Everolimus (EVE), an mTOR inhibitor, is widely used in solid organ transplantation (SOT) because of its immunosuppressive properties. Due to its narrow therapeutic window and significant pharmacokinetic variability, therapeutic drug monitoring (TDM) is essential for achieving optimal outcomes. We developed and thoroughly validated a robust LC-MS/MS method to measure EVE levels in venous whole blood (WB) and capillary blood collected using two microsampling devices: Mitra™ (volumetric absorptive microsampling, VAMS) and Capitainer® (quantitative dried blood spot, qDBS). The validation followed EMA and IATDMCT guidelines, assessing linearity (1.27–64.80 ng/mL for WB and 0.50–60 ng/mL for VAMS/qDBS), as well as selectivity, accuracy, precision, matrix effects, recovery, stability, and incurred sample reanalysis. Clinical validation involved 66 matched samples from 33 adult SOT recipients. The method demonstrated high accuracy and precision across all matrices, with no significant carryover or matrix interference. Statistical analysis using Passing–Bablok regression and Bland–Altman plots showed excellent agreement between the microsampling methods and the venous reference. Hematocrit effects were tested both in laboratory conditions and on clinical samples and were found to be negligible. This study provides the first comprehensive analytical and clinical validation of the Mitra and Capitainer devices for EVE monitoring. The validated LC-MS/MS microsampling method supports decentralized, patient-centred TDM, offering a reliable alternative to conventional blood sampling in transplant care. Full article

Conventional methods for the synthesis of porous carbons are typically time- and energy-consuming and often contribute to the excessive accumulation of waste solvents. An alternative approach is to employ environmentally friendly procedures, such as mechanochemical synthesis, which holds great potential for large-scale production of advanced carbon-based materials in coming years. This review covers mechanochemical syntheses of highly porous carbons, with a particular focus on new adsorbents and catalysts that can be obtained from biomass. Mechanochemically assisted methods are well suited for producing highly porous carbons (e.g., ordered mesoporous carbons, hierarchical porous carbons, porous carbon fibers, and carbon–metal composites) from tannins, lignin, cellulose, coconut shells, nutshells, bamboo waste, dried flowers, and many other low-cost biomass wastes. Most mechanochemically prepared porous carbons are proposed for applications related to adsorption, catalysis, and energy storage. This review aims to offer researchers insights into the potential utilization of biowastes, facilitating the development of cost-effective strategies for the production of porous carbons that meet industrial demands. The development of slow-release fertilizers (SRFs) based on production residues is a promising strategy to improve nutrient use efficiency and promote circular economy practices in agriculture. In this study, a series of experimental formulations were designed and tested using pumice scraps, liquid and dried blood, and bone meal, aiming at producing sustainable and low-cost N-P-K SRFs. These were processed through mixing and granulation, both in the laboratory and on a semi-industrial scale. The formulations were evaluated through release tests in 2% citric acid solution simulating the acidic conditions of the rhizosphere, and in acetic acid to assess potential nutrient leaching under acid rain conditions. The results showed a progressive cumulative release of macronutrients (NPKs), ranging from approximately 8% at 24 h to 73% after 90 days for the most effective formulation (WBF6). Agronomic trials on lettuce confirmed the effectiveness of WBF6, resulting in significant biomass increases compared with both the untreated control and a conventional fertilizer. The use of livestock waste and minerals facilitated the development of a scalable product aligned with the principles of sustainable agriculture. The observed release behavior, combined with the simplicity of production, positions these formulations as a promising alternative to conventional slow-release fertilizers. Full article

The authenticity of Protected Designation of Origin (PDO) Feta cheese is critical for consumer confidence and market integrity, particularly in light of widespread concerns over economically motivated adulteration. This study evaluated the potential of Attenuated Total Reflectance–Fourier Transform Infrared (ATR-FTIR) spectroscopy combined with chemometric modeling to differentiate authentic Feta from non-Feta white brined cheeses. A total of 90 cheese samples, consisting of verified Feta and cow milk cheeses, were analyzed in both freeze-dried and fresh forms. Spectral data from raw, first derivative, and second derivative spectra were analyzed using principal component analysis–linear discriminant analysis (PCA-LDA) and Partial Least Squares Discriminant Analysis (PLS-DA) to distinguish authentic Feta from non-Feta cheese samples. Derivative processing significantly improved classification accuracy. All classification models performed relatively well, but the PLS-DA model applied to second derivative spectra of freeze-dried samples achieved the best results, with 95.8% accuracy, 100% sensitivity, and 90.9% specificity. The most consistently highlighted discriminatory regions across models included ~2920 cm−1 (C–H stretching in lipids), ~1650 cm−1 (Amide I band, corresponding to C=O stretching in proteins), and the 1300–900 cm−1 range, which is associated with carbohydrate-related bands. These findings support ATR-FTIR spectroscopy as a rapid, non-destructive tool for routine Feta authentication. The approach offers promise for enhancing traceability and quality assurance in high-value dairy products. Full article

Mabroom dates (Phoenix dactylifera L.) are recognized as one of the most important crops in Qatar. Fresh fruit dates are susceptible to mould and post-harvest spoilage, resulting in a significant financial loss. Octanoic fatty acid (OFA) has been shown to regulate the growth of mould-causing organisms such as fungi and bacteria. It is known to have antibacterial properties. The objective of the current study was to evaluate the in vitro effect of OFA on the post-harvest pathogens of Mabroom fruits. Fresh, apparently healthy, and fully ripe Mabroom dates were obtained from the National Agriculture and Food Corporation (NAFCO). The chosen fruits were packed in sterile, well-ventilated plastic boxes and transported to the lab under controlled conditions. The fruits were distributed into five groups (G1 to G5). The groups G1, G2, and G3 received 1%, 2%, and 3.5% OFA, respectively, while G4 was left untreated and G5 was washed only with tap water as a positive control treatment. Each group contained 200 g of fresh and healthy semi-soft dates. The samples were then dried and incubated in a humidity chamber at 25 °C ± 2 for seven days. The signs and symptoms of decay were monitored and recorded. The presence of pathogens was confirmed via phenotypic and microscopic-based methods. The results showed a significant difference (p ≤ 0.05) among the groups. OFA at 3.5% had the strongest inhibitory action against post-harvest pathogens, followed by OFA2%. However, there were no differences (p ≤ 0.05) between OFA1% and the control groups. Aspergillus spp., Penicillium spp., Rhizopus spp., and Botrytis spp. were most abundant in the control group, followed by OFA2% and OFA1%, respectively. In conclusion, octanoic fatty acid at 3.5% may improve the quality of date fruits through its high antimicrobial activity, reduce the effect of post-harvest decay, minimize the loss of date fruits during storage, and improve the sustainability of date fruits. Further experiments are necessary to confirm the effectiveness of OFA as a green solution for sustainable date fruit production. Full article

A nanoparticle formulation was generated from distiller dried grains with solubles (DDGS), and its effect on the production of anthraquinones (AQs) was evaluated on Rubia tinctorum hairy roots. The DDGS material was washed with water and ethyl acetate to remove mainly the soluble organic/inorganic molecules and reduce the fat content, respectively, followed by an alkaline treatment to remove the polysaccharides. The resulting alkaline solutions were then lyophilized and redispersed in deionized water to generate a monodispersed nanoparticulate formulation (DDGS-NP) with a hydrodynamic diameter and zeta potential of 227 ± 42 nm and −53 ± 7 mV, respectively. The formulation demonstrated good colloidal stability over time, and sterilized DDGS-NPs maintained comparable physicochemical properties. The nanoparticles were enriched in protein fractions, unsaturated fatty acids, and orthophosphate anion components from DDGS, as determined by solid-state Nuclear Magnetic Resonance (NMR), X-ray photoelectron spectroscopy (XPS), organic elemental analysis (OEA), and inductively coupled plasma optical emission spectrometry (ICP-OES) techniques. The DDGS-NPs were tested at different concentrations on Rubia tinctorum hairy roots, in comparison to or in combination with methyl jasmonate (MeJ), for their capacity to induce the production of AQs. All DDGS-NP concentrations increased the production of specific AQs to 7.7 (100 mg L−1), 7.8 (200 mg L−1), and 9.3 µmol/gFW (500 mg L−1), with an extracellular AQ accumulation of 18 µM for the highest DDGS-NP concentration, in comparison with the control hairy roots (~2 µM AQ). The plant growth was not affected at any of the tested nanoparticle concentrations. Interestingly, the combination of DDGS-NPs and MeJ resulted in the highest extracellular AQ accumulation in R. tinctorum root cultures. Full article

Enhanced Protein Extraction from Auxenochlorella protothecoides Through Synergistic Mechanical Cell Disruption and Alkaline Solubilization

Microalgae proteins are increasingly recognized in the food and nutraceutical industries for their functional versatility and high nutritional value. Mild alkaline treatment is commonly used for cell wall degradation and intracellular protein solubilization, consequently enhancing the protein extraction yield. The findings of this study reveal that alkaline treatment alone, even at higher NaOH concentration (up to 0.3 M) and treatment time (up to 90 min), was ineffective (max. 2.4% yield) for the extraction of protein from Auxenochlorella protothecoides biomass. This challenge was significantly reduced through synergistic application of mechanical cell disruption using high-pressure homogenization (HPH) and alkaline solubilization. Single-pass HPH (35 k psi) alone without alkaline treatment led to 52.3% protein solubilization from wet biomass directly harvested from culture broth, while it was only 18.5% for spray-dried biomass. The combined effect of HPH and alkaline (0.1 M NaOH) treatment significantly increased protein extraction yield to 68.0% for a spray-dried biomass loading of 50 g L−1. Through replacing spray-dried biomass with wet biomass, the requirement of NaOH was reduced by 5-fold to 0.02 M to achieve a similar yield of 68.1%. The process integration of HPH with the mild alkaline solubilization and utilization of wet biomass from culture broth showed high potential for industrialization of microalgae protein extraction. This method achieves high extraction yield while reducing alkaline waste and eliminating the need for energy-consuming drying of biomass, thereby minimizing the environmental impact. Full article

Luffa fruit is an underutilized and novel ingredient in North America. To increase the shelf life of luffa fruit, this study evaluated the creation of luffa fruit powder using three different drying temperatures (40 °C, 50 °C, and 60 °C). The objective of this study was to evaluate the sensory properties and acceptability of luffa fruit powder with unfamiliar consumers (those who do not regularly eat luffa fruit). Participants (n = 88) evaluated the luffa fruit powders mixed into couscous, as well as a control (couscous without luffa fruit powder) using check-all-that-apply and hedonic scales. Furthermore, the participants were asked how they felt about luffa fruit powder after evaluating the samples (comment question). The hedonic scores were not significantly affected by the addition of the luffa seed powders dried at different temperatures. However, the luffa seed powder was associated with a mild flavour, as well as being described as earthy and vegetal. The participants did indicate that the luffa seed powder added moistness to the couscous. As the drying temperature increased so did the intensity of the flavour. Furthermore, participants indicated they would be interested in luffa seed powder if it has nutritional benefits. Overall, this study investigated the sensory properties of a novel ingredient, luffa seed powder, and future studies should continue to explore its sensory properties and chemical components. Dried lemon slices (LSs) have become increasingly popular as a healthful beverage when infused in hot water. This study examined the effects of freeze drying (FD), hot air drying (HAD), heat pump drying (HPD), and far-infrared drying (FID) on the quality of dried LSs and their brewed beverages. The results show that FD-LSs and their corresponding beverages have the most appealing appearance and maximum levels of ascorbic acid (2.47 and 0.80 mg/g, respectively), synephrine (8.15 and 0.94 mg/g, respectively), and the overwhelming majority of natural and available phenolic compounds, as well as the strongest antioxidant activity, although numerous volatile compounds in FD-LSs were in the lowest abundances. HPD-LSs exhibited similar trends to FD-LSs but contained the peak concentrations of limonene (2258.87 μg/g), γ-terpinene (704.19 μg/g), β-pinene (502.92 μg/g), and α-pinene (188.91 μg/g), which were the four most abundant volatile compounds in dried LSs. Additionally, active ingredients in HPD-LSs generally featured relative high levels of available amounts. In contrast, HAD- and FID-LSs typically displayed unfavorable coloration and low retention levels of natural and available active ingredients. Consequently, FD and HPD demonstrate superior suitability for the commercial-scale production of dried LSs. Full article

Chronic lung infection is the main predictor of morbidity and mortality in cystic fibrosis (CF), and current pharmacological alternatives are ineffective against Pseudomonas aeruginosa infections. We developed ciprofloxacin (CIP) for inhalation, aiming at improving its solubility through the formation of an amorphous solid dispersion (ASD) using gelatin (GA). CIP and GA were dissolved in varying ratios and then spray-dried, obtaining CIP-GA microspheres in a single step. The dissolution rate, size distribution, morphology, and aerodynamic properties of CIP-GA microspheres were studied, as well as their antimicrobial activity on P. aeruginosa biofilms. Microspheres formulated with a higher GA ratio increased the dissolution of CIP ten-fold at 6 h compared to gelatin-free CIP. Formulations with 75% GA or more could form ASDs and improve CIP’s dissolution rate. CIP-GA microspheres outperformed CIP in eradicating P. aeruginosa biofilm at 24 h. The spray-drying process produced CIP-GA microspheres with good aerodynamic properties, as indicated by a fine particle fraction (FPF) of 67%, a D50 of 3.52 μm, and encapsulation efficiencies above 70%. Overall, this study demonstrates the potential of gelatin to enhance the solubility of poorly soluble drugs by forming ASDs. As an FDA-approved excipient for lung delivery, these findings are valuable for particle engineering and facilitating the rapid translation of technologies to the market. Full article

Microbial contamination is a global concern with impacts on a variety of industries ranging from marine to biomedical applications. Recent research on hydrophilic polymer-based coatings is focused on combining antifouling polymers with nanomaterials to enhance mechanical, optical, and stimuli-responsive properties, yielding colour changing, self-healing, and super hydrophilic materials. This study combines the hydrophilic and antifouling properties of vitamin B5 analogous methacrylamide (B5AMA)-based polymers with stimuli-responsive anthocyanin-dye-loaded cellulose nanocrystals (CNCs) to develop antifouling materials with colour changing capabilities upon bacterial contamination. Poly(B5AMA)-grafted CNCs were prepared through surface-initiated photoiniferter reversible addition fragmentation chain transfer (SP-RAFT) polymerization and characterized through proton nuclear magnetic resonance (1H-NMR), transmission electron microscopy (SEM/TEM), and X-ray photon spectroscopy (XPS) to confirm the formation of surface-grafted polymer chains. The bare CNCs and poly(B5AMA)-grafted CNCs were loaded with anthocyanin dye and evaluated for pH-dependent colour changing capabilities. Interestingly, anthocyanin-loaded CNCs demonstrated vibrant colour changes in both solution and dried film form upon bacterial contamination; however, limited colour changing capabilities of the composites, specifically in dried film form, were attributed to the enhanced dispersibility and antifouling capabilities of the polymer-coated CNCs. Xerostomia, the subjective complaint of a dry mouth, is frequently associated with salivary flow reduction and/or salivary gland hypofunction. This condition significantly impacts an individual’s quality of life and oral health, including difficulties in speaking, chewing, and swallowing. Xerostomia may be caused by autoimmune diseases, xerogenic medications, and radiation therapy. Our objective was to identify differentially expressed proteins in the saliva of patients with medication and autoimmune disease-associated xerostomia compared to non-xerostomic control subjects. Two groups of individuals (N = 45 total) were recruited: non-xerostomic subjects (NX-group; n = 18) and xerostomic patients (XP-group; n = 27). Dried saliva spot samples were collected from major salivary glands, i.e., parotid (left and right) and submandibular glands. Proteomic analysis was performed by deep nanoLC-MS/MS. Differential protein expression in the XP-group relative to the NX-group was determined by the Mann–Whitney U-test with FDR Benjamini–Hochberg correction (padj < 0.05). The Search Tool for Recurring Instances of Neighboring Genes (STRINGv12.0) was used to generate interaction networks and perform pathway analysis. A total of 1407 proteins were detected. Of these, 86 from the left parotid gland, 112 from the right parotid gland, and 73 from the submandibular gland were differentially expressed proteins (DEPs). Using STRING analysis, we identified, for the first time, several neurodegenerative disease-associated networks, primarily involving the downregulation of the 20S proteasome core complex and glyoxalase proteins across salivary glands. In this study, we determined neuronal dysregulation and impaired methylglyoxal (MGO) detoxification, possibly through reduced protein expression of glyoxalase Parkinson’s Disease (PD) Protein 7 (encoded by the PARK7 gene) in major salivary glands of xerostomic patients. Indeed, impaired MGO detoxification has been previously shown to cause salivary gland dysfunction in a mouse model of type 2 diabetes. Based on other DEPs associated with neurodegenerative disorders, our results also suggest a possible deficiency in the parasympathetic nervous system innervation of salivary glands, warranting further investigation. Nutritional Enhancement of Crackers Through the Incorporation of By-Products from the Frozen Pumpkin Industry

The agri-food sector faces the challenge of valorizing by-products and reducing waste. The frozen pumpkin industry generates substantial amounts of by-products rich in nutritional value, especially β-carotene. This study evaluates the nutritional and physical impact of incorporating pumpkin pulp flour (dehydrated and freeze-dried) obtained from by-products into cracker formulation. Crackers were prepared by replacing 10% and 20% of wheat flour with pumpkin flour, assessing the effects based on drying method. Physical parameters (expansion, color, and texture parameters) were measured, in the dough and in the baked products. Furthermore, β-carotene content was analyzed by HPLC-DAD, antioxidant capacity was measured with DPPH, ABTS, and ORAC, and total phenolic content was evaluated with the Folin–Ciocalteu method. Proximate composition and mineral content were also analyzed. Additionally, a preliminary sensory evaluation was conducted with 50 untrained consumer judges to assess acceptability of external appearance, texture, and taste. The inclusion of pumpkin flour significantly increased β-carotene content (up to 2.36 mg/100 g), total phenolics, and antioxidant activity of the baked crackers. Proximate analysis showed a marked improvement in fiber content and a slight reduction in energy value compared to wheat flour. Mineral analysis revealed that pumpkin flours exhibited significantly higher levels of K, Ca, Mg, and P, with improved but not always statistically significant retention in the final crackers. Freeze-dried flour retained more bioactive compounds and enhanced color. However, it also increased cracker hardness, particularly with dehydrated flour. Only the 10% freeze-dried formulation showed mechanical properties similar to those of the control. Sensory analysis indicated that all formulations were positively accepted, with the 10% freeze-dried sample showing the best balance in consumer preference across all evaluated attributes. Frozen pumpkin by-products can be effectively valorized through their incorporation into bakery products such as crackers, enhancing their nutritional and functional profile. Freeze-drying better preserves antioxidants and β-carotene, while a 10% substitution offers a balance between nutritional enrichment and technological performance and sensory acceptability. Aim: To prepare two different kinds of wound care creams containing plant extracts and examine their effectiveness in comparison with a placebo cream and a commercial wound care cream, Madecassol®. Methods: The two cream formulations were developed using the same placebo cream (PC) as base cream. One formulation contained balsam of oriental sweet gum, or Levant storax, named as Levant Storax Cream (LSC); the other contained oil of Calendula, extract of St. John’s Wort, aescin (an extract of horse chestnut), and freeze-dried powder from Aloe vera (L.) Burm. f. leaf juice, designated as Complex Cream (CC). In the characterization of the creams, organoleptic properties, pH, viscosity, size distribution, and zeta potential of oil globules were measured. Furthermore, the stability of the creams was assessed under different environmental conditions. In vitro studies were performed by using an excisional wound model in rats to assess the potential of the creams for stimulating wound healing. The efficacy of LSC and CC was compared with a commercial reference cream, Madecassol® (M), and the placebo control. The study was also designed with a negative control group of rats that were not treated but handled the same way as the other treatment groups. The wound contraction rate, total skin thickness recovery, and results of histopathological parameter examinations were used to compare the effectiveness of the treatments. Results: The stability of formulated creams confirmed that they were stable for the duration of the study. In vivo studies showed that rats treated with LSC achieved the highest wound healing rates when compared with the other groups. A better response was recorded for the CC-treated population when compared to both control and placebo groups, but there was no significant difference seen in healing score between CC and M groups. Impact of Spectral Irradiance Control on Bioactive Compounds and Color Preservation in Solar-Dried Papaya

The quality effects of spectral irradiance conditions during papaya (Carica papaya L.) drying were investigated using three different dryers: a solar dryer with dynamic irradiance control (SDIC), a cylindrical solar dryer (CSD), and a solar simulator dryer (SSD). This study builds upon previous PDLC film applications in solar drying by specifically examining its impact on phytochemical preservation and color degradation, addressing gaps in spectral-specific effects on food quality parameters. The drying conditions were as follows: a temperature of 50 °C for each method, 700 w/m2 for both SDIC and solar simulator dryers (SSD), and full solar irradiance for the cylindrical solar dryer (CSD). The cylindrical solar dryer exhibited 210 min of drying time due to higher solar irradiance than SDIC (300 min), while SSD lasted 180 min. Drying rates were highest for CSD (0.056 g H2O/g d.m. min−1), followed by SDIC (0.027 g H2O/g d.m. min−1). Color analysis revealed that CSD resulted in the most significant color degradation, followed by SSD and SDIC. This was attributed to the varying spectral composition of radiation in each method. The CSD, with a full solar spectrum, including higher UV and visible radiation, induced more pronounced color changes than SDIC, which received lower intensity radiation in these ranges. Chemical analyses showed that SSD samples had the highest antioxidant activity (1432.91 µmol TE/g dw by ABTS) and phenolic content (58.92 mg GAE/100 g), suggesting simulated conditions may better preserve certain phytochemicals. SDIC maintained better carotenoid-related color parameters while showing intermediate antioxidant levels (1084.09 µmol TE/g dw). These results demonstrate that irradiance control significantly impacts drying efficiency and quality parameters. Full article
