The aim of this study was to prevent the development of microorganisms in the refrigerated storage of tench by releasing allyl isothiocyanate (AITC) produced by black mustard seeds. Tench reared in an aquaculture centre were sacrificed and the fillets were separated. Different amounts of defatted mustard seed (300, 400 and 500 mg) were added to hermetic polypropylene trays. Microbiological, sensory, and gas chromatography with MS detection analysis were done. AITC release increased progressively until the third day of storage, significantly delaying the development of microorganisms in samples with higher mustard seed content. The tasting panel detected positive aromas at the beginning of the study, but these decreased and negative aromas appeared. The mustard seed treatment resulted in a higher positive aroma at the end of the storage, reducing rotting and ammonia odours. A total of 31 volatile compounds were detected and grouped into hydrocarbon, alcohol, benzenoid, isothiocyanate, ketone, acetate, aldehyde, and others. Butylated hydroxytoluene, an indicator of bacterial contamination, was the major aromatic compound found during storage. The release of AITC resulted in fewer organic compounds with negative aromas appearing during storage. PCA analysis allowed us to classify the assays during storage according to their volatile profiles, confirming the differences observed between treatments. Thus, adding mustard seed to fish packaging could be a viable alternative to extending the product’s shelf life and ensuring food safety. Full article

Dyes are widely used in various industries, but their removal from wastewater remains a challenge due to their resistance to biodegradation. While substantial research exists regarding the removal of individual dyes, there is much less about the removal of their mixtures. The aim of the research was to determine the possibility of using the immobilised mycelium of Pleurotus ostreatus strains to remove three-component mixtures of dyes from different classes. Efficiency of the removal in the continuously aerated reactor was similar to that obtained in a periodically aerated reactor and was over 90% at the end of each cycle. Despite the addition of subsequent portions of dyes, no increase in the toxicity of post-process samples was observed, and even a decrease in zootoxicity was noticed. The results of the study therefore indicate that an immobilised biomass can be used to remove the dyes, without the need to constantly inject air into the reactor. Full article

To investigate the effect of Botrytis cinerea infection severity on the flavor characteristics of Petit Manseng noble rot wine, this study analyzed wines produced from Petit Manseng grapes grown in the eastern foothills of Helan Mountain, Ningxia, China. The grapes were categorized into three groups based on infection status: uninfected, mildly infected, and severely infected with Botrytis cinerea. Headspace solid-phase microextraction coupled with gas chromatography-mass spectrometry (HS-SPME-GC-MS) and an electronic nose were employed to detect and analyze the aroma components of wines under the three infection conditions. Additionally, trained sensory panelists conducted sensory evaluations of the wine aromas. The results revealed that wines made from severely infected grapes exhibited the richest and most complex aroma profiles. A total of 70 volatile compounds were identified, comprising 32 esters, 17 alcohols, 5 acids, 8 aldehydes and ketones, 4 terpenes, and 4 other compounds. Among these, esters and alcohols accounted for the highest contents. Key aroma-active compounds included isoamyl acetate, ethyl decanoate, phenethyl acetate, ethyl laurate, hexanoic acid, linalool, decanoic acid, citronellol, ethyl hexanoate, and methyl octanoate. Sensory evaluation indicated that the “floral aroma”, “pineapple/banana aroma”, “honey aroma”, and “overall aroma intensity” were most pronounced in the severely infected group. These findings provide theoretical support for the harvesting of severely Botrytis cinerea-infected Petit Manseng grapes and the production of high-quality noble rot wine in this region. Full article

Catalytic and bioelectrocatalytic properties of four white rot fungal laccases (Trametes hirsuta, ThL; Coriolopsis caperata, CcL; Steccherinum murashkinskyi, SmL; and Antrodiella faginea, AfL) from different orthologous groups were comparatively studied in homogeneous reactions of electron donor substrate oxidation and in a heterogeneous reaction of dioxygen electroreduction. The ThL and CcL laccases belong to high-redox-potential enzymes (E0T1 = 780 mV), while the AfL and SmL laccases are medium-redox-potential enzymes (E0T1 = 620 and 650 mV). We evaluated the efficiency of laccases in mediatorless bioelectrocatalytic dioxygen reduction by the steady-state potential (Ess), onset potential (Eonset), half-wave potential (E1/2), and the slope of the linear segment of the polarization curve. A good correlation was observed between the T1 center potential of the laccases and their electrocatalytic characteristics; however, no correlation with the homogeneous reactions of electron donor substrates’ oxidation was detected. The results obtained are discussed in the light of the known data on the three-dimensional structures of the laccases studied. Full article

In chili cultivation, obstacles to continuous cropping significantly compromise crop yield and soil health, whereas crop rotation can enhance the microbial environment of the soil and reduce disease incidence. However, its effects on the diversity of rhizosphere soil microbial communities are not clear. In this study, we analyzed the composition and characteristics of rhizosphere soil microbial communities under chili continuous cropping (CC) and chili–cotton crop rotation (CR) using high-throughput sequencing technology. CR treatment reduced the alpha diversity indices (including Chao1, Observed_species, and Shannon index) of bacterial communities and had less of an effect on fungal community diversity. Principal component analysis (PCA) revealed distinct compositional differences in bacterial and fungal communities between the treatments. Compared with CC, CR treatment has altered the structure of the soil microbial community. In terms of bacterial communities, the relative abundance of Firmicutes increased from 12.89% to 17.97%, while the Proteobacteria increased by 6.8%. At the genus level, CR treatment significantly enriched beneficial genera such as RB41 (8.19%), Lactobacillus (4.56%), and Bacillus (1.50%) (p < 0.05). In contrast, the relative abundances of Alternaria and Fusarium in the fungal community decreased by 6.62% and 5.34%, respectively (p < 0.05). Venn diagrams and linear discriminant effect size analysis (LEfSe) further indicated that CR facilitated the enrichment of beneficial bacteria, such as Bacillus, whereas CC favored enrichment of pathogens, such as Firmicutes. Fusarium solani MG6 and F. oxysporum LG2 are the primary chili root-rot pathogens. Optimal growth occurs at 25 °C, pH 6: after 5 days, MG6 colonies reach 6.42 ± 0.04 cm, and LG2 5.33 ± 0.02 cm, peaking in sporulation (p < 0.05). In addition, there are significant differences in the utilization spectra of carbon and nitrogen sources between the two strains of fungi, suggesting their different ecological adaptability. Integrated analyses revealed that CR enhanced soil health and reduced the root rot incidence by optimizing the structure of soil microbial communities, increasing the proportion of beneficial bacteria, and suppressing pathogens, providing a scientific basis for microbial-based soil management strategies in chili cultivation. Postharvest storage of kiwifruit requires the implementation of precise environmental conditions to maintain fruit quality and reduce decay. In this research, conducted over two years, we examined whether the storage conditions, characterized by low temperature (1 ± 1 °C) and ultra-high relative humidity (higher than 99%, close to saturation), generated by the Xedavap® machine from Xeda International, were effective in maintaining the fruit quality and reducing postharvest rots compared to standard storage conditions, characterized by involved low temperature (1 ± 1 °C) and high relative humidity (98%). Kiwifruits preserved under the experimental conditions exhibited a significantly lower rot incidence after 60 days of storage, with the treated fruits showing 4.48% rot compared to 23.03% under the standard conditions in the first year, using inoculated fruits, and 6.30% versus 9.20% in the second year using naturally infected fruits, respectively. After shelf life (second year only), rot incidence remained significantly lower in the treated fruits (12.80%) compared to the control (42.30%). Additionally, quality analyses showed better parameters when using the Xedavap® system over standard methods. The ripening process was effectively slowed down, as indicated by changes in the total soluble solids, firmness, and titratable acidity compared to the control. These results highlight the potential of ultra-high relative humidity conditions to reduce postharvest rot, extend the shelf life, and enhance the marketability of kiwifruit, presenting a promising and innovative solution for the horticultural industry. Alperujo, a solid by-product from the two-phase olive oil extraction process, poses significant environmental challenges due to its high organic load, phytotoxicity, and phenolic content. At the same time, it represents a promising feedstock for recovering value-added compounds such as phenols and volatile fatty acids (VFAs). When used as a substrate for white rot fungi (WRF), it also produces ligninolytic enzymes. This study explores the use of two native WRF, Anthracophyllum discolor and Stereum hirsutum, for the biotransformation of alperujo under solid-state fermentation conditions, with and without supplementation of copper and manganese, two cofactors known to enhance fungal enzymatic activity. S. hirsutum stood out for its ability to release high concentrations of phenolic compounds (up to 6001 ± 236 mg gallic acid eq L−1) and VFAs (up to 1627 ± 325 mg L−1) into the aqueous extract, particularly with metal supplementation. In contrast, A. discolor was more effective in degrading phenolic compounds within the solid matrix, achieving a 41% reduction over a 30-day period. However, its ability to accumulate phenolics and VFAs in the extract was limited. Both WRF exhibited increased enzymatic activities (particularly Laccase and Manganese Peroxidase) with the addition of Cu-Mn, highlighting the potential of the aqueous extract as a natural source of biocatalysts. Phytotoxicity assays using Solanum lycopersicum seeds confirmed a partial detoxification of the treated alperujo. However, none of the fungi could entirely eliminate inhibitory effects on their own, suggesting the need for complementary stabilization steps before agricultural reuse. Overall, the results indicate that S. hirsutum, especially when combined with metal supplementation, is better suited for valorizing alperujo through the recovery of bioactive compounds. Meanwhile, A. discolor may be more suitable for detoxifying the solid phase strategies. These findings support the integration of fungal pretreatment into biorefinery schemes that valorize agroindustrial residues while mitigating environmental issues. Full article

Root rot, mainly caused by Fusarium oxysporum, is one of the most destructive diseases and leads to significant economic loss of Astragalus membranaceus. To develop an effective strategy for the management of this serious disease, a bacterial strain 2-12 was screened from A. membranaceus rhizosphere soil and identified as Bacillus paralicheniformis based on the phylogenetic analyses of gyrase subunit B gene (gyrB) and RNA polymerase gene (rpoB) sequences. Interestingly, the volatile organic compounds (VOCs) produced by B. paralicheniformis 2-12 exhibited potent antifungal activities against F. oxysporum, as well as fifteen other plant pathogens. Under scanning electron microscopy observation, hyphae treated with the VOCs exhibited abnormal variation such as distortion, twist, and vesiculation, leading to distinctive protoplasm shrinkage. After treatment with B. paralicheniformis 2-12 VOCs, the lesion diameter and disease incidence both reduced significantly compared to control (p < 0.05), thus demonstrating prominent biological efficiency. Moreover, B. paralicheniformis 2-12 VOCs were composed of 17 VOCs, including 9 alkanes, 3 alcohols, 3 acids and esters, 1 aromatic compound, and 1 alkyne compound. A total of 1945 DEGs, including 1001 up-regulated and 944 down-regulated genes, were screened via transcriptome analysis. These DEGs were mainly associated with membranes and membrane parts, amino acid metabolism, and lipid metabolism. The findings in this work strongly suggested that B. paralicheniformis 2-12 VOCs could be applied as a new candidate for the control of A. membranaceus root rot. Full article

Oregano (Origanum vulgare) cultivation is of great economic importance in Peru. Tacna stands out as its main producer. However, the presence of phytopathogenic fungi represents a challenge for its production. This study aimed to characterize both the morphological and molecular levels of the causal agent of crown and root rot in a crop field in the Camilaca district, Candarave, Tacna. To this end, systematic sampling was carried out using the five-gold method, collecting plants with typical symptoms. Fungi were isolated from diseased roots and characterized using macroscopic and microscopic morphological analysis as well as sequencing and multilocus phylogenetic analysis (ITS, 28S, HIS3, TEF1, TUB2). In addition, pathogenicity tests were performed on healthy plants to confirm the infectivity of the isolates. The results demonstrated that root rot was caused by a complex of phytopathogenic fungi through phylogenetic analysis of Dactylonectria torresensis, Fusarium oxysporum, F. iranicum, and F. redolens. These findings represent the first report of these species as causal agents of oregano root rot in Peru, highlighting the need for integrated management strategies that reduce the economic impact of these diseases and contribute to the sustainability of the crop in key producing regions such as Tacna. Chilling injury (CI) frequently occurs in postharvest flat peach fruit during cold storage, leading to quality deterioration and a reduced shelf life. Therefore, investigating the key factors involved in alleviating CI and developing effective preservatives are vital scientific issues for the industry. 2,4-Epibrassinolide (EBR) is a crucial endogenous hormone involved in plant response to both biological and environmental stressors. At present, most studies focus on the mechanisms of mitigating CI using a single concentration of EBR treatment, while few studies focus on the effects varying EBR concentrations have on CI. The purpose of this research is to explore the effects of varying concentrations of EBR on the postharvest quality and cold resistance of peach fruit, thereby establishing a basis for refining a technical framework of environmentally sustainable strategies to mitigate postharvest CI. The results show that EBR treatment effectively inhibits the generation of reactive oxygen species (ROS) and malondialdehyde (MDA) by maintaining the activities of antioxidant enzymes such as superoxide dismutase (SOD), peroxidase (POD), and catalase (CAT), thereby delaying the internal browning process of postharvest peaches. In addition, EBR treatment reduced the consumption of total phenolics by inhibiting the activities of polyphenol oxidase (PPO) and phenylalanine ammonia lyase (PAL). Experimental results identify that 5 μmol L−1 EBR treatment emerged as the most effective concentration for maintaining core postharvest quality attributes. It significantly delayed the decrease in firmness, reduced weight loss, effectively inhibited the production of H2O2 and O2·, particularly during the early storage period, strongly restrained the activity of PAL, and maintained lower rot rates and internal browning indexes. While the 15 μmol L−1 EBR treatment enhanced antioxidant activity, increased total phenolic content at certain stages, and maintained higher soluble solids and acid content, its effects on key physical quality parameters, like firmness and weight loss, were less pronounced compared to the 5 μmol L−1 treatment. Penicillium astrolabium is a primary pathogenic fungus that causes grape blue mold during postharvest, leading to substantial losses in the grape industry. Nevertheless, hypovirulence-associated mycoviruses can attenuate the virulence of postharvest grape-rot pathogens, thereby offering a promising biocontrol tool. Characterizing the mycovirus repertoire of P. astrolabium is imperative for grape protection, yet remains largely unexplored. Here, we screened six strains harboring viruses in 13 P. astrolabium isolates from rotted grapes. Using high-throughput sequencing, four novel dsRNA viruses and two +ssRNA viruses were identified from the six P. astrolabium strains. The dsRNA viruses belonged to two families—Chrysoviridae and Partitiviridae—and were designated to Penicillium astrolabium chrysovirus 1 (PaCV1), Penicillum astrolabium partitivirus 1′ (PaPV1′), Penicillum astrolabium partitivirus 2 (PaPV2), and Penicillum astrolabium partitivirus 3 (PaPV3). For the +ssRNA viruses, one was clustered into the Alphaflexiviridae family, while the other one was clustered into the Narnaviridae family. The two +ssRNA viruses were named Penicillium astrolabium alphaflexivirus 1 (PaAFV1) and Penicillium astrolabium narnavirus 1 (PaNV1), respectively. Moreover, several viral genomic contigs with non-overlapping and discontinuous sequences were identified in this study, which were probably representatives of five viruses from four families, including Discoviridae, Peribunyaviridae, Botourmiaviridae, and Picobirnaviridae. Taken together, our findings could expand the diversity of mycoviruses, advance the understanding of mycovirus evolution in P. astrolabium, and provide both potential biocontrol resources and a research system for dissecting virus–fungus–plant interactions. Complete Genome Analysis and Antimicrobial Mechanism of Burkholderia gladioli ZBSF BH07 Reveal Its Dual Role in the Biocontrol of Grapevine Diseases and Growth Promotion in Grapevines

Burkholderia gladioli is a multifaceted bacterium with both pathogenic and beneficial strains, and nonpathogenic Burkholderia species have shown potential as plant growth-promoting rhizobacteria (PGPRs) and biocontrol agents. However, the molecular mechanisms underlying their beneficial functions remain poorly characterized. This study systematically investigated the antimicrobial mechanisms and plant growth-promoting properties of B. gladioli strain ZBSF BH07, isolated from the grape rhizosphere, by combining genomic and functional analyses, including whole-genome sequencing, gene annotation, phylogenetic and comparative genomics, in vitro antifungal assays, and plant growth promotion evaluations. The results showed that ZBSF BH07 exhibited broad-spectrum antifungal activity, inhibiting 14 grape pathogens with an average inhibition rate of 56.58% and showing dual preventive/curative effects against grape white rot, while also significantly promoting grape seedling growth with increases of 54.9% in plant height, 172.9% in root fresh weight, and 231.34% in root dry weight. Genomic analysis revealed an 8.56-Mb genome (two chromosomes and one plasmid) encoding 7431 genes and 26 secondary metabolite biosynthesis clusters (predominantly nonribosomal peptide synthetases), supporting its capacity for antifungal metabolite secretion, and functional analysis confirmed genes for indole-3-acetic acid (IAA) synthesis, phosphate solubilization, and siderophore production. These results demonstrate that ZBSF BH07 suppresses pathogens via antifungal metabolites and enhances grape growth through phytohormone regulation and nutrient acquisition, providing novel insights into the dual mechanisms of B. gladioli as a biocontrol and growth-promoting agent and laying a scientific foundation for developing sustainable grapevine disease management strategies. Microorganisms in their natural ecological niches are constantly challenged by other inhabitants. Antagonisms exhibited by interacting microbial species are directed towards survival and increasing of their fitness. The Soft Rot Pectobacteriaceae (SRP) is a good model to study these complex microbial interactions. Along with being present in various environments, SRPs are often transferred between environments, allowing the bacteria to encounter members of other species. In this study, we investigated interactions between Dickeya solani, a representative of SRPs and a causative agent of potato soft rot, and Bacillus subtilis, which is known to be a potent producer of secondary metabolites mediating antibiosis. We have found that the soil isolate B. subtilis MB73/2 not only suppresses in vitro soft-rotting of infected potato tubers but is also able to cause directional, coordinated escape of natural isolates D. solani IFB0102 and IPO2222. While this coordinated movement of D. solani depends on surfactin produced by B. subtilis MB73/2, we show that both Dickeya strains exhibit different antagonistic interaction phenotypes toward the competing Bacillus. We prove that this antagonism depends on a single nucleotide polymorphism in one of transcriptional regulators of D. solani belonging to the LysR family. Full article

Maize (Zea may L.) is one of the most important crops worldwide, but ear rot poses a significant threat to its production. Diverse pathogens cause ear rot in China, with Fusarium spp. being predominant, especially Fusarium graminearum and Fusarium verticillioides. Current methods for the control of ear rot are limited, making the use of resistant germplasm resources an effective and economical management strategy. Earlier research focused on resistance to Fusarium ear rot (FER; caused by F. verticillioides) and Gibberella ear rot (GER; caused by F. graminearum), but assessing maize resistance to multiple major Fusarium spp. is critical in ensuring maize production. Thus, the resistance of 343 maize germplasm resources to ear rot caused by six Fusarium spp. (F. verticillioides, F. graminearum, F. proliferatum, F. meridionale, F. subglutinans, and F. temperatum) was evaluated in this study. Over three years, 69 and 77 lines resistant to six and five ear rot diseases, respectively, and 139 lines resistant to both FER and GER were identified. Moreover, the 343 germplasm resources were divided into eight heterotic groups, of which PH4CV was the most resistant one, whereas NSS and Pioneer Female were the least resistant ones. These findings provide a basis for the development of maize cultivars with broad-spectrum ear rot resistance. Full article

Oat sheath rot disease significantly reduces commercial oat yields, yet research on its incidence, causative pathogens, and control strategies remains limited, particularly in China. This study investigated the occurrence of oat sheath rot in major oat-producing regions of Northern China. Here, we isolated and identified two species of primary pathogenic fungi, Scopulariopsis brevicaulis and Alternaria alternata. Next, we conducted pathogenicity tests to confirm their role in the progression of oat sheath rot disease. Subsequently, we screened putative biocontrol fungi and identified Trichoderma harzianum and Trichoderma koningii as effective antagonistic biocontrol fungi. Both species demonstrated strong inhibitory effects against two primary pathogens through competitive interactions, with T. koningii achieving 100% inhibition in one test. Overall, T. harzianum and T. koningii both exerted strong inhibitory effects against pathogenic fungi via different forms of competition. Most importantly, infection experiments showed that T. harzianum and T. koningii both exerted strong antifungal effects against the pathogenic fungi that cause oat sheath rot. Taken together, our findings provide a foundation for developing biological control strategies to mitigate oat sheath rot in oat cultivation in China. Full article
