Salinization hinders the restoration of vegetation in salt-affected soils by negatively impacting plant growth and development. Halophytes play a key role in the restoration of saline and degraded lands due to unique features explaining their growth aptitude in such extreme ecosystems. Suaeda fruticosa is an euhalophyte well known for its medicinal properties and its potential for saline soil phytoremediation. However, excessive salt accumulation in soil limits the development of this species. Research findings increasingly advocate the use of extremophile rhizosphere bacteria as an effective approach to reclaim salinized soils, in conjunction with their salt-alleviating effect on plants. Here, a pot experiment was conducted to assess the role of a halotolerant plant growth-promoting actinobacterium, Glutamicibacter sp., on the growth, nutritional status, and shoot content of proline, total soluble carbohydrates, and phenolic compounds in the halophyte S. fruticosa grown for 60 d under high salinity (600 mM NaCl). Results showed that inoculation with Glutamicibacter sp. significantly promoted the growth of inoculated plants under stress conditions. More specifically, bacterial inoculation increased the shoot concentration of proline, total polyphenols, potassium (K+), nitrogen (N), and K+/Na+ ratio in shoots, while significantly decreasing Na+ concentrations. These mechanisms partly explain S. fruticosa tolerance to high saline concentrations. Our findings provide some mechanistic elements at the ecophysiological level, enabling a better understanding of the crucial role of plant growth-promoting rhizobacteria (PGPRs) in enhancing halophyte growth and highlight their potential for utilization in restoring vegetation in salt-affected soils. Bamboo shoot boiled liquid (BSBL), a processing byproduct containing soluble proteins, peptides, amino acids, carbohydrates, and phenolics, is typically discarded, causing resource waste and environmental issues. This study analyzed metabolic changes in BSBL during Pediococcus pentosaceus B49 fermentation. The result of partial least squares discriminant analysis (PLS-DA) revealed significant metabolite profile differences across fermentation times (0 h, 24 h, 48 h, 72 h, 96 h). The most substantial alterations occurred within the first 24 h, followed by stabilization. Compared to unfermented BSBL, fermented samples exhibited significantly elevated signal intensities for 5,7-dimethoxyflavone, cinnamic acid, 3,4-dihydro-2H-1-benzopyran-2-one, 6,8-dimethyl-4-hydroxycoumarin, and 2-hydroxycinnamic acid (p < 0.05), showing upward trends over time. Conversely, (+)-gallocatechin intensity decreased gradually. Bitter peptides, such as alanylisoleucine, isoleucylisoleucine, leucylvaline, and phenylalanylisoleucine, in BSBL exhibited a significant reduction following fermentation with P. pentosaceus B49 (p < 0.05). KEGG enrichment indicated tyrosine metabolism (ko00350) and arginine/proline metabolism (ko00330) as the most impacted pathways. These findings elucidate metabolic regulation in BSBL fermentation, supporting development of functional fermented bamboo products. Full article

Soil phosphorus (P) availability is an important determinant of productivity in Pinus massoniana (Masson pine) forests. The mechanistic bases governing the physiological and growth responses of Masson pine to varying soil P conditions remain insufficiently characterized. This study aims to decipher the adaptive strategies of Masson pine to different soil P levels, focusing on root morphological–architectural plasticity and the allocation dynamics of nutrient elements and photosynthetic assimilates. One-year-old potted Masson pine seedlings were exposed to four P addition treatments for one year: P0 (0 mg kg−1), P1 (25 mg kg−1), P2 (50 mg·kg−1), and P3 (100 mg kg−1). In July and December, measurements were conducted on seedling organ biomass, root morphological indices [root length (RL), root surface area (RSA), root diameter (RD), specific root length (SRL), and root length ratio (RLR) for each diameter grade], root architectural indices [number of root tips (RTs), fractal dimension (FD), root branching angle (RBA), and root topological index (TI)], as well as the content of nitrogen (N), phosphorus (P), carbon (C), and non-structural carbohydrates (NSCs) in roots, stems, and leaves. Compared with the P0 treatment, P2 and P3 significantly increased root biomass, root–shoot ratio, RL, RSA, RTs, RLR of finer roots (diameter ≤ 0.4 mm), nutrient accumulation ratio in roots, and starch (ST) content in roots, stems and leaves. Meanwhile, they decreased soluble sugar (SS) content, SS/ST ratio, C and N content, and N/P and C/P ratios in stems and leaves, as well as nutrient accumulation ratio in leaves. The P3 treatment significantly reduced RBA and increased FD and SRL. Our results indicated that Masson pine adapts to low P by developing shallower roots with a reduced branching intensity and promoting the conversion of ST to SS. P’s addition effectively alleviates growth limitations imposed by low P, stimulating root growth, branching, and gravitropism. Although a sole P addition promotes short-term growth and P uptake, it triggers a substantial consumption of N, C, and SS, leading to significant decreases in N/P and C/P ratios and exacerbating N’s limitation, which is detrimental to long-term growth. Under high-P conditions, Masson pine strategically prioritizes allocating limited N and SS to roots, facilitating the formation of thinner roots with low C costs. Full article

Noncommunicable diseases (NCDs) like diabetes and cancer drive demand for therapeutic functional foods. This study developed freeze-dried fermented camel milk (FCM) with Ajwa date pulp (ADP), evaluating its physical and functional properties, probiotic survival, and potential benefits for diabetes and cancer. To achieve this target, six FCM formulations were prepared using ABT-5 starter culture (containing Lactobacillus acidophilus, Bifidobacterium bifidum, and Streptococcus thermophilus) with or without Lacticaseibacillus rhamnosus B-1937 and ADP (12% or 15%). The samples were freeze-dried, and their functional properties, such as water activity, dispersibility, water absorption capacity, water absorption index, water solubility index, insolubility index, and sedimentation, were assessed. Reconstitution properties such as density, flowability, air content, porosity, loose bulk density, packed bulk density, particle density, carrier index, Hausner ratio, porosity, and density were examined. In addition, color and probiotic survivability under simulated gastrointestinal conditions were analyzed. Also, antidiabetic potential was assessed via α-amylase and α-glucosidase inhibition assays, while cytotoxicity was evaluated using the MTT assay on Caco-2 cells. The results show that ADP supplementation significantly improved dispersibility (up to 72.73% in FCM15D+L). These improvements are attributed to changes in particle size distribution and increased carbohydrate and mineral content, which facilitate powder rehydration and reduce clumping. All FCM variants demonstrated low water activity (0.196–0.226), indicating good potential for shelf stability. The reconstitution properties revealed that FCM powders with ADP had higher bulk and packed densities but lower particle density and porosity than controls. Including ADP reduced interstitial air and increased occluded air within the powders, which may minimize oxidation risks and improve packaging efficiency. ADP incorporation resulted in a significant decrease in lightness (L*) and increases in redness (a*) and yellowness (b*), with greater pigment and phenolic content at higher ADP levels. These changes reflect the natural colorants and browning reactions associated with ADP, leading to a more intense and visually distinct product. Probiotic survivability was higher in ADP-fortified samples, with L. acidophilus and B. bifidum showing resilience in intestinal conditions. The FCM15D+L formulation exhibited potent antidiabetic effects, with IC50 values of 111.43 μg mL−1 for α-amylase and 77.21 μg mL−1 for α-glucosidase activities, though lower than control FCM (8.37 and 10.74 μg mL−1, respectively). Cytotoxicity against Caco-2 cells was most potent in non-ADP samples (IC50: 82.22 μg mL−1 for FCM), suggesting ADP and L. rhamnosus may reduce antiproliferative effects due to proteolytic activity. In conclusion, the study demonstrates that ADP-enriched FCM is a promising functional food with enhanced probiotic viability, antidiabetic potential, and desirable physical properties. This work highlights the potential of camel milk and date synergies in combating some NCDs in vitro, suggesting potential for functional food application. Full article

Advanced glycation end-products (AGEs) are formed by the non-enzymatic glycation of proteins, lipids, and nucleic acids due to the consumption of high-carbohydrate diets; their production is also promoted by a sedentary lifestyle as well as cigarette smoking. Elevated levels of AGEs in the circulatory system and internal organs of the body are commonly observed in a number of cardiovascular diseases such as hypertension, diabetes, atherosclerosis, coronary artery disease, aortic aneurysm, atrial fibrillation, myocardial infarction, and heart failure, which are associated with the development of oxidative stress and myocardial inflammation. The adverse effects of AGEs on the cardiovascular system are elicited by both non-receptor mechanisms involving the cross-linking of extracellular and intracellular proteins, and by receptor-mediated mechanisms involving the binding of AGEs with advanced glycation end-product receptors (RAGEs) on the cell membrane. AGE–RAGE interactions along with the cross-linking of proteins promote the generation of oxidative stress, the production of inflammation, the occurrence of intracellular Ca2+-overload, and alterations in the extracellular matrix leading to the development of cardiovascular dysfunction. AGEs also bind with two other protein receptors in the circulatory system: soluble RAGEs (sRAGEs) are released upon the proteolysis of RAGEs due to the activation of matrix metalloproteinase, and endogenous secretory RAGEs (esRAGEs) are secreted as a spliced variant of endogenous RAGEs. While the AGE–RAGE signal transduction axis serves as a pathogenic mechanism, both sRAGEs and esRAGEs serve as cytoprotective interventions. The serum levels of sRAGEs are decreased in ischemic heart disease, vascular disease, and heart failure, as well as in other cardiovascular diseases, but are increased in chronic diabetes and renal disease. Several interventions which can reduce the formation of AGEs, block the AGE–RAGE axis, or increase the levels of circulating sRAGEs have been shown to exert beneficial effects in diverse cardiovascular diseases. These observations support the view that the AGE–RAGE axis not only plays a critical role in pathogenesis, but is also an excellent target for the treatment of cardiovascular disease. Full article

Triticale (Triticosecale Wittmack) is a versatile forage crop valued for its high yield, balanced nutrition, and environmental adaptability. However, the dough-stage triricale has higher dry matter and starch content but lower water-soluble carbohydrate levels than earlier stages, posing fermentation challenges that may impair silage quality. This study aimed to investigate the effects of lactic acid bacteria inoculation on the fermentation quality, bacterial community, and metabolome of whole-plant triticale silage at the dough stage. Fresh triticale was ensiled for 30 days without or with an inoculant containing Lactiplantibacillus plantarum and Streptococcus bovis. Fermentation quality, bacterial succession, and metabolic profiles were analyzed at multiple time points. Inoculation significantly improved fermentation quality, characterized by a rapid pH drop, increased lactic acid production, and better preservation of fiber components. Microbial analysis revealed that inoculation successfully established Lactobacillus as the dominant genus while suppressing spoilage bacteria like Enterobacter and Clostridium. Metabolomic analysis on day 30 identified numerous differential metabolites, indicating that inoculation primarily altered pathways related to amino acid and purine metabolism. In conclusion, inoculating dough-stage triticale with this LAB combination effectively directs the fermentation trajectory. It enhances silage quality not only by optimizing organic acid profiles and microbial succession but also by modulating key metabolic pathways, ultimately leading to improved nutrient preservation. Climate change has contributed to a decline in winter chilling accumulation, a critical requirement for budbreak in temperate fruit crops. Its consequence has been a reduction in fruit production. To compensate for insufficient chilling, hydrogen cyanamide (HC) is widely applied, though its effectiveness remains limited. This study investigated the effect of HC application on budbreak in low-chill kiwifruit under warm conditions by correlating phenological responses with changes in carbohydrate and nitrogen concentrations in bark tissues across bud positions. Phenological observations revealed the highest budbreak percentage and total flower buds at the apical position. HC significantly increased budbreak by 58.82% at the apical position and by 375% at the middle position, with corresponding increases in total flower buds by 148.78% and 1066.67%, respectively. Additionally, shoot lengths were uniform among bud positions in HC-treated canes, whereas non-treated canes showed shoot length heterogeneity. Moreover, HC treatment triggered an earlier and more pronounced reduction in soluble sugars (sucrose and hexoses) concentrations along the gradient from apical to basal bud positions, where the response was strongest at the apical position, which was strongly associated with enhanced budbreak percentages and total flower bud formation. While total nitrogen content was highest in the apical position, it was unaffected by HC application. These findings indicate that HC may promote budbreak by enhancing the mobilization and consumption of soluble sugars for bud growth, thereby improving budbreak performance, flower bud production, and uniform shoot development in low-chill kiwifruit under warm conditions. Nitrogen is an essential element for plant growth, and both insufficient and excessive use of nitrogen have been shown to negatively affect sweet potato production. Nitrogen supply can affect carbon metabolism in plant storage organs; however, limited studies have examined its effects on the accumulation of non-structural carbohydrates (soluble sugar and starch) during the formation of sweet potato storage roots. Two pot trials were conducted to evaluate the effects of different nitrogen application levels and timings on the accumulation of non-structural carbohydrates during the formation of sweet potato storage roots. In the first experiment, plants were supplied with 0, 50, 100, or 200 mg/L of nitrogen. In the second experiment, the optimum nitrogen rate (100 mg/L) for storage root formation from the previous experiment was applied at five different times: nil N supply and nitrogen applied at planting or 3, 7, or 14 days after planting. A significant highest starch accumulation in roots during the first 35 days after transplanting was recorded in the 100 mg/L treatment. However, sweet potato required more nitrogen after storage root formation, as indicated by higher non-structural carbohydrate accumulation in roots (1905 mg/plant) in the 200 mg/L treatment at 49 days after planting. Earlier nitrogen applications promoted soluble sugar and starch accumulation in plants during storage root formation, with up to 5697 mg of non-structural carbohydrate accumulated in a plant. The study provided agronomic indicators that moderate nitrogen should be available in soil before or on planting day. Full article

This study chemically characterized three Pleurotus ostreatus fruiting bodies cultivated in the Iberian Peninsula under different conditions (biological and industrial), with emphasis on polysaccharide analysis. Comprehensive comparative data on cultivation-dependent nutritional variations will potentially improve their nutritional and therapeutic applications. Industrial mushrooms (POC and POA) contained significantly higher carbohydrate content (74%), while the biologically cultivated mushroom (POL) exhibited more protein (22.6%), fat (4.2%), and ashes (8.0%). Monosaccharide analysis showed glucose dominance (28.7–45.5%), with mannose, galactose, xylose, and arabinose also present. Trehalose was the primary free sugar (4.8–14.9%). The (1→3)(1→6)-β-glucans varied significantly across samples (POL: 20.5%; POC: 29.3%; POA: 34.3%). Nuclear magnetic resonance analysis suggested complex polysaccharide arrangements. Water-soluble carbohydrates and proteins showed molecular weight distributions of 0.18–21 kDa and 0.20–75 kDa, respectively. All mushrooms were rich in essential amino acids, phosphorus (2.79–3.07%), potassium (0.56–0.68%), linoleic acid (0.82–1.14%), and oleic acid (0.22–0.31%). Fourier transform infrared confirmed a mushroom-specific biochemical profile. These findings corroborate the high nutritional value of POL, POC, and POA, with a significant contribution to the daily requirements of fiber, protein, and minerals (phosphorus, potassium, magnesium, iron, zinc, copper, and selenium), making them suitable for functional foods and nutraceuticals with cultivation-dependent nutritional profiles. Amid global environmental change, the intensification of nitrogen (N) deposition exerts critical impacts on the growth of forest vegetation and the structure and function of ecosystems in subtropical China. However, the physiological and growth response mechanisms of subtropical tree species remain poorly understood. This study explored adaptive mechanisms of typical subtropical tree species to N deposition, analyzing biomass accumulation, root plasticity, and nutrient/photosynthate allocation strategies. One-year-old potted seedlings of Phyllostachys edulis (moso bamboo) and Cunninghamia lanceolata (Chinese fir) were subjected to four N-addition treatments (N0: 0, N1: 6 g·m−2·a−1, N2: 12 g·m−2·a−1, N3: 18 g·m−2·a−1) for one year. In July and December, measurements were conducted on seedling organ biomass, root morphological and architectural traits, as well as nutrient elements (N and phosphorus(P)) and non-structural carbohydrate (soluble sugars and starch) contents in roots, stems, and leaves. Our results demonstrate that the Chinese fir exhibits stronger tolerance to N deposition and greater root morphological plasticity than moso bamboo. It adapts to N deposition by developing root systems with a higher finer root (diameter ≤ 0.2 mm) ratio, lower construction cost, greater branching intensity and angle, and architecture approaching dichotomous branching. Although N deposition promotes short-term biomass and N accumulation in both species, it reduces P and soluble sugars contents, leading to N/P imbalance and adverse effects on long-term growth. Under conditions of P and photosynthate scarcity, the Chinese fir preferentially allocates soluble sugars to leaves, while moso bamboo prioritizes P and soluble sugars to roots. In the first half of the growing season, moso bamboo allocates more biomass and N to aboveground parts, whereas in the second half, it allocates more biomass and P to roots to adapt to N deposition. This study reveals that Chinese fir enhances its tolerance to N deposition through the plasticity of root morphology and architecture, while moso bamboo exhibits dynamic resource allocation strategies. The research identifies highly adaptive root morphological and architectural patterns, demonstrating that optimizing the allocation of elements and photosynthates and avoiding elemental balance risks represent critical survival mechanisms for subtropical tree species under intensified N deposition. Integrated application of chemical fertilizers with organic manure might improve crop yields and N-use efficiency (NUE, grain yield per unit N uptake), but the underlying physiological mechanisms are unclear. In this study, we aimed to examine the effects of combined inorganic and organic fertilizers on wheat biomass allocation, root growth, water-soluble carbohydrates (WSCs) translocation, leaf senescence, N uptake, and their relationship with yield and NUE. We established a 2-year factorial field experiment with five nutrient treatments with ratios of inorganic: organic fertilizers from 0 to 1, and three varieties—two new: Weilong169 and Zhongmai578; and one reference: Xiaoyan22. The yield ranged from 3469 to 8095 kg ha−1, and it generally declined in response to a higher proportion of organic fertilizer. The NUE increased when there was a higher proportion of organic fertilizer. Weilong169 exhibited higher NUE than Zhongmai578, and both new cultivars outperformed the reference variety in the N harvest index. The yield correlated with leaf senescence traits and harvest index, and NUE was associated with WSC translocation and N uptake. The combination of fertilizers with a low portion of organic maintained yield and improved NUE; Weilong169 had the highest yield, NUE, and N harvest index. A low portion of organic manure substitution for chemical fertilizer suited all varieties. A new variety with a higher yield, N harvest index, and NUE highlights the importance of N traits in breeding programs. Full article

Pear buds exhibit inherent dormancy, during which carbohydrates play a pivotal role in dormancy release and germination. In this study, Pyrus pyrifolia ‘Cuiguan’ was employed as the experimental material to investigate the molecular mechanisms underlying flower bud dormancy release. The results revealed that the dynamic balance between starch and soluble sugar is critical for promoting dormancy release and germination in P. pyrifolia ‘Cuiguan’ flower buds. Through transcriptomic and proteomic profiling, a total of 4035 differentially expressed genes (DEGs) and 1596 differentially expressed proteins (DEPs) were identified, which were predominantly associated with carbohydrate metabolism, particularly sugar metabolism pathways. Their changes were coordinately regulated at both transcriptional and translational levels. Key structural genes involved in maltose and sucrose biosynthesis, including BAM (LOC103949270), AAM (LOC125479337, LOC103940334, and LOC103941903), SPS (LOC125475683), and INV (LOC125478747), were significantly upregulated during the germination stage, facilitating flower bud sprouting. Integrated multi-omic analysis demonstrated that starch–sugar interconversion may govern dormancy release and sustained bud growth by modulating sugar metabolism-related genes and proteins. These findings provide novel insights into the molecular mechanisms of carbohydrate biosynthesis and associated protein regulation during dormancy release and development of P. pyrifolia ‘Cuiguan’ under natural conditions. Global climate change has intensified the heterogeneity of precipitation regimes in subtropical regions, and the increasing frequency of extreme drought events poses a significant threat to biogeochemical cycling in forest ecosystems. Yet, the pathways by which reduced precipitation regulates deadwood decomposition and thereby influences soil nutrient pools remain poorly resolved. Here, we investigated a Cunninghamia lanceolata (Lamb.) Hook. plantation in subtropical China under ambient precipitation (CK) and precipitation reduction treatments of 30%, 50%, and 80%, systematically examining how reduced precipitation alters the chemical properties of deadwood substrates and, in turn, soil nutrient status. Our findings reveal that (1) as precipitation declined, soil water content decreased significantly (p < 0.01), while deadwood pH declined and total organic carbon (TOC), nonstructural carbohydrates (NSCs), and lignin content markedly accumulated (p < 0.01); (2) these shifts in deadwood chemistry affected feedback mechanisms, leading to the suppression of soil nutrient pools: extreme drought (80% reduction) significantly reduced soil TOC, dissolved organic carbon (DOC), total nitrogen (TN), and total phosphorus (TP) (p < 0.01) and inhibited N and P mineralization, whereas the 30% reduction treatment elicited a transient increase in soil microbial biomass carbon (MBC), indicative of microbial acclimation to mild water stress; and (3) principal component analysis (PCA) showed that the 80% reduction treatment drove lignin accumulation in deadwood, while the 30% reduction treatment exerted the greatest influence on soil DOC, TOC, and MBC; partial least squares path modeling (PLS-PM) further demonstrated that soil water content and deadwood substrate properties (pH, lignin, soluble sugars, TOC, C/N, and lignin/N) were strongly negatively correlated (r = −0.9051, p < 0.01), and that deadwood chemistry was, in turn, negatively correlated with soil nutrient variables (pH, TOC, DOC, MBC, TP, TN, and dissolved organic nitrogen [DON]; r = −0.8056, p < 0.01). Together, these results indicate that precipitation reduction—by drying soils—profoundly modifies deadwood chemical composition (lignin accumulation and NSC retention) and thereby, via slowed organic-matter mineralization, constrains soil nutrient release and accumulation. This work provides a mechanistic framework for understanding forest carbon–nitrogen cycling under climate change. Full article

Molybdenum fertilization represents a viable alternative for improving forage quality, potentially complementing or enhancing the effects of nitrogen fertilization. This study aimed to determine whether foliar or soil application of molybdenum would increase the crude protein content and digestibility of sorghum cultivated as a monoculture or intercropped with cowpea. The first experiment followed a 2 × 2 + 2 factorial design, including two application methods (foliar or soil), two cropping systems (monoculture or intercropping), and two additional control treatments (with and without molybdenum). In the second experiment, a split-plot design was used to assess the effects of molybdenum fertilization on the in situ digestibility of sorghum from monoculture and intercropping systems. Molybdenum fertilization increased the levels of crude protein, total carbohydrates, and soluble fractions. It also enhanced the disappearance rate, potential degradability, and effective degradability of sorghum, regardless of the application method or cropping system. Foliar or soil application of molybdenum is recommended to optimize the crude protein content and in situ digestibility of sorghum cultivated either as a monoculture or intercropped with cowpea. The plant microbiome plays a crucial role in the health of the tea plant, while Bacillus thuringiensis (Bt) is widely utilized as a biological pesticide in tea gardens, promoting sustainable agricultural practices. However, the effects of Bt spraying on tea quality and the structure and function of the phyllosphere microbiome remain unclear. This study evaluated the effects of Bt spraying on tea quality, microbiome composition, diversity, and potential functions using tea leaf quality measurements and high-throughput sequencing of the 16S/ITS rDNA genes. Results showed that spraying Bt1 significantly increased the contents of free amino acids (by 15.27%), flavonoids (by 18.00%), soluble sugars (by 62.55%), and key compounds such as epicatechin gallate (by 10.50%), gallocatechin gallate (by 122.52%), and epigallocatechin gallate (by 61.29%), leading to improved leaf quality. Co-occurrence network analysis indicated that the community structure of both epiphytic and endophytic microbes became more complex after Bt treatment. The abundance of beneficial bacteria, such as Novosphingobium, Methylobacterium, and Sphingomonas, increased significantly, while pathogenic fungi like Aspergillus and Phyllosticta decreased. Functional prediction indicated enhanced amino acid metabolism, secondary metabolism, and carbohydrate metabolism, particularly the biosynthesis of flavonoids, which supports disease resistance and boosts secondary metabolite levels. Furthermore, Bt application reduced pathogenic fungi, enhancing the tea plant’s resistance to diseases. Overall, foliar spraying of Bt can positively alter the phyllosphere microbiome by enriching beneficial bacteria and improving metabolic functions, ultimately enhancing tea plant resistance and quality, and providing a scientific basis for sustainable pest management in tea cultivation.
