Dear Colleagues,
Due to your published record in the field, we would like to extend an invitation to contribute to a Special Issue on “mTOR Signaling in Cancer”. Both research articles and reviews will be accepted for consideration. We feel that a Special Issue will be more impactful and increase the visibility of individual papers, and we look forward to receiving your contribution.
The mTOR pathway is a key signaling hub that senses cellular levels of energy, nutrients, and growth factor stimulation to either promote cell growth under favorable conditions by upregulating translation and biosynthesis of macromolecules or to suppress catabolic processes like autophagy. The mTOR kinase nucleates two functionally distinct complexes. mTOR complex 1 (mTORC1) is activated by nutrient sufficiency and growth factors, and promotes anabolic processes to increase cellular biomass and cell growth. mTORC2 is growth factor-stimulated and activates anti-apoptotic survival and is also involved in cytoskeletal remodeling.
Hyperactivated mTOR signaling is a common event in tumors, as the promotion of translation and other anabolic processes is used to support unrestrained cell proliferation. Indeed, many upstream negative regulators of mTOR are major tumor suppressors (PTEN, TSC2), while its downstream effectors include oncogenes like Akt.
mTOR signaling in cancer is an active field of study, and this Special Issue will address the many roles of mTOR in cancer, particularly in tumor metabolism, autophagy, and apoptosis. Also covered are clinical developments of mTOR inhibitors such as rapamycin, and the promise of next-generation non-rapalog mTOR inhibitors.

All manuscripts are thoroughly refereed through a single-blind peer-review process. Cancers is an international peer-reviewed open access semimonthly journal published by MDPI.
