This study investigates the combined effect of electrodeposited gold nanoparticles (AuNPs) and AuNP–polypyrrole (PPy)-modified Saccharomyces cerevisiae on electrochemical glucose sensing. AuNPs were deposited onto electrode surfaces by cyclic voltammetry, and the resulting interfaces were characterized using atomic force microscopy, cyclic voltammetry, and electrochemical impedance spectroscopy. AFM analysis confirmed increased surface roughness and height variability after deposition, indicating substantial restructuring of the electrode interface. Electrochemical measurements showed that AuNP deposition altered interfacial charge storage and transfer and increased the measured charge-transfer resistance. Glucose sensing was evaluated in a ferricyanide-mediated system using yeast layers with or without AuNP and PPy modification over a 0–60 mM concentration range. All configurations exhibited saturating, non-linear glucose responses described by Hill fitting. Among the evaluated yeast-modified electrodes, the AuNP–PPy modified yeast produced the strongest glucose-induced current increase and the best low-concentration performance, achieving a limit of detection of 0.540 mM, compared with 1.016 mM and 1.330 mM for single-modified layers and 3.360 mM for unmodified yeast. These results show that combining AuNP electrodeposition with AuNP–PPy yeast modification improves interfacial properties and enhances mediator-assisted electrochemical glucose sensing. Full article

This study investigates microbial fuel cell (MFC) performance through the modification of Saccharomyces cerevisiae with gold nanoparticles (AuNPs) and polypyrrole (PPy). The yeast/AuNP-modified electrodes generated the highest median current of 2.57 nA, significantly outperforming the yeast/PPy-modified (0.82 nA) electrodes. Power density measurements further confirmed the superior performance of the yeast/AuNP-modified electrodes, showcasing a notable improvement in current densities and power outputs. Yeast/AuNP-modified graphite electrodes produced a higher power density of 22.8 mW/m2, while exhibiting a lower current density compared to electrodes modified solely with yeast, which achieved a power density of 5.7 mW/m2. These findings highlight the potential of AuNPs in significantly enhancing the electrochemical performance of yeast-based MFCs, providing a promising approach for the development of more efficient bioelectrochemical systems. Gold nanoparticles–cuprous oxide/reduced graphene oxide/polypyrrole (AuNPs-Cu2O/rGO/PPy) hybrid nanocomposites were synthesized for surface acoustic wave (SAW) sensors, achieving high sensitivity (2 Hz/ppb), selectivity, and fast response (~2 min) at room temperature. The films, deposited via spin-coating, were characterized by SEM, EDS, and XRD, revealing a rough, wrinkled morphology beneficial for gas adsorption. The sensor showed significant frequency shifts to NH3, enhanced by AuNPs, Cu2O, rGO, and PPy. It had a 6.4-fold stronger response to NH3 compared to CO2, H2, and CO, confirming excellent selectivity. The linear detection range was 12–1000 ppb, with a limit of detection (LOD) of 8 ppb. Humidity affected performance, causing negative frequency shifts, and sensitivity declined after 30 days due to resistivity changes. Despite this, the sensor demonstrated excellent NH3 selectivity and stability across multiple cycles. In simulated breath tests, it distinguished between healthy and patient-like samples, highlighting its potential as a reliable, non-invasive diagnostic tool. Full article

Dopamine (DA) is a neurotransmitter that works in the brain. It plays several important roles in executive functions, including motor control, memory, mood, motivation, and reward. DA imbalances are associated with diseases in the nervous system such as Parkinson’s disease, schizophrenia, Alzheimer’s disease, and attention deficit hyperactivity disorder (ADHD). Therefore, the development of a biosensor for the detection of precise amounts of DA is of great interest. In this research, polypyrrole-3-carboxylic acid/polypyrrole/gold nanoparticle (PP3C/PPy/AuNPs) composites were developed for the electrochemical detection of DA. Firstly, a PP3C/PPy/AuNPs composite thin film was synthesized by electropolymerization on a fluorine-doped tin oxide (FTO)-coated glass substrate. Subsequently, cyclic voltammetry (CV), scanning electron microscopy (SEM), and differential pulse voltammetry (DPV) were used for the characterization and study of the efficiency of the obtained conducting polymer–gold nanoparticle composite thin film for the detection of DA. The proposed electrochemical sensor showed good sensitivity and selectivity for the detection of DA with a wide detection linear range from 5 to 180 μM (R2 = 0.9913). The limit of detection (LOD) and limit of quantitation (LOQ) values were 9.72 nM and 0.032 μM, respectively. Therefore, it can be concluded that the electrochemically fabricated PP3C/PPy/AuNPs composite thin film can be applied as an electrochemical biosensor for the detection of dopamine for the early diagnosis of various neurological disorders in the future. Microbial fuel cells (MFCs) are a candidate for green energy sources due to microbes’ ability to generate charge in their metabolic processes. The main problem in MFCs is slow charge transfer between microorganisms and electrodes. Several methods to improve charge transfer have been used until now: modification of microorganisms by conductive polymers, use of lipophilic mediators, and conductive nanomaterials. We created an MFC with a graphite anode, covering it with 9,10-phenatrenequinone and polypyrrole-modified Saccharomyces cerevisiae with and without 10 nm sphere gold nanoparticles. The MFC was evaluated using cyclic voltammetry and power density measurements. The peak current from cyclic voltammetry measurements increased from 3.76 mA/cm2 to 5.01 mA/cm2 with bare and polypyrrole-modified yeast, respectively. The MFC with polypyrrole- and nanoparticle-modified yeast reached a maximum power density of 150 mW/m2 in PBS with 20 mM Fe(III) and 20 mM glucose, using a load of 10 kΩ. The same MFC with the same load in wastewater reached 179.2 mW/m2. These results suggest that this MFC configuration can be used to improve charge transfer. Full article

This study addresses the necessity to monitor the presence of glyphosate (Gly) in waters, highlighting the need for on-site detection of Gly by using electrochemical sensors in environmental and agricultural monitoring programs. Two approaches were employed: (1) modification with graphene decorated with gold nanoparticles (AuNPs-Gr) and dispersed in either dimethylformamide (DMF) or a solution containing Nafion and isopropanol (NAF), and (2) molecularly imprinted polymers (MIPs) based on polypyrrole (PPy) deposited on gold SPEs (AuSPE). Electrochemical characterization revealed that sensors made of AuNPs-Gr/SPCE exhibited enhanced conductivity, larger active area, and improved charge transfer kinetics compared to unmodified SPEs and SPEs modified with graphene alone. However, the indirect detection mechanism of Gly via complex formation with metallic cations in AuNPs-Gr-based sensors introduces complexities and compromises sensitivity and selectivity. In contrast, MIPPy/AuSPE sensors demonstrated superior performance, offering enhanced reliability and sensitivity for Gly analysis. The MIPPy/AuSPE sensor allowed the detection of Gly concentrations as low as 5 ng/L, with excellent selectivity and reproducibility. Moreover, testing in real surface water samples from the Olt River in Romania showed recovery rates ranging from 90% to 99%, highlighting the effectiveness of the detection method. Future perspectives include expanding the investigation to monitor Gly decomposition in aquatic environments over time, providing insights into the decomposition’s long-term effects on water quality and ecosystem health, and modifying regulatory measures and agricultural practices for mitigating its impact. This research contributes to the development of robust and reliable electrochemical sensors for on-site monitoring of Glyphosate in environmental and agricultural settings. Full article

This paper describes the study of an amperometric glucose biosensor based on an enzymatic biofuel cell consisting of a bioanode and a biocathode modified with the same enzyme—glucose oxidase (GOx). A graphite rod electrode (GRE) was electrochemically modified with a layer of Prussian blue (PB) nanoparticles embedded in a poly(pyrrole-2-carboxylic acid) (PPCA) shell, and an additional layer of PPCA and was used as the cathode. A GRE modified with a nanocomposite composed of poly(1,10-phenanthroline-5,6-dione) (PPD) and gold nanoparticles (AuNPs) entrapped in a PPCA shell was used as an anode. Both electrodes were modified with GOx by covalently bonding the enzyme to the carboxyl groups of PPCA. The developed biosensor exhibited a wide linear range of 0.15–124.00 mM with an R2 of 0.9998 and a sensitivity of 0.16 μA/mM. The limit of detection (LOD) and quantification (LOQ) were found to be 0.07 and 0.23 mM, respectively. The biosensor demonstrated exceptional selectivity to glucose and operational stability throughout 35 days, as well as good reproducibility, repeatability, and anti-interference ability towards common interfering substances. The studies on human serum demonstrate the ability of the newly designed biosensor to determine glucose in complex real samples at clinically relevant concentrations. Full article

Exhaled human breath analysis has great potential for the diagnosis of diseases in non-invasive way. The 13C-Urea breath test for the diagnosis of Helicobacter pylori infection indicates the ammonia concentration of 50–400 ppb in the breath. This work successfully developed a surface acoustic wave (SAW) resonator based on graphene/polypyrrole composite films decorated by gold nanoparticles (AuNPs–G/PPy) with sensitivity and selectivity to detect ammonia in parts-per-billion concentrations, which is promising for the accurate diagnosis of H. pylori infection. XRD, EDS, and SEM characterized the AuNPs–G/PPy nanocomposites, providing comprehensive insights into their structural, compositional, and morphological properties. The gas-sensing capabilities of the fabricated SAW sensors were extensively investigated, focusing on their response to NH3 gas at ambient temperature. The concentration of ammonia gas was effectively quantified by monitoring the frequency shift of the SAW device. Notably, our developed SAW sensor demonstrated outstanding sensitivity, selectivity, repeatability, and reproducibility for 50–1000 ppb NH3 in dry air. The excellent sensing performance of the AuNPs–G/PPy hybrid composite film can be attributed to the synergistic effects of graphene’s superior conductivity, the catalytic properties of gold nanoparticles, and the conductivity sensitization facilitated by electron-hole recombination on the polypyrrole surface. Bone marrow mesenchymal stem cells (BMMSCs) possess a strong ability to differentiate into the chondrogenic lineage, which is important for cartilage regeneration. External stimuli, such as electrical stimulation (ES), are frequently studied for chondrogenic differentiation of BMMSCs; however, the application of conductive polymers such as polypyrrole (Ppy), has never been used for stimulating BMMSCs chondrogenesis in vitro before. Thus, the aim of this study was to evaluate the chondrogenic potential of human BMMSCs after stimulation with Ppy nanoparticles (Ppy NPs) and compare them to cartilage-derived chondrocytes. In this study, we tested Ppy NPs without and with 13 nm gold NPs (Ppy/Au) for BMMSCs and chondrocyte proliferation, viability, and chondrogenic differentiation for 21 days, without the use of ES. The results demonstrated significantly higher amounts of cartilage oligomeric matrix protein (COMP) in BMMSCs stimulated with Ppy and Ppy/Au NPs, as compared to the control. The expression of chondrogenic genes (SOX9, ACAN, COL2A1) in BMMSCs and chondrocytes were upregulated by Ppy and Ppy/Au NPs, as compared to controls. Histological staining with safranin-O indicated higher extracellular matrix production in Ppy and Ppy/Au NPs stimulated samples, as compared to controls. In conclusion, Ppy and Ppy/Au NPs stimulate BMMSC chondrogenic differentiation; however, BMMSCs were more responsive to Ppy, while chondrocytes possessed a stronger chondrogenic response to Ppy/Au NPs. In this study, polypyrrole nanotubes (PPy-NT) and gold nanoparticles (AuNPs) were electrochemically synthesized to form a hybrid material and used as an electroactive layer for the attachment of proteins for the construction of a high-performance biosensor. Besides the enhancement of intrinsic conductivity of the PPy-NT, the AuNPs act as an anchor group for the formation of self-assembly monolayers (SAMs) from the gold–sulfur covalent interaction between gold and Mercaptopropionic acid (MPA). This material was used to evaluate the viability and performance of the platform developed for biosensing, and three different biological approaches were tested: first, the Avidin-HRP/Biotin couple and characterizations were made by using cyclic voltammetry (CV) and electrochemical impedance spectroscopy (EIS), wherein we detected Biotin in a linear range of 100–900 fmol L−1. The studies continued with folate group biomolecules, using the folate receptor α (FR-α) as a bioreceptor. Tests with anti-FR antibody detection were performed, and the results obtained indicate a linear range of detection from 0.001 to 6.70 pmol L−1. The same FR-α receptor was used for Folic Acid detection, and the results showed a limit of detection of 0.030 nmol L−1 and a limit of quantification of 90 pmol L−1. The results indicate that the proposed biosensor is sensitive and capable of operating in a range of clinical interests. Full article

This study reports a new protocol for the management of Hidradenitis Suppurativa (HS), depending on the synergistic photodynamic and photothermal effect of eosin yellow-gold-polypyrrole hybrid nanoparticles (E-G-Ppy NPs). E-G-Ppy NPs and gold-polypyrrole NPs (G-Ppy NPs) were synthesized, characterized, and formulated in topical hydrogels. Then, in vivo trans-epidermal permeation study, under both dark and white light-irradiation conditions, was done on albino mice. The E-G-Ppy hydrogel was then applied on a twenty-four years old female with recurrent axillary HS lesions pretreated with fractional CO2 laser. Thereafter, the treated lesions were irradiated sequentially, using an IPL system, in the visible (~550 nm) and NIR band (630–1100 nm) to activate the synthesized nanoparticles. Results showed that, upon application to mice skin, E-G-Ppy exhibited good tolerance and safety under dark conditions and induced degenerative changes into dermal layers after white-light activation, reflecting deep penetration. Photo-activation of E-G-Ppy hydrogel to a severe Hidradenitis Suppurativa case showed an improvement of 80% of the lesions according to average HS-LASI scores after 4 sessions with no recurrence during a follow-up period of six months. In summary, the dual photodynamic/photothermal activation of E-G-Ppy NPs can represent a promising modality for management of HS. Further expanded clinical studies may be needed. Full article

Polypyrrole (PPy) is an interesting conducting polymer due to its good environmental stability, high conductivity, and biocompatibility. The association between PPy and metallic nanoparticles has been widely studied since it enhances electrochemical properties. Polypyrrole (PPy) is an interesting conducting polymer due to its good environmental stability, high conductivity, and biocompatibility. The association between PPy and metallic nanoparticles has been widely studied since it enhances electrochemical properties. In this context, gold ions are reduced to gold nanoparticles (AuNPs) directly on the polymer surface as PPy can be oxidized to an overoxidized state. This work proposes the PPy electrochemical synthesis followed by the direct reduction of gold on its surface in a fast reaction. The modified electrodes were characterized by electronic microscopic and infrared spectroscopy. The effect of reduction time on the electrochemical properties was evaluated by the electrocatalytic properties of the obtained material from the oxidation of ascorbic acid (AA) and electrochemical impedance spectroscopy studies. The presence of AuNPs improved the AA electrocatalysis by reducing oxidation potential and lowering charge transfer resistance. EIS data were fitted using a transmission line model. The results indicated an increase in the electronic transport of the polymeric film in the presence of AuNPs. However, PPy overoxidation occurs when the AuNPs’ deposition is higher than 30 s. In PPy/AuNPs 15 s, smaller and less agglomerated particles were formed with fewer PPy overoxidized, confirming the observed electrocatalytic behavior. Full article

A new electrochemical DNA biosensor based on mercaptopropionic acid (MPA)-capped ZnS quantum dots (MPA-ZnS QDs) immobilization matrix for covalent binding with 20-base aminated oligonucleotide has been successfully developed. Prior to the modification, screen-printed carbon paste electrode (SPE) was self-assembled with multilayer gold nanoparticles (AuNPs) and cysteamine (Cys). The inclusion of MPA-ZnS QDs semiconducting material in modified electrodes has enhanced the electron transfer between the SPE transducer and DNA leading to improved bioanalytical assay of target biomolecules. Electrochemical studies performed by cyclic voltammetry (CV) and differential pulsed voltammetry (DPV) demonstrated that the MPA-ZnS QDs modified AuNPs electrode was able to produce a lower charge transfer resistance response and hence higher electrical current response. Under optimal conditions, the immobilized synthetic DNA probe exhibited high selectivity towards synthetic target DNA. Based on the DPV response of the reduction of anthraquinone monosulphonic acid (AQMS) redox probe, the MPA-ZnS QDs-based electrochemical DNA biosensor responded to target DNA concentration from 1 × 10−9 μM to 1 × 10−3 μM with a sensitivity 1.2884 ± 0.12 µA, linear correlation coefficient (R2) of 0.9848 and limit of detection (LOD) of 1 × 10−11 μM target DNA. The DNA biosensor exhibited satisfactory reproducibility with an average relative standard deviation (RSD) of 7.4%. The proposed electrochemical transducer substrate has been employed to immobilize the aminated Arowana fish (Scleropages formosus) DNA probe. The DNA biosensor showed linearity to target DNA from 1 × 1011 to 1 × 106 µM (R2 = 0.9785) with sensitivity 1.1251 ± 0.243 µA and LOD of 1 × 1011 µM. The biosensor has been successfully used to determine the gender of Arowana fish without incorporating toxic raw materials previously employed in the hazardous processing conditions of polypyrrole chemical conducting polymer, whereby the cleaning step becomes difficult with thicker films due to high levels of toxic residues from the decrease in polymerization efficacy as films grew. Bone injuries and fractures are often associated with post-surgical failures, extended healing times, infection, a lack of return to a normal active lifestyle, and corrosion associated allergies. In this regard, this review presents a comprehensive report on advances in nanotechnology driven solutions for bone tissue engineering. The fabrication of metals such as copper, gold, platinum, palladium, silver, strontium, titanium, zinc oxide, and magnetic nanoparticles with tunable physico-chemical and opto-electronic properties for osteogenic scaffolds is discussed here in detail. Furthermore, the rational selection of a polymeric base such as chitosan, collagen, poly (L-lactide), hydroxyl-propyl-methyl cellulose, poly-lactic-co-glycolic acid, polyglucose-sorbitol-carboxymethy ether, polycaprolactone, natural rubber latex, and silk fibroin for scaffold preparation is also discussed. These advanced materials and fabrication strategies not only provide for appropriate mechanical strength but also render integrity, making them appealing for orthopedic applications. Further, such scaffolds can be functionalized with ligands or biomolecules such as hydroxyapatite, polypyrrole (PPy), magnesium, zinc dopants, and growth factors to stimulate osteogenic differentiation, mineralization, and neovascularization to aid in rapid healing. Future directions to co-incorporate bioceramics, biogenic nanoparticles, and fourth generation biomaterials to enhance biocompatibility, mechanical properties, and rapid recovery are also included in this review. Hence, the further development of such biomimetic metal-based nano-scaffolds at a lower cost with reduced risks and greater efficacy at regrowing bone can revolutionize the future of orthopedics. Biosensors for the determination of glucose concentration have a great significance in clinical diagnosis, and in the food and pharmaceutics industries. In this research, short-chain polyaniline (PANI) and polypyrrole (Ppy)-based nanocomposites with glucose oxidase (GOx) and 6 nm diameter AuNPs (AuNPs(6 nm)) were deposited on the graphite rod (GR) electrode followed by the immobilization of GOx. Optimal conditions for the modification of GR electrodes by conducting polymer-based nanocomposites and GOx were elaborated. The electrodes were investigated by cyclic voltammetry and constant potential amperometry in the presence of the redox mediator phenazine methosulfate (PMS). The improved enzymatic biosensors based on GR/PANI-AuNPs(6 nm)-GOx/GOx and GR/Ppy-AuNPs(6 nm)-GOx/GOx electrodes were characterized by high sensitivity (65.4 and 55.4 μA mM−1 cm−2), low limit of detection (0.070 and 0.071 mmol L−1), wide linear range (up to 16.5 mmol L−1), good repeatability (RSD 4.67 and 5.89%), and appropriate stability (half-life period (τ1/2) was 22 and 17 days, respectively). The excellent anti-interference ability to ascorbic and uric acids and successful practical application for glucose determination in serum samples was presented for GR/PANI-AuNPs(6 nm)-GOx/GOx electrode.
