Dear Colleagues,
Myeloid cells are central players in innate immunity, inflammation, tissue repair, and the regulation of adaptive immune responses. In cancer, however, they can act as drivers of malignancy or as key modulators of the tumor microenvironment (TME), influencing disease initiation, progression, and therapeutic resistance.
For instance, in acute and chronic myeloid leukemias, myelodysplastic syndromes, and myeloproliferative neoplasms, malignant transformation within the myeloid lineage leads to bone marrow failure, immune dysregulation, and, in severe cases, multiorgan impairment. Despite major therapeutic advances, myeloid neoplasms remain difficult to cure, with frequent relapses due to the persistence of minimal residual disease.
Conversely, myeloid cells play a crucial role in solid cancers as well. Tumor-associated macrophages (TAMs), myeloid-derived suppressor cells (MDSCs), and neutrophils shape the TME toward a pro-tumorigenic and immunosuppressive milieu, promoting angiogenesis, inhibition of cytotoxic T-cell activity, and extracellular matrix (ECM) remodeling.
This Special Issue of Cells aims to bring together original research articles and comprehensive reviews highlighting novel mechanisms and therapeutic strategies that target myeloid cells—either as the malignant counterpart in hematologic cancers or as critical regulators of tumor development and immune escape in solid malignancies.
