Different Responses to Salinity of Pythium spp. Causing Root Rot on Atriplex hortensis var. rubra Grown in Hydroponics

Atriplex hortensis var. rubra (red orache, RO) is a halotolerant species rich in nutraceutical compounds, which makes it a valuable crop for human nutrition. This plant could also be exploited for phytoremediation of contaminated soil and wastewater, and for saline aquaponics. A root rot disease was observed on hydroponically grown RO plants, caused by Pythium deliense and Pythium Cluster B2a sp. Identification was based on morphology, molecular analysis (ITS and COI), and phylogenetic analysis. We assessed disease severity in plants grown in a growth chamber with nutrient solutions containing different NaCl concentrations (0, 7, and 14 g L−1 NaCl). In vitro growth at different salinity levels and temperatures was also evaluated. Both Pythium species were pathogenic but showed different responses. Pythium deliense was significantly more virulent than Pythium Cluster B2a sp., causing a steady reduction in root dry weight (RDW) of 70% across all salinity levels. Pythium Cluster B2a sp. reduced RDW by 50% at 0 and 7 g L−1 NaCl while no symptoms were observed at 14 g L−1 NaCl. Pythium deliense grew best at 7 and 14 g L−1 NaCl, while Pythium Cluster B2a sp. growth was reduced at 14 g L−1 NaCl. Both pathogens had an optimum temperature of 30 °C. This is the first report of Pythium spp. causing root rot on RO grown hydroponically. The effective use of halophytic crops must consider pathogen occurrence and fitness in saline conditions. In this study, the impact of moderate and high CO2 and O2 levels was compared to low and moderate gas combinations during prolonged storage on the quality of Regina sweet cherries harvested in different maturity stages, particularly in terms of decreasing internal browning. Fruits were harvested in two different maturity stages (Light and Dark Mahogany skin color) and stored in CA of 15% CO2 + 10% O2; 10% CO2 + 10% O2; 10% CO2 + 5% O2; 5% CO2 + 5% O2 and MA of 4 to 5% CO2 + 16 to 17% O2 for 30 and 40 days at 0 °C and 90% RH, followed by a marketing period. After the storage, both maturity stages significantly reduced internal browning, decay, and visual quality losses in CA with 10–15% CO2 and 10% O2. In addition, it preserved luminosity, total soluble solids (TSSs), titratable acidity (TA), and bioactive compounds such as anthocyanins and phenols. This treatment also maintained the visual appearance of the sweet cherries, favoring their market acceptance. At the same time, the light red fruits showed a better general quality compared to darker color after the storage. In conclusion, a controlled atmosphere with optimized CO2 and O2 concentrations, together with harvesting with a Light Mahogany external color, represents an effective strategy to extend the shelf life of Regina sweet cherries up to 40 days plus the marketing period, maintaining their physical and sensory quality for export markets. Rhabditis (Rhabditella) axei is a predominantly free-living nematode commonly found in sewage systems and decomposing organic matter. While primarily saprophytic, it has been documented as an opportunistic pathogen in human urinary and gastrointestinal tracts. The Chinese red panda (Ailurus styani), a rare and protected species in China, has not previously been reported as a host for Rhabditis (Rhabditella) spp. infections. This study reports the first documented occurrence of R. axei in red panda feces, unambiguously confirmed through integrative taxonomic approaches combining morphological and molecular analyses. The nematodes exhibited key morphological features consistent with R. axei, including a cylindrical rhabditiform esophagus, sexually dimorphic tail structures, and diagnostic spicule morphology. Molecular analysis based on 18S-ITS-28S rDNA sequencing confirmed their identity, showing >99% sequence similarity to R. axei reference strains (GenBank: PP135624.1, PP135622.1). Phylogenetic reconstruction using 18S rDNA and ITS rDNA sequences placed the isolate within a well-supported R. axei clade, clearly distinguishing it from related species such as R. blumi and R. brassicae. The findings demonstrate the ecological plasticity of R. axei as a facultative parasite capable of infecting non-traditional hosts and further highlight potential zoonotic risks associated with environmental exposure in captive wildlife populations. Our results emphasize the indispensable role of molecular diagnostics in accurately distinguishing morphologically similar nematodes within the Rhabditidae family, while providing essential baseline data for health monitoring in both in situ and ex situ conservation programs for this endangered species. Sport Participation and Gender Differences in Dietary Preferences: A Cross-Sectional Study in Italian Adults

Background: The relationship between sports participation and food preferences in adults, as well as the influence of gender, is still unclear. Objective: The objective of this study was to investigate the association between sports participation and individual food preferences and to explore potential gender differences among sports participants in a large group of Italian adults. Methods: This cross-sectional study involved 2665 adults (aged ≥ 18 years) who lead normal lives and underwent a routine lifestyle and dietary assessment at a clinical centre specialising in nutrition, metabolic health, and lifestyle counselling in Rome. Participants completed an online questionnaire on food preferences (19 foods) and sports practice. Multivariate logistic regression models, adjusted for age, sex, and smoking, were used to assess associations. Results: Sports participation was defined as engaging in structured physical activity at least once per week and was reported by 53.5% of subjects (men: 60.1%; women: 49.0%; p < 0.0001). After adjustment, active individuals were significantly more likely to prefer plant-based drinks, low-fat yoghurt, fish, cooked and raw vegetables, fruit, whole grains, tofu, and dark chocolate (all p < 0.05) and less likely to prefer cow’s milk (p = 0.018). Among sport participants, males were more likely to prefer meat (general, white, red, processed) and eggs, while females preferred plant-based drinks. No significant gender differences were observed for dairy products, legumes, or fish. Differences in food preferences were also observed according to the type of sport, with bodybuilders showing higher preference for tofu and dark chocolate. The strongest associations were found in the 25–44 age group. Conclusions: Sports participation is independently associated with specific food preferences, characterised by greater preference for plant-based and fibre-rich foods, and gender differences in food choices persist even among active adults. These findings highlight the need to consider both sports participation and gender when designing nutritional interventions. Full article

AMPK-Targeting Effects of (−)-Epicatechin Gallate from Hibiscus sabdariffa Linne Leaves on Dual Modulation of Hepatic Lipid Accumulation and Glycogen Synthesis in an In Vitro Oleic Acid Model

Metabolic dysfunction-associated steatotic liver disease (MASLD) begins with hepatic lipid accumulation and triggers insulin resistance. Hibiscus leaf extract exhibits antioxidant and anti-atherosclerotic activities, and is rich in (−)-epicatechin gallate (ECG). Despite ECG’s well-known pharmacological activities and its total antioxidant capacity being stronger than that of other catechins, its regulatory effects on MASLD have not been fully described previously. Therefore, this study attempted to evaluate the anti-MASLD potential of ECG isolated from Hibiscus leaves on abnormal lipid and glucose metabolism in hepatocytes. First, oleic acid (OA) was used as an experimental model to induce lipid dysmetabolism in human primary hepatocytes. Treatment with ECG can significantly (p < 0.05) reduce the OA-induced cellular lipid accumulation. Nile red staining revealed, compared to the OA group, the inhibition percentages of 29, 61, and 82% at the tested doses of ECG, respectively. The beneficial effects of ECG were associated with the downregulation of SREBPs/HMGCR and upregulation of PPARα/CPT1 through targeting AMPK. Also, ECG at 0.4 µM produced a significant (p < 0.01) decrease in oxidative stress by 83%, and a marked (p < 0.05) increase in glycogen synthesis by 145% on the OA-exposed hepatocytes with insulin signaling blockade. Mechanistic assays indicated lipid and glucose metabolic homeostasis of ECG might be mediated via regulation of lipogenesis, fatty acid β-oxidation, and insulin resistance, as confirmed by an AMPK inhibitor. These results suggest ECG is a dual modulator of lipid and carbohydrate dysmetabolism in hepatocytes. Full article

Background/Objectives: Possessing red and white ecotypes, and utilized in traditional Guyanese medicine, Doliocarpus dentatus’ red ecotype is preferred locally for its purported superior therapeutic efficacy. Although therapeutic metabolites were detected in D. dentatus previously, phytohormones remain largely unexplored, until now. Cytokinins, phytohormones responsible for plant cell division, growth and differentiation, are gaining traction for their therapeutic potential in human health. This study screened and quantified endogenous cytokinins and correlated detected cytokinins with selected secondary metabolites. Methods: Liquid chromatography–mass spectrometry was used to acquire phytohormone and metabolite data. Bioinformatics tools were used to assess untargeted metabolomics datasets via statistical and pathway analyses, and chemical groupings of putative metabolites. Results: In total, 20 of the 35 phytohormones were detected and quantified in both ecotypes, with the red ecotype displaying higher free base and glucoside cytokinin concentrations and exhibited 6.2 times the total CK content when compared to the white ecotype. Pathway analysis revealed flavonoid and monoterpenoid biosynthesis in red and white ecotypes, respectively. Positive correlations between specific cytokinins and alkaloids, and between trans-Zeatin and isopentenyladenosine riboside with phenolic compounds were observed. Conclusions: These results suggest that the red ecotype’s elevated cytokinin levels coupled with flavonoid biosynthesis enrichment support its preference in Guyanese traditional medicine. Red deer fat makes up approximately 7–10% of the animal’s weight and is not currently used. Regarding sustainability in the food industry, it is desirable to look for opportunities for its processing and use, not only in the food industry. The aim of this study is the enzymatic modification of red deer fat, leading to modification of its physicochemical properties, and the study of changes in phase transitions of modified fat, its structure, color, and texture. Hydrolysis was performed using sn-1,3-specific lipase at different water concentrations (10–30%) and reaction times (2–6 h). The results showed that there was a significant decrease in melting and crystallization temperatures with an increasing degree of hydrolysis, which was confirmed by differential scanning calorimetry. FTIR spectra revealed a decrease in the intensity of the ester bonds, indicating cleavage of triacylglycerols. Texture analysis of the modified fats confirmed a decrease in hardness of up to 50% and an increase in spreadability. The color parameter values remained within an acceptable range. The results show that enzymatic modification is an effective tool for targeted modification of red deer fat properties, and this expands the possibilities of its application in cosmetic matrices and food applications as functional lipids. In this work, lightweight aggregates (LWAs) were prepared from an Italian red clay, pumice scraps, and spent coffee grounds. Chemical and physical characterization was first performed on the raw materials and then on the finished products. By studying the thermal behavior of the materials, the correct firing temperature was evaluated. The obtained aggregates were fired in two different modes: in a rotary kiln and in a static kiln; the influence of the firing processes on the finished products was assessed. This study can be useful for industrially scaling up this process. Firing in a rotary kiln reduced the average diameter of the aggregates (negative expansion index), resulting in a higher compressive strength and dry particle density compared to an aggregate containing only clay. The pH and electrical conductivity values address their use in agronomy without causing problems to crops, while the higher compressive strength, density, and porosity values could allow their use in construction. The Effects of the Red River Jig on the Wholistic Health of Adults in Saskatchewan

The Red River Jig is a traditional Métis dance practiced among Indigenous and non-Indigenous Peoples. While exercise improves physical health and fitness, the impacts of cultural dances on wholistic health are less clear. This study aimed to investigate the psychosocial (cultural and mental), social, physical function, and physical fitness benefits of a Red River Jig intervention. In partnership with Li Toneur Nimiyitoohk Métis Dance Group, Indigenous and non-Indigenous adults (N = 40, 39 ± 15 years, 32 females) completed an 8-week Red River Jig intervention. Social support, cultural identity, memory, and mental wellbeing questionnaires, seated blood pressure and heart rate, weight, pulse-wave velocity, heart rate variability, baroreceptor sensitivity, jump height, sit-and-reach flexibility, one-leg and tandem balance, and six-minute walk test were assessed pre- and post-intervention. Community, family, and friend support scores, six-minute walk distance (553.0 ± 88.7 m vs. 602.2 ± 138.6 m, p = 0.002), jump, leg power, and systolic blood pressure low-to-high-frequency ratio increased after the intervention. Ethnic identity remained the same while affirmation and belonging declined, leading to declines in overall cultural identity, as learning about Métis culture through the Red River Jig may highlight gaps in cultural knowledge. Seated systolic blood pressure (116.5 ± 7.3 mmHg vs. 112.5 ± 10.7 mmHg, p = 0.01) and lower peripheral pulse-wave velocity (10.0 ± 2.0 m·s−1 vs. 9.4 ± 1.9 m·s−1, p = 0.04) decreased after the intervention. Red River Jig dance training can improve social support, physical function, and physical fitness for Indigenous and non-Indigenous adults. Full article

Cyborg insects offer a biologically powered solution for locomotion control, but conventional methods typically rely on invasive electrical stimulation. Here, we introduce a noninvasive, phototaxis-based strategy to steer walking Endebius florensis beetles using light-emitting diode (LED) stimuli. Electroretinogram recordings revealed spectral sensitivity to blue, green, and yellow light, with reduced response to red. Behavioral assays demonstrated robust positive phototaxis to blue light and negative phototaxis to yellow. Using these findings, we built a wireless microcontroller-based backpack emitting directional blue light to induce steering. The beetles reliably turned toward the activated light, achieving angular deflections over 60° within seconds. This approach enables repeatable, trauma-free insect control and establishes a new paradigm for biohybrid locomotion systems. Full article

Phenology-Aware Machine Learning Framework for Chlorophyll Estimation in Cotton Using Hyperspectral Reflectance

Accurate and non-destructive monitoring of leaf chlorophyll content (LCC) is essential for assessing crop photosynthetic activity and nitrogen status in precision agriculture. This study introduces a phenology-aware machine learning framework that combines hyperspectral reflectance data with various regression models to estimate leaf chlorophyll content (LCC) in cotton at six key reproductive stages. Field experiments utilized synchronized spectral and SPAD measurements, incorporating spectral transformations—such as vegetation indices (VIs), first-order derivatives, and trilateration edge parameters (TEPs, a new set of geometric metrics for red-edge characterization)—for evaluation. Five regression approaches were evaluated, including univariate and multivariate linear models, along with three machine learning algorithms: Random Forest, K-Nearest Neighbor, and Support Vector Regression. Random Forest consistently outperformed the other models, achieving the highest R2 (0.85) and the lowest RMSE (4.1) during the bud stage. Notably, the optimal prediction accuracy was achieved with fewer than five spectral features. P3MA: A Promising Mycobacteriophage Infecting Mycobacterium abscessus

Background/Objectives: Mycobacterium abscessus is an opportunistic pathogen causing infections mainly in patients with immunosuppression and chronic pulmonary pathologies. Extended treatment periods are needed to tackle this pathogen, bacterial eradication is rare, and recurrence can take place with time. New alternative treatments are being investigated, such as bacteriophage therapy. This work describes the characterization of the mycobacteriophage P3MA, showing its ability to infect clinical and standard M. abscessus strains. Methods: Phylogenetic analysis, electron microscopy, growth curves, biofilm assays, checkerboard, and granuloma-like medium studies were performed. Results: P3MA inhibited the growth of clinical samples in both planktonic and biofilm states as well as in a granuloma-like model. The study of the interaction with antibiotics revealed that P3MA exhibited an antagonistic effect combined with clarithromycin, indifference with amikacin, and synergy with imipenem. Conclusions: All these results suggest that, after genetic engineering, P3MA could be a promising candidate for phage therapy in combination with imipenem, including lung infections. Full article

Corn is one of the most consumed cereals in the Mexican diet. In this country, there are multiple varieties that exhibit nutraceutical potential due to their content of different metabolites with biological activity, such as blue corn. Another variety that has received little study is the red pigmented corn variety Chilac from Puebla, Mexico, which is being studied for its nutraceutical potential. A differential extraction using the Soxhlet method was carried out to evaluate the phenolic content, total flavonoid content, and monomeric anthocyanins, and free radical scavenging test was performed using the DPPH reagent. A proximate analysis was also conducted to identify the main macronutrients. The results of the proximate analysis were comparable to those of other traditional corn varieties, with carbohydrates being the macronutrient present in the highest amount at 77.9%. Regarding phenolic content and the presence of anthocyanins, the best extractions were obtained using alcoholic solvents; for example, ethanol for phenols, yielding 1368.420 ± 104.094 mg of gallic acid equivalents (GAE)/kg plant. In contrast, the flavonoid content was higher in the aqueous extract, with 833.984 ± 65.218 mg QE/Kg. In the case of the DPPH assay, the best result was obtained with ethyl acetate (73.81 ± 5.31%). These findings provide a foundation for expanding the use of corn varieties with nutraceutical potential, opening the possibility of studies focused on deeper characterization. Full article

Micronutrients play a prominent role in plant growth and development, and their bioavailability is a growing global concern. Zinc is one of the most important micronutrients in the plant life cycle, acting as a metallic cofactor for numerous biochemical reactions within plant cells. Zinc deficiency in plants leads to various physiological abnormalities, ultimately affecting nutritional quality and posing challenges to food security. Biofortification methods have been adopted by agronomists to increase Zn concentrations in crops through optimal foliar and soil applications. Changing climatic conditions and conventional agricultural practices alter edaphic factors, reducing zinc bioavailability in soils due to abrupt weather changes. Precision agriculture emphasizes need-based and site-specific technologies to address these nutritional deficiencies. Nanoscience, a multidimensional approach, reduces particle size to the nanometer (nm) scale to enhance their efficiency in precise amounts. Nanoscale forms of Zn+2 and their broad applications across crops are gaining attention in agriculture under varied application methods. This review focuses on the significance of Zn oxide (ZnO) nanoparticles (ZnONPs) and their extensive application in crop production. We also discuss optimum dosage levels, ZnONPs synthesis, application methods, toxicity, and promising future strategies in this field. Full article

In this work, the structures of the isolated anion and anhydrous and monohydrated sodium salts of alizarin red S (ARS) have been theoretically investigated within the density functional theory framework (B3LYP/6-311++G** calculations). The combination of calculations with the scaled quantum mechanics force field (SQMFF) methodology has allowed the assignment of the experimental infrared spectrum of ARS in the solid phase and the determination of the corresponding force constants. The structural analysis also included the investigation of the NMR and UV-visible spectra of the compound in solution in light of the undertaken quantum chemical calculations, the obtained theoretical data being in good agreement with the corresponding experimental ones. The impact of the presence of the Na+ counterion and hydration water on the properties of the organic ARS fragment was evaluated. Atoms in molecules theory (AIM) analysis was also undertaken to obtain further details on the electronic structure of the investigated species, and the HOMO-LUMO gap was determined to evaluate their relative reactivity. Globally, the results obtained in this work extend the available information on alizarin red S and may also be used for the fast identification of the three studied species of the compound investigated (anhydrous and monohydrated sodium salts and isolated anion).
