Immunogenetics provides a mechanistic framework to link genetic diversity with immune function. Within this field, a myriad of immune cells and molecules interact inside a dynamic, complex network to precisely recognize and eliminate a vast array of foreign pathogens. The B and T lymphocytes, along with their respective receptors, serve as a key model system. The diversity of both immunoglobulins and T cell receptors arises from evolutionary processes acting on germline gene repertoires and somatic V(D)J recombination, with somatic hypermutation additionally driving immunoglobulin diversification.
This genetic variability translates into structural diversity, particularly within the complementarity-determining regions (CDRs), an evolutionary and structural hotspot, enabling adaptive recognition of diverse antigens.
Understanding how gene evolution constraints and shapes antibody and T cell receptor structures is essential for deciphering antigen recognition and its dysregulation in disease. This Special Issue aims to integrate evolutionary, genetic, and structural perspectives to elucidate the principles governing antibody/receptor–antigen interactions.
