to the description of pathways that could be the substrate of novel therapeutic interventions. Critical reappraisal of approved disease-modyfing drugs, failed trials and perspectives on new molecular targets to act on neurodegeneration and foster myelin repair are also welcome.
