The aim of the research was to analyse the effect of different extraction temperatures on the colligative, hydrodynamic, and rheological properties of a water-soluble AXs fractions. The research material consisted of raw water extracts of arabinoxylans obtained from the husk at the following temperatures: 40 °C (AX40), 60 °C (AX60), 80 °C (AX80), and 100 °C (AX100). These were characterised in terms of their hydrodynamic, osmotic, and rheological properties, as well as the average molecular mass of the polysaccharide fractions. An increase in extraction temperature resulted in an increase in weight-average molecular mass, from 2190 kDa (AX40) to 3320 kDa (AX100). The values of the osmotic average molecular mass were higher than those obtained from GPC, and decreased with increasing extraction temperature. The dominance of biopolymer–biopolymer interactions was evident in the shape of the autocorrelation function, which did not disappear as the extraction temperature and concentration increased. Furthermore, the values of the second virial coefficient were negative, which is indicative of the tendency of biopolymer chains to aggregate. The rheological properties of the extracts changed from being described by a power-law model (AX40 and AX60) to being described by the full non-linear De Kee model (AX80 and AX100). Full article

Decoctions of stem barks from Aucoumea klaineana, Canarium schweinfurthii, Pentadesma butyracea and Scorodophloeus zenkeri are used against affections of irritable bowel syndrome in Gabonese traditional medicine. In the present study, we aim to determine whether the bark polysaccharides may contribute to the activity of these plants against the symptoms of gastrointestinal disorders. To this end, we investigated the structure and the pharmacological activity of polysaccharides extracted from their stem barks. The pectic and hemicellulose polysaccharides were isolated, and their sugar compositions were determined by gas chromatography. In addition, analysis by MALDI-TOF mass spectrometry of oligosaccharides released after digestion with an endo-xylanase indicated that glucuronoarabinoxylans are the main hemicellulose of stem barks. We then evaluated the influence of the polysaccharide fractions on the spontaneous contractile activity of rat ileal smooth muscle and the cholinergic system. Spasmolytic activity of pectic fractions from all stem barks, as well as lemon polygalacturonic acid, were observed, indicating that these extracts exhibit a myorelaxant activity. In contrast, the bark hemicellulose fractions, as well as commercially available beechwood glucuronoxylan and wheat arabinoxylan, were demonstrated to be able to increase the basal contractile activity of smooth muscle. These data show that, beyond physicochemical effects affecting the bowel water content, plant polysaccharides have also an impact on the spontaneous smooth muscle contractility, the main mechanism involved in the pathophysiology of gastrointestinal disorders. A nanoparticle formulation was generated from distiller dried grains with solubles (DDGS), and its effect on the production of anthraquinones (AQs) was evaluated on Rubia tinctorum hairy roots. The DDGS material was washed with water and ethyl acetate to remove mainly the soluble organic/inorganic molecules and reduce the fat content, respectively, followed by an alkaline treatment to remove the polysaccharides. The resulting alkaline solutions were then lyophilized and redispersed in deionized water to generate a monodispersed nanoparticulate formulation (DDGS-NP) with a hydrodynamic diameter and zeta potential of 227 ± 42 nm and −53 ± 7 mV, respectively. The formulation demonstrated good colloidal stability over time, and sterilized DDGS-NPs maintained comparable physicochemical properties. The nanoparticles were enriched in protein fractions, unsaturated fatty acids, and orthophosphate anion components from DDGS, as determined by solid-state Nuclear Magnetic Resonance (NMR), X-ray photoelectron spectroscopy (XPS), organic elemental analysis (OEA), and inductively coupled plasma optical emission spectrometry (ICP-OES) techniques. The DDGS-NPs were tested at different concentrations on Rubia tinctorum hairy roots, in comparison to or in combination with methyl jasmonate (MeJ), for their capacity to induce the production of AQs. All DDGS-NP concentrations increased the production of specific AQs to 7.7 (100 mg L−1), 7.8 (200 mg L−1), and 9.3 µmol/gFW (500 mg L−1), with an extracellular AQ accumulation of 18 µM for the highest DDGS-NP concentration, in comparison with the control hairy roots (~2 µM AQ). The plant growth was not affected at any of the tested nanoparticle concentrations. Interestingly, the combination of DDGS-NPs and MeJ resulted in the highest extracellular AQ accumulation in R. tinctorum root cultures. Full article

Alpinia officinarum Hance exhibits various bioactivities, with polysaccharides being one of its key bioactive components. However, the relationship between the structural characteristics of these polysaccharides and their bioactivities remains unclear and underexplored. In this study, to optimize the extraction process, a Response Surface Methodology-based design combined with single-factor experiments was applied to determine the optimal conditions for the ultrasonic-microwave-assisted extraction of polysaccharides from A. officinarum. The primary structural characteristics and antioxidant activities of two polysaccharide fractions, PAOR-1 extracted by ultrasonic-microwave-assisted extraction and PAOR-2 extracted by hot reflux extraction (HRE), were systematically compared. The optimal extraction conditions, including a liquid–solid ratio of 1:50, extraction time of 19 mins, and ultrasonic power of 410 W, yielded a maximum polysaccharide extraction rate of 18.28% ± 2.23%. The extracted polysaccharides were characterized as acidic polysaccharides with a three-dimensional structure. PAOR-1 and PAOR-2 have different monosaccharide compositions, surface morphologies, and thermal stabilities. The antioxidant activity in vitro studies suggest that PAOR-1 may have higher antioxidant activity than PAOR-2 due to its higher content of uronic acids, lower relative molecular mass, and a more closely packed spatial configuration. These findings provide a theoretical basis for the development and utilization of AOR. Full article

Limonium gmelini (Willd.) Kuntze, a plant widely used in traditional medicine, has garnered increasing attention for its diverse pharmacological activities, including anti-inflammatory, hepatoprotective, antioxidant, and antimicrobial effects. This study aimed to explore the chemical composition and biological activities of polysaccharides and polyphenolic compounds extracted from both the roots and aboveground parts of Limonium gmelini. Several methods of extraction, including ultrasound-assisted extraction (UAE), conventional maceration (CM), and supercritical fluid extraction (SFE), were employed to obtain bioactive fractions. Chemical profiling, primarily represented by monosaccharides and polyphenolic compounds, was characterized and analyzed using proton nuclear magnetic resonance spectroscopy (1H-NMR) and gas chromatography-mass spectrometry (GC-MS) techniques. While polyphenol-rich fractions exhibited significant antibacterial activity, particularly against Staphylococcus epidermidis, polysaccharide-rich aqueous fractions showed minimal antibacterial activity. Among the methods, CM and UAE yielded higher polyphenol content, whereas SFE provided more selective extractions. Notably, methanolic SPE fractions derived from the roots were especially enriched in active polyphenols such as gallic acid, myricetin, and naringenin, and they exhibited the highest antibacterial activity against Staphylococcus epidermidis. In contrast, extracts from the aboveground parts showed more moderate activity and a partially different chemical profile. These findings underscore the importance of plant part selection and support the targeted use of root-derived polyphenol-enriched fractions from L. gmelini as promising candidates for the development of natural antibacterial agents. Further investigation is needed to isolate and validate the most active constituents for potential therapeutic applications. Full article

The fast-disintegrating capsules rapidly disintegrate in various physiological environments, ensuring therapeutic efficacy. The formulation of plant-based capsules with balanced mechanical and fast disintegration characteristics continues to present technical challenges in pharmaceutical development. In this study, natural marine polysaccharides were utilized to achieve both rapid disintegration and excellent mechanical properties by combining κ-Carrageenan (κ-C) and ι-Carrageenan (ι-C). Additionally, the selection of KCl + NaCl mixed coagulants, along with the evaluation of their types, mass fractions, and ratios, enhanced the mechanical properties and transmittance of the capsules. FTIR analysis revealed that the membrane with a 5:5 κ-C/ι-C ratio formed hydrogen bonds, which were beneficial to its fast disintegration. SEM analysis revealed a dense microstructure in this formulation, contributing to its improved mechanical properties. Finally, this study hypothesizes that the disintegration behaviors of the capsules exhibited significant pH dependence, with ion exudation predominating in pH 1.2 and pH 7.0 media, while swelling dominated under pH 4.5 and pH 6.8 media. The prepared carrageenan blend-based capsules exhibited fast disintegration properties while maintaining excellent mechanical and barrier properties, thereby broadening the application of plant-based capsules in the field of medicine. This study aimed to optimize the enzyme-assisted extraction of polysaccharides (RTFPs) from Rosa roxburghii fruit using response surface methodology. Under the optimal extraction conditions, the yield of RTFPs reached 14.02%, which was close to the predicted value of 13.96%. The primary structural characteristics and the antioxidative and immunomodulatory activities of RTFPs were also examined. Structural characterization revealed that RTFPs comprise 36.38% neutral sugar, 48.83% uronic acid, and 7.29% protein. Their heteropolysaccharide structure features two distinct molecular weight fractions (1.87 × 105 Da and 4.75 × 103 Da) and a monosaccharide composition dominated by glucose (38.93%), arabinose (20.66%), galactose (20.58%), galacturonic acid (10.94%), and xylose (6.52%). Antioxidant assays demonstrated potent radical scavenging activity, with IC50 values of 11 μg/mL (DPPH) and 150 μg/mL (ABTS), comparable to conventional antioxidants. Immunomodulatory studies on RAW264.7 macrophages revealed that RTFPs (100–400 μg/mL) significantly enhanced phagocytosis by 12.61–76.63% and stimulated the secretion of nitric oxide (NO) and tumor necrosis factor-α (TNF-α). These bioactivities are attributed to RTFPs’ high uronic acid content, moderate molecular weight distribution, unique monosaccharide profile, and highly branched conformation. Full article

Low-molecular-weight polypeptides (<3 kDa) were prepared from Periplaneta americana via enzymatic hydrolysis and ultrafiltration, yielding 3.53 ± 0.01 mg/g of peptide-rich extract. The extract was primarily composed of peptides, proteins, polysaccharides, phenolics, and flavonoids. HPLC-MS analysis identified 1402 peptide sequences, 80.51% of which were below 1000 Da, predominantly consisting of tri-, tetra-, and octapeptides. Monosaccharide profiling detected D-(+)-galactose, and quantitative assays determined the contents of total phenolics (12.28 mg/g), flavonoids (15.50 mg/g), proteins (85.84 mg/g), and total sugars (17.62 mg/g). The biological activities of the extract were systematically evaluated. The peptide fraction inhibited hyaluronidase activity by 58% at 5 mg/mL, suggesting protection of extracellular matrix integrity. In HaCaT keratinocytes, it promoted cell proliferation by 62.6%, accelerated scratch wound closure by 54%, upregulated Wnt-10b and β-catenin expression, and reduced intracellular ROS levels under oxidative stress. In LPS-stimulated RAW 264.7 macrophages, the extract decreased TNF-α, IL-6, and IL-1β production by 30%, 25%, and 28%, respectively, reduced MDA levels by 35.2%, and enhanced CAT and SOD activities by 12.3% and 60.3%. In vivo, complete closure of full-thickness skin wounds in mice was achieved by day 14. Safety evaluations using the chick chorioallantoic membrane assay and human patch tests confirmed the extract to be non-irritating and non-toxic. These findings highlight Periplaneta americana extract as a promising multifunctional bioactive ingredient for cosmetic and dermatological applications. Further studies on its active components, mechanisms of action, and clinical efficacy are warranted to support its development in skin health and aesthetic medicine. Full article

Dendrobium, a prominent genus in the Orchidaceae family, has generated significant research attention due to its demonstrated biological potential, particularly its notable anti-inflammatory and antioxidant activities. In this study, two fractions of fermented Dendrobium officinale polysaccharides (FDOPs) were successfully isolated through a multi-stage purification strategy including gradient ethanol precipitation, gel column chromatography, and ion exchange chromatography with Lactobacillus reuteri CCFM863. Structural characterization revealed that both Dendrobium officinale polysaccharide fractions consisted of (1→4)-β-D-Manp, (1→4)-β-D-Glcp, and (1→4)-α-D-Glcp residues. The anti-inflammatory efficacy and keratinocyte-protective potential of FDOPs (FDOP-1A and FDOP-2A) were investigated by using lipopolysaccharide (LPS)-induced RAW264.7 and HaCaT cells models, which showed significant inhibitions on the inflammatory factors of monocyte chemoattractant protein-1 (MCP-1), tumor necrosis factor-alpha (TNF-α), nitric oxide (NO), and interleukin-1 beta (IL-1β); recovered levels of filaggrin (FLG), aquaporin 3 (AQP3), transient receptor potential vanilloid 4 (TRPV4), cathelicidin antimicrobial peptide (CAMP)/LL-37, and adiponectin (ADIPOQ); and the reduced protein expression of the TLR4/IκB-α/NF-κB/NLRP3 pathway. Notably, the FDOPs exhibited a remarkable reactive oxygen species (ROS) scavenging capacity, demonstrating superior antioxidant activity. Therefore, FDOPs show dual anti-inflammatory and antioxidant properties, making them suitable as active ingredients for modulating epidermal inflammation and promoting skin barrier repair. This study highlights waste bread (WB) as a novel, cost-effective, and nutrient-rich substrate for microbial growth, offering a sustainable alternative to conventional media. As a renewable resource, WB promotes the circular economy by reducing food waste and encouraging biotechnological innovation. The incorporation of WB into microbial culture media enhanced the growth of various reference strains (E. coli, E. faecalis, P. aeruginosa, and S. aureus), with at least a two-fold increase compared to conventional Luria-Bertani (LB) medium. Moreover, combining 2% WB with diluted LB (1/10) reduced medium costs by up to 90%. Furthermore, it was confirmed that 1% WB can effectively replace starch during the screening of amylolytic strains. Applying a fractional factorial design, the production of amylase by Bacillus sp. BSS (Amy-BSS) was enhanced 15-fold. An analysis of the Pareto diagram revealed that WB was the most significant factor. Additionally, Amy-BSS was applied to hydrolyze polysaccharides in WB, enabling the generation of high-value-added products in food processing. This hydrolysis process yielded 4.6 g/L of fermentable sugars from 1% WB. Evaluating the economic feasibility of WB valorization into value-added products elucidates potential pathways for cost reduction and enhanced environmental sustainability, thereby positioning WB as a viable tool for sustainable development. Olive mill wastewater (OMW), a byproduct of the olive oil industry, is a potential source of natural bioactive phenolic polymers. In this work, a column chromatography technique was used for the isolation of a new complex polymer (named OMW-2000XAD) from OMW via fractionation on Amberlite® XAD16 resin. The developed procedure was simple and proved to be reproducible using OMW from two different sources. OMW-2000XAD was further characterized by elemental, glycosidic, and amino acid composition analysis, as well as spectroscopic techniques. The polymer’s molecular size, which was estimated via gel filtration chromatography, was 1960 kDa, which is significantly larger than other high-molecular weight fractions previously isolated from OMW or other agro-industrial wastes. OMW-2000XAD was mainly composed of phenolic compounds (89.8%). It also contained polysaccharides (16.1%) and proteins (10.3%), with glucose (12.25%) and cysteine (1.71%) being the most abundant sugar and amino acid, respectively, as well as metals (1.29%, primarily potassium). However, due to its low solubility, complexity, and heterogeneous composition, it was not possible to identify all phenolic compounds or elucidate a definitive structure via MS, FTIR, and NMR. OMW-2000XAD exhibited strong radical scavenging antioxidant capacity (ABTS•+, DPPH and peroxyl radicals), with results up to 7415 µmol Trolox equivalent/mol (ORAC method), but showed no antiproliferative effects, highlighting the need for further research. The protein fraction of slaughterhouse blood remains underutilized primarily due to challenges associated with its instability during processing and storage. This study aimed to develop stable bovine blood plasma gels using selected lactic acid bacteria and flaxseed oil cake flour. Various lactic acid bacteria strains were incorporated at concentrations of 5–20% (w/w), and gel properties such as pH, gelation time, yield stress, and freeze–thaw syneresis were evaluated. Optimal gelation was achieved at 20% inoculum, producing fibrin networks with yield stresses (372 Pa) comparable to recalcified controls (410 Pa), but accompanied by high serum loss and undesired acidic aromas at higher bacterial densities. Incorporating 5% hydrated flaxseed oil cake flour successfully reduced syneresis below 10%, improved water-holding capacity (135%), and prevented development of off-flavors, demonstrating beneficial interactions between flaxseed polysaccharides and blood plasma proteins. Thus, combining a 20% mixed lactic starter with 5% flaxseed cake flour yielded a stable plasma gel suitable for meat product applications, balancing rapid gel formation, high moisture retention, desirable rheological properties, and neutral sensory characteristics. Aluminum (Al) dissolves from soil at low pH and is absorbed by plants, inhibiting their growth. Since most of the Al absorbed by plants is present in the cell wall, it is thought that the binding of Al to cell wall polysaccharides alters the properties of the cell wall and inhibits cell elongation. However, it remains unclear in which component of the cell wall Al accumulates. In this study, we determined the distribution of Al in rice root cell wall fractions under Al stress conditions. The results show that Al accumulates predominantly in the hemicellulose fraction, with star1 mutants accumulating significantly more Al than WT plants. An analysis of cell wall sugars revealed an increase in xyloglucan content under Al stress, which influenced the inhibition of root elongation. OsXTH19, a member of the xyloglucan endotransglucosylase/hydrolase (XTH) family, exhibits only xyloglucan endohydrolase (XEH) activity and lacks endotransglucosylase (XET) activity. OsXTH19 overexpressor rice (OsXTH19-OX) enhances the degradation of xyloglucan. Furthermore, OsXTH19-OX rice with reduced xyloglucan levels exhibited reduced Al accumulation and enhanced root growth under Al stress. Full article

This study presents an investigation on the potential of using one-year-old field-stored Medicago sativa (alfalfa) as a raw material for a multi-output biorefinery. The main objective was to fractionate the biomass into valuable components—crude protein, hemicellulose-derived polysaccharides, lignin, and cellulose—and to explore the latter’s suitability in papermaking. To this end, three pretreatment strategies (water, alkaline buffer, and NaOH solution) were applied, followed by soda pulping under varying severity conditions. Both solid and liquid fractions were collected and chemically characterized using FTIR, HPLC, and standardized chemical methods. Water-based pretreatment was most effective for protein extraction, achieving over 40% protein content in precipitated fractions. The harshest pulping conditions (20% NaOH, 160 °C, 60 min) yielded cellulose-rich pulp with high glucan content, while also facilitating lignin and hemicellulose recovery from black liquor. Furthermore, the pulps derived from alfalfa stems were tested for papermaking. When blended with old corrugated cardboard (OCC), the fibers enhanced tensile and burst strength by 35% and 70%, respectively, compared to OCC alone. These findings support the valorization of unexploited alfalfa deposits and suggest a feasible biorefinery approach for protein, fiber, and polymer recovery, aligned with circular economy principles. The Chemical Profiling and Immunological Activity of Polysaccharides from the Rhizome of Imperata cylindrica Using Hot Water Extraction

To investigate the immunomodulatory activity of polysaccharides derived from the rhizome of Imperata cylindrica, polysaccharides (IRPs-H) were extracted using hot water extraction and further purified via DEAE-52 ion-exchange chromatography, yielding three fractions: IRPs-H1, IRPs-H2, and IRPs-H3. The structural features of these fractions were characterized by Fourier-transform infrared spectroscopy (FT-IR), high-performance gel permeation chromatography (HPGPC), atomic force microscopy (AFM), and thermogravimetric analysis (TGA). Their immunological activities were evaluated in vitro. All three fractions were identified as neutral pyranose-type polysaccharides, primarily composed of glucose and xylose, exhibiting good thermal stability and lacking long-chain structures. In vitro assays using RAW264.7 macrophages demonstrated that these polysaccharides promoted cell proliferation (50–800 μg/mL), enhanced phagocytic activity, and induced morphological changes characteristic of macrophage activation, including irregular shapes and pseudopod formation. ELISA and flow cytometry analyses revealed dose-dependent increases in nitric oxide (NO), interleukin-6 (IL-6), tumor necrosis factor-α (TNF-α), and reactive oxygen species (ROS) levels. Notably, the IRPs-H3 fraction stimulated TNF-α and IL-6 production to levels of 438.02 ± 14.14 pg/mL and 30.13 ± 1.27 pg/mL, respectively, which were comparable to those induced by lipopolysaccharide (LPS), the positive control (460.83 ± 16.10 pg/mL and 31.87 ± 1.72 pg/mL, respectively). These results suggest that polysaccharides extracted from the rhizome of Imperata cylindrica possess significant immunostimulatory properties and hold potential for development as functional food ingredients or immune-enhancing agents.
