Special Issue Information

Dear Colleagues,

Following the success of the first edition of the Special Issue entitled “The Janus Face of Oxidative Stress in Normal and Pathological Conditions”, which resulted in the publication of 13 high-quality contributions, we are pleased to announce the launch of a second edition devoted to this timely and rapidly evolving field.

Oxidative stress represents one of the most fascinating and complex biological phenomena, characterized by a dual nature that fully reflects its “Janus face”. On the one hand, reactive oxygen and nitrogen species (ROS/RNS) are essential mediators of physiological processes, including intracellular signaling, immune defense, cellular adaptation, metabolism, differentiation and tissue homeostasis. On the other hand, when their production exceeds the capacity of antioxidant and repair systems, or when redox signaling becomes dysregulated, these same molecules may contribute to molecular damage, chronic inflammation, cellular dysfunction and the onset or progression of numerous pathological conditions.

This second edition aims to further explore the intricate balance between the beneficial and detrimental effects of oxidative stress in both normal physiology and disease. Particular attention will be given to the molecular and cellular mechanisms through which redox imbalance influences pathological processes, including cancer, neurodegenerative diseases, cardiovascular disorders, metabolic dysfunctions, immune-endocrine alterations, addiction-related conditions, aging and inflammatory diseases. At the same time, this Special Issue aims to highlight the emerging role of controlled redox modulation as a potential therapeutic strategy, moving beyond the simplistic concept of antioxidant supplementation toward a more precise understanding of context-dependent redox biology.

We welcome original research articles, clinical and preclinical studies and comprehensive reviews addressing the mechanisms, biomarkers and therapeutic implications of oxidative stress. Contributions investigating ROS/RNS signaling, mitochondrial dysfunction, antioxidant responses, redox-sensitive pathways, nutraceutical and pharmacological interventions and novel translational approaches are particularly encouraged.

Through this second edition, we aim to provide an updated and multidisciplinary overview of oxidative stress as both a physiological regulator and a pathological initiator. By gathering contributions from different areas of biomedical research, this Special Issue will potentiate the knowledge of how the same redox events may lead to profoundly different outcomes depending on the biological context, intensity, duration and cellular or systemic environment in which they occur.
