Cover Story: Diabetes, which affects over 530 million adults worldwide, is characterized by the loss of functional pancreatic β-cells, primarily through apoptosis. This review examines the roles of the anti-apoptotic proteins Bcl-2 and Bcl-xL, which are critical regulators of the intrinsic apoptosis pathway. In addition to their role in apoptosis, these proteins influence intracellular Ca2+ homeostasis and mitochondrial metabolism, making them essential for β-cell survival and function. Dysregulation of Bcl-2 and Bcl-xL contributes to β-cell dysfunction, thereby accelerating the progression of diabetes. This article emphasizes the importance of maintaining proper levels of Bcl-2 and Bcl-xL in β-cells and explores potential therapeutic strategies targeting these proteins to preserve functional β-cell mass and combat diabetes. View this paper
