Dear Colleagues,

Glucocorticoids have an essential role in regulating development and homeostasis. The effect of these hormones is mediated by the glucocorticoid receptor (GR), a member of the ligand-controlled nuclear receptor family. Once activated by glucocorticoids, the GR protein is translocated to the nucleus, where it will bind thousands of sites across the genome. There, GR will recruit various co-regulators to ultimately modulate gene expression in a positive or negative manner. The signaling of GR is a complex output of its interaction with chromatin, co-regulators, and other transcription factors. Of particular interest is the interaction with other steroid hormone receptors which leads to altered signaling.

Unraveling the mechanisms that underpin GR action is of high importance, as glucocorticoids are involved in various pathologies, including cancer. While the role of GR in combating liquid cancers is well established, in terms of solid cancers, such as breast and lung cancer, the effects of GR action seem to be cancer type dependent.

This Special Issue of Life welcomes in vitro and in vivo studies as well as comprehensive reviews on glucocorticoid receptor molecular mechanisms, crosstalk with other steroid hormone receptors, diagnostic and therapeutic advances in the treatment of solid cancers.
