Pine needles are traditional herbal remedies used for centuries to treat various ailments, including rheumatism, bronchitis, burns, inflammation, and infections. This study aimed to evaluate the antioxidant, analgesic (peripheral and central), and wound-healing activities of Pinus pinaster (PPN) and Pinus halepensis (PAN) needles while identifying the bioactive compounds responsible for these effects. Phytochemical analysis revealed several phenolic compounds, including p-coumaroylquinic acid, quercetin, narcissin, and myricetin-3-O-glucoside. Both extracts showed strong antioxidant activity, with high total phenolic content (TPC: 384.84 ± 0.84 and 524.46 mg GAE/g DM for PPN and PAN, respectively) and flavonoid content (TFC: 109.44 ± 0.62 and 111.64 ± 0.62 mg QE/g DM, respectively). Peripheral analgesic activity, assessed using the acetic acid-induced writhing test, revealed that PAN (300 mg/kg) significantly reduced pain by 72.3%, while central analgesic effects, evaluated by the tail immersion test, were comparable to the reference drug for both extracts. In vivo wound-healing tests showed accelerated wound contraction and complete closure by day 21, indicating strong regenerative potential. Overall, this study demonstrates that PPN and PAN needle extracts possess significant antioxidant, analgesic, and wound-healing activities, supporting their traditional use and highlighting their potential as natural therapeutic agents for managing oxidative stress, pain, and skin injuries. Climate change can impact various facets of a region’s fire regime, such as the frequency and timing of fire ignitions. This study examines the temporal trends of monthly fire counts in the Northwest and Northeast Regions of Ontario, Canada, between 1960 and 2023. Fires ignited by human activities or lightning are analyzed separately. The significance of historical trends is investigated using the Cochrane–Orcutt method, which identifies decreasing trends in the number of human-caused fires for several months, including May through July. A complementary trend analysis of total area burned is also conducted. The forecasting of future months’ fire counts is explored using a Negative Binomial Autoregressive (NB-AR) model suitable for count time series data with overdispersion. In the NB-AR model, the use of climate teleconnections at a range of temporal lags as predictors is investigated, and their predictive skill is quantified through cross-validation estimates of Mean Absolute Error on a testing dataset. Considered teleconnections include the El Niño-Southern Oscillation (ENSO), Pacific Decadal Oscillation (PDO), Arctic Oscillation (AO), North Atlantic Oscillation (NAO), and Atlantic Multidecadal Oscillation (AMO). The study finds the use of teleconnection predictors promising, with a notable benefit for forecasting human-caused fire counts but mixed results for forecasting lightning-caused fire counts. Background/Objectives: Ash made from juniper trees and added to cornmeal-based dishes may have provided calcium (Ca) to traditional Indigenous diets. Few studies have quantified the mineral content of juniper ash, including its Ca content. The objective of this study was to determine whether juniper ash could serve as a safe source of non-dairy Ca in an intervention study. Methods: Branches from two varieties of Juniper (Rocky Mountain Juniper, or Juniperus scopulorum and Eastern Red Cedar, or Juniperus virginiana) were harvested and burned to ash in a laboratory setting. Juniper ash from the southwestern U.S. available for retail purchase was used for comparison. All samples were tested for content of 10 nutritive elements (Ca, copper, iron, potassium, magnesium, manganese, sodium, phosphorus, selenium, and zinc) and 20 potentially toxic elements (silver, aluminum, arsenic, barium, beryllium, cadmium, cobalt, chromium, mercury, lithium, molybdenum, nickel, lead, antimony, tin, strontium, thallium, uranium, and vanadium) as well as n = 576 pesticide residues. Results: All samples contained both nutritive and potentially toxic elements. Each teaspoon of ash contained an average of 445 ± 141 mg Ca. However, the samples also contained lead in amounts ranging from 1.09 ppm to 15 ppm. Conclusions: Information on the nutritive and potentially toxic elemental content of juniper ash and how it may interact within a food matrix is insufficient to determine its safety as a Ca source. Further investigation is needed on the bioavailability of calcium oxide and its interaction with other dietary components to clarify the potential role of juniper ash in contemporary food patterns. Background: This study investigates the influence of emotional processing on flexor reflex responses in the upper limbs, focusing on cutaneomuscular reflexes (CMRs) and the cutaneous silent period (CSP) in patients with chronic neuropathic pain. The modulation of motor reflexes by emotions remains unclear. Methods: Fifty-one patients with chronic upper limb neuropathic pain (carpal tunnel syndrome, other neuropathies, post-burn hypertrophic scars) and twenty healthy controls underwent standardized electrodiagnostic signal acquisition. Neurophysiological assessments (CMRs, CSP, standard nerve conduction tests) and psychological evaluations (anxiety, depression, emotion processing) were conducted. Neurophysiological signal acquisition included median and ulnar nerve conduction studies recorded with an electrodiagnostic system (48 kHz sampling rate; 30–3000 Hz bandpass). CSP and CMRs were recorded from the abductor pollicis brevis using surface electrodes (bipolar belly–tendon montage) and were evoked by electrical stimulation delivered through ring electrodes, with individualized perceptual-threshold calibration. Statistical analyses examined correlations between neurophysiological and psychological measures. Results: Patients showed significantly longer duration and higher intensity of CMRs and CSP than controls (p < 0.01). CMR and CSP durations correlated positively with anxiety, depression, and alexithymia scores, and negatively with facial emotion recognition. General Linear Model analyses indicated these relations were mediated by tactile and pain perception thresholds. Conclusions: The findings support that spinal reflex responses in the upper limbs are modulated by emotional and cognitive-affective processes, especially in chronic pain contexts. This highlights the complex interaction between emotion regulation and motor control in neuropathic pain conditions. Full article

The characteristics of litter combustion have a significant impact on the spread of surface fires in the central Yunnan Province, a high-risk forest fire zone. The burning behavior of individual and mixed-species litter samples from five dominant tree species (Pinus yunnanensis Franch., Keteleeria evelyniana Mast., Quercus variabilis Blume., Quercus aliena var. acutiserrata, and Alnus nepalensis D. Don.) was assessed in this study using cone calorimeter tests. Fern fronds and fine branches were included in additional tests to evaluate their effects on specific combustion parameters, such as Fire Performance Index (FPI), Flame Duration (FD), Time to Ignition (TTI), Mass Loss Rate (MLR), Residual Mass Fraction (RMF), Peak Heat Release Rate (PHRR), and Total Heat Release (THR). There were remarkable differences in the burning properties of the three types of litter (broadleaf, pine needles, and short pine needles). The THR and PHRR values of P. yunnanensis were the highest, whereas the PHRR of the other species varied very little. Short pine needle litter showed incomplete combustion and a long flame duration. When measured against pure pine needle litter, mixtures of P. yunnanensis and broadleaf litter showed lower PHRR. When set side by side to pure pine needle litter, P. yunnanensis and broadleaf litter showed lower PHRR. THR rose when fine branches were included, underlining the significance of fine woody fuels in fire behavior. The insertion of ferns increases the percentage of unburned biomass, prolongs TTI, and dramatically reduces PHRR. Full article

On 2021, in the intensive care unit of a County Emergency Hospital where oxygen therapy treatment was applied to COVID patients, located in the municipality of Ploiesti, Prahova County, a fire occurred that resulted in the destruction by burning of the ICU room, the death of two people, and the injury of a medical professional. In order to elucidate the accelerating causes of the combustion phenomenon of materials in the ICU room, a combustion stand was designed whose atmosphere can be controlled in terms of achieving high oxygen concentrations of 40% vol., in accordance with the treatment schemes applied to the patients and with the configuration of the room and the frequency of use of the access door. In this experimental stand, a series of combustion tests of flexible polyurethane foam samples were performed, which highlighted the acceleration of combustion and the complete consumption of the mass. The purpose of this work is to explain the rapidity of the fire in a hospital ward, both with experimental methods and with the help of FDS. Full article

Microbial polysaccharides are promising components for wound-care products. This study reports the development of wound-healing antimicrobial hydrogels, based on pullulan from Aureobasidium pullulans, combined with mesenchymal cell-derived conditioned medium. Structural characterization of pullulan was confirmed by FTIR and NMR. Twenty-three formulations containing pullulan, chitosan, gelatin, citric acid, and antimicrobial agents were prepared. Physicochemical screening identified optimal hydrogels: No. 22 (1.2% pullulan, 1.2% chitosan, 0.2% citric acid, 2.4% gelatin, 0.1% conditioned medium, 0.4% glutaraldehyde) and No. 23 (2.4% pullulan, no chitosan, the remaining components identical to those in No. 22). Both exhibited pH values of 5.34 and 5.49, moisture content of 92%, swelling capacities of 175% and 213%, and dynamic viscosity between 58–120 mPa·s. Cytotoxicity testing with human mesenchymal stem cells showed no significant toxicity, with both hydrogels supporting cell adhesion and proliferation. Antimicrobial assays demonstrated inhibitory activity against Staphylococcus aureus and Escherichia coli for both formulations; only hydrogel No. 23 inhibited Pseudomonas aeruginosa. In vitro scratch assays revealed that hydrogel No. 23 significantly promoted fibroblast migration, achieving 30.25% scratch closure after 24 h. The developed formulations combine favorable physicochemical properties with antimicrobial efficacy and regenerative potential, supporting further evaluation as advanced wound-healing and anti-burn dressings. Prescribed fire is a common habitat management tool for white-tailed deer (Odocoileus virginianus Zimm.) that can influence browse quantity and quality. We tested effects of time since burn and number of burns within a decade on browse forage productivity in forested stands in the Pineywoods ecoregion of Texas. We utilized 46 plots on sites managed by the United States Forest Service National Forests and Grasslands in Texas, The Nature Conservancy, and a private landowner. Preferred browse forage species were sampled and analyzed for nutrient content, and years since last prescribed burn and the number of burns within the last 10 years were compared. Deer had strong preferences for plants with greater crude protein, magnesium, and potassium. Crude protein and net energy for maintenance were generally greater with a more frequent burn regime. Different nutrients peaked at different burn intervals. Frequent fires resulted in higher crude protein (x¯  = 14.0%) than infrequently burned sites. At four burns per decade, net maintenance energy was highest (x¯ = 0.6 Mcal Kg−1). Linear regression models only explained between 28% and 41% of utilization, although some preferences for some nutrients, such as crude protein and magnesium, were detected. To improve the nutritional carrying capacity for white-tailed deer, long-term management regimes should incorporate site-specific burn plans that include fire frequency. Timing and burn frequency are critical to achieving optimum results that improve browse forage availability, quality, and utilization. Full article

Background and Clinical Significance: Paroxysmal extreme pain disorder (PEPD) is an extremely rare autosomal dominant sodium channelopathy caused by SCN9A gain-of-function variants. It is characterized by infantile-onset excruciating paroxysmal pain, typically in rectal, ocular, or mandibular regions, triggered by innocuous stimuli and accompanied by autonomic flares. Carbamazepine is dramatically effective in most reported cases. To date, only two genetically confirmed cases have been documented in Chinese patients, and fewer than 20 disease-causing variants are reported worldwide. We report the third Chinese case harboring a novel likely pathogenic SCN9A variant (p.Leu1623Gln), notable for its unusually severe, progressive, and carbamazepine-refractory phenotype, as well as life-threatening psychiatric sequelae, highlighting phenotypic heterogeneity and the devastating impact when standard therapy fails. Case Presentation: A Chinese male proband with positive family history presented with lifelong trigger-induced catastrophic burning and tearing pain in the perineum and lower limbs, associated with erythema, swelling, and occasional non-epileptic seizures. Attacks worsened with age despite escalating polypharmacy, including high-dose opioids, benzodiazepines, topical lidocaine and carbamazepine. Both the proband and his father developed profound psychosocial sequelae including severe depression and suicidal attempts. Next-generation sequencing in the proband revealed a novel heterozygous likely pathogenic variant NM_001365536.1 (SCN9A): c.4868T>A p.(Leu1623Gln). Conclusions: This third reported ethnic Chinese PEPD case expands the genotypic and phenotypic spectrum of SCN9A-related channelopathies, demonstrating that some variants can produce carbamazepine-refractory, progressive, and profoundly disabling disease with high suicidality risk. Early genetic diagnosis is critical in family planning and cascade testing, and has the potential in guiding targeted therapy that is under active research. Full article

Background: Burns cause about 180,000 deaths annually and lead to substantial morbidity, especially in low- and middle-income countries. Clinical assessment of burn depth and TBSA relies on visual and bedside examination and remains subjective. Convolutional neural networks (CNNs) have been proposed to improve objectivity in image-based burn assessment, but clinical generalizability and acceptance remain uncertain. Aims: To map current evidence on CNN performance for burn TBSA, burn depth and treatment-related tasks and to explore whether a large language model (LLM) can organize extracted findings into a transparent, literature-derived orientation decision tree. Methods: We performed a scoping review following PRISMA-ScR. PubMed, Web of Science and Cochrane were searched on 5 April 2025. Eligible studies reported CNN analysis of 2D burn images and quantitative performance metrics. We summarized reported values descriptively. We then provided a structured summary of extracted findings to ChatGPT to draft a one-page orientation decision tree. Two consultant burn surgeons reviewed the figure for clarity and plausibility. Results: Of 659 records, 24 studies were included. Across studies, reported performance for TBSA and depth assessment was often high, but study designs, datasets, labels, imaging modalities and validation strategies varied substantially. High reported performance does not necessarily imply clinical robustness or real-world accuracy. A single study reported high test-set accuracy for graft versus non-graft using heavily expanded data. This value should not be generalized. Conclusions: CNNs show promise for image-based burn TBSA and depth assessment, but heterogeneity, dataset limitations and limited external validation restrict interpretation and clinical transfer. The LLM-derived decision tree is a literature-synthesis orientation figure, not a clinical decision-support tool. Full article

Transient receptor potential vanilloid 1 (TRPV1) channels are critical mediators of cutaneous allergic inflammation, contributing to pruritus, erythema, and hypersensitivity in allergic skin disorders. Despite their therapeutic potential, clinically available TRPV1 inhibitors remain limited, leaving effective treatment options lacking. Here, we focused on a self-constructed virtual pentapeptide library and identified a highly selective TRPV1 inhibitor that demonstrated pronounced anti-allergic effects in human skin assays. Through structure-based virtual screening of approximately 200,000 peptide conformations, five candidate pentapeptides, especially P5 (DQKNC), exhibited the inhibition. Electrophysiological recordings showed that P5 inhibited TRPV1 currents with nanomolar potency, while exhibiting negligible effects on major cardiac and neuronal ion channels, highlighting its favorable selectivity and safety profile. In capsaicin-induced human skin hypersensitivity tests, topical P5 significantly reduced burning pain, erythema, and pruritus, with simultaneous application providing the most robust relief. These findings reveal a short peptide with strong TRPV1 selectivity and demonstrable efficacy in alleviating skin inflammation and allergic responses, supporting the notion that rationally designed pentapeptides may represent promising topical therapeutics for allergic skin disorders. Plantain (Musa paradisiaca L.) is a tropical monocotyledonous, succulent plant of the Musaceae family commonly grown for food in the tropical regions of the African, Asian, and South American continents, where its parts are also sought for ethnomedicinal purposes in the treatment of burns, inflammation, and diabetes, among others. In the present preliminary exploratory study, the ethanol extract of the underutilized Musa paradisiaca peel (MPE) was evaluated for its in vitro inhibitory effects on α-amylase and α-glucosidase, as well as its in vivo hypoglycemic activity and potential biochemical toxicity. MPE (100, 200, 400 mg/kg) was orally administered to normal experimental rats for 30 days, following which the lipid profile, antioxidant status, and serum/tissue indices of hepatic, renal, and cardiac functions were evaluated. MPE produced significant inhibition (p < 0.05) of α-amylase (37%) and α-glucosidase (46%) at 120 µg/mL in vitro. The effect was lower than that of acarbose (IC50 = 44.4 ± 1.14 and 15.60 ± 0.01 µg/mL, respectively). A modest blood glucose-lowering effect of MPE was observed at the highest tested dose (400 mg/kg) following subacute oral administration. During this treatment period, no biochemical alterations of toxicological importance were caused by MPE, as the organ–body weight ratio and serum/tissue indicators of organ function/damage were not adversely altered. In conclusion, MPE demonstrated inhibitory activity against both α-amylase and α-glucosidase, which may contribute to its potential hypoglycemic effects. Additionally, the findings indicate that the peel extract is non-toxic in rats following sub-acute administration at doses up to 400 mg/kg body weight. Further studies involving diabetic models and chronic exposure will substantiate and extend these preliminary observations. Full article

Aerogels are recognized as exceptional thermal insulation materials, but poor mechanical strength and flammability problems hinder their application in high-temperature environments. Thermal management materials that combine high mechanical strength with superior flame retardancy are, therefore, critically important for thermal insulation. Herein, ultra-lightweight aerogels were facilely fabricated using chitosan (CS) and acidified attapulgite (SATP) as the primary components. The optimal composite, CS-SATP30%, exhibited a compressive strength of 633.15 kPa at 80% strain, demonstrating significant improvement in mechanical properties. Structural analysis revealed that the hydroxyl groups and amino groups of CS molecules formed hydrogen bonds with SATP, ensuring excellent interfacial affinity among the constituents. Compared to pure CS aerogel, the total heat release (THR) and peak heat release rate (PHRR) of CS-SATP30% were substantially reduced to 3.83 MJ/m2 and 37.00 kW/m2, respectively. Furthermore, the limiting oxygen index (LOI) of CS-SATP30% increased to 34% and passed the vertical burning test (UL-94). Background/Objectives: Chronic wounds are considered a silent epidemic, affecting a significant portion of the global population and often leading to severe complications. In particular, wounds resulting from burns or trauma can give rise to squamous cell carcinoma (SCC), a form of skin cancer that arises under chronic inflammatory conditions. This study aims to develop and evaluate pH-responsive gelatin-based hydrogel films incorporating 5-fluorouracil (5-FU) for targeted treatment of SCC in chronic wound environments. Methods: Hydrogel films were formulated using gelatin and loaded with 5-FU. The design leveraged the pH differences between healthy skin and chronic wounds to enable stimuli-responsive drug release. The hydrofilms were characterized by evaluating their surface properties, including transparency, contact angle, and nanoscale morphology. In vitro swelling and dissolution behaviors were analyzed under varying pH conditions. Hemocompatibility was assessed through standard blood interaction assays. Cytotoxicity and selective toxicity were tested using both non-tumoral and tumoral representative skin cell lines. Results: The hydrogel films demonstrated pH-dependent swelling and dissolution, aligning with the neutral and basic environment of chronic wounds. Surface analysis revealed suitable transparency, wettability, and nanoscale uniformity for wound application. In vitro studies showed excellent hemocompatibility. Cytotoxicity assays confirmed good selective toxicity against the A431 skin carcinoma cell line, with minimal effects on healthy cells. Conclusions: The developed gelatin-based hydrogel films exhibit promising characteristics for targeted SCC therapy in chronic wounds. Their pH responsiveness, biocompatibility, and selective antitumor activity support their potential as effective and safe delivery systems. This platform may offer a novel therapeutic approach for managing malignancies arising in non-healing wound environments. Full article

The pulp and paper mill industry in Corner Brook, Newfoundland and Labrador, Canada produces 150 Mg/day of pulp and paper mill sludge (PPMS) with a high moisture content (70–80%). Instead of the current practice of burning, PPMS may be repurposed as vermicompost. However, the optimum population density of Eisenia fetida to maximize the bioconversion process and its influence on population dynamics are largely unknown. The current study aimed at determining how stocking densities affect the vermicomposting time, vermicompost quality, and population dynamics of E. fetida. By using three stocking densities—1.13 g (TL), 2.04 g (TM), and 3.01 g (TH)—the E. fetida per kg of PPMS and the quality of the vermicompost were determined by chemical analysis followed by a germination test and bioassay on Raphanus sativus seedlings. The total vermicompost produced in TL (66.3%) and TM (68.8%) made the bioconversion process quicker in 60 days. The addition of more E. fetida (TH) delayed the process of vermicomposting by up to 90 days (p < 0.003). Overall, 0.87 (TM) E. fetida/L of PPMS was found to be the optimum population density for obtaining the best quantity and quality of vermicompost. The highest earthworm biomass was harvested in TL, followed by TM and TH, as 3.2, 1.3, and 0.9-fold, respectively, compared to the introduced biomass in 60 days (p < 0.008). The mean growth rate (2.6 mg/worm/day), biomass gain (3.49 mg/g), and reproduction rate (3.9 cocoon/worm) were also significantly higher (p < 0.023) in TL compared to TM and TH in 60 days. Therefore, the present study shows the importance of using an optimum stocking density to maximize the bioconversion process in PPMS, especially in cooler regions. Full article
