Dear Colleagues,

Oxidative stress is increasingly recognized as a crucial mechanism influencing brain development and the etiology of neurodevelopmental disorders (NDDs). Disparities between reactive oxygen species production and antioxidant defenses can disrupt synaptogenesis, neurogenesis, and myelination during precarious developmental windows, thus contributing to behavioral, cognitive, and motor impairments observed across conditions, including attention‑deficit/hyperactivity disorder, fetal alcohol spectrum disorders, autism spectrum disorder, and intellectual disability. Emerging evidence discloses a role for oxidative stress in maladaptive neuroinflammation, microglial activation, and altered neuronal connectivity, linking molecular redox disturbances to circuit-level dysfunctions. The heterogeneity of NDDs mirrors multifactorial interactions among genetic susceptibilities, prenatal and perinatal conditions, metabolic dysregulation, and environmental exposures that modulate redox homeostasis; this complication emphasizes the necessity for integrative studies that combine neuroimaging, biochemical markers, and functional outcomes. Translational research exploring metabolic modulation, antioxidant strategies, and timing-specific interventions holds promise but requires rigorous validation in well-characterized cohorts.

This Special Issue aims to collect biomarker investigations, mechanistic studies, and therapeutic trials that elucidate how oxidative stress could shape neurodevelopment and to disclose reliable targets for early intervention and prevention.
