Background: Non-small cell lung carcinoma (NSCLC) is the most prevalent form of lung cancer, and its progression is closely associated with constitutive activation of signal transducer and activator of transcription 3 (STAT3). This study used surface plasmon resonance (SPR) technology to develop a STAT3-targeting recognition system and identify natural STAT3-targeting compounds from the traditional Chinese medicine Psoralea corylifolia and to evaluate their anti-NSCLC activities, with particular attention to reactive oxygen species (ROS) regulation. Methods: The SPR biosensor immobilized with STAT3 was used to screen and enrich STAT3-binding constituents of Psoralea corylifolia, and to determine ligand-STAT3 affinities. Molecular docking was performed to characterize interactions within the STAT3 SH2 domain. Functional effects were assessed in A549 cells using proliferation and scratch migration assays. Antioxidant capacity was evaluated via hydroxyl radical and superoxide anion scavenging assays, and intracellular ROS levels were measured in hydrogen peroxide (H2O2)-induced oxidative stress models in human umbilical vein endothelial cells (HUVECs) and A549 cells. Results: SPR analysis showed that psoralen and isopsoralen bind to STAT3, with equilibrium dissociation constants (KD) of 80.92 µM and 28.11 µM, respectively. Molecular docking further confirmed their interaction with the STAT3 SH2 domain. Both compounds inhibited A549 proliferation and reduced migration. Beyond direct STAT3 inhibition, both compounds demonstrated notable free radical scavenging activity. In a H2O2-induced oxidative stress model, pretreatment with psoralen or isopsoralen significantly reduced ROS levels in HUVECs, while increasing ROS accumulation in A549 lung cancer cells. Conclusions: This work identifies psoralen and isopsoralen as novel dual-function STAT3 inhibitors that exert anti-NSCLC effects through combined STAT3 suppression and context-dependent ROS modulation, and demonstrates the utility of SPR for screening bioactive natural products. By analyzing the influence of the titanium–sapphire (Ti:S) crystal thermal effect on the laser resonator during the generation of a 689 nm laser, the thermal characteristics of the Ti:S crystal operating near the gain edge were investigated in this letter. On this basis, a Ti:S laser with high conversion efficiency suitable for operation at the wavelength of 689 nm was designed. Benefiting from the quantification of thermal effects, the beam waist size at the center of the Ti:S crystal was precisely controlled. Finally, a single-frequency continuous-wave 689 nm laser with an output power of 3.65 W was achieved, and the corresponding optical-to-optical conversion efficiency was up to 23.1%. Then, after locking the transmission peak of the inserted etalon to the resonance frequency of the resonator, the continuous-frequency tuning range of 17 GHz around 689 nm was realized by scanning the voltage applied to the piezoelectric transducer (PZT) mounted on the cavity mirror. Furthermore, based on the realized single-frequency continuous-wave tunable 689 nm laser source, the absorption spectra of strontium atoms near 689 nm were obtained, which established a promising method for preparing 689 nm laser sources designed for strontium atomic ensembles. This study first assessed the friction and wear properties of two polytetrafluoroethylene materials sliding against electroplated chrome and high-velocity oxy-fuel-sprayed WC-10Co-4Cr coatings. Subsequently, the sealing performance of three different structure seals made from these two polytetrafluoroethylene materials was investigated on both electroplated chrome and high-velocity oxy-fuel-sprayed WC-10Co-4Cr coatings. The study results indicate the following: in terms of changes in the counter-face surface roughness, both the electroplated chrome and high-velocity oxy-fuel-sprayed WC-10Co-4Cr surfaces exhibited an increase in surface roughness after sliding, demonstrating the phenomenon of “soft material wearing hard material.” Moreover, the changes in surface roughness were greater after sliding against wollastonite mineral-filled polytetrafluoroethylene than against polyether ether ketone-filled polytetrafluoroethylene, indicating that wollastonite mineral-filled polytetrafluoroethylene was more likely to cause damage to the metal surface. Regarding the friction coefficient and wear amount, under dry friction conditions, both materials exhibited higher friction coefficients but lower wear rates on high-velocity oxy-fuel-sprayed WC-10Co-4Cr surfaces, while showing lower friction coefficients but higher wear rates on electroplated chrome surfaces. This behavior was related to the ease of transfer film formation and the stability of the transfer films formed by polytetrafluoroethylene materials on the two surfaces. In terms of the products’ sealing performance, test results showed that, for composite seals with polytetrafluoroethylene as the counter-face, sealing performance was better on high-velocity oxy-fuel-sprayed WC-10Co-4Cr surfaces than on electroplated chrome surfaces. For seals with rubber as the counter-face, there was little difference in sealing performance between high-velocity oxy-fuel-sprayed WC-10Co-4Cr and electroplated chrome surfaces. Full article

Although particulate matter has been associated with sleep problems, the effects of PM2.5-bound organophosphate esters (OPEs) on children’s sleep remain unclear. OPEs have neurotoxic and endocrine-disrupting effects that may disrupt sleep–wake regulation during neurodevelopment, supporting biological plausibility for sleep impacts. In this study, we quantified the individual and mixture effects of PM2.5-bound OPEs on the sleep disorder domain. This cross-sectional study included 110,169 children aged 6–18 years from primary and secondary schools in the Pearl River Delta (PRD), China. Sleep disorders were evaluated using the validated Sleep Disturbance Scale for Children (SDSC). Elastic net and mixed effect models identified specific OPE–sleep associations, while weighted quantile sum regression evaluated mixture effects. All odds ratios indicate a change in the likelihood of sleep disorders per interquartile range (IQR) increase in OPE concentrations. The strongest individual associations were observed for TDCIPP with short sleep duration (OR = 1.56–1.61; moderate association), TEHP with short sleep duration (OR = 1.59–1.64; moderate association), and TPHP with overall sleep disorder (OR = 1.32–1.42; modest association). Combined OPE exposure was positively associated with all sleep disorder domains (ORs = 2.02–2.85; moderate-to-large associations). These results indicate that inhaling PM2.5-bound OPE mixtures could negatively impact children’s sleep health. This emphasizes a critical developmental period and highlights the importance of public health concerns related to emerging airborne contaminants. Full article

Against the backdrop of China’s “Dual Carbon” strategy (peak carbon emissions and carbon neutrality), timber forests serve the dual function of wood supply and carbon sink enhancement. In this study, we employed the Kuenm package in R to optimize Maximum Entropy model (MaxEnt) parameters. Based on the distribution data of six timber tree species in Sichuan Province and 43 environmental factors, we utilized the MaxEnt outputs and ArcGIS 10.8 software to map the geographic distribution of the suitable habitats for these species from the present day into the future (2061–2080) under different climate scenarios (SSP126 and SSP585). Furthermore, we analyzed the migration trend of their future distribution centers. The model optimization significantly improved both fit and predictive performance, with AUC values ranging from 0.8552 to 0.9637 and TSS values ranging from 0.6289 to 0.84, indicating high predictive capability and stability of the model. Analysis of environmental factors, including altitude, precipitation, and temperature, revealed that altitude plays a dominant role in species distribution. Future climate scenario simulations indicated that climate change will significantly alter the distribution of suitable habitats for these timber tree species. The suitable areas for some species contracted, with changes being particularly pronounced under the SSP585 scenario, in which the high-suitability area for Phoebe zhennan is projected to increase from 12,788 km2 to 20,004 km2, whereas the high-suitability area for Eucalyptus robusta is expected to contract from 8706 km2 to 7715 km2. The migration distances of suitable habitats for timber tree species in Sichuan range from 5 km to 101 km southwestward under different climate scenarios, and these shifts are statistically significant (p < 0.01), with shifts in elevation and precipitation patterns, reflecting species-specific responses to climate change. This study aims to predict future suitable habitats of timber tree species in Sichuan, providing scientific support for forestry planning, forest quality improvement, and climate risk mitigation. The Hedgehog (Hh) signaling pathway is a key regulator of adipogenesis and lipid metabolism. However, the specific role of its receptor, Patched2 (Ptch2), in these processes remains unclear. Here, using a CRISPR/Cas9-mediated ptch2 homozygous mutation model in Nile tilapia (Oreochromis niloticus), we found that Ptch2 deficiency induced visceral and perirenal lipomatosis characterized by small, multinucleated adipocytes. Comparative adipose transcriptomics revealed pronounced adipogenic reprogramming, with marked upregulation of genes governing de novo lipogenesis (e.g., acaca, fasn), fatty acid desaturation (e.g., scd, fadsd6), and triglyceride synthesis (e.g., dgat2, lpl). Biochemically, mutants exhibited elevated blood glucose and liver transaminases (alanine aminotransferase, aspartate aminotransferase) activity, and reduced alkaline phosphatase activity, indicating systemic metabolic dysregulation and hepatic stress. Our findings demonstrate that loss of Ptch2 triggers lipoma formation and adipogenic transcriptome reprogramming, highlighting its essential role in maintaining adipose tissue homeostasis. Traditional homogeneous copper foils suffer from a trade-off between strength and ductility, while gradient or heterogeneous structures are mostly based on deformation processing, making it difficult to achieve controllable construction within a thickness of ≤10 μm. This study aims to directly construct a layered structure with a “fine–coarse–fine” (A-B-A) gradient grain distribution, denoted as 3L-ABA in an 8 μm copper foil via direct current electrodeposition, which utilizes composite additives to regulate electrochemical polarization and nucleation modes. Through systematic characterization and mechanical testing, it was found that the 3L-ABA copper foil exhibits a tensile strength of 604 ± 18 MPa, an elongation of 3.6 ± 0.25%, and low surface roughness Rz of 0.46 μm. Microscopic mechanism analysis demonstrates that the gradient structure achieves synergistic strengthening and toughening through surface fine-grain strengthening, intermediate coarse-grain coordinated plastic deformation, combined with dislocation density and twin strengthening. Electrochemical tests confirm that Additive A (containing collagen, bis-(3-sulfopropyl)-disulfide (SPS), thiourea and 2-mercapto-5-benzimidazolesulfonic acid sodium salt (2M5S)) induces strong cathodic polarization, promoting instantaneous nucleation and grain refinement, whereas Additive B (containing collagen and bis-(3-sulfopropyl)-disulfide (SPS) shows weaker polarization and promotes grain growth. This research provides a scalable electrodeposition solution for the microstructural design and performance regulation of ultra-thin copper foils. Full article

In this paper, a new type of actuator was presented. The actuator has two modes, stepping and scanning modes. The stepping mode can achieve large stroke through motion accumulation, while the scanning mode can enable high-resolution driving. The mechanical structure and the operating principles of the actuator were described. The structure and the model of the clamping units and driving unit were designed by theoretical modeling and finite element simulation analysis. Finally, a prototype was fabricated, and the experiment results revealed that the actuator achieves a maximum speed of 367 μm/s and a resolution of 0.02 μm. Full article

Background/Objectives: The rhizome of Gastrodia elata Blume (Tianma) is a functional food with medicinal value in China, used to improve the health of the central nervous system and reported to exhibit anti-cellular senescent activity. P-hydroxybenzaldehyde (P-HBA) is a key aromatic compound isolated from Tianma; however, its potential to mitigate cellular senescence remains unclear. Methods: We employed ultra-performance liquid chromatography-mass spectrometry to identify the chemical characterization of Tianma extract. Cell viability assay, senescence-associated-β-galactosidase (SA-β-Gal) assay, and immunofluorescence staining and autophagy analysis were used to evaluate the anti-senescent activity of P-HBA and other Tianma components. Results: Our findings demonstrate that Tianma methanol extract (TME) and P-HBA significantly reduce cellular senescent inducer hydroxyurea (HU)-induced DNA damage, SA-β-Gal activity increase, and autophagic dysfunction in human SH-SY5Y cells. Notably, an autophagy inhibitor, chloroquine, can reduce anti-cellular senescent activity of P-HBA. Conclusions: These results suggest that P-HBA exhibits the effect of reducing cellular senescent phenotypes, and its effect is achieved by enhancing autophagy. Full article

Magmatic activity is crucial for identification of the tectonic framework of the ancient oceanic crust. In this study, systematic investigation, including a field survey, zircon LA-ICP-MS U-Pb dating, and whole-rock geochemical analysis, has been carried out on the intrusive quartz- and granodiorites within the Meso-Tethyan Shiquanhe Ophiolitic Mélange (SQM), Tibet. Zircon U-Pb dating yields the weighted mean ages of 174.7 ± 1.4 Ma (quartz diorite) and 178.9 ± 1.2 Ma (granodiorite), respectively, demonstrating the Early Jurassic formation age. The quartz diorite samples are metaluminous (A/NKC = 0.77–0.95) (molar/Al2O3/(CaO + Na2O + K2O)), while the granodiorite samples are weakly peraluminous (A/NKC = 0.95–1.21), and both of them exhibit tholeiitic to calc-alkaline geochemical characteristics and can be classified as I-type granites. The right-dipping rare-earth element (REE) patterns, enrichment in large ion lithophile elements (LILEs: Rb, Ba, Th), and depletion in high-field-strength elements (HFSEs: Nb, Ta, Ti), as well as relatively high (La/Yb)N ratios, are features compatible with an island arc setting. Combined with previous works, we suggest that the Shiquanhe ophiolitic mélange not only preserves records of mid-late Jurassic island arc magmatic activity but also contains evidence of island arc magmatism from the late Early Jurassic. Fast-response liquid crystal (LC) displays have attracted attention for use as gaming displays with high frame rate and field-sequential displays. This work presents an LC display mode with vertical alignment and horizontal electric field driving, which achieves millisecond-scale response time. The proposed LC display mode may achieve an average grayscale-to-grayscale response time of 1.42 ms using low-rotational-viscosity LC material, optimized device architecture, and overdrive, offering a potential application for gaming displays and color-sequential displays. This study analyzed regional and sectoral PM2.5 sources across 17 Shandong cities during the 2017–2018 winter heating season, which is selected because it is representative of severe air pollution with an average PM2.5 of 65.75 μg/m3 and hourly peak exceeding 250 μg/m3. This air pollution episode aligned with key control policies, where seven major cities implemented steel capacity reduction and coal-to-gas/electric heating, as a baseline for evaluating emission reduction effectiveness. The particulate matter source apportionment technology in the Comprehensive Air Quality Model with extensions (CAMx) was applied to simulate the source contributions to PM2.5 in 17 cities from different regions and sectors including industry, residence, transportation, and coal-burning power plants. The meteorological fields required for the CAMx model were generated using the Weather Research and Forecasting (WRF) model. The results showed that all cities besides Dezhou city in Shandong Province contributed PM2.5 locally, varying from 39% to 53%. The emissions from Hebei province have a large impact on the PM2.5 concentrations in Shandong Province. The non-local industrial and residential sources in Shandong Province accounted for the prominent proportion of local PM2.5 in all cities. The contribution of non-local industrial sources to PM2.5 in Heze City was up to 56.99%. As for Zibo City, the largest contribution of PM2.5 was from non-local residential sources, around 56%. Additionally, the local industrial and residential sources in Jinan and Rizhao cities had relatively more contributions to the local PM2.5 concentrations compared to the other cities in Shandong Province. Finally, the emission reduction effects were evaluated by applying different reduction ratios of local industrial and transportation sources, with decreases in PM2.5 concentrations ranging from 0.2 to 26 µg/m3 in each city. Disulfidptosis is a recently identified form of regulatory cell death (RCD), Which has emerged as a research hotspot due to its distinctive feature of accumulating protein disulfide bonds, setting it apart from other RCD mechanisms. This discovery may offer new therapeutic strategies for cancer and various chronic diseases. This review aims to summarize the molecular mechanisms, inhibitors, regulatory networks, distinctions and connections between disulfidptosis and other regulatory death pathways, and the application of disulfidptosis in tumors and other chronic diseases. It also identifies unresolved issues and provides an outlook on future prospects. Full article

Rheumatoid arthritis (RA) is a chronic autoimmune disorder characterized by persistent synovitis, in which fibroblast-like synoviocytes (FLSs) serve as the primary effector cells that drive the destruction of joints. Baicalin has previously demonstrated efficacy in significantly ameliorating joint symptoms in rats with CIA. As such, this study aims to investigate its underlying molecular mechanisms and impact on the FLSs of rats with CIA through an integrated proteomics and transcriptomics analysis. A Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis was conducted based on two datasets; it revealed that the retrograde endocannabinoid signaling pathway—associated with susceptibility to RA—is the only one involved in both the signaling and metabolic processes modulated by baicalin. Nineteen differentially expressed proteins (DEPs) downregulated by baicalin comprise seventeen subunits of NADH dehydrogenase and two receptors, glutamate receptor 2 (GRIA2) and γ-aminobutyric acid receptor subunit alpha-5 (GABRA5). Three differential metabolites (DMs) were also affected by baicalin: γ-aminobutyric acid (GABA) and phosphatidylcholine (PC) were upregulated and phosphatidylethanolamine (PE) was downregulated. Our findings suggest that the baicalin-mediated alleviation of joint synovitis is closely related to the upregulation of GABA and PC; downregulation of GRIA2, GABRA5, and PE; and preservation of mitochondrial homeostasis within the retrograde endocannabinoid signaling pathway in FLSs.
