Cover Story: Nicotinamide is a derivate of vitamin B3 and is the precursor of NAD, a key coenzyme involved in several cellular reactions. NAM is often consumed daily and is endogenously produced by tryptophan. Additionally, NAM supplementation (500 mg–1g/die) can efficiently protect cells from oxidative stress, DNA damages, inflammation and aging. Indeed, NAM inhibits two energy-demanding enzymes—PARP1 and SIRT1—leading to NAD restoration, which is fundamental for the DNA damage repair system. Moreover, NAM can reduce the release of proinflammatory cytokines and reactive oxygen species (ROS), enhancing the repair of DNA and the recovery of the cell cycle. View this paper
