In this research, we investigate the interaction between the redox mediator ferrocene carboxylic acid (Fc-COOH) and glucose oxidase (GOD) in order to determine the thermodynamics parameters Kint, ΔGint, ΔHint, and ΔSint using simple UV–visible experiments at different temperatures. Positive values of ΔHint, ΔSint, together with a negative value of ΔGint indicate an entropy-driven hydrophobic interaction typical of spontaneous association processes. The homogeneous electron transfer rate constants between the oxidized organometallic mediator and the reduced enzyme (ks), along with their activation parameters (ΔGET, ΔHET and ΔSET), were calculated using data obtained from foot of the wave analysis (FOWA) of cyclic voltammetry experiments performed at variable temperature. According to transition state theory, the obtained parameters indicate a low activation enthalpy that reflects minimal energetic requirements for electron transfer, while the large negative activation entropy suggests the formation of an ordered transition state. The positive activation free energy falls within the expected range for biological electron transfer processes. Variable temperature cyclic voltammetry experiments of ferrocene carboxylic acid (Fc-COOH) were also performed. The obtained ΔG°, ΔH°, and ΔS° parameters indicate strong stabilization of the redox pair, consistent with a small difference in solvation energy. Circular dichroism, UV–vis spectroscopy, and combined CD and UV–Vis Spectroelectrochemistry measurements performed during redox mediation demonstrate that no significant structural alterations occur in either the enzyme or the redox mediator before or during the electron transfer processes. (1) Background: Subclinical neck pain is mild-to-moderate neck pain that has not yet been treated, and where individuals experience pain-free days. Alterations in sensorimotor integration, motor control, proprioception, and cerebellar inhibition have been observed in individuals with subclinical neck pain. Upregulation of the cervico-ocular reflex is documented in subclinical neck pain, with no difference in the gain of the vestibulo-ocular reflex. Vestibulo-ocular reflex gain adaptation and associated differences in visuo-motor control have not been successfully measured in this population. This study aims to investigate the vestibulo-ocular reflex gain adaptation and visuo-motor control in individuals with subclinical neck pain. (2) Methods: 30 right-hand-dominant participants (19 healthy controls: 10 male and 9 female; 16 subclinical neck pain: 6 male and 10 female) aged 18 to 35 performed an eye tracking task. Participants were seated 90cm away from a monitor and instructed to hold their gaze on a stationary or moving target projected onto a screen while performing active head rotations. Trials were divided into 12 blocks (pre-adaptation, 10 adaptation, and post-adaptation) for a total of 192 trials. During adaptation, the target would move at increasing speeds during each block, increasing by 10% of active head velocity up to a maximum of 100%. (3) Results: The subclinical neck pain group demonstrated significantly higher total saccades (p = 0.006, ƞ2 = 0.240) and overt catch-up saccades (p = 0.041, ƞ2 = 0.141) than the healthy control group. (4) Conclusion: Subclinical neck pain alters the visual–vestibular interaction. Full article

Background: Adjuvant anti-PD-1 therapy improves recurrence-free survival (RFS) in high-risk melanoma, but many patients experience adverse events. 2-deoxy-2-[18F]fluoro-D-glucose positron emission tomography with computed tomography [18F]FDG-PET/CT has been proposed to identify biomarkers that may predict outcome of treatment. Objectives: The aim of this register-based study was to investigate the prognostic value of immune-related adverse events (irAEs), spleen-to-liver ratio (SLR), and bone marrow-to-liver ratio (BLR), detected by [18F]FDG-PET/CT. Methods: This retrospective, register-based cohort study included 122 patients with radically resected stage III–IV melanoma treated with adjuvant anti-PD-1. Patient data were extracted from a Danish register, and measurements for SLR and BLR were made using an AI model. Cox regression models were made on irAEs and BLR to assess associations with RFS and overall survival (OS). Results: Over half of the patients experienced recurrence, and one quarter died during follow-up of 4 ¾ years. Seventy-four percent exhibited at least one PET-detected irAE. This study found no association between irAEs and OS. Regarding RFS, our findings suggest an increased risk of recurrence for the presence of irAEs within the first 1.5 years of follow-up (HR: 2.93, CI: 1.10–7.84, p = 0.032). BLR and SLR were not associated with RFS or OS in multivariable models. Conclusions: This study did not confirm the findings of a positive association between irAEs and survival found in previous studies. PET-detected irAEs were common in the study population, but did not predict OS, while early-onset irAEs were linked to increased recurrence risk. Neither SLR nor BLR demonstrated prognostic value. Further research is needed to clarify the clinical utility of PET-derived biomarkers, especially in the adjuvant setting. Full article

Background and Clinical Significance: Although rupture of aneurysms at the internal carotid-posterior communicating artery (ICA-PCom) junction accounts for a small percentage of all ruptured intracranial aneurysms, they are clinically relevant due to their proximity to perforator-rich cisterns, the optic-carotid-oculomotor pathways and flow-diverting zones, as well as their high likelihood for causing early neurological instability. Additionally, ruptured ICA-PCom aneurysms that have multiple lobulations are associated with increased variability in wall shear stress, local inflammatory remodeling and higher propensity for rupture at smaller sizes compared to other types of aneurysms. Due to the rapidity of early physiological destabilization in most patients with ruptured ICA-PCom aneurysms, clinical–anatomical correlations in these cases are often obscured by neurological deterioration; therefore, the presentation of this patient provides a unique opportunity to correlate the minimal early symptoms, tri-lobulation of the aneurysm and confined cisternal hemorrhage, to better understand rupture behavior, surgical decision-making in an anatomically challenging area, and postoperative recovery. Case Presentation: A 48-year-old hypertensive female experienced an acute “thunderclap” headache accompanied by intense photophobia and focal meningeal irritation, but, unexpectedly, retained a normal neurologic examination. She did exhibit some minor ocular motor micro-latencies, early cortical attentional strain and lateralized pain sensation that suggested localized cisternal involvement despite lack of generalized neurologic impairment. Digital subtraction angiography and three-dimensional CT angiography revealed a ruptured, tri-lobulated aneurysm originating from the communicating portion of the left internal carotid artery proximal to its origin from the posterior communicating artery, oriented toward the perimesencephalic cisterns. The aneurysm was surgically clipped using a standard left pterional craniotomy with direct visualization, after careful dissection through the carotid and optic windows to preserve the anterior choroidal artery, oculomotor nerve, and surrounding perforators. The neck of the aneurysm was reconstructed with a single straight clip, without compromise to the parent vessel lumen. The patient had an uneventful postoperative course without vasospasm or neurologic deficit. At both 3 and 9 months postoperatively the patient remained free of clinical neurologic deficit, and imaging demonstrated continued aneurysm exclusion, preserved ICA-PCom anatomy, and no evidence of delayed ischemic injury or hydrocephalus. Conclusions: The goal of this report is to demonstrate how a ruptured, morphologically complex ICA-PCom aneurysm may present with preserved neurologic function, thereby enabling the study of clinical–anatomical associations before secondary injury mechanisms intervene. The relationship between the configuration of the patient’s symptoms, geometry of the aneurysm and pattern of hemorrhage within the cisterns offers insight into a rare rupture pattern observed during routine clinical experience. Through complete anatomical analysis, timely microsurgical reconstruction and consistent follow-up, the authors were able to achieve long-term recovery of this particular patient. Continued advancements in vascular imaging techniques, aneurysmal wall modeling, and postoperative monitoring will likely help clarify the underlying mechanism(s) responsible for such presentations. Background: The senescence of testicular Leydig cells (LCs) is a key cause of age-related testosterone deficiency, in which oxidative stress (OS) and mitochondrial dysfunction are critical driving mechanisms. We explore whether the bioactive peptide C248 of PRDX4, an intracellular antioxidant, exerts mitochondrial protection to ameliorate LCs’ function. Methods: Based on the antioxidant domains of the PRDX4 protein, small molecular peptides were designed, and bioactive peptide C248 stood out from the crowd. An OS-induced senescence model of LCs was constructed by treating the MLTC-1 cell line with hydrogen peroxide (H2O2). C248 peptide or nicotinamide mononucleotide (NMN), as the positive control, was administered in the culture medium. The cellular function-related indicators, including DPPH free radical scavenging rate, cell viability, testosterone level, hydrogen peroxide (H2O2) content, senescence-associated β-galactosidase (SA-β-gal) activity, 8-hydroxy-2′-deoxyguanosine (8-OHDG) level, and 4-hydroxynonenal (4-HNE) level, were evaluated. The mitochondrial function and structural indicators, such as mitochondrial membrane potential, ATP production, mitochondrial morphology, and mitochondrial DNA (mtDNA) copy number, were subsequently tested. Results: In vitro experiments confirmed that C248 could scavenge DPPH free radicals in a dose-dependent manner, reduce the levels of reactive oxygen species, and increase antioxidant enzyme activity in LCs (p < 0.01). Both C248 and NMN increased testosterone secretion and improved cell viability (p < 0.01). Both C248 and NMN increased mitochondrial morphology and quantity, mitochondrial membrane potential (p < 0.01), ATP production (p < 0.01), and mitochondrial DNA (mtDNA) copy number (p < 0.01). Conclusion: This study reveals that the small molecular C248, a bioactive peptide of PRDX4, is a new candidate molecule for intervening in LC senescence and confirms that mitochondrial protection is a key strategy for improving age-related testicular dysfunction. High-quality, interoperable biodiversity information is a prerequisite for effective conservation policy, compliance with European Union (EU) reporting obligations, and efficient environmental decision-making. Greece’s LIFE EL-BIOS (LIFE20 GIE/GR/001317) developed the first National Biodiversity Information System, aiming to aggregate, standardise, and disseminate spatial and non-spatial data for species, habitats, pressures, and trends. This paper provides an economic valuation of this information system as a public, non-market good. We designed a two-stage stated-preference study: (i) a short pre-survey to calibrate initial bids and (ii) the main survey employing double-bounded dichotomous choice (DBDC) contingent valuation with a spike-logit specification. The payment vehicle was a hypothetical monthly subscription in a post-LIFE scenario. The instrument measured time savings (hours), perceived reliability (Likert 1–5), and key demographics/roles. A total of 167 valid responses were collected in September 2025. Participants reported an average of 5.2 h saved per use (median 4; max 14). Among those expressing willingness to pay (WTP), 77% rated EL-BIOS reliability as “High/Very high”. Econometric results indicate time savings as the strongest positive determinant of WTP; perceived reliability is positive and marginally significant; years of experience are negatively associated with acceptance; and cost has a strong negative effect. Mean WTP is estimated at €6.7 per month (median €3.5). Notably, 64% of those unwilling to pay declared protest motives (data should remain public and free). Accordingly, non-payment is decomposed into true zero WTP versus protest-based refusal, i.e., refusal to pay despite acknowledging value. This high protest share reflects principled opposition to paying for public biodiversity data rather than low perceived value of the system. The EL-BIOS database generates measurable productivity gains and social value both through positive WTP and principled protest responses supporting open public data. These findings inform policy on sustainable financing, governance, and long-term operation of national biodiversity information systems. Full article

Accurate group target association is essential for multi-sensor multi-target tracking, particularly in heterogeneous radar systems where systematic biases, asynchronous observations, and dense formations frequently cause ambiguous or incorrect associations. Existing approaches often rely on strict spatial assumptions or pre-trained models, limiting their robustness when measurement distortions and sensor-specific deviations are present. To address these challenges, this work proposes a robust association framework that integrates deep feature embedding, density-adaptive clustering, and global graph-theoretic matching. The method first applies an autoencoder–HDBSCAN clustering scheme to extract stable latent representations and obtain adaptive group structures under nonlinear distortions and non-uniform target densities. A weighted bipartite graph is then constructed, and a global optimal matching strategy is employed to compensate for heterogeneous systematic errors while preserving inter-group structural consistency. A mutual-support verification mechanism further enhances robustness against random disturbances. Monte Carlo experiments show that the proposed method maintains over 90% association accuracy even in dense scenarios with a target spacing of 1.4 km. Under various systematic bias conditions, it outperforms representative baselines such as Deep Association and JPDA by more than 20%. These results demonstrate the method’s robustness, adaptability, and suitability for practical multi-radar applications. The framework is training-free and easily deployable, offering a reliable solution for group target association in real-world multi-sensor fusion systems. Full article

This study addresses the fabrication challenges associated with producing diverse geometries for concrete dry connections, particularly regarding cost, time, and geometric limitations. The research investigates methods for fabricating precise, rebar-free dry connections in concrete, focusing on stamping and green-state computer numerical control (CNC) milling. These methods are evaluated using metrics such as dimensional accuracy, tool abrasion, and energy consumption. In the stamping process, a design of experiments (DOE) approach varied water content, concrete age, stamping load, and operational factors (vibration and formwork) across cone, truncated cone, truncated pyramid, and pyramid geometries. An optimal age range of 90 to 105 min, within a broader operational window of 90 to 120 min, was identified. Geometry-specific exceptions, such as approximately 68 min for the truncated cone and 130 min for the pyramid, were attributed to interactions between shape and age rather than deviations from general guidance. Within the tested parameters, water fraction primarily influenced lateral geometric error (diameter or width), while age most significantly affected vertical error. For green-state milling, both extrusion- and shotcrete-printed stock were machined at 90 min, 1 day, and 1 week. From 90 min to 1 week, the total milling energy increased on average by about 35%, and at one week end-face (head) passes caused substantially higher tool wear, with mean circumference losses of about 3.2 mm for head engagement and about 1.0 mm for side passes. Tool abrasion and energy demand increased with curing time, and extrusion required marginally more energy at equivalent ages. Milling was conducted in two engagement modes: side (flank) and end-face (head), which were evaluated separately. End-face engagement resulted in substantially greater tool abrasion than side passes, providing a clear explanation for tolerance drift in final joint geometries. Additionally, soil-based forming, which involves imprinting the stamp into soft, oil-treated fine sand to create a reversible mold, produced high-fidelity replicas with clean release for intricate patterns. This approach offers a practical alternative where friction and demolding constraints limit the effectiveness of direct stamping. Full article

Objective: To explore the association between uric acid and the prevalence of hyperlipidemia via the NHANES database, a combined hyperuricemia–hyperlipidemia (HUA-HLP) quail model was subsequently established to investigate the pathological characteristics of the model. Methods: In the NHANES database, information on patients with hyperuricemia is collected, and the association between serum uric acid levels and the prevalence of hyperlipidemia is analyzed by adjusting for confounding variables. A high-purine and high-fat diet was prepared with a ratio of regular feed–yeast extract powder–lard = 15:2:3. By measuring uric acid and blood lipid levels, and observing the activities of uric acid-producing enzymes and enzymes related to lipid metabolism synthesis and decomposition, the metabolic disorder and pathological characteristics of the model were evaluated. Results: By adjusting for confounding variables, it is found that as serum uric acid levels increase, the prevalence of hyperlipidemia rises significantly. The high-purine and high-fat diet successfully induced a quail model of hyperuricemia combined with hyperlipidemia. During the first week, serum uric acid, triglyceride, total cholesterol, and low-density lipoprotein cholesterol levels were significantly elevated and remained high until the end of the experiment. Serum free fatty acid levels were significantly increased from the second week and remained at a high level. Serum high-density lipoprotein cholesterol levels were significantly reduced from the third week and remained stable thereafter. In addition, the enzymes involved in uric acid synthesis as well as those related to lipid metabolism (including synthesis and decomposition) also exhibited significant abnormalities. Conclusions: In the human body, uric acid and lipid metabolism interact with each other and exacerbate one another’s abnormalities. A high-purine and high-fat diet can induce a quail model of hyperuricemia combined with hyperlipidemia. Uric acid and lipid metabolism are simultaneously disturbed, and the activities of uric acid-producing enzymes as well as enzymes related to lipid metabolism synthesis and decomposition are also altered. Alterations in the expression of the Klotho gene have been associated with chronic kidney disease (CKD), and its potential as an early diagnostic biomarker is currently under active investigation. In this work, we report the development of a highly sensitive, label-free electrochemical DNA-based biosensor for the detection of a 100 mer DNA fragment corresponding to a partial region of Klotho mRNA. The proposed bioplatform integrates mixed self-assembled monolayers (SAMs) and gold nanoparticles for efficient DNA immobilization within a sandwich-type configuration, coupled with impedimetric detection. Different SAM architectures were evaluated by cyclic voltammetry and electrochemical impedance spectroscopy, with the binary monolayer composed of 1-hexadecanethiol (HDT) and the capture probe (CP) exhibiting the best analytical performance. The use of gold nanoparticle-modified screen-printed carbon electrodes (AuNPs–SPCEs) resulted in a 1.4-fold increase in the signal-to-noise ratio compared to screen-printed gold electrodes. Additionally, the incorporation of a blocking step using bovine serum albumin (BSA–HDT–CP–AuNPs–SPCE) enhanced the sensitivity by 1.6-fold compared to the unblocked system. The genosensor displayed a linear response in the concentration range of 3 × 10−10 to 7.5 × 10−8 M, achieving a detection limit of 0.09 nM. Relative standard deviations below 7.5% were obtained for different Klotho concentrations, confirming high intra-assay and intermediary precision. Selectivity assays demonstrated negligible signals for non-complementary sequences, while recovery experiments in spiked human serum samples yielded satisfactory values between 96.5% and 103.4%. Eggshell cracks are a critical factor affecting egg quality and food safety, with traditional detection methods often struggling to detect fine cracks, especially under multi-colored shells and complex backgrounds. To address this issue, we propose a non-destructive detection approach based on an enhanced three-dimensional convolutional neural network (3D-CNN), named 3D-CrackNet, integrated with hyperspectral imaging (HSI) for high-precision identification and localization of eggshell cracks. Operating within the 1000–2500 nm spectral range, the proposed framework employs spectral preprocessing and optimal band selection to improve discriminative feature representation. A residual learning module is incorporated to mitigate gradient degradation during deep joint spectral-spatial feature extraction, while a parameter-free SimAM attention mechanism adaptively enhances crack-related regions and suppresses background interference. This architecture enables the network to effectively capture both fine-grained spatial textures and contiguous spectral patterns associated with cracks. Experiments on a self-constructed dataset of 400 egg samples show that 3D-CrackNet achieves an F1-score of 75.49% and an Intersection over Union (IoU) of 60.62%, significantly outperforming conventional 1D-CNN and 2D-CNN models. These findings validate that 3D-CrackNet offers a robust, non-destructive, and efficient solution for accurately detecting and localizing subtle eggshell cracks, demonstrating strong potential for intelligent online egg quality grading and micro-defect monitoring in industrial applications. Neutrophil gelatinase-associated lipocalin (NGAL) has emerged as a critical biomarker for the early diagnosis of acute kidney injury (AKI). The development of novel detection platforms that combine rapid analysis with high sensitivity is essential for improving clinical outcomes. In this study, we established an antibody-based detection system for NGAL using biolayer interferometry (BLI), a label-free optical biosensing technique that monitors real-time interference patterns generated by white light reflected from biomolecular binding events on a biosensor surface. A panel of six anti-NGAL monoclonal antibodies was generated and characterized for its binding properties, identifying candidates with high specificity for NGAL. For robust sensor functionalization, selected monoclonal antibodies were biotinylated and immobilized onto streptavidin-coated biosensor tips, establishing a stable and efficient detection interface. The optimized BLI platform demonstrated a limit of detection (LOD) of 46.1 ng/mL with wild dynamic range of 19 to 40,000 ng/mL. The platform’s accuracy was validated using human serum samples, with spike-and-recovery experiments yielding recovery rates of 96.6–104.6%. This demonstrates the capability to accurately quantify NGAL under physiologically relevant conditions with minimal matrix interference. Furthermore, the real-time kinetic measurements enabled rapid analysis, with the entire assay completed in less than half an hour. These findings establish a proof-of-concept for a BLI-based biosensor for NGAL detection, demonstrating sensitivity and specificity that show potential for clinical applications. Full article

Biological control is an effective and environmentally friendly strategy for managing plant diseases. In this study, a broad-spectrum antagonistic bacterium, designated strain AN6, was isolated from rice plants and exhibited potent inhibitory activity against a variety of phytopathogens. In Oxford cup assays, AN6 suppressed the growth of Xanthomonas oryzae pv. oryzae (Xoo) by 73.60%, and its cell-free culture filtrate caused pronounced morphological deformation in the bacterial cells. Further in vitro assays, including dual-culture assays, volatile organic compound (VOC) assays, and cell-free supernatant (CFS) assays, demonstrated that AN6 also exerted strong antifungal effects against several pathogenic fungi. In addition, the strain was found to produce proteases and siderophores, which may contribute to its antagonistic capabilities. Taxonomic identification based on morphological traits, 16S rRNA and gyrA gene sequencing, average nucleotide identity (ANI), in silico DNA–DNA hybridization (isDDH), and phylogenetic analysis classified strain AN6 as Bacillus velezensis. Whole-genome sequencing revealed that AN6 harbors a 3,929,788 bp genome comprising 4025 protein-coding genes with a GC content of 46.50%. Thirteen biosynthetic gene clusters (BGCs) associated with the production of secondary metabolites—such as nonribosomal peptides, polyketides, and dipeptide antibiotics—were identified. The pot experiment further validated the biocontrol potential of AN6, achieving an 80.49% reduction in rice bacterial blight caused by Xanthomonas oryzae pv. oryzae. Collectively, these results indicate that B. velezensis AN6 is a promising candidate for development as a highly effective biocontrol agent for the integrated management of diverse plant diseases. Background: Human papillomavirus (HPV) vaccination remains a critical preventive strategy against HPV-related cancers, yet uptake among young adults in Poland remains suboptimal. Objectives: This study aimed to assess HPV vaccination status, determinants, and perceived barriers to vaccination among healthcare students. Methods: This retrospective survey was conducted among 1062 students of the Medical University of Lodz, including those studying Medicine, Nursing, Midwifery, and Public Health. Results: Overall, 20% of respondents reported HPV vaccination, with the highest coverage among midwifery students (26.8%) and the lowest among medical students (16.8%). The major barriers to vaccination were found to be cost and misconceptions regarding vaccination age limits. As most respondents were above 14 years old when receiving the first dose, they were not eligible for the national free vaccination program. The significant motivators were parental influence and guidance from the medical university; however, recommendations for vaccination were infrequent. Multivariable logistic regression analysis found marital status (p = 0.029), paternal medical education (p = 0.003), and prior sexual experience (p = 0.037) to be significantly associated with vaccination status. Adverse events were reported by 45% of vaccinated respondents, most commonly reactions at the injection site. Nursing and midwifery students more often perceived adverse events as moderate or severe, but none discontinued vaccination. Conclusions: These findings underscore the need for financial support mechanisms and targeted educational interventions to enhance HPV vaccine uptake among future healthcare professionals in Poland. Background/Objectives: The use of intraoperative venoarterial extracorporeal life support (VA ECLS) has traditionally been used to support unstable patients undergoing complex lung transplantation. More evidence is emerging that the use of intraoperative VA ECLS may be beneficial for all patients undergoing lung transplantation. The aim of this study was to report the safety and feasibility of lung transplantation with the routine use of central VA ECLS. Methods: In this single-center retrospective observational study, all consecutive patients undergoing lung transplantation from April 2021 until September 2025 were included. Early outcomes and the incidence of primary graft dysfunction were evaluated with the International Society for Heart and Lung Transplantation criteria at 72 h after transplantation. Survival and chronic lung allograft dysfunction (CLAD)-free survival were reported with Kaplan–Meier estimates and 95% confidence intervals (CIs). Results: During the study period, 35 patients were successfully transplanted with the aid of central VA ECLS. There were no complications associated with intraoperative ECLS. One revision surgery was performed for immediate postoperative bleeding, and one for bronchial anastomosis air leak. Operative mortality occurred in three patients (8.6%). The median in-hospital stay was 30 (25–43) days. Severe primary graft dysfunction at 72 h was observed in four (11.4%) patients. Survival and CLAD-free survival at 1-, 3-, and 5-years following surgery were 85% (95% CI [74–98]), 74% (95% CI [59–92]), 67% (95% CI [28–82]), and 82% (95% CI [70–96]), 52% (95% CI [37–74]), 36% (95% CI [11–59]), respectively. Conclusions: Lung transplantation can safely be performed with the aid of central VA ECLS, with a low rate of primary graft dysfunction and favorable long-term outcomes. Further follow-up studies and greater experience are needed to make inferences on the long-term outcomes. This technique is relatively recent and evolving, representing an innovative intersection of advanced supportive technology with transplant surgery, potentially broadening indications and improving success rates.
