Background: The aesthetic performance of single-shade polymer-based restorative materials (SPRs) can be compromised by extrinsic stains. Understanding the effects of novel whitening interventions on SPRs is crucial. Objective: This in vitro study aimed to evaluate the effects of different whitening interventions, including a novel purple tooth serum and charcoal-based whitening toothpaste with and without in-office bleaching, on the color of a new coffee-stained SPR. Materials and Methods: Seventy disc-shaped SPR specimens were prepared, stained, and then divided into seven groups (n = 10). Three groups were subjected directly to 2500 cycles of brushing simulation with regular toothpaste (control), charcoal toothpaste, or purple tooth serum. The rest were divided into bleaching groups, and the four groups underwent a simulation of bleaching and then brushing with the three products. The color parameters were recorded at the stained baseline, after brushing, after bleaching, and after post-bleaching brushing. The color change (ΔE00) was calculated, and the data were analyzed statistically using the Kruskal–Wallis test and Dunn–Bonferroni pairwise comparisons (p < 0.05). Results: In-office bleaching without brushing had a statistically significantly higher ΔE00 value than all other groups (p < 0.001). Post hoc tests indicated that the ΔE00 values of the brushed specimens were not significantly different from each other when assessed with and without bleaching (p > 0.05). When using the charcoal toothpaste, the post-bleaching brushed specimens had a noticeable color change above the PT. Conclusions: Bleaching improved the stained SPR color initially, but other treatments may offer longer-lasting aesthetics. The charcoal toothpaste showed promising results when combined with bleaching. The purple serum showed limited effectiveness. Full article

Background/Objectives: In-office bleaching commonly employs high concentrations of hydrogen peroxide (HP) or carbamide peroxide (CP), which may compromise enamel integrity. This in vitro paired-design study aimed to compare the chemical and mechanical effects of three commercial bleaching agents—Opalescence Boost (40% HP), Opalescence Quick (45% CP), and BlancOne Ultra+ (35% HP)—on human enamel. The null hypothesis assumed no significant differences between the control and treated samples. Given the ongoing debate over pH, active ingredients, and enamel impact, comparing whitening systems remains clinically important. Methods: Forty-two extracted teeth were assigned to three experimental groups (n = 14) with matched controls. Each underwent a single bleaching session per manufacturer protocol: Opalescence Boost (≤60 min), Opalescence Quick (15–30 min), and BlancOne Ultra+ (three light-activated cycles of 8–10 min). Enamel chemical changes were analyzed by Fourier transform infrared (FTIR) spectroscopy (phosphate and carbonate bands), and surface hardness by Vickers microhardness testing. Paired t-tests (α = 0.05) assessed statistical significance. Results: FTIR analysis revealed alterations in phosphate and carbonate bands for all agents, most notably for Opalescence Boost and BlancOne Ultra+. Microhardness testing showed significant reductions in enamel hardness for Opalescence Boost (control: 37.21 ± 1.74 Hv; treated: 34.63 ± 1.70 Hv; p = 0.00) and Opalescence Quick (control: 45.82 ± 1.71 Hv; treated: 39.34 ± 1.94 Hv; p < 0.0001), whereas BlancOne Ultra+ showed no significant difference (control: 51.64 ± 1.59 HV; treated: 51.60 ± 2.34 Hv; p = 0.95). Conclusions: HP-based agents, particularly at higher concentrations, caused greater enamel alterations than CP-based products. While clinically relevant, the results should be interpreted cautiously due to in vitro limitations and natural enamel variability. Full article

Toothpaste is an essential oral hygiene product commonly used to sustain oral health due to its incorporation of antimicrobial agents. Numerous functional toothpastes enriched with antimicrobial agents have been developed and are available to consumers. This study evaluates the antimicrobial and antibiofilm efficacy of 12 commercially available toothpaste products, including those with specialized functions. Statistical significance was assessed to validate the differences observed among the toothpaste samples. Their effects on Streptococcus mutans, the primary pathogen responsible for dental caries, were evaluated. The minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) were determined, and bacterial growth was measured to compare antimicrobial activities. Toothpaste containing 1000 μg/mL fluoride and whitening toothpaste exhibited the strongest antimicrobial effects, effectively inhibiting S. mutans growth. Additionally, bamboo salt-enriched and tartar-control toothpaste demonstrated inhibitory effects on bacterial growth. Assays to evaluate the ability of cells to form biofilms and the expression of genes involved in biofilm formation revealed a partial correlation between biofilm formation and spaP, gtfB, gtfC, and gtfD expression, although some showed opposite trends. Collectively, this study provides valuable insights into the antimicrobial and biofilm inhibition capabilities of commercial toothpastes against S. mutans, offering a foundation for evaluating the efficacy of functional toothpaste products. Background: The rising demand for aesthetic dental treatments has spurred interest in peroxide-free color correctors as alternatives to traditional hydrogen peroxide formulations, which are associated with tooth sensitivity and potential enamel demineralization. This systematic review evaluates the whitening efficacy and safety profile of hydrogen peroxide-free color corrector (HPFCC) products, focusing on color change metrics, enamel and dentin integrity, and adverse effects. Methods: Following PRISMA guidelines, we searched PubMed, Scopus, and Web of Science throughout January 2025 for randomized controlled trials, observational studies, and in vitro experiments comparing HPFCC to placebo or peroxide-based agents. The data extraction covered study design, sample characteristics, intervention details, shade improvement (ΔE00 or CIE Lab), enamel/dentin mechanical properties (microhardness, roughness, elastic modulus), and incidence of sensitivity or tissue irritation. Risk of bias was assessed using the Cochrane tool for clinical studies and the QUIN tool for in vitro research. Results: Six studies (n = 20–80 samples or subjects) met the inclusion criteria. In vitro, HPFCC achieved mean ΔE00 values of 3.5 (bovine incisors; n = 80) and 2.8 (human molars; n = 20), versus up to 8.9 for carbamide peroxide (p < 0.01). Across studies, HPFCC achieved a mean ΔE00 of 2.8–3.5 surpassing the perceptibility threshold of 2.7 and approaching the clinical acceptability benchmark of 3.3. Surface microhardness increased by 12.9 ± 11.7 VHN with HPFCC (p < 0.001), and ultramicrohardness rose by 110 VHN over 56 days in prolonged use studies. No significant enamel erosion or dentin roughness changes were observed, and the sensitivity incidence remained below 3%. Conclusions: These findings derive from one clinical trial (n = 60) and five in vitro studies (n = 20–80), encompassing violet-pigment serums and gels with differing concentrations. Due to heterogeneity in designs, formulations, and outcome measures, we conducted a narrative synthesis rather than a meta-analysis. Although HPFCC ΔE00 values were lower than those of carbamide peroxide, they consistently exceeded perceptibility thresholds while maintaining enamel integrity and causing sensitivity in fewer than 3% of subjects, supporting HPFCCs as moderate but safe alternatives for young patients. Tooth bleaching is a widely used esthetic treatment; however, bleaching agents can temporarily alter the surface morphology of enamel, increasing surface roughness and porosity, which may lead to increased susceptibility to discoloration. This in vitro study investigated the effectiveness of fluoride varnish in preventing immediate discoloration of bovine incisors after bleaching. Specimens were bleached with 35% hydrogen peroxide and treated with either Clinpro White Varnish (CW) or Enamelast Fluoride Varnish (EN), whereas control specimens received no treatment after bleaching. The samples were immersed in coffee for 24 h, and the color difference (ΔE00) was calculated using the CIEDE2000 formula. The surface morphology of enamel was examined using SEM. The fluoride varnish groups showed significantly lower color difference values than the control group (p < 0.05), with ΔE00 reduced by approximately two-thirds in both the CW and EN groups. SEM observations showed that the enamel surfaces in the varnish-treated groups exhibited reduced surface irregularities compared to the untreated group, suggesting remineralization. These results suggest that the immediate application of fluoride varnish after bleaching can effectively reduce short-term discoloration by providing physical protection and promoting remineralization. Fluoride varnish may serve as a simple and effective strategy to maintain whitening outcomes and minimize early discoloration. Full article

Background/Objectives: Tooth bleaching is a widely requested aesthetic procedure in modern dentistry. However, the structural effects of high-concentration peroxide-based bleaching agents on enamel remain insufficiently understood. This study aims to evaluate and compare the effects of three in-office bleaching agents—Opalescence Boost (40% hydrogen peroxide [HP]), Opalescence Quick (45% carbamide peroxide [CP]), and BlancOne Ultra + (35% hydrogen peroxide [HP])—on enamel surface characteristics and color change using confocal laser scanning microscopy (CLSM) and spectrophotometric analysis. Methods: Forty-two extracted human teeth were sectioned and divided into experimental and control halves. Each experimental specimen underwent bleaching according to the manufacturer’s protocol. Color measurements were conducted at baseline, immediately post-treatment, at 3 days, 7 days, and 6 months following treatment using the Vita Easyshade® spectrophotometer. Color differences were calculated using the CIEDE2000 (ΔE00) formula. Enamel surface morphology was assessed by CLSM. Data were analyzed using Wilcoxon and Kruskal–Wallis tests (p < 0.05), performed with SPSS v23. Results: All bleaching agents produced clinically perceptible color changes (ΔE00 > 3.3). Opalescence Boost achieved the highest and most consistent whitening effect (mean ΔE00 > 11), while Opalescence Quick showed moderate efficacy (ΔE00 ~6–8), and BlancOne Ultra+ induced milder changes (ΔE00 ~4–5). CLSM imaging revealed surface alterations in all bleached samples, with more pronounced changes observed in specimens treated with higher peroxide concentrations. Conclusions: All three bleaching systems were effective in improving enamel color, with Opalescence Boost delivering the most substantial and durable effect. CLSM analysis confirmed morphological changes in enamel without evidence of severe damage. These results underscore the importance of selecting bleaching protocols that balance efficacy with enamel safety. Further in vivo studies are recommended to validate long-term structural effects and support clinical decision-making. Full article

Objective: To evaluate the color stability and microhardness of resin-based bioactive composites after brushing with over-the-counter whitening toothpastes. Methods: A conventional resin (Luna 2) and two bioactive resins (Stela Mix and Activa Presto) were tested. Four toothpastes were used: Colgate Fresh Gel (control), Colgate Max White, Yotuel, and Crest 3D White. Forty disks of each material were prepared and divided into four subgroups. The disks were brushed to simulate three months of daily brushing. Microhardness and color changes were measured before and after brushing. Color variation was calculated using the ΔEab, ΔE00, and ΔWID indexes. Wilcoxon and two-way ANOVA tests were performed (p < 0.05). Results: In the Luna 2 and Stela groups, the b* parameter decreased significantly with all toothpastes (p < 0.05), while lightness and a* remained stable with no significant differences (p > 0.05). Stela Mix and Activa Presto exhibited color changes within the “moderately unacceptable” range according to ΔE00 (>1.8 and ≤3.6). Based on the ΔWID index, Luna 2 showed the highest variation when treated with Colgate Max (2.14 ± 1.33) and the lowest in the control group (1.08 ± 0.56A), remaining within acceptable margins with all treatments. The microhardness values before/after treatment remained for Luna 2 between 77.44 and 76.97; for Stela Mix between 76.24 and 74.13; and for Activa presto between 74.5 and 71.33. Differences were not significant for any of the pastes within each composite (p > 0.05). Conclusions: The evaluated bioactive resins exhibited color changes within the moderately unacceptable range. Colgate Max White induced the most significant color changes. Microhardness was not affected by treatment with whitening toothpastes. Full article

American ginseng (Panax quinquefolium L.) residue from the extraction industry can be dried and mixed with rice bran as media for black truffle solid-state fermentation to enhance reuse and bioactive functions. Different ratios of rice bran (R) and American ginseng residue (G) mixtures were used as solid-state media for 5 weeks of black truffle fermentation, and then their bio-component contents and whitening effects were analyzed. Finally, four drying methods—hot air drying (HA), microwave drying (MW), hot air-assisted radio frequency (HARF) drying, and radio frequency vacuum (RFV) drying—were assessed to optimize drying efficiency for fermented medium. The results showed that using a 3:1 ratio of rice bran and American ginseng residue as the medium increased the crude polysaccharide and flavonoid contents by approximately threefold and enhanced the ginsenoside Rg3 content about twelvefold. Additionally, the 100 µg/mL ethanol extract of the fermented product inhibited 70% of tyrosinase activity and reduced the melanin area on zebrafish embryos by 42.74%. In the drying study, RFV drying R2G1 required only 13 min without exceeding 70 °C, demonstrating superior drying efficiency, temperature control, and low energy consumption. Overall, this study demonstrates the potential of black truffle fermentation of solid-state media from rice bran and American ginseng residue mixtures for whitening applications and highlights RFV drying as an efficient method for by-products. This study investigated the potential cosmetic properties of the ethyl acetate (EtOAc) fraction obtained from the roots of Astilboides tabularis (Hemsl.) Engl., focusing on skin-whitening, antiwrinkle, and moisturizing effects using cell-based assays and three-dimensional (3D) artificial skin models (Neoderm-ED and Neoderm-ME). The EtOAc fraction showed significant dose-dependent inhibitory activity against tyrosinase (TYR) (72.0% inhibition at 50 µg/mL), comparable to that of kojic acid. In α-melanocyte-stimulating hormone (α-MSH)-stimulated Neoderm-ME artificial skin containing melanocytes, the EtOAc fraction reduced melanin synthesis at concentrations of 50 and 75 µg/mL and decreased melanogenesis-related gene expression, including TYR, microphthalmia-associated transcription factor (MITF), tyrosinase-related protein-1 (TRP-1) and TRP-2. In the antiwrinkle assays, the EtOAc fraction effectively inhibited elastase activity (41.5% inhibition at 10 µg/mL), exceeding the efficacy of ursolic acid. In the Neoderm-ED artificial skin model, the EtOAc fraction reversed structural damage induced by particulate matter (PM10), restoring epidermal thickness and dermal density. This improvement was supported by the increased expression of skin barrier and antiwrinkle genes, including filaggrin, hyaluronic acid synthase-1 (HAS-1), HAS-2, aquaporin-3 (AQP-3), collagen type I alpha 1 chain (COL1A1), elastin, tissue inhibitor of metalloproteinases-1 (TIMP-1), and TIMP-2, as well as decreased expression of matrix metalloproteinases (MMP-1, MMP-3, and MMP-9). Our results indicate that the EtOAc fraction from A. tabularis root has considerable potential as a multifunctional cosmetic. Full article

Maintaining the cleanliness of orthodontic aligners is crucial for oral hygiene and preserving the optical properties of aligners. In this randomized clinical trial, we compared the effectiveness of different cleaning methods for the maintenance of Invisalign clear aligners. Twelve adult patients received five aligners, each worn for 10 days. The aligners were divided based on the cleaning method: tooth brushing with whitening toothpaste, vinegar, Fittydent Super Cleansing Tablets, Invisalign cleaning crystals, and only water. Scanning electron microscopy (SEM) was used to detect surface morphology changes; color changes (ΔE) were evaluated using a spectrophotometer. Fourier transform infrared spectroscopy (FTIR) with a diamond hemisphere was used to study the aligners’ chemical compositions. Nanoindentation testing was used to assess changes in the elastic modulus. SEM confirmed the effectiveness of Invisalign cleaning crystals in maintaining cleanliness, revealing a surface similar to that of the control group with no adverse effects. Color stability analysis revealed significant ΔE value differences; whitening toothpaste had significantly lower ΔE values than water and Invisalign cleaning crystals. The elastic modulus and FTIR analyses indicated no significant differences between the cleaning methods. Allium genus (Amaryllidaceae) is widely distributed in the Northern hemisphere. Some species, including garlic and onion, have been used since ancient times as both food ingredients and medicinal plants. Many reviews deal with the chemical constituents, particularly the typical sulfur compounds, as well as with Allium pharmacological properties, such as antimicrobial, anti-inflammatory, antioxidant, and cytotoxic activities. The bibliographic search performed in this review is mainly focused on the potential role of Allium species in inhibiting melanogenesis, which has been mainly assessed through the evaluation of the inhibitory properties on tyrosinase, the key enzyme in melanin biosynthesis. Two well established models for identifying potential skin-whitening agents have been used to assess the anti-melanogenic effects of Allium species, the mushroom tyrosinase and the murine melanoma B16 cell line. Here, a literature search from Scopus, Web of Science, and PubMed databases has been performed using the keywords “Allium”, “tyrosinase”, “anti-melanogenic”, and “melanogenesis”, combined by means of Boolean operators. Based on selected inclusion criteria, 32 eligible papers have been selected. The aim of this systematic review is to offer an overview of the species for which the ability to affect melanogenesis has been demonstrated to date, highlighting a new and emerging perspective on the potential therapeutic use of Allium species. The biological properties of isolated pure compounds and the negative outcomes have been also considered. Full article

Peptides are molecules composed of two or more amino acids linked by peptide bonds, and they play essential biological roles. In recent decades, peptides have become pivotal bioactive ingredients in pharmaceuticals and cosmetics due to their unique features. Originally developed for therapeutic purposes, peptides have gained popularity in the cosmetic field, providing solutions for anti-aging, whitening, moisturizing, and skin repair. Moreover, innovations such as artificial intelligence-assisted peptide design, efficient delivery systems, and the integration of multifunctional ingredients have significantly contributed to the industry’s rapid evolution. This review explores the historical milestones of peptides in medicine and cosmetics, delves into cutting-edge synthesis technologies, and dissects the molecular mechanisms behind their cosmetic properties. Research in medicinal peptides has promoted the development of cosmetic peptides. Despite their potential, challenges such as stability, bioavailability, and cost-effective production remain barriers to widespread adoption. Future studies should focus on enhancing peptide stability, developing synergistic formulations, and conducting large-scale clinical trials to validate long-term efficacy. With continuous innovation, peptides are poised to redefine the cosmetic industry, bridging the gap between pharmaceuticals and skincare for safer and more effective solutions. Full article

Effects of Resveratrol Derivatives on Melanogenesis and Antioxidant Activity in B16F10 Cells

Excessive melanogenesis causes abnormal pigmentation and a higher risk of skin disorders (e.g., melanoma). Resveratrol (RSV), a natural polyphenol, exerts antioxidant and anti-aging effects. However, the effects of RSV and its derivatives on melanogenesis remain unclear. This study investigated their effects on melanogenesis and antioxidant activity in B16F10 cells. After measuring cell viability, B16F10 cells were incubated with 50 µM of RSV, dihydroresveratrol (DIRSV), and other RSV derivatives for 24 h. The relative melanin content and tyrosinase activity were quantified. The protein and mRNA levels of melanogenesis-related genes (MITF, CREB, TYR, and TRP) and the binding affinity of RSV derivatives to their target proteins were measured. The antioxidant activity was evaluated using ABTS and DPPH assays. RSV and DIRSV (50 µM) significantly reduced melanin content and tyrosinase activity, respectively. However, other derivatives had no significant effects. RSV, DIRSV, and other derivatives significantly suppressed MITF and CREB levels. Additionally, DIRSV significantly reduced p-CREB and TYR protein levels and showed a higher affinity for CREB than RSV, despite no significant changes in MITF, TYR, or TRP mRNA levels. In the antioxidant assays, RSV and DIRSV exhibited significant ABTS and DPPH radical scavenging activities. DIRSV, like RSV, inhibits melanogenesis and exhibits antioxidant effects in B16F10 cells. However, RSV derivatives demonstrate partial antioxidant activity and inhibit melanogenesis-related proteins but do not significantly affect melanogenesis. DIRSV’s practical applications as a skin-protective and -whitening agent warrant further exploration. Full article

Background: Tooth whitening is a widely sought-after cosmetic procedure, with various at-home and professional treatments available. This study compares the whitening efficacy of an activated charcoal toothpaste and a 6% hydrogen peroxide whitening pen under controlled in vitro conditions. Methods: Twenty freshly extracted human teeth were stained with a coffee solution and divided into two groups. Group A underwent daily applications of activated charcoal toothpaste for 30 days, while Group B received a single 5 min application of a 6% hydrogen peroxide whitening pen. Tooth color was assessed using the VITA Classical A1-D4 Shade Guide at baseline, mid-treatment, and post-treatment for Group A and at baseline and immediately after treatment for Group B. Results: The activated charcoal toothpaste exhibited a gradual whitening effect, with the most significant improvements occurring within the first two weeks (p < 0.01), after which the whitening effect plateaued. In contrast, the hydrogen peroxide whitening pen produced immediate and substantial whitening (p < 0.001). Statistical analysis using the Wilcoxon signed-rank test and Mann–Whitney U test confirmed the superior efficacy of the hydrogen peroxide treatment. Conclusions: The hydrogen peroxide whitening pen was significantly more effective in achieving rapid and substantial whitening compared to the activated charcoal toothpaste, which provided gradual but limited improvements. Further clinical studies are needed to evaluate the long-term color stability. Full article

This study proposes a comprehensive model for identifying mine water inrush sources in coal mines throughout the full mining cycle, utilizing an improved whitening function and the CRITIC-weighted grey situational decision method. Traditional water source identification methods often fail to account for the dynamic changes in water sources during the mining process, which can be influenced by geological and hydrological conditions. The model integrates an exponential whitening function with the CRITIC weighting approach to address the high variability and correlations between variables. Through the analysis of 244 groundwater samples from the Sunan mining area, the model demonstrated significant improvements in accuracy across different mining stages. The results showed overall classification accuracies exceeding 85%, indicating the model’s effectiveness in providing real-time early warnings for water hazards. This model not only optimizes traditional methods but also offers a robust tool for dynamic water source identification, thereby supporting safer and more efficient coal mining operations.
