In the artificial cultivation of Morchella sextelata (Black Morel), exogenous nutrient bags (ENBs) commonly employ wheat grains as the primary substrate raw material. However, this approach is costly and runs counter to the “non-grain” development direction advocated by the edible mushroom industry. Under controlled field conditions, twelve self-made formulations were set up and compared with a conventional market formulation to comprehensively analyze their impacts on the agronomic traits, yield, soil physicochemical properties, and economic benefits of M. sextelata fruiting bodies. The research findings indicate that the nutrient bag formulations have a significant effect on soil available nutrients. Specifically, the contents of alkali-hydrolysable nitrogen (AN) and available potassium (AK) exhibit a significantly negative correlation with M. sextelata yield (r = −0.60, p < 0.05; r = −0.72, p < 0.01, respectively). Among all the treatment groups, the KY1 formulation (comprising 30% wheat grains, 5% rice bran, 60% corncobs, 2% rice husks, 1% lime, and 1% gypsum) achieved the highest yield of 915.13 kg per 667 m2, which was 16.1% higher than that of the control group. The net economic benefit per unit area (667 m2) reached CNY 75,282.15, representing a 20.7% increase compared to the traditional wheat grains-based formulation. In conclusion, partially substituting wheat grains with rice bran in ENBs can not only reduce reliance on staple food resources but also enhance yield and economic efficiency. Due to the differences in cultivated strains and environmental conditions, the impact on morel yield is substantial; therefore, the results of this study need further validation through pilot trials. This study evaluated six agrobyproducts (oak, jujube, apple, pear, peach, black locust) as alternative substrates for Lentinula edodes cultivation to mitigate oak dependency. Twelve substrate formulations were tested, including individual and mixed sawdust combinations. Results demonstrated successful mycelial colonization across all treatments, with treatment PAS (78% pear sawdust, 20% wheat bran, 1.5% gypsum, and 0.5% lime) exhibiting the fastest mycelial growth (4.70 mm/day) and full colonization in 105 days. Treatment BLS (78% black locust sawdust, 20% wheat bran, 1.5% gypsum, and 0.5% lime) achieved the highest biological efficiency (97.26%) and productivity (0.85 kg/bag). Nutrient analysis revealed substrate-specific enhancements: PAS maximized vitamin C (4.88 mg/100 g) and iron, while PAS + OS (39% peach sawdust, 39% oak sawdust, 20% wheat bran, 1.5% gypsum, and 0.5% lime) elevated protein (3.88%), phosphorus, and zinc. PCA highlighted distinct nutritional profiles for BLS- and jujube-based mushrooms. Correlation analyses identified the third (r = 0.838) and fourth flushes (r = 0.922) as critical for total yield, with selenium and zinc significantly linked to growth rates. Black locust and peach substrates outperformed or complemented oak, offering sustainable alternatives. These findings underscore the potential of agrowaste utilization to reduce ecological strain while maintaining high yields and nutritional quality, aligning with global agricultural sustainability goals. Safe antibiotic substitutes are needed given the rise in antimicrobial resistance, environmental contamination, and stringent antibiotic regulations. Insect-derived antimicrobial peptides (AMPs) are promising candidates due to their antimicrobial activity, stability, and safety. This study investigates the antimicrobial mechanism of crude AMP extracts and their physicochemical characteristics in black soldier fly larvae (BSFL). The results indicated that BSFL reared on a wheat bran diet exhibited significantly improved growth performance and AMP production when compared to the other three diets. AMP extracts showed enhanced antimicrobial activity and physicochemical stability, including temperatures and metal ions except Cu+. Moreover, AMP extracts disrupted the cell membrane and inhibited the cell cycle of Staphylococcus aureus (S. aureus), thus exhibiting antimicrobial activity. Furthermore, transcriptomic and KEGG enrichment analyses identified 509 differentially expressed genes (DEGs) related to the Toll and IMD signaling pathways. STRING and GeneMANIA analyses confirmed the association of these pathways with immune response and AMP secretion. qRT-PCR results showed elevated expression of immune genes (GNBP3, NFKBIA, GADD45, and Spz) in BSFL following S. aureus immunization, consistent with RNA-seq findings. These findings offer a valuable reference for using AMPs as antibiotic substitutes in animal feeds and highlight the need for further research on AMP purification and the synergistic regulation of protein synthesis and AMP production in BSFL. Black fungus is the second-most consumed edible fungus in China. The establishment of a risk assessment mechanism for heavy metals in black fungus is particularly critical to the safety of edible fungi. To clarify a risk assessment mechanism of heavy metal pollution of edible fungi in northeast China, in this study, the contents of Pb, Cr, CD and As in 415 samples were determined, and a total of 1660 valid data were obtained. Then, based on Monte Carlo simulation, a non-parametric probability assessment system for heavy metals in black fungus was established and improved. The results showed that the residual amounts of As, Pb, Cd and Cr in black fungus were in the order of Cr > Pb > As > Cd. The background content of four heavy metals in the main raw materials was preliminarily clarified. Among them, the content of As is between 0.010–0.320 mg·kg−1, Pb is between 0.051–0.792 mg·kg−1, Cd is between 0.019–0.236 mg·kg−1, and Cr is between 0.06–3.41 mg·kg−1. These results indicate that the dietary exposure risk of heavy metals ingested by dried black fungus in Chinese minors and adults is basically safe, but at the high exposure levels of 97.5% sites and 99% sites, Cr is at the light pollution level, and the comprehensive pollution of four heavy metals is at the moderate pollution level. In addition, this study found that raw materials can cause heavy metal accumulation in black fungus, mainly from sawdust, followed by rice bran and wheat bran. Full article

Insect larval frass has been proposed as a fertilizer and amendment, but methods for testing its effects on plants are poorly developed and need standardization. We obtained different types of black soldier fly (Hermetia illucens) frass via the factorial combination of (a) two insect diets, as follows: G (Gainesville = 50% wheat bran, 30% alfalfa meal, 20% maize meal) and W (43% sheep whey + 57% seeds); (b) two frass thermal treatments: NT = untreated and T = treated at 70 °C for 1 h. We tested the effects on the germination of cress (Lepidium sativum L.) and wheat (Triticum durum Desf.) by applying 1:2 w:w water extracts at 0, 25, 50, 75 and 100% concentration. Standardizing frass water content before extraction affected chemical composition. Frass extracts showed high electrical conductivity (8.88 to 13.78 mS cm−1). The W diet was suppressive towards Escherichia coli and showed a lower content of nitrates (e.g., WNT 40% lower than GNT) and a concentration-dependent phytotoxic effect on germinating plants. At 25% concentration, germination indices of G were 4.5 to 40-fold those at 100%. Root and shoot length and root hair area were affected by diet and concentration of frass extracts (e.g., root and shoot length in cress at 25% were, respectively, 4.53 and 2 times higher than at 100%), whereas the effects of the thermal treatment were few or inconclusive. On barley (Hordeum vulgare L.) grown in micropots on a silty loam soil, root mass was reduced by 37% at high extract concentration. A quick procedure for root hair surface area was developed based on the geographic information system (GIS) and may provide a fast method for incorporating root hair phenotyping in frass evaluation. The results indicate that below-ground structures need to be addressed in research on frass effects. For this, phyotoxicity tests should encompass different extract dilutions, and frass water content should be standardized before extraction in the direction of canonical procedures to allow comparisons. Beyond their nutritional benefits, vitamins could decrease the risk of chronic diseases due to their potent antioxidant capacity. The present work is aimed at reviewing the state of the art regarding (1) the vitamins involved in oxidative stress prevention in accordance with the requirements established by the European Food Safety Authority (EFSA) and (2) the foods of plant origin that are sources of those vitamins and have potential benefits against oxidative stress in humans. According to the European regulations based on EFSA scientific evidence, riboflavin, vitamin C, and vitamin E are those vitamins subjected to the approved health claim “contribute to the protection of cells from oxidative stress”. Scientific studies conducted in humans with some natural food sources of riboflavin (almonds, wheat germ, mushrooms, oat bran), vitamin C (guava, kale, black currant, Brussels sprouts, broccoli, orange), and vitamin E (hazelnuts, almonds, peanuts, pistachio nuts, extra virgin olive oil, dates, rye) have been performed and published in the literature. However, no food of plant origin has obtained a favorable EFSA opinion to substantiate the approval of health claims related to its potential properties related to oxidative stress prevention. Further studies (concretely, well-controlled human intervention studies) must be carried out in accordance with EFSA requirements to provide the highest level of scientific evidence that could demonstrate the potential relationship between foods of plant origin and antioxidant capacity. This review could be useful for the scientific community to study the application of health claims referring to the antioxidant capacity potentially exerted by foods of plant origin. Full article

Black soldier fly (BSF)-based bioconversion can reduce significant volumes of biosolids and other organic waste while generating high-value BSF larvae (BSFL) and frass. While the mass of frass is greater than the BSFL biomass, its end use is less explored, especially when the bioprocessed waste, such as biosolids, contains high concentrations of contaminants. We assessed the potential to use frass from bioconverted biosolids as fertiliser by analysing chemical parameters and conducting phytotoxicity germination tests. We included frass from bioconverted food waste and wheat bran as comparisons. The chemical composition and phytotoxicity of the frass was related to the type of feedstock. Frass originating from biosolids and from wheat bran had the highest phytotoxicity, which was correlated with increased NH4+-N and EC. Initially, these feedstocks had significantly higher total N compared to food waste. Frass derived from food waste showed the lowest phytotoxicity, which was related to low NH4+-N and EC. This study demonstrates that frass from BSF-based bioconversion could be used as fertiliser; however, the original feedstock will dictate how this by-product has to be used. In this study, frass from food waste was most suitable as fertiliser. Frass originating from bioconverted biosolids needs to be applied at similar rates as unconverted biosolids to avoid phytotoxicity. Full article

Black wheat bran (BWB) is an important source of dietary fiber (DF) and phenolic compounds and has stronger nutritional advantages than ordinary WB. However, the low content of soluble dietary fiber (SDF) negatively influences its physicochemical properties and nutritive functions. To obtain a higher content of SDF in BWB, we evaluated the impact of co-modification by extrusion and enzymes (cellulase, xylanase, high-temperature α-amylase, and acid protease) on water extractable arabinoxylan (WEAX) in BWB. An optimized co-modification method was obtained through single-factor and orthogonal experiments. The prebiotic potential of co-modified BWB was also evaluated using pooled fecal microbiota from young, healthy volunteers. The commonly investigated inulin served as a positive control. After co-modification, WEAX content was dramatically increased from 0.31 g/100 g to 3.03 g/100 g (p < 0.05). The water holding capacity, oil holding capacity, and cholesterol adsorption capacity (pH = 2.0 and pH = 7.0) of BWB were increased by 100%, 71%, 131%, and 133%, respectively (p < 0.05). Scanning electron microscopy demonstrated a looser and more porous microstructure for co-modified BWB granules. Through in vitro anerobic fermentation, co-modified BWB achieved a higher content of Bifidobacterium and Lactobacillus than inulin fermentation. In addition, co-modified BWB induced the highest butyric acid production, indicating high potential as prebiotics. The results may contribute to improving technologies for developing high-fiber-content cereal products. Full article

Various types of functional yogurts were obtained from normalized milk (with normalized lipid content) and a standardized probiotic consortium of probiotic bacteria named ABY3. All the types of yogurts obtained contained prebiotics from black or red rice; malt of barley, rye, wheat; or wheat bran. The physico-chemical analyses of all the functionalized products obtained showed that all of them met the quality standard for yogurt products. However, the sensorial analyses showed that the products obtained from black and red rice were of very good quality. The biological analyses indicated that all the types of products contained live probiotic bacteria, but wheat bran and red rice could increase their numbers. Tests performed on tumor cell line Caco-2 with corresponding postbiotics revealed cytotoxicity greater than 30% after 48 h of exposure in the case of yogurts obtained from milk with 0.8% lipid content and red rice or blond malt of barley. In the case of yogurts derived from milk with 2.5% lipid content, only the variants that contained blond malt of rye or wheat became cytotoxic against the Caco-2 cell line. Full article

The proper treatment of cannabis agricultural wastes can reduce the environmental impact of its cultivation and generate valuable products. This study aimed to test the potential of cannabis agricultural wastes as a substrate for the rearing of black soldier fly larvae (BSFL) and yellow mealworms (MW). In the case of BSFL, replacing the fibre component (straw) in the substrate with the hemp waste can increase the nutritional value of the substrate and led to bigger larvae. The bigger larvae had lower P and Mg, and higher Fe and Ca. Crude protein also varied based on the size of larvae and/or the content of protein in the initial substrate, which was boosted by replacing straw with hemp material. No other cannabinoids than cannabidiolic acid (CBDA), cannabigerolic acid (CBGA), and cannabidiol (CBD) were found in significant amounts in the larvae. In the case of MW, the larvae grew less on the hemp material in comparison to wheat bran. Replacing wheat bran with the hemp material led to smaller larvae with higher Ca, Fe, K, and crude protein content, but lower Mg and P values. No cannabinoids were detected in the MW fed with the hemp material. Full article

In anaerobic soil disinfestation (ASD), soil amended with a carbon source undergoes anaerobic conditions accompanied by changes in microbial community composition and an increase in the concentration of organic acids, primarily acetic and butyric acids, and gases that are deleterious to plant pathogens, insects, and potentially to weeds. The purpose of this study was to explore the efficacy of ASD with different carbon sources on inactivation of propagules of a variety of weed species. Germination and viability of propagules of common lambsquarters (Chenopodium album L.), black nightshade (Solanum nigrum L.), yellow nutsedge (Cyperus esculentus L.), common pokeweed (Phytolacca decandra L.), barnyardgrass (Echinochloa crus-galli L.), dandelion (Taraxacum officinale (Weber)), and redroot pigweed (Amaranthus retroflexus L.) were tested using different rates of four carbon sources: molasses, wheat bran, mustard greens biomass, and raw chicken manure. Wheat bran was the most effective carbon source for inactivation of all weed propagules, followed by molasses and mustard greens biomass. Carbon sources were mixed with soil, which was irrigated to saturation and covered with plastic to promote anaerobic conditions for three weeks. Chicken manure inactivated all the tested species except A. retroflexus compared to the anaerobic control. Rates of carbon sources applied in soil were weakly but significantly correlated (r > 0.42 and p < 0.04) with weed seed mortality or germination inhibition (E. crus-galli) for all the tested carbon sources and weed species except for chicken manure for C. album, A. retroflexus, and S. nigrum. Laboratory tests to confirm organic acid toxicity showed that 1000 µL L−1 acetic plus 1000 µL L−1 butyric acids for at least 60 h inhibited of sprouting of C. esculentus tubers and E. crusgalli seeds, whereas 120 h or exposure was required to inhibit C. album seed germination. Anaerobic soil disinfestation effectively suppressed weed seed germination and viability when wheat bran, molasses and mustard greens biomass were used as carbon sources. This approach could be adopted for weed management in organic systems or any farming system where priority is given to environmentally sustainable practices. Full article

Among the insect species reared as alternative protein sources, Hermetia illucens (black soldier Fly, BSF) has shown a huge potential mostly due to its high protein content, its bioconversion rates, and versatility in using different feeding substrates. Insect rearing may use continuous or batch feeding regimes and, among the used substrates, supermarket feedstock waste has gained recent interest under a circular economy perspective, but several uncertainties remain regarding the heterogeneity and the potential effects of the quantity and quality of these substrates on BSF larvae (BSFL) development. In this experimental work, five replicates of a hundred BSFL were fed in a continuous feeding regime, using seven different isolated vegetables as substrates (wheat bran, pumpkin, apple, grape pomace, red onion, red cabbage, and spinach), at three different temperatures (20, 25, and 30 °C) and two substrate moisture conditions (natural and 70% substrate moisture), until 50% of the larvae achieved the prepupal stage. BSFL performance and bioconversion parameters were evaluated. Our results show that some substrates should be avoided when rearing Hermetia illucens on feedstocks. Among these, apple feed led to poorer and slower development performances with more than 100 days of larval stage, while grape pomace and spinach showed higher mortality rates, which may be due to some anti-nutritional compounds. Larvae fed on pumpkin, red cabbage, and red onion presented good bioconversion results with higher values of efficiency of conversion of digested feed between 14.4 and 25. This work delivers relevant results for black soldier fly reared on a continuous feeding system using vegetable feedstock substrates and their potential trade-offs. Full article

Comparison and Optimization of Different Extraction Methods of Bound Phenolics from Jizi439 Black Wheat Bran

Diet rich in phenolics would potentially associate with multiple health benefits. Response surface methodology (RSM) was introduced to optimize the process of ultrasound- and microwave-assisted extraction of bound phenolics from the bran of a newly developed black wheat breeding line Jizi439 and then compared with the traditional alkaline method. The optimum conditions were found to be 66 °C, 48 min, and power 240 W for ultrasound-assisted extraction (UAE), and 120 s, power 420 W for microwave-assisted extraction (MAE), respectively. Total bound phenolic contents (TBPCs), determined by Folin-Ciocalteu reagent, were 8466.7 ± 240.9 μg gallic acid equivalents per gram (μg GAE/g) bran for UAE and 8340.7 ± 146.7 μg GAE/g bran for MAE under optimized conditions, which were both significantly higher than that of the traditional method (5688.9 ± 179.6 μg GAE/g) (p < 0.05). Antioxidant activities (AAs) were determined by DPPH and ABTS methods. UAE extracts showed the highest DPPH scavenging activity (77.5 ± 0.9%), while MAE extracts showed the highest ABTS scavenging activity (72.1 ± 0.6%). Both were significantly higher than that of the traditional method (69.6 ± 1.1% for DPPH and 65.9 ± 0.5% for ABTS) (p < 0.05). Total bound phenolics (TBPs) profiles were further analyzed by HPLC, and results indicated that ferulic acid was dominant, followed by vanillic acid and p-coumaric acid. The contents of each identified individual phenolics were significantly increased by ultrasound and microwave. In conclusion, UAE and MAE were comparable with each other in TBP yields and AAs; however, when taking operation time and energy consumption into consideration, MAE was more efficient than UAE. Our study suggested efficiency extraction methods for further use of bound phenolics as a healthy food ingredient. Full article

Jizi439, a newly developed black wheat breeding line, was reported to effectively regulate blood glucose, which may potentially be associated with its intrinsic high level of phenolic compounds (PCs). To maximize the PCs yield and thereby enhance their antioxidant activity, orthogonal experiments were designed in sequence for extrusion of Jizi439 black wheat bran (BWB) powder and followed by the extraction of PCs assisted with ultrasound technique. White wheat bran was used as a control. The optimum condition for extrusion was 110 °C, 25% feed water content, 140 rpm screw speed; meanwhile, 50 °C, 40 min, 35 kHz ultrasonic frequency, 300 W ultrasonic power for ultrasound-assisted extraction (UAE). Total phenolic content (TPC) as determined by Folin–Ciocalteu method was 2856.3 ± 57.7 μg gallic acid equivalents (GAE) per gram of dry weight (DW) of phenolic extract; meanwhile, antioxidant activity (AA) in terms of DPPH radical scavenging ratio was 85.5% ± 1.1% under optimized conditions, which were both significantly higher than the control. Phenolic acids except for gallic acid, as well as flavonoids, including luteolin and apigenin were increased by extrusion and ultrasound, as suggested by HPLC results. In conclusion, our study would provide a valuable reference for processing Jizi439 BWB before making or commercially utilize it into health-related food products. Full article

Recent interest in the mass production of black soldier fly (BSF) larvae has resulted in many studies being generated. However, a majority of the studies are benchtop, or small-scale, experiments. Results generated from such studies may not translate to large-scale/industrial production. The current study was conducted at a conventional large-scale (10,000 larvae/treatment fed seven kg) to determine the impact on selected life-history traits when BSF were fed seven kg of manure (swine, dairy, or poultry) or a control diet (Gainesville diet: 50% wheat bran, 30% alfalfa meal, and 20% corn). Results showed larvae fed dairy manure took one to two days longer to develop to prepupation, with lower survivorship (45%) compared to those fed poultry or swine manure (>70%). Furthermore, the maximum larval weight was reached on day six for those fed swine manure, while other treatments achieved the maximum weight on day seven. However, larvae fed swine manure averaged 150 mg, while those fed the other diets ranged between 175 and 200 mg. Data from this study may be valuable for the industrialization of BSF. Companies using a scale varying from previously published work, including this study, should conduct pilot studies to optimize their system prior to implementation. Full article
