Common bean (Phaseolus vulgaris L.) is a critical protein-rich legume supporting food and nutritional security globally. However, Fusarium wilt, caused by Fusarium solani, remains a major constraint to production, with yield losses reaching up to 84%. While biocontrol strategies have been explored, most microbial agents are sourced from mesophilic environments and show limited effectiveness under abiotic stress. Here, we report the isolation and characterization of extremophilic Bacillus spp. from the hypersaline Lake Bogoria, Kenya, and their biocontrol potential against F. solani. From 30 isolates obtained via serial dilution, 9 exhibited antagonistic activity in vitro, with mycelial inhibition ranging from 1.07–1.93 cm 16S rRNA sequencing revealed taxonomic diversity within the Bacillus genus, including unique extremotolerant strains. Molecular screening identified genes associated with the biosynthesis of antifungal metabolites such as 2,4-diacetylphloroglucinol, pyrrolnitrin, and hydrogen cyanide. Enzyme assays confirmed substantial production of chitinase (1.33–3160 U/mL) and chitosanase (10.62–28.33 mm), supporting a cell wall-targeted antagonism mechanism. In planta assays with the lead isolate (B7) significantly reduced disease incidence (8–35%) and wilt severity (1–5 affected plants), while enhancing root colonization under pathogen pressure. These findings demonstrate that extremophile-derived Bacillus spp. possess robust antifungal traits and highlight their potential as climate-resilient biocontrol agents for sustainable bean production in arid and semi-arid agroecosystems. The purpose of this work was to obtain specialized enrichment cultures from an original extreme acidophilic consortium of extremely acidophilic microorganisms and to study their microbial community composition and biotechnological potential. At temperatures of 25, 35, 40 and 50 °C, distinct enrichments of extremely acidophilic microorganisms used in the processes of bioleaching sulfide ores were obtained using nutrient media containing ferrous sulfate, elemental sulfur and a copper sulfide concentrate as nutrient inorganic substrates, with and without the addition of 0.02% yeast extract. The microbial community composition was studied using the sequencing of the V3–V4 hypervariable region of the 16S rRNA genes. The different growth conditions led to changes in the microbial composition and relative abundance of mesophilic and moderately thermophilic, strict autotrophic and mixotrophic microorganisms in members of the genera Acidithiobacillus, Sulfobacillus, Leptospirillum, Acidibacillus, Ferroplasma and Cuniculiplasma. The dynamics of the oxidation of ferrous iron, sulfur, and sulfide minerals (pyrite and chalcopyrite) by the enrichments was also studied in the temperature range of 25 to 50 °C. The study of enrichment cultures using the molecular biological method using the metabarcoding method of variable V3–24 V4 fragments of 16S rRNA genes showed that enrichment cultures obtained under different conditions differed in composition, which can be explained by differences in the physiological properties of the identified microorganisms. Regarding the dynamics of the oxidation of ferrous ions, sulfur, and sulfide minerals (pyrite and chalcopyrite), each enrichment culture was studied at a temperature range of 25 to 50 °C and indicated that all obtained enrichments were capable of oxidizing ferrous iron, sulfur and minerals at different rates. Reducing post-harvest losses of cereal crops is a key challenge for ensuring global food security amid the limited arable land and growing population. This study investigates the effectiveness of electron beam irradiation (5 MeV, ILU-10 accelerator) as a physical decontamination method for various cereal crops cultivated in Kazakhstan. Samples were irradiated at doses ranging from 1 to 5 kGy, and microbiological indicators—including Quantity of Mesophilic Aerobic and Facultative Anaerobic Microorganisms (QMAFAnM), yeasts, and molds—were quantified according to national standards. Experimental results demonstrated an exponential decline in microbial contamination, with a >99% reduction achieved at doses of 4–5 kGy. The modeled inactivation kinetics showed strong agreement with the experimental data: R2 = 0.995 for QMAFAnM and R2 = 0.948 for mold, confirming the reliability of the exponential decay models. Additionally, key quality parameters—including protein content, moisture, and gluten—were evaluated post-irradiation. The results showed that protein levels remained largely stable across all doses, while slight but statistically insignificant fluctuations were observed in moisture and gluten contents. Principal component analysis and scatterplot matrix visualization confirmed clustering patterns related to radiation dose and crop type. The findings substantiate the feasibility of electron beam treatment as a scalable and safe technology for improving the microbiological quality and storage stability of cereal crops. Full article

Tryptophan is synthesized in microorganisms via a five-step enzymatic pathway originating from chorismate, which is a product of the shikimate pathway. As a biosynthetic precursor to a wide range of high-value compounds such as indole-3-acetic acid, indigo, indirubin, and violacein, this pathway has been a central target for metabolic engineering to enhance microbial production. Anthranilate phosphoribosyltransferase (AnPRT) catalyzes the second step of the pathway by transferring a phosphoribosyl group from PRPP to anthranilate, forming phosphoribosyl anthranilate (PRA). AnPRT, the sole member of class IV phosphoribosyltransferases, adopts a unique fold and functions as a homodimer. While the structural basis of AnPRT activity has been elucidated in several organisms, thermostable variants remain underexplored despite their relevance for high-temperature bioprocessing. In this study, the crystal structure of AnPRT from the thermophilic archaeon Methanocaldococcus jannaschii (MjAnPRT) was determined at a 2.16 Å resolution. The enzyme exhibits a conserved dimeric architecture and key catalytic motifs. Comparative structural analysis with mesophilic and hyper thermophilic homologs revealed that MjAnPRT possesses enhanced local stability in catalytically important regions and strengthened inter-subunit interactions. These features likely contribute to its thermostability and provide a valuable framework for the rational design of robust AnPRTs for industrial and synthetic biology applications. Full article

In this study, the effects of graphene-based nanomaterials—specifically graphene nanoplatelets (GNPs) and graphene oxide (GO) nanosheets—on methane (CH4) production during anaerobic digestion (AD) of thermally hydrolyzed sewage sludge were investigated. Anaerobic digestion was carried out over a 40-day period under mesophilic conditions in batch digesters with a volume of 2.65 L. The influence of various dosages of GNPs and GO nanosheets on methane yields was assessed, including a comparison between GNPs with different specific surface areas (320 m2/g and 530 m2/g). The highest CH4 yield (194 mL/g-VSadded) was observed with a GNP dosage of 5 mg/g-TS and a surface area of 530 m2/g, showing an increase of 3.08% compared to the control. This treatment group had the greatest positive effect also on the degradation of organic matter, with total solids (TS) and volatile solids (VS) removal reaching 34.35% and 44.18%, respectively. However, the GO dosages that significantly decreased cumulative CH4 production were determined to be 10–15 mg/g-TS. Graphene oxide at dosages of 10 and 15 mg/g-TS reduced specific cumulative CH4 yields by 4.03% and 5.85%, respectively, compared to the control, indicating CH4 yield inhibition. This lab-scale study highlights the potential for integrating GNPs into full-scale, continuously operated wastewater treatment anaerobic digesters for long-term use in future applications. This study aims to evaluate the effect of adding a chitosan/perlite (Ch/P) carrier to anaerobic digestion (AD) on the efficiency and kinetics of the process, as well as the directional changes in the bacterial microbiome. A carrier with this composition was applied in the AD process for the first time. A laboratory experiment using wafer waste (WF) and cheese (CE) waste was conducted under mesophilic conditions. The analysis of physico-chemical properties confirmed the suitability of the tested carrier material for anaerobic digestion. Both components influenced the microstructural characteristics of the carrier: perlite contributed to the development of specific surface area, while chitosan determined the porosity of the system. Using next-generation sequencing (NGS), the study examined how the additive affected the genetic diversity of bacterial communities. Fourier-transform infrared spectroscopy (FTIR) revealed that the degradation rate depended on both the carrier and the substrate type. Consequently, the presence of the carrier led to an increase in the volume of biogas and methane produced. The volume of methane for the wafer waste (WF–control) increased from 351.72 m3 Mg−1 (VS) to 410.74 m3 Mg−1 (VS), while for the cosubstrate sample (wafer and cheese, WFC–control), it increased from 476.84 m3 Mg−1 (VS) to 588.55 m3 Mg−1 (VS). Organic amendments provide a sustainable strategy to enhance soil quality in degraded environments while also helping to reduce greenhouse gas emissions, for example, by improving soil structure, minimizing the use of synthetic fertilizers, and promoting a green economy. This study assessed the comparative effects of two organic amendments—vermicompost leachate and biochar—on the performance of popcorn maize (Zea mays L. var. everta) cultivated in saline soil conditions. Four treatments were evaluated: T0 (Control), T1 (Vermicompost leachate), T2 (Biochar), and T3 (Vermicompost leachate + Biochar), each with 10 replicates arranged in a Completely Randomized Design (CRD). Although various soil physicochemical, microbiological, and agronomic parameters displayed no significant differences compared to the control, the application of biochar resulted in considerable improvements in soil total organic carbon, the microbial community (mesophilic aerobic bacteria, molds, and yeasts), and increased seed length and diameter. In contrast, vermicompost leachate alone negatively impacted plant growth, leading to decreases in leaf area, stem thickness, and grain yield. Specifically, grain yield declined by 46% with leachate alone and by 31% when combined with biochar, compared to the control. These findings emphasize the superior effectiveness of biochar over vermicompost leachate as a soil amendment under saline conditions and highlight the potential risks of widely applying compost teas in stressed soils. It is recommended to conduct site-specific assessments and screenings for phytotoxins and phytopathogens prior to use. Additionally, the combined application of leachate and biochar may not be advisable given the tested soil characteristics. Full article

Sewage sludge, characterized by its high organic matter and nutrient content, as well as the presence of microbial pathogens and other contaminants, requires proper management due to its significant generation rate. The table olive sector, which is highly significant in Spain as a global leader in production and export, generates various waste streams such the Organic Solid By-Products from Table Olive Processing (OSBTOP), which are mainly derived from the olive pit after the pitting process. The main aim of this study was to enhance the methane production performance of sewage sludge through co-digestion with OSBTOP as a co-substrate. Batch assays demonstrated that employing OSBTOP as a co-substrate increased methane content by 35–41% across all tested mixtures. While the highest methane yield was produced at a 40:60 (sludge:OSBTOP) ratio, a 60:40 mixture proved to be a more advantageous option for scale-up and practical application. This is attributed to factors such as the higher availability of sludge and its inherent buffering capacity, which counteracts the accumulation of volatile fatty acids and promotes process stability, thereby contributing to the study’s objective of significantly enhancing methane production from sewage sludge through co-digestion. In semi-continuous operation, methane yields in the co-digestion scenario exceeded those of mixed sludge digestion, showing a yield of 180 versus 120 LCH4−1 · kgVSadded−1, representing a 50% improvement. This study highlights the potential of anaerobic digestion as a strategy for valorizing OSBTOP, a by-product with no prior studies, while demonstrating that its co-digestion with sewage sludge enhances methane generation, offering a sustainable approach to organic waste treatment. Thermostable cellulases and xylanases have broad acceptance in food, feed, paper and pulp, and bioconversion of lignocellulosics. Thermophilic fungi serve as an excellent source of thermostable enzymes. This study characterized four endo-β-1,4-glucanases (two glycoside hydrolase (GH) family 5 and two GH7 members) and four endo-β-1,4-xylanases (two GH10 and two GH11 members) from thermophilic fungus Thielavia terrestris, along with one GH10 endo-β-1,4-xylanase each from thermophilic fungus Chaetomium thermophilum and mesophilic fungus Chaetomium globosum. Comparative analysis was conducted against three previously reported GH10 endoxylanases: two thermostable enzymes from the thermophilic fungus Humicola insolens and thermophilic bacterium Halalkalibacterium halodurans, and one mesophilic enzyme from model fungus Neurospora crassa. The GH10 xylanase TtXyn10C (Thite_2118148; UniProt G2R8T7) from T. terrestris demonstrated high thermostability and activity, with an optimal temperature of 80–85 °C. It retained over 60% of its activity after 2 h at 70 °C, maintained approximately 30% activity after 15 min at 80 °C, and showed nearly complete stability following 1 min of exposure to 95 °C. TtXyn10C exhibited specific activity toward beechwood xylan (1130 ± 15 U/mg) that exceeded xylanases from H. insolens and H. halodurans while being comparable to N. crassa xylanase activity. Furthermore, TtXyn10C maintained stability across a pH range of 3–9 and resisted trypsin digestion, indicating its broad applicability. The study expands understanding of enzymes from thermophilic fungi. The discovery of the TtXyn10C offers a new model for investigating the high activity-stability trade-off and structure-activity relationships critical for industrial enzymes. Full article

This study evaluated the efficacy of acclimated cow manure as a seed microbiome to enhance food waste hydrolysis. Anaerobic hydrolysis was performed on simulated food waste in a hydrolytic–acidogenic leach bed reactor (LBR) operated in batch mode under mesophilic conditions (35 °C) for 16 days. The acclimation process involved three sequential runs: Run-1 utilized 20% (w/w) cow manure as seed, Run-2 employed the digestate from Run-1 (day 5), and Run-3 used the digestate from Run-1 (day 10). Run-3 achieved 70.4% removal of volatile solids (VSs), surpassing Run-1 (47.1%) and Run-2 (57.1%). Compared with the first run, the production of chemical oxygen demand (COD) and total soluble products (TSPs) increased by 48.7% and 75.9%, respectively, in Run-3. The hydrolysis rate of proteins was 48.4% in Run-1, while an increase of 16.9% was achieved in Run-3 with the acclimatized consortium. A molecular analysis of the microbial community existing in the reactors of Run-2 and Run-3 indicated that the improvement in process performance was closely related to the selection and enrichment of specific hydrolytic–acidogenic bacteria in the reactor. A functional analysis showed that the gene copy numbers for pyruvate synthesis and fatty acid synthesis and metabolism pathways were higher in all bacterial species in Run-3 compared to in those of the other two runs, indicating improved capacity through acclimation in Run-3. The experimental results demonstrate that the hydrolysis of food waste can be enhanced through the acclimation of seed microbes from cow manure. Full article

Adequate treatment of the organic fraction of municipal solid waste (OFMSW) in co-digestion with sewage sludge (SS) through dark fermentation (DF) technologies has been widely studied and recognized. However, there is little experience with a high-solids approach, where practical and scalable conditions are established to lay the groundwork for further development of feasible industrial-scale projects. In this study, the biochemical hydrogen potential of OFMSW using a 7 L batch reactor at mesophilic conditions was evaluated. Parameters such as pH, redox potential, temperature, alkalinity, total solids, and substrate/inoculum ratio were adjusted and monitored. Biogas composition was analyzed by gas chromatography. The microbial characterization of SS and post-reaction percolate liquids was determined through metagenomics analyses. Results show a biohydrogen yield of 38.4 NmLH2/gVS OFMSW, which forms ~60% of the produced biogas. Aeration was proven to be an efficient inoculum pretreatment method, mainly to decrease the levels of methanogenic archaea and metabolic competition, and at the same time maintain the required total solid (TS) contents for high-solids conditions. The microbial community analysis reveals that biohydrogen production was carried out by specific anaerobic and aerobic bacteria, predominantly dominated by the phylum Firmicutes, including the genus Bacillus (44.63% of the total microbial community), Clostridium, Romboutsia, and the phylum Proteobacteria, with the genus Proteus. Effect of Lyophilised Sumac Extract on the Microbiological, Physicochemical, and Antioxidant Properties of Fresh Carrot Juice

The lyophilised sumac (Rhus coriaria L) extract (LSE), in amounts of 0.25, 0.5, 1.0, 1.25, and 1.5 g/100 ml of juice, was incorporated into carrot juice, and its properties were assessed after 24, 48, and 72 h. A product without the lyophilised sumac extract served as the control. The highest supplementation level enhanced the physicochemical characteristics of carrot juice, increasing carotenoid and polyphenolic contents by 22% and 70% on the first day. The LSE significantly boosted antioxidant activity, yielding over a tenfold increase, while reducing capacity was elevated more than sevenfold. LC-MS analysis confirmed the presence of bioactive compounds, such as chalcones, flavonols, flavones, and phenolic acids, further validating the extract’s functional potential. Acidity and redness exhibited a proportional increase with the rising concentrations of the additive used. Additionally, microbial growth, including aerobic mesophiles, yeasts, and moulds, was markedly suppressed. After 72 h, the total count of aerobic microorganisms and yeasts/mould was reduced by 5.64 log and 4.94 log, respectively, compared to the control. The lyophilised sumac extract, rich in valuable bioactive compounds with antioxidant properties, effectively preserved freshly pressed carrot juice, mitigating spoilage and extending its shelf life. This form of sumac serves as a sustainable beverage additive, minimises food waste, and aligns with clean-label trends. Full article

Analysis of Biomorphs in the Teleorman County of Romania

The study of flora is crucial for conserving natural resources and assessing human impact on the environment. This paper explores floristic diversity, the role of plants, and the integration of technology in botanical research. In the studied area, 462 plant species were identified. Bioform analysis revealed a predominance of hemicryptophytes (45.45%) and therophytes (26.19%), suggesting a warm climate and significant zoo-anthropogenic influences. Other bioforms were present in lower percentages. Most plant species in Teleorman County are mesophilic (39.39%) and mesoxerophilic (30.95%), indicating adaptation to moderate or slightly dry environments. Regarding temperature affinity, the majority are micromesotherms (62.98%), suited for mild thermal conditions. Soil reaction analysis showed a preference for weakly acidic neutrophilic (39.82%) and euryionic (33.76%) soils, indicating tolerance for neutral to slightly acidic pH levels. The research evaluates the structure and diversity of flora in Teleorman County and emphasizes the influence of climate factors such as humidity, temperature, and soil pH on species distribution. Using the transect method and fractal analysis, this study concludes that temperature is the dominant climatic factor shaping local biodiversity. Full article

High Pressure Processing of Raw Ewe’s Cheese Promotes Microbiological Safety and Quality During Prolonged Storage

Serra da Estrela cheese (a raw ewe’s milk) ripened for 45 days was treated at 600 MPa for 6 min (P1) and at 450 MPa for 6 (P2) and 9 min (P3) and kept in refrigerated storage for 15 months. Lactobacillus and Lactococcus viable cell numbers were reduced in 3.2–3.6 and 2.7–3.6 log cycle units, respectively. Lower reductions were verified for total aerobic mesophilic and Enterococcus viable cell numbers in cheeses treated at 450 MPa (2.4–2.5 and 1.2 log reductions, respectively). In HPP cheeses, yeasts and moulds were below the enumeration limit up to 6 months of storage, but at 15 months 3.6–4.2 log cfu/g were quantified in all cheeses, while Enterobacteriaceae were inactivated to below the quantification limit. The increment of pressure treatment caused a greater impact on the microbiota’s viability than the increase in time under pressure. During storage, minor total colour differences were determined for HPP P3 cheese surface relative to control cheeses, ChC, at 45 days of ripening. HPP can thus be a good process to apply after cheese manufacture, since it offers a good potential to render raw-milk cheese microbiologically safer, with minimal changes in quality. Full article

Identification of Restoration Pathways for the Climate Adaptation of Wych Elm (Ulmus glabra Huds.) in Türkiye

Ulmus glabra Huds. is a mesophilic, montane broadleaf tree with high ecological value, commonly found in temperate riparian and floodplain forests across Türkiye. Its populations in Türkiye have declined due to anthropogenic disturbances and climatic pressures that cause habitat fragmentation and threaten the species’ long-term survival. In this research, we used Maximum Entropy (MaxEnt) to build species distribution models (SDMs) and applied the Restoration Planner (RP) tool to identify and prioritize critical restoration sites under both current and projected climate scenarios (SSP245, SSP370, SSP585). The SDMs highlighted areas of high suitability, primarily along the Black Sea coast. Future projections show that habitat fragmentation and shifts in suitable areas are expected to worsen. To systematically compare restoration options across different future scenarios, we derived and applied four spatial network status indicators using the RP tool. Specifically, we calculated Restoration Pixels (REST_PIX), Average Distance of Restoration Pixels from the Network (AVDIST_RP), Change in Equivalent Connected Area (ΔECA), and Restoration Efficiency (EFFIC) using the RP tool. For the 1 <-> 2 restoration pathways, the highest efficiency (EFFIC = 38.17) was recorded under present climate conditions. However, the largest improvement in connectivity (ΔECA = 60,775.62) was found in the 4 <-> 5 pathway under the SSP585 scenario, though this required substantial restoration effort (REST_PIX = 385). Temporal analysis noted that the restoration action will have most effectiveness between 2040 and 2080, while between 2081 and 2100, increased habitat fragmentation can severely undermine ecological connectivity. The result indicates that incorporation of habitat suitability modeling into restoration planning can help to design cost-effective restoration actions for degraded land. Moreover, the approach used herein provides a reproducible framework for the enhancement of species sustainability and habitat connectivity under varying climate conditions. Full article
