Special Issue Information

Dear Colleagues,

Cancer progression is driven not only by genetic mutations but also by widespread changes in epigenomic regulation that alter transcriptional programs and cellular identity. Recent studies have revealed that tumor cells can dynamically rewire enhancer landscapes, transcription factor networks, and chromatin architecture to support proliferation, metastasis, and therapy resistance. Advances in genomic technologies, including single-cell sequencing, chromatin profiling, and 3D genome mapping, now enable detailed characterization of regulatory elements that control oncogenic transcriptional programs. These approaches have identified super-enhancers, enhancer hubs, and regulatory variants that play key roles in tumor development across multiple cancer types. In parallel, epigenetic regulators and transcriptional dependencies are emerging as promising therapeutic targets. This Special Issue will highlight recent advances in understanding how epigenomic plasticity and enhancer reprogramming contribute to tumor heterogeneity, disease progression, and treatment response. We welcome original research and review articles that integrate experimental and computational approaches to investigate chromatin regulation, gene regulatory networks, and epigenomic mechanisms underlying cancer biology.

Accepted papers will be published continuously in the journal (as soon as accepted) and will be listed together on the special issue website. Research articles, review articles as well as communications are invited. For planned papers, a title and short abstract (about 250 words) can be sent to the Editorial Office for assessment.

Submitted manuscripts should not have been published previously, nor be under consideration for publication elsewhere (except conference proceedings papers). All manuscripts are thoroughly refereed through a single-blind peer-review process. Cancers is an international peer-reviewed open access semimonthly journal published by MDPI.
