Special Issue Information

Dear Colleagues,

The therapeutic landscape of hematological malignancies has been profoundly transformed by the development of adoptive cell therapies. In particular, chimeric antigen receptor (CAR)-T cell therapy has demonstrated remarkable efficacy in treating refractory B-cell malignancies and multiple myeloma. Despite these advances, important challenges remain, including variability in manufacturing outputs, limited persistence, immune escape mechanisms, and severe treatment-associated toxicities.

The identification and validation of robust translational biomarkers are essential to bridge the gap between product manufacturing, immune dynamics, and durable clinical responses. This Special Issue focuses on the intersection of product characterization, longitudinal immune profiling, and clinical outcomes, aiming to improve patient stratification, therapeutic monitoring, and outcome prediction. A deeper understanding of cellular fitness, host immune context, and molecular mechanisms of resistance is critical for advancing precision immunotherapy.

This Special Issue welcomes contributions addressing, but not limited to, the following areas:

  • Innovative manufacturing platforms and their impact on cellular product functionality.
  • High-dimensional immune profiling approaches (e.g., scRNA-seq, spectral flow cytometry) for monitoring in vivo expansion, persistence, and exhaustion.
  • Predictive biomarkers for the early detection of toxicity and long-term clinical response.
  • Mechanisms of resistance, including antigen loss and interactions with the immunosuppressive tumor microenvironment.
  • The integration of multi-omics data and machine learning approaches to support personalized cellular immunotherapy.

By garnering advances across these domains, this Special Issue aims to provide a comprehensive framework for optimizing cellular immunotherapies, improving their safety, efficacy, and accessibility for patients with hematological malignancies.
