Impact of Donor Age on Graft Failure After Deceased Donor Liver Transplantation by Donor-Recipient Sex Combinations: An Analysis of the UNOS OPTN Database

Background Sex disparity has been highlighted in personalized medicine for various human diseases including acute/chronic liver diseases. In the transplant community, greater graft failure risk in female-to-male liver transplantation (LT) has been repeatedly reported, and a recent study in living donor LT reported that the inferiority of female-to-male LT is shown only when donor age is ≤40 y. We aimed to analyze the United Network for Organ Sharing (UNOS) database to test if the poorer outcome of female-to-male LT changes by donor age of 40 y in deceased donor LT, as shown in living donor LT. Methods In this retrospective cohort study, 11,752 adult patients in the UNOS registry who underwent deceased donor LT between 2000–2023 were analyzed. Multivariable analysis was performed to adjust the effects from transplant years, graft ischemia time, disease severity, and others. The primary outcome was graft failure. Results Within the subgroup of recipients with ≤40 y donors, graft failure risk was significantly greater in female-to-male LT than others (vs. female-to-female, HR = 1.43 [1.16–1.76], p < 0.001; vs. male-to-female, HR = 1.46 [1.18–1.81], p < 0.001; vs. male-to-male, HR = 1.26 [1.16–1.49], p = 0.009). In contrast, within the subgroup of recipients with >40 y donors, the risk was comparable between female-to-male LT and other donor-recipient sex groups (vs. female-to-female, p = 0.907; vs. male-to-female, p = 0.781; vs. male-to-male, p = 0.937). We tested various cutoff donor ages and determined that 40 y is the best cutoff value to define the risk subgroup in female-to-male LT. Conclusions In the current study, we found that the sex disparity shown in living donor LT is also observed in deceased donor LT. That is, post-transplant graft failure risk was greater in female-to-male LT than other donor–recipient sex groups only when donor age was ≤40 y. In contrast, graft failure risk was comparable irrespective of donor-recipient sex combinations when donor age was >40 y. Full article

Background/Objectives: Delayed gastric emptying (DGE) is a well-known complication of laparoscopic pylorus-preserving gastrectomy (LPPG). Patients who underwent LPPG in the KLASS-04 trial, which was a multicenter prospective randomized control trial comparing LPPG and laparoscopic distal gastrectomy (LDG), showed an unneglectable incidence of long-term DGE compared to patients who underwent LDG. This study aimed to identify the multifactorial risk factors associated with DGE and to analyze the quality of life (QoL) of patients with DGE following LPPG. Methods: DGE was defined as “nearly normal diet residue” at least once in the endoscopic follow-up at 1, 2, and 3 years after the surgery. Clinicopathological features, surgical outcomes, and QoL were compared between the DGE and non-DGE groups. Results: DGE was observed in 21/124 patients (16.3%) who underwent LPPG. Patients without previous abdominal surgery had a higher incidence of DGE in the univariate (32% vs. 4.8%, p = 0.011) and logistic regression analyses (odds ratio: 0.106, 95% confidence interval: 0.014–0.824, p = 0.032). Patients with DGE reported more symptoms of nausea and vomiting (p = 0.004), constipation (p = 0.04), and a dry mouth (p = 0.005). Conclusions: Despite the strict protocol used to avoid well-known risk factors for DGE, such as damage to the hepatic branch of the vagus nerve, infrapyloric artery and vein, and short antral cuff, the LPPG group of the KLASS-04 trial exhibited a considerable incidence of DGE. No clinicopathological or surgical factors, other than the absence of a previous surgical history, were identified as multifactorial risk factors for DGE. However, DGE had a negative impact on the QoL of patients. Full article

The Inula japonica flower is traditionally used to alleviate lung inflammatory symptoms. While the therapeutic effect of the I. japonica flower on lung diseases has been suggested, the efficacy of the I. japonica flower in treating atopic dermatitis (AD) remains unknown. We investigated the effects of a water extract of the I. japonica flower (WEIF) on Dermatophagoides farinae extract (DfE)-induced AD-like inflammation in NC/Nga mice. Histological analysis of the epidermal structure, mast cell infiltration, and barrier protein expression were examined. Serum inflammatory mediator levels were assessed. To elucidate the regulatory pathway of WEIF, the effects of 1,5-dicaffeoylquinic acid (DCQA) and 1-O-acetylbritannilactone (ABL) in WEIF on the JAK/STAT pathway were evaluated in interferon-γ/tumor necrosis factor (TNF)-α-stimulated human adult epidermal keratinocytes. WEIF ameliorated DfE-induced skin inflammation by reducing dermatitis scores, mast cell infiltration, skin structural damage, and serum inflammatory mediator levels. Additionally, DCQA and ABL significantly inhibited JAK/STAT activation in interferon-γ/TNF-α-treated keratinocytes. Furthermore, ligand-binding analysis revealed high binding affinities of DCQA and ABL for JAK. These results suggest the pharmacological potential of WEIF to alleviate DfE-induced skin inflammation by inhibiting the JAK/STAT signaling pathway. In conclusion, these findings support the development of WEIF as a therapeutic treatment for AD-like skin inflammatory diseases. Full article

This review explores the dual role of reactive oxygen species (ROS) and free radicals in the pathogenesis of endometriosis, aiming to deepen our understanding of these processes through a systematic literature review. To assess the induction and involvement of ROS in endometriosis, we conducted a comprehensive literature review using Cochrane Libraries, EMBASE, Google Scholar, PubMed, and SCOPUS databases. Of 30 qualifying papers ultimately reviewed, 28 reported a significant contribution of ROS to the pathogenesis of endometriosis, while two found no association. The presence of ROS in endometriosis is associated with infertility, irregular menstrual cycles, painful menstruation, and chronic pelvic discomfort. Among individual ROS types studied, hydrogen peroxide was most frequently investigated, followed by lipid peroxides and superoxide radicals. Notable polymorphisms associated with ROS in endometriosis include those for AT-rich interactive domain 1A (ARID1A) and quinone oxidoreductase 1 (NQO1) isoforms. Key enzymes for ROS scavenging and detoxification include superoxide dismutase, glutathione, and glutathione peroxidase. Effective inhibitors of ROS related to endometriosis are vitamins C and E, astaxanthin, fatty acid-binding protein 4, cerium oxide nanoparticles (nanoceria), osteopontin, sphingosine 1-phosphate, N-acetyl-L-cysteine, catalase, and a high-antioxidant diet. Elevated levels of ROS and free radicals are involved in the pathogenesis of endometriosis, suggesting that targeting these molecules could offer potential therapeutic strategies. Full article

Hallux valgus is one of the common causes of forefoot pain in the field of foot and ankle surgery. This condition is characterized by valgus and pronation deformities of the first ray, leading to bunion pain, metatarsalgia, callus formation, and gait disturbances. Conventional open osteotomy of the first metatarsal and proximal phalanx of the first toe has been widely performed. Recently, with increasing reports of favorable radiologic and clinical outcomes of minimally invasive surgery, this technique has been performed by many surgeons. Despite the various surgical methods available, there is still no consensus on the optimal treatment of hallux valgus, and the advantages and disadvantages of open versus minimally invasive techniques remain a topic of debate. CKD is a leading cause of illness in older cats, but early detection is challenging due to the limitations of conventional biomarkers. This study evaluated the clinical utility of the UAC for identifying early-stage renal dysfunction in cats and proposed a diagnostic matrix incorporating the UAC with other biomarkers. Blood and urine samples from 59 healthy cats and 190 cats with CKD were analyzed, and UAC levels were compared with symmetric dimethylarginine (SDMA), creatinine, and blood urea nitrogen (BUN). UAC values were significantly elevated in cats with CKD, including those in stage 1. Receiver operating characteristic analysis identified a UAC cut-off of 16.3 mg/g, yielding 100% specificity and 43.7% sensitivity for early-stage CKD classification. The UAC showed significant correlations with other renal biomarkers and was incorporated into a multi-parameter matrix to support disease staging. These findings suggest that the UAC may be a promising supplementary biomarker for evaluating renal function in cats and could aid clinical decision-making when interpreted in conjunction with established diagnostic parameters. Sarcopenia, the age-related loss of skeletal muscle mass and function, is associated with inflammation, mitochondrial dysfunction, and gut barrier impairment. This study investigates the postbiotic effects of heat-killed Lactiplantibacillus plantarum HY7715 (HY7715) and its extracellular vesicles (EVs) on muscle health and intestinal integrity. In C2C12 myotubes, both treatments enhanced myogenic differentiation by upregulating Myf5 and MYOG, and improved mitochondrial activity and biogenesis via increased PGC1α and mTOR expression. Under TNFα-induced muscle atrophy, they suppressed expression of atrophy-related markers (Fbox32, MuRF1, and myostatin). EVs showed stronger anti-inflammatory effects by reducing IL6 expression in muscle cells. In intestinal Caco-2 cells, HY7715-derived EVs improved barrier function by upregulating tight junction proteins (ZO-1, occludin, and claudins), and effectively reduced LPS-induced inflammation. These findings suggest that heat-killed HY7715 and its EVs may alleviate sarcopenia by enhancing muscle regeneration and maintaining intestinal homeostasis, highlighting their potential as safe, gut–muscle axis-targeting postbiotic interventions for healthy aging. This study evaluated the therapeutic potential of 70% ethanol extracts from six medicinal plants (Chenopodium album, Cassia tora, Cudrania tricuspidata, Dioscorea polystachya, Lonicera japonica, Solidago virgaurea subsp. gigantea) through their antibacterial, antioxidant, cytotoxic, and immunomodulatory activities, targeting applications in aquaculture. All extracts exhibited potent antibacterial activity (MIC ≤ 10 μg/mL) against Aeromonas spp. and Photobacterium damselae subsp. damselae, but limited efficacy against Streptococcus parauberis. C. tricuspidata (CTR) and C. tora (CTO) demonstrated superior antioxidant activity (IC50 = 1292 μg/mL and IC50 = 227 μg/mL, respectively), correlating with high polyphenol content (1498 and 1409 mg GAE/g). CTR displayed significant concentration-dependent cytotoxicity (IC50 = 904.2 μg/mL), while C. album (CA) promoted cell proliferation (132.3% viability). In LPS-stimulated CHSE-214 cells, D. polystachya (DP) induced the highest IL-8 expression (207-fold), followed by Chenopodium album (CA) (194-fold IL-8, 49-fold TNF-α) and CTR (245-fold RIPK2), activating NF-κB, MAPK, and NOD-like receptor pathways critical for teleost immunity. Lonicera japonica (LJ) suppressed TNF-α (0.4-fold) and IRF1 (0.3-fold), indicating anti-inflammatory potential, while S. virgaurea subsp. gigantea (SV) showed biphasic TNF-α modulation (79-fold at 10 μg/mL, 5-fold at 100 μg/mL). These diverse bioactivities, particularly the robust immunomodulatory effects, highlight the promise of these extracts as natural therapeutic agents for fish health management in aquaculture. Background: This retrospective, multi-reader, blinded, pivotal trial assessed the performance of artificial intelligence (AI)-based clinical decision support system used to improve the clinician detection of emergent large vessel occlusion (ELVO) using brain non-contrast computed tomography (NCCT) images. Methods: We enrolled 477 patients, of which 112 had anterior circulation ELVO, and 365 served as controls. First, patients were evaluated by the consensus of four clinicians without AI assistance through the identification of ELVO using NCCT images. After a 2-week washout period, the same investigators performed an AI-assisted evaluation. The primary and secondary endpoints in ELVO prediction between unassisted and assisted readings were sensitivity and specificity and AUROC and individual-level sensitivity and specificity, respectively. The standalone predictive ability of the AI system was also analyzed. Results: The assisted evaluations resulted in higher sensitivity and specificity than the unassisted evaluations at 75.9% vs. 92.0% (p < 0.01) and 83.0% vs. 92.6% (p < 0.01) while also resulting in higher accuracy and AUROC at 81.3% vs. 92.5%, (p < 0.01) and 0.87 [95% CI: 0.84–0.90] vs. 0.95 [95% CI: 0.93–0.97] (p < 0.01). Furthermore, the AI system improved sensitivity and specificity for three and four readers, respectively, and had a standalone sensitivity of 88.4% (95% CI: 81.0–93.7) and a specificity of 91.2% (95% CI: 87.9–93.9). Conclusions: This study shows that an AI-based clinical decision support system can improve the clinical detection of ELVO using NCCT. Moreover, the AI system may facilitate acute stroke reperfusion therapy by assisting physicians in the initial triaging of patients, particularly in thrombectomy-incapable centers. Full article

In the baculovirus expression system, recombinant viruses generated via bacmids often exhibit reduced expression and genetic stability of target genes during serial passages. This instability is thought to arise from the proximity of non-essential exogenous genes to the target gene insertion site. This study investigated the impact of the target gene insertion locus on its expression and stability within the recombinant viral genome. In addition to the conventional polyhedrin (polh) locus, we evaluated four additional loci located distal to the non-essential exogenous genes. Among them, the ODV-e56 and ChiA/v-cath loci maintained target gene expression and genetic stability more effectively than the polh and p10 loci, even after ten consecutive undiluted viral passages. Notably, essential or functionally important viral genes were located near the ODV-e56 and ChiA/v-cath loci, whereas such genes were absent near the p10 locus. These findings suggest that enhanced stability and expression are associated with the proximity to essential viral genes. Therefore, the strategic selection of target gene insertion sites in close proximity to essential viral elements, rather than simply avoiding non-essential exogenous regions, represents a promising strategy to enhance recombinant protein production in baculovirus expression systems. Located within the subpolar front (SPF) zone, the eastern coast of Korea is subject to frequent environmental changes on both spatial and temporal scales. In this study, we analyzed seasonal changes in the algal community structure and related environmental factors at two sites, Chodo (CD) and Sageunjin (SG), which were influenced by cold and warm-water masses, respectively, in the SPF zone. Sea surface temperature (SST) exhibited greater seasonal change in CD, whereas significant wave height was higher in SG. The salinity and nutrient concentrations decreased during summer and increased in winter at both sites. Seasonal shifts in the dominant species were distinct, with turf species positively correlated with SST dominating the intertidal zone of SG during winter, and canopy species were associated with nutrient concentrations dominating the intertidal zone of CD. Bryopsidales, which was positively correlated with the N:P ratio, dominated the SG subtidal zone in summer, whereas Fucales, which was positively correlated with the SST, dominated the CD subtidal zone. These results enhance our understanding of seaweed responses to environmental changes in SPF zones and provide a basis for predicting future changes in coastal ecosystems. Full article

Lemon Verbena Extract Enhances Sleep Quality and Duration via Modulation of Adenosine A1 and GABAA Receptors in Pentobarbital-Induced and Polysomnography-Based Sleep Models

This study investigated the effects of lemon verbena extract (LVE) on sleep regulation using both a pentobarbital-induced sleep model and an EEG-based sleep assessment model in mice. To elucidate its potential mechanisms, mice were randomly assigned to five groups: control, positive control (diazepam, 2 mg/kg b.w.), and three LVE-treated groups receiving 40, 80, or 160 mg/kg b.w. via oral administration. In the pentobarbital-induced sleep model, mice underwent a two-week oral administration of LVE, followed by intraperitoneal pentobarbital injections. The results demonstrated that LVE significantly shortened sleep latency and prolonged sleep duration compared to the control group. Notably, adenosine A1 receptor expression, both at the mRNA and protein levels, was markedly upregulated in the brains of LVE-treated mice. Furthermore, LVE’s administration led to a significant increase in the mRNA expression of gamma-aminobutyric acid type A (GABAA) receptor subunits (α2 and β2) in brain tissue. In the electroencephalography (EEG)/electromyogram (EMG)-based sleep model, mice underwent surgical implantation of EEG and EMG electrodes, followed by one week of LVE administration. Quantitative EEG analysis revealed that LVE treatment reduced wakefulness while significantly enhancing REM and NREM sleep’s duration, indicating its potential sleep-promoting effects. These findings suggest that LVE may serve as a promising natural sleep aid, improving both the quality and duration of sleep through the modulation of adenosine and GABAergic signaling pathways. The fried potato market is a high-value industry, exceeding USD 55 billion and still growing. However, the genetic mechanisms underlying key frying traits remain poorly understood. In this study, we conducted a transcriptome analysis on two types of potatoes with distinct end-use purposes to identify the gene expression profiles related to desirable frying qualities, focusing on texture and appearance after frying. Key genes encoding starch synthase 1 and 3, sucrose synthase 4, invertases, and pectin methyl-esterase inhibitors were found to be differentially regulated in waxy and mealy potatoes based on their frying characteristics. Notably, mealy potatoes exhibited a higher expression of starch synthesis-related genes and a lower expression of invertase genes. These expression patterns may enhance glucose-to-starch conversion, thereby reducing glucose levels and minimizing sugar-induced browning, which results in a lighter fried appearance. Additionally, we identified two transcription factors, StbZIP2 and StbZIP35, that are potentially co-expressed with two starch synthases. These transcription factors are responsive to abscisic acid, a key hormonal regulator involved in tuber development. This study provides transcriptomic insights for processing quality improvement and identifies key candidate genes for marker-assisted breeding. Further studies across more diverse samples with integrative multi-omics approaches will strengthen the application of these insights in breeding programs. Full article

A novel deployable reflector antenna for small satellites has been designed, fabricated, and experimentally validated. The reflector utilizes a doubly curved flexible surface manufactured from a triaxially woven fabric-reinforced silicone (TWFS) composite. By leveraging high-strain composite materials, the design enables a highly compact stowed configuration while maintaining precise surface accuracy upon deployment. The deployment mechanism is proposed to accommodate a 0.6 m diameter parabolic reflector within a minimal stowed volume, optimizing space efficiency for satellite integration. To validate this concept, a prototype of the reflector antenna has been fabricated and demonstrated the feasibility and effectiveness of the proposed approach. Full article

A baculovirus expression system (BES) for the production of recombinant proteins requires rapid and easy virus titer determination. In this study, a novel direct titration method was developed using a novel Sf9-QE cell line to easily and economically determine virus titers in a short time. This direct titration method can determine virus titers by directly counting the initially infected cells. This method requires the rapid identification of the initial virus-infected cells. The genome of Sf9-QE cells, which fluoresce upon virus infection, was found to contain at least seven copy numbers of the enhanced green fluorescent protein (EGFP) transgene. This result suggests that Sf9-QE cells in the early stages of virus infection can be identified by the high expression of EGFP. It was also shown that for accurate virus titration using the direct titration method, Sf9-QE cells should be used within 3 d of subculturing. Additionally, counting fluorescent cells to establish virus infection should be performed within 15 to 30 h after virus infection for reliable virus titration. The direct titration method using Sf9-QE cells provides a rapid, reliable, and cost-effective alternative for determining baculovirus titers in BES research. Full article
