Brown algae such as Ascophyllum nodosum (AN) and Fucus vesiculosus (FV) are gaining considerable attention as functional feed additives due to their health-beneficial properties. This study evaluated the antioxidant potential of AN and FV extracts in intestinal epithelial cells and the in vivo model Caenorhabditis elegans (C. elegans). Aqueous AN and FV extracts were characterized for total phenolic content (TPC), antioxidant capacity (TEAC, FRAP), and phlorotannin composition using LC-HRMS/MS. Antioxidant effects were assessed in vitro, measuring AAPH-induced ROS production in Caco-2 and IPEC-J2 cells via H2DCF-DA, and in vivo, evaluating the effects of paraquat-induced oxidative stress and AN or FV treatment on worm motility, GST-4::GFP reporter expression, and gene expression in C. elegans. FV exhibited higher total phenolic content, antioxidant capacity (TEAC, FRAP), and a broader phlorotannin profile (degree of polymerization [DP] 2–9) than AN (DP 2–7), as determined by LC-HRMS/MS. Both extracts attenuated AAPH-induced oxidative stress in epithelial cells, with FV showing greater efficacy. In C. elegans, pre-treatment with AN and FV significantly mitigated a paraquat-induced motility decline by 22% and 11%, respectively, compared to PQ-stressed controls. Under unstressed conditions, both extracts enhanced nematode healthspan, with significant effects observed at 400 µg/g for AN and starting at 100 µg/g for FV. Gene expression analysis indicated that both extracts modulated antioxidant pathways in unstressed worms. Under oxidative stress, pre-treatment with AN and FV significantly reduced GST-4::GFP expression. In the nematode, AN was more protective under acute stress, whereas FV better supported physiological function in the absence of stressors. These findings demonstrate that AN and FV counteract oxidative stress in intestinal epithelial cells and in C. elegans, highlighting their potential as stress-reducing agents in animal feed. Allergy is recognized as a public health problem with pandemic consequences and is estimated to affect more than 50% of Europeans in 2025. Prebiotic and probiotic food implementation has recently emerged as an alternative strategy to promote immunomodulatory beneficial effects in allergic patients. Among prebiotics, Phaeophyceae algae represent a niche of research with enormous possibilities. The present study aims to evaluate the in vitro prebiotic potential of fucoidan from Fucus vesiculosus, Macrocystis pyrifera, and Undaria pinnatifida algae, to promote the growth of Limosilactobacillus reuteri and Lacticaseibacillus rhamnosus GG as probiotic bacteria added to the formulation of a novel yogurt. Concentrations of fucoidan of 100 and 2000 µg/mL were added to reference growth media and kinetic growth curves for both microorganisms were fitted to the Gompertz equation. Optimized prebiotic conditions for fucoidan were selected to validate in vitro results by means of the formulation of a novel fermented prebiotic yogurt. Conventional yogurts (including Streptococcus thermophilus and Lactobacillus delbrueckii subs. bulgaricus) were formulated with the different fucoidans, and production batches were prepared for L. rhamnosus and L. reuteri. Increased L. reuteri and L. rhamnosus populations in 1.7–2.2 log10 cycles just after 48 h of in vitro exposure were detected in fucoidan supplemented yogurt. M. pyrifera and U. pinnatifida fucoidans were the most effective ones (500 µg/mL) promoting probiotic growth in new formulated yogurts (during the complete shelf life of products, 28 days). Diet supplementation with fucoidan can be proposed as a strategy to modulate beneficial microbiota against allergy. Full article

The technology of fucoidan extraction significantly affects its properties. This study aimed to evaluate the impact of dynamic maceration (DM) and ultrasound-assisted extraction (UAE) on the antioxidant and anticancer properties of fucoidan from Arctic brown algae. Fucus vesiculosus (Fv), Fucus serratus (Fs), Fucus distichus (Fd), and Ascophyllum nodosum (An) were collected from the Barents Sea. The average yield of fucoidan and uronic acid was higher (by 43.2% and 22.0%, respectively) after UAE, while phlorotannin content decreased by 53.7% compared with DM. The fucose level for all algae increased after UAE, while the molecular weight of fucoidans was lower. The highest antioxidant activity was noted for the fucoidan from An and Fv, which were obtained by DM and can be associated with the high concentrations of phlorotannins. The treatment of HeLa G-63 cells with all studied fucoidans for 48 h increased concentration-dependently the number of dead cells. The most promising were Fv and Fs fucoidans with high phlorotannins, low sulfates, and uronic acid extracted by DM. The co-administration of paclitaxel and fucoidan caused cell cycle arrest in the G2/M phase. The calculated for the first time combinatory effect showed that the simultaneous use of paclitaxel and fucoidan exposure leads to a synergistic interaction. Our results support the rationality of fucoidan use in complex chemotherapy to improve survival, quality of life and immunity in patients with cervical carcinoma. Full article

Natural plant- and algae-based extracts used in crop cultivation offer numerous advantages, including the potential to positively affect plant growth, exhibit hormonal activity, increase stress resistance, improve crop quality as environmentally benign alternatives to synthetic agrochemicals and help combat oxidative stress. The presented experiments aimed to compare the effectiveness of extracts from brown algae such as Ascophyllum nodosum and Fucus vesiculosus, as well as the plant Sideritis scardica, on the germination and initial growth of red kale (Brassica napus var. Pabularia) microgreens. Microgreens treated with aqueous extracts of A. nodosum, F. vesiculosus, as well as the control group, had the highest growth, whereas the lowest growth was observed in plants treated with water–ethanol extracts at the highest tested concentration (10%). The 10% water–ethanol extracts of brown algae reduced plant biomass, while aqueous extracts increased it. Applying water extracts of algae at concentrations (10, 1, 0.1%), as well as the water extract of S. scardica (10, 1%), led to an increase in the total phenolic content in the tested experimental groups. A significant influence on increasing total flavonoid content was noted for water extracts of F. vesiculosus at concentrations ranging from 0.1% to 10%. An opposite effect was observed for the water–ethanol extracts, where the lowest TFC was found in plants grown on mats soaked with 0.1% F. vesiculosus and 1% A. nodosum. All water–ethanol extracts tended to reduce the antioxidant activity of the tested red kale microgreens. In microgreens treated with water extracts of F. vesiculosus at concentrations of 1% and 10%, an increase in antioxidant activity was observed. Examining the impact of plant and algae extracts on kale germination and growth may provide valuable information on ways to improve the quality and health-promoting properties of kale microgreens. Antioxidants are indispensable in protecting the skin from oxidative stress caused by environmental factors such as ultraviolet (UV) radiation, pollution, and lifestyle-related influences. This review examines the essential role of antioxidants in modern cosmetology, highlighting their dual functionality as protective agents and active components in skincare formulations. Oxidative stress, primarily driven by an imbalance between reactive oxygen species (ROS) production and the skin’s defense mechanisms, accelerates aging processes, damages cellular structures, and compromises skin integrity. Antioxidants, whether natural or synthetic, act by neutralizing ROS, reducing inflammation, and promoting cellular repair, effectively mitigating these harmful effects. This comprehensive analysis synthesizes findings from 280 studies accessed via key databases, including PubMed, Scopus, and ScienceDirect. It investigates the biochemical mechanisms of antioxidant activity, emphasizing compounds such as vitamins (C, E, A), carotenoids, polyphenols, peptides, and minerals, alongside bioactive extracts derived from algae, fungi, lichens, and plants. Carotenoids, including ꞵ-carotene, lutein, lycopene, and astaxanthin, demonstrate potent antioxidant activity, making them crucial for photoprotection and anti-aging. Phenolic compounds, such as ferulic acid, resveratrol, hesperidin, and xanthohumol, play a significant role in neutralizing oxidative stress and improving skin health. This review also highlights bioactives from algae, fungi, and lichens. Algae, particularly microalgae like Haematococcus pluvialis, known for astaxanthin production, are highlighted for their extraordinary photoprotective and anti-aging properties. Brown algae (Fucus vesiculosus) and red algae (Porphyra) provide polysaccharides and bioactive molecules that enhance hydration and barrier function. Fungi contribute a wealth of antioxidant and anti-inflammatory compounds, including polysaccharides, ꞵ-glucans, and enzymes, which support cellular repair and protect against oxidative damage. Lichens, through unique phenolic metabolites, offer potent free-radical-scavenging properties and serve as effective ingredients in formulations targeting environmental stress. Plant-derived antioxidants offer a diverse range of benefits. Plant-derived antioxidants, such as flavonoids, phenolic acids, and carotenoids, further amplify skin resilience, hydration, and repair mechanisms, aligning with the growing demand for nature-inspired solutions in cosmetics. The integration of these diverse natural sources into cosmetic formulations reflects the industry’s commitment to sustainability, innovation, and efficacy. By harnessing the synergistic potential of bioactives from algae, fungi, lichens, and plants, modern cosmetology is advancing toward multifunctional, health-conscious, and eco-friendly products. Future research directions include optimizing delivery systems for these bioactives, enhancing their stability and bioavailability, and expanding their applications to meet evolving dermatological challenges. The optimization of bioactive compound extraction from Fucus vesiculosus using ultrasound-assisted extraction (UAE) via sonotrode was investigated to maximize phenolic recovery and antioxidant activity while promoting a sustainable process. Optimal conditions (40% v/v ethanol in water, 38 min, 36% amplitude) were selected to maximize phenolic recovery while considering environmental and energy sustainability by optimizing extraction efficiency and minimizing solvent and energy usage. HPLC-ESI-QTOF-MS analysis tentatively identified 25 phenolic compounds, including sulfated phenolic acids, phlorotannins, flavonoids, and halophenols, with some reported for the first time in F. vesiculosus, underscoring the complexity of this alga’s metabolome. The antioxidant activity of the optimized extract was evaluated through FRAP (143.7 µmol TE/g), DPPH (EC50 105.6 µg/mL), and TEAC (189.1 µmol Trolox/g) assays. The optimized process highlights F. vesiculosus as a valuable source of natural antioxidants, with potential applications in biotechnology, cosmetics, and food industries. Full article

Algae extracts may be a promising alternative to harmful chemicals and pesticides used commercially in the cultivation of plants with higher nutritional and health-promoting values. The cultivation of barley microgreens (Hordeum vulgare L.) was facilitated by the use of aqueous extracts from Fucus vesiculosus algae, which served as a biostimulant. Seeds for experiments were produced in accordance with EU standards, certified as organic and used to grow plants in a controlled pot experiment. A qualitative analysis of the extract, which was used to irrigate the plants, was also performed in this study, as well as stimulating properties by activating the system protecting against oxidative stress. Total phenolic content (TPC), total flavonoid content (TFV) and enzymes involved in their formation such as phenylalanine ammonia lyase (PAL) and polyphenol oxidase (PPO), as well as enzymes involved in the removal of reactive oxygen species such as catalase (CAT) and superoxide dismutase (SOD), were determined in the obtained microgreen samples. Antioxidant activity against DPPH (2,2-diphenyl-1-picrylhydrazyl) was also evaluated. A noticeable increase in SOD content and antioxidant activity against DPPH was observed in barley microgreen samples after extract treatment. These results suggest that the use of extracts of this beneficial alga can enhance the antioxidant activity of the barley microgreens. Full article

Recent agricultural research has prioritized the development of environmentally friendly management strategies to ensure food security, among which the application of biostimulants such as brown algae extracts, arbuscular mycorrhizal fungi (AMF), and their combination are included. The experimental protocol was based on the factorial combination of two planting times (4 May and 1 June) and seven biostimulant treatments (three brown algae species, Cystoseria tamariscifolia—C.t.; Fucus vesiculosus—F.v.; Padina pavonica—P.p.; arbuscular mycorrhizal fungi—AMF; C.t. + AMF; F.v. + AMF; P.p. + AMF) plus an untreated control. The earlier transplant resulted in a higher yield, due to the higher number of fruits per plant, and a higher plant fresh and dry biomass. The treatments with P.p. and F.v. extracts and the combination P.p. + AMF led to the highest yields (56.7 t ha−1), mainly due to the highest fruit number per plant. The earlier planting time led to higher values of dry residue, soluble solids, firmness, and colour component ‘a’. The highest values of fruit dry residue were recorded under the F.v. and P.p. extracts, and the combinations F.v. + AMF and P.p. + AMF, the highest soluble solid content with P.p. treatment, and firmness under P.p. + AMF. The highest levels of ‘L’ and ‘a’ fruit colour components were obtained under the P.p. extract treatment, of ‘b’ upon the application of P.p. and F.v. extract, and AMF + P.p. and AMF + F.v. The later planting time led to significantly higher values of the antioxidant parameters, as did the application of the P.p. extract and P.p. + AMF. CAT activity was more intense corresponding to the later tomato crop cycle, P.p. extract, and AMF + P.p. Overall, our study highlights the potential of biostimulants, particularly brown algae extracts and their combination with AMF, to improve tomato yield, antioxidant properties, and biochemical activities. The global food industry faces a critical challenge in ensuring sustainable practices to meet the demands of a growing population while minimizing environmental impact. At the same time, consumer awareness and the demand for quality products drive innovation and inspire positive changes in the food supply chain. Aiming to create a more sustainable and nutrient-rich alternative, this study is summarized by characterizing the physical and chemical characteristics of algae-enriched chickpea hummus: an innovative approach to popular food products. The algae-enriched hummuses were developed with an incorporation (6% w/w) of Gelidium corneum and Fucus vesiculosus seaweeds and Chlorella vulgaris (hetero and autotrophic) microalgae to reveal their technological potential and evaluate the nutritional and rheological characteristics relative to a control hummus (without algae). From a nutritional perspective, the main results indicated that hummus enriched with microalgae showed an increase in protein content and an improved mineral profile. This was particularly notable for the seaweed F. vesiculosus and the autotrophic microalga C. vulgaris, leading to claims of being a “source of” and “rich in” various minerals. Additionally, the antioxidant activity of hummus containing F. vesiculosus and C. vulgaris increased significantly compared to the control. From a rheological perspective, incorporating algae into the humus strengthened its structure. The microalgae further enhanced the dish’s elasticity and firmness, thus improving this chickpea-based dish´s overall texture and quality. Full article

Newly synthesized cerium oxide was successfully obtained by the hard templating route. The optimal As(III) and As(V) adsorption onto the studied adsorbent was reached for the initial pH of 4.0 and a contact time of 10 h. The highest static adsorption capacities for As(III) and As(V) were 92 mg g−1 and 66 mg g−1, respectively. The pseudo-second-order model was well fitted to the As(III) and As(V) experimental kinetics data. The Langmuir model described the As(III) and As(V) adsorption isotherms on synthesized material. The adsorption mechanism of the studied ions onto the synthesized cerium oxide was complex and should be further investigated. The optimal solid–liquid ratio during the proposed aqueous extraction of inorganic As from the Fucus vesiculosus algae was 1:50. The optimal dosage of the synthesized cerium oxide (0.06 g L−1) was successfully applied for the first time for inorganic As removal from the aqueous algal extract. Full article

Although there are supplements based on algae that claim to aid in weight loss, there is a notable scarcity of scientific evidence supporting their effectiveness, and their regular consumption safety remains inadequately addressed. In this work, commercially available Arthrospira (Spirulina) platensis Gomont and/or Fucus vesiculosus L. supplements showed moderate capacity to inhibit the activity of carbohydrate-metabolizing enzymes, and to scavenge biologically relevant reactive species. IC25 values varying between 4.54 ± 0.81 and 66.73 ± 5.91 µg of dry extract/mL and between 53.74 ± 8.42 and 1737.96 ± 98.26 µg of dry extract/mL were obtained for α-glucosidase and aldose reductase, respectively. A weaker effect towards α-amylase activity was observed, with a maximum activity of the extracts not going beyond 33%, at the highest concentrations tested. Spirulina extracts showed generally better effects than those from F. vesiculosus. Similar results were observed concerning the antiradical capacity. In a general way, the extracts were able to intercept the in vitro-generated reactive species nitric oxide (NO) and superoxide anion (O2•−) radicals, with better results for O2•−scavenging with the spirulina samples (IC25 values of 67.16 and 122.84 µg of dry extract/mL). Chemically, similar pigment profiles were observed between spirulina supplements and the authenticated counterpart. However, fucoxanthin, the chemotaxonomic marker of brown seaweeds, was not found in F. vesiculosus samples, pointing to the occurrence of a degradation phenomenon before, during, or after raw material processing. Our findings can contribute to providing data to allow regulatory entities (e.g., EFSA and FDA) to better rule these products in a way that can benefit society. Full article

Brown macroalgae are rich sources of nutrients and health-promoting compounds. Nevertheless, their consumption is still limited by their strong organoleptic characteristics, thus requiring the development of extraction strategies to profit from their nutritional value. To fulfil this, two sequential extraction approaches were developed, differing in the solvent used in the first extraction step, water in approach 1 or food-grade ethanol in approach 2, to obtain economic and affordable extracts rich in specific compounds from Fucus vesiculosus. The use of water in the first step of extraction allowed us to recover water-soluble phlorotannins, laminarans and mannuronic-rich alginates, making the subsequent 70% ethanol extract richest in fucoxanthin (0.07% algae DW), and the hot water fractions purest in fucoidans and alginates with a lower mannuronic-to-guluronic (M/G) ratio (2.91). Conversely, when beginning extraction procedures with 96% ethanol, the recovered yields of phlorotannins increased (0.43 g PGE/100 g algae DW), but there was a concomitant seven-fold decrease in the recovery of fucoxanthin in the subsequent 70% ethanol extract. This approach also led to less pure hot water fractions containing fucoidans, laminarans and alginates with a higher M/G ratio (5.50). Overall, this work unveiled the potential of the first extraction steps in sustainable and holistic cascade strategies to modulate the composition of food-grade extracts, creating prospects of their application as tailored functional ingredients in food products. Osteosarcoma is a bone cancer primarily affecting teenagers. It has a poor prognosis and diminished quality of life after treatment due to chemotherapy side effects, surgical complications and post-surgical osteoporosis risks. The sulphated polysaccharide fucoidan, derived from brown algae, has been a subject of interest for its potential anti-cancer properties and its impact on bone regeneration. This study explores the influence of crude, low-molecular-weight (LMW, 10–50 kDa), medium-molecular-weight (MMW, 50–100 kDa) and high-molecular-weight (HMW, >100 kDa) fractions from Sargassum filipendula, harvested from the Colombian sea coast, as well as crude fucoidan from Fucus vesiculosus, on a specific human osteoprogenitor cell type, human embryonic-derived mesenchymal stem cells. Fourier transform infrared spectroscopy coupled with attenuated total reflection (FTIR-ATR) results showed the highest sulphation levels and lowest uronic acid content in crude extract from F. vesiculosus. There was a dose-dependent drop in focal adhesion formation, proliferation and osteogenic differentiation of cells for all fucoidan types, but the least toxicity was observed for LMW and MMW. Transmission electron microscopy (TEM), JC-1 (5,50,6,60-tetrachloro-1,10,3,30-tetraethylbenzimi-dazolylcarbocyanine iodide) staining and cytochrome c analyses confirmed mitochondrial damage, swollen ER and upregulated autophagy due to fucoidans, with the highest severity in the case of F. vesiculosus fucoidan. Stress-induced apoptosis-like cell death by F. vesiculosus fucoidan and stress-induced necrosis-like cell death by S. filipendula fucoidans were also confirmed. LMW and MMW doses of <200 ng/mL were the least toxic and showed potential osteoinductivity. This research underscores the multifaceted impact of fucoidans on osteoprogenitor cells and highlights the delicate balance between potential therapeutic benefits and the challenges involved in using fucoidans for post-surgery treatments in patients with osteosarcoma. The brown algal genus Fucus provides essential ecosystem services crucial for marine environments. Macroalgae (seaweeds) release dissolved organic matter, hence, are under strong settlement pressure from micro- and macrofoulers. Seaweeds are able to control surface epibionts directly by releasing antimicrobial compounds onto their surfaces, and indirectly by recruiting beneficial microorganisms that produce antimicrobial/antifouling metabolites. In the Kiel Fjord, in the German Baltic Sea, three distinct Fucus species coexist: F. vesiculosus, F. serratus, and F. distichus subsp. evanescens. Despite sharing the same habitat, they show varying fouling levels; F. distichus subsp. evanescens is the least fouled, while F. vesiculosus is the most fouled. The present study explored the surface metabolomes and epiphytic microbiota of these three Fucus spp., aiming to uncover the factors that contribute to the differences in the fouling intensity on their surfaces. Towards this aim, algal surface metabolomes were analyzed using comparative untargeted LC-MS/MS-based metabolomics, to identify the marker metabolites influencing surface fouling. Their epiphytic microbial communities were also comparatively characterized using high-throughput amplicon sequencing, to pinpoint the differences in the surface microbiomes of the algae. Our results show that the surface of the least fouling species, F. distichus subsp. evanescens, is enriched with bioactive compounds, such as betaine lipids MGTA, 4-pyridoxic acid, and ulvaline, which are absent from the other species. Additionally, it exhibits a high abundance of the fungal genera Mucor and Alternaria, along with the bacterial genus Yoonia-Loktanella. These taxa are known for producing antimicrobial/antifouling compounds, suggesting their potential role in the observed fouling resistance on the surface of the F. distichus subsp. evanescens compared to F. serratus and F. vesiculosus. These findings provide valuable clues on the differential surface fouling intensity of Fucus spp., and their importance in marine chemical defense and fouling dynamics. Full article

Arctic brown algae are considered a promising industrial-scale source of bioactive sub-stances as polysaccharides, polyphenols, and low-molecular secondary metabolites. Conventional technologies for their processing are focused mainly on the isolation of polysaccharides and involve the use of hazardous solvents. In the present study a “green” approach to the fractionation of brown algae biomass based on the dissolution in ionic liquids (ILs) with 1-butil-3-methylimidazolium (bmim) cation with further sequential precipitation of polysaccharides and polyphenols with acetone and water, respectively, is proposed. The effects of IL cation nature, temperature, and treatment duration on the dissolution of bladderwrack (Fucus vesiculosus), yields of the fractions, and their chemical composition were studied involving FTIR and NMR spectroscopy, as well as size-exclusion chromatography and monosaccharide analysis. It was shown that the use of bmim acetate ensures almost complete dissolution of plant material after 24 h treatment at 150 °C and separate isolation of the polysaccharide mixture (alginates, cellulose, and fucoidan) and polyphenols (phlorotannins) with the yields of ~40 and ~10%, respectively. The near-quantitative extraction of polyphenolic fraction with the weight-average molecular mass of 10–20 kDa can be achieved even under mild conditions (80–100 °C). Efficient isolation of polysaccharides requires harsh conditions. Higher temperatures contribute to an increase in fucoidan content in the polysaccharide fraction. Full article
