Dear Colleagues,
Acute myeloid leukemia (AML) remains one of the most formidable hematologic malignancies, characterized by its biological heterogeneity, frequent treatment resistance, and persistently poor outcomes in older or unfit patient populations. Despite decades of reliance on conventional cytarabine-based induction regimens, the past several years have witnessed a paradigm shift driven by a deepening understanding of AML's molecular landscape and the emergence of targeted, immunologic, and combination strategies informed by rigorous clinical investigation.
This Special Issue of Cancers brings together insights from recent clinical trials exploring innovative therapeutic approaches across the AML treatment continuum—from newly diagnosed disease through relapsed and refractory settings. Contributions examine advances in FLT3 and IDH inhibition, venetoclax-based combinations, menin inhibitors, antibody–drug conjugates, and novel immunotherapeutic platforms. Together, these studies illuminate both the promise and the complexity of modern AML therapy, offering a timely resource for clinicians and researchers navigating this rapidly evolving field.
