Glutamate Supplementation Ameliorated Growth Impairment and Intestinal Injury in High-Soya-Meal-Fed Epinephelus coioides

This research assessed the efficacy of glutamate (Glu) supplementation to feed in counteracting growth restriction and intestinal stress-induced injury in juvenile groupers (Epinephelus coioides; initial weight 15.11 ± 0.03 g). The study comprised five isonitrogenous and iso-lipidic diets: a fish-meal-based (FM) diet, a soya-meal-based (SBM) diet, and SBM diets containing varying Glu levels of 1.0% (G-1), 2.0% (G-2), or 3.0% (G-3). The trial employed a randomized design with five treatment groups. Each group was housed in triplicate aquariums and received assigned diets for 56 consecutive days. Supplementation with Glu resulted in dose-dependent enhancements in weight gain, specific growth rate, serum high-density lipoprotein cholesterol, intestinal superoxide dismutase activity, digestive enzyme activity (trypsin, lipase, amylase), amino acid metabolic enzyme activity (glutaminase, GLS; glutamine synthetase), and intestinal mRNA levels of GLS, IL-10, and TGF-β1. Maximal values of the G-3 diet were restored to the levels of the FM diet (p > 0.05). Serum total cholesterol, intestinal total antioxidant capacity, and catalase activity followed a similar increasing trend with Glu level, attaining maxima in diet G-3, yet these values remained markedly lower than those of the FM diet (p < 0.05). Conversely, intestinal malondialdehyde content and mRNA levels of genes IL-8, IL-12, IL-1β, and TNF-α exhibited a significant dose-dependent decrease, reaching minimal levels in diet G-3 that were restored to the levels of the FM diet (p > 0.05). The results above demonstrate that Glu addition enhances nutritional status and intestinal structural integrity by augmenting antioxidant and digestive capacity and mitigating inflammatory responses, consequently enhancing growth performance and intestinal health. Optimizing Rooster Semen Preservation: Effect of Oxygen Exposure, Sample Rotation, and HEPES Buffer Supplementation

This study aimed to investigate physical and biochemical strategies to optimize the preservation and fertilizing capacity of rooster semen during chilled storage and after artificial insemination (AI), respectively. Two semen extenders—0.9% sodium chloride (NaCl) and IGGKPh—were evaluated through two factorial experiments. In Experiment 1, a 2 × 2 factorial design was used to examine the effects of oxygen exposure (aerobic vs. reduced-oxygen conditions) and tube rotation (rotated vs. non-rotated) on semen quality during 24 h of storage at 5 °C. Sperm quality was evaluated based on progressive motility, viability, pH, and malondialdehyde (MDA) concentration. IGGKPh was significantly more effective than NaCl in preserving sperm function, maintaining motility above 70% and viability near 90%. Aerobic conditions and tube rotation improved motility but also increased MDA levels, indicating a tradeoff between enhanced metabolic activity and oxidative stress. Semen stored in NaCl lost its fertilizing capacity after 22 h, whereas IGGKPh under aerobic and rotated conditions resulted in significantly higher fertility rates (91.77%) compared with non-rotated samples. In Experiment 2, the effects of HEPES buffer supplementation (present vs. absent) and handling temperature (5 °C vs. 25 °C) were evaluated under simulated AI conditions. Semen extended in IGGKPh was stored at 5 °C for 22 h prior to handling, while NaCl samples were used immediately after dilution. Sperm quality was assessed at 0, 30, and 60 min of exposure. HEPES significantly reduced MDA levels and improved motility and viability in both extenders. Fertility rates were highest in the HEPES-supplemented groups, especially under chilled handling. In conclusion, optimal preservation of rooster semen requires a combination of metabolic support, adequate oxygen availability, gentle mixing, and pH stabilization. While IGGKPh was effective for storage up to 24 h, its performance was further enhanced by HEPES buffer. These findings offer practical recommendations for AI programs in poultry, particularly under field conditions where temperature fluctuations and delayed insemination are common. Full article

Spent mushroom substrate (SMS) is abundant in nutrients, including polysaccharides, essential amino acids, dietary fiber (DF), and other bioactive compounds. The compound microorganism preparation (CMP), comprising multiple beneficial bacterial groups, is widely used as a feed additive to modulate intestinal microbiota and enhance nutrient absorption in animals. This study investigated the effects of supplementing young sika deer’s concentrated feed with SMS and CMP on total intestinal digestibility, growth performance, serum immune indicators, biochemical parameters, and intestinal microbial composition. Ninety animals were randomly assigned to three dietary treatments: control (basal diet), LE1 (basal diet + 5% SMS), and LE2 (basal diet + 5% SMS + 100 mg CMP/deer/day). Results showed that the LE2 group had significantly higher total weight gain (TWG) and average daily gain (ADG) than both the control group and the LE1 group. Levels of crude protein (CP), total protein (TP), albumin (ALB), blood glucose (GLU), and immunoglobulins (IgG, IgM, IgA) were significantly higher in the LE2 group than in the other groups, with LE1 also showing improvement over the control group. Additionally, ether extract (EE) and high-density lipoprotein (HDL) levels were found to be elevated in both the LE1 and LE2 groups when compared to the control group. In contrast, low-density lipoprotein (LDL) levels as well as urea nitrogen concentrations showed a significant increase in the LE1 group compared to both the control and LE2 groups. Notably, when compared to the control group, there was a significant reduction in the abundance of certain pathogenic bacteria within both the LE1 and LE2 groups; conversely, there was a substantial increase in the relative abundance of beneficial bacterial communities. These results demonstrate that combined SMS and CMP supplementation effectively improves growth performance, nutrient utilization, and intestinal health in young sika deer, suggesting its potential as a functional feed additive. Wild Fauna in Oman: Foot-and-Mouth Disease Outbreak in Arabyan Oryx (Oryx leucorix)

The Sultanate of Oman boasts remarkable biodiversity, exemplified by such species as the Arabian leopard (Panthera pardus nimr) and the Arabian oryx (Oryx leucoryx), national symbols that highlight the extensive conservation efforts required to protect the country’s natural heritage. During decades, Omani authorities have taken significant measures to safeguard wildlife and preserve the natural environment. A sanctuary dedicated to the reintroduction of the Arabian Oryx, after extinction in nature in 1972, was established in 1980 in the Al Wusta Governorate under the patronage of the Royal Diwan and currently administrated by the recently established Environment Authority. During the almost 40 years since the reintroduction and the creation of the sanctuary, the oryx population has grown slowly but constantly. In 2021, the sanctuary hosted 738 oryx, allowing the start of the reintroduction of the species into the natural environment. Small groups of animals were released into the wild in selected areas. No animal health adverse events were recorded, and mortality was generally due to injuries received as a consequence of fighting, in particular during mating season. Standard veterinary care, including control of internal and external parasites, was regularly provided. In some occasions, immunization against certain diseases, such as clostridial infections, pasteurellosis, or mycoplasmosis, was also applied. In 2023, an FMD outbreak in cattle reported in Dhofar, about 500 km from the Al Wusta sanctuary, motivated specific prophylactic actions to prevent the risk of diffusion to oryx. From December 2023 to January 2024, an immunization program was undertaken using an FMD vaccine against serotypes A, O, and SAT 1, mostly in male oryx, while pregnant oryx were avoided for abortion risk due to handling. The following year, in January 2025, a severe outbreak occurred in oryx herds held in the sanctuary. The rapid onset and the spread of clinical symptoms among animals (100% morbidity in the second day after the first appearance of signs in some individuals) were suggestive of a highly contagious disease. The animals suffered from severe depression and inappetence, rapidly followed by abundant salivation, erosions of the oral mucosa and tongue, and diarrhea, with a short course characterized by prostration and death of the animal in the most severe cases. Therapeutical attempts (administration of antibiotics and rehydration) were mostly ineffective. Laboratory investigations (ELISA and PCR) ruled out contagious bovine pleuropneumonia (CBPP), Johne’s disease and Peste des petits ruminants (PPR). Both serology and antigen detection showed positiveness to foot-and-mouth disease (FMD). Out of a total population of 669 present in the sanctuary at the beginning of the outbreak, 226 (33.78%) oryx died. Despite the vaccinal status, the 38.49% of dead animals resulted being vaccinated against FMD. Taking into account the incalculable value of the species, the outbreak represented a very dangerous event that risked wiping out the decades of conservation efforts. Therefore, all the available means, such as accrued biosecurity and adequate prophylaxis, should be implemented to prevent the recurrence of such health risks. The delicate equilibrium of wild fauna in Oman requires study and support for an effective protection, in line with the national plan “Vision 2040”, targeting the inclusion of the Sultanate within the 20 best virtuous countries for wildlife protection. Interactive Effects of Dietary Protein Levels and Magnetic Water Treatment on Water Quality, Growth Metrics, Carcass Composition, Redox Balance, Enzymatic Functions, and Immune Responses in Oreochromis niloticus

The present study was conducted to evaluate the individual and combined effects of dietary crude protein levels and magnetic water treatment on the growth performance, water quality, body composition, physiological responses, and immunity of Oreochromis niloticus. Using a 3 × 2 factorial design, three levels of dietary crude protein (25%, 30%, and 35%) and two water types (magnetized and non-magnetized) were tested. A total of 180 juvenile tilapia (average initial weight: 4.13 ± 0.004 g) were randomly assigned to six treatment groups and reared for 10 weeks. Results showed that magnetic water treatment significantly improved dissolved oxygen and pH, while reducing ammonia, nitrite, and nitrate concentrations. Growth performance indicators, including final weight, specific growth rate, feed conversion ratio, and average daily gain, were significantly improved by both magnetic water and increased dietary protein. Carcass crude protein content improved with both the higher dietary protein level and magnetic water, while lipid content decreased. Liver and kidney function indicators (AST, ALT, ALP, and urea) were significantly improved by magnetic treatment and higher protein levels. Blood biochemical markers (TP, ALB, and GLO) were elevated, while glucose, cholesterol, and triglycerides were reduced by magnetic water; significant interactions were observed for globulin, triglycerides, and total protein. Antioxidant enzyme activities (SOD, CAT, and GPx) increased, and MDA decreased in response to magnetic water and high-protein diets. Similarly, digestive enzyme activities (lipase, protease, and amylase) and immune parameters (IgM and lysozyme) were significantly improved, with the best values recorded in the 35% protein + magnetic water group. In conclusion, magnetic water treatment and dietary protein level independently and interactively affect the physiological performance and health of Nile tilapia, with the best outcomes obtained at 35% protein under magnetic water conditions. Effects of Dietary Metabolizable Energy and Crude Protein Levels on the Nutrient Metabolism, Gut Development and Microbiota Composition in Jingyuan Chicken

The effects of varying dietary metabolizable energy (ME) and crude protein (CP) levels, along with their interactive effects, on the apparent nutrient metabolism, development of digestive organs, intestinal morphology, and microbiota composition in Jingyuan chickens during the growing phase were evaluated. A total of 540 seven-week-old male Jingyuan chickens were randomly assigned to nine groups, with six replicates per group and 10 chickens per replicate. The trial lasted for 11 weeks. A 3 × 3 factorial design was adopted, comprising three levels of ME, namely, low (11.28 MJ/kg, LE group), medium (11.70 MJ/kg, ME group), and high (12.12 MJ/kg, HE group) and three levels of CP, namely, low (14.00%, LP group), medium (15.50%, MP group), and high (17.00%, HP group). The levels of ME and CP, along with their interactions, had significant effects on the average daily gain (ADG), average daily feed intake, feed conversion ratio (FCR), apparent metabolizable rate of CP, gizzard weight, duodenal lengths, jejunal villus height (VH), crypt depth (CD), and muscle layer thickness (MLT) (p < 0.05). The combination of medium level ME (11.70 MJ/kg) and medium level CP (15.50%) (MEMP group) exhibited the best performance, with the highest ADG and the lowest FCR (p < 0.05). Moreover, this group showed significantly higher duodenal length, jejunal CD, VH/CD and MLT compared with the other groups (p < 0.05). Dietary ME and CP levels greatly influenced cecal microbiota composition. Chickens in the MEMP group exhibited an increased abundance of Erysipelotrichaceae, Syntrophomonadaceae, Akkermansia, and Clostridia_vadinBB60_group, and there was an obvious decrease in the relative abundance of Desulfobacterota (p < 0.05). This study demonstrated that dietary ME and CP levels, along with their interactions, could significantly influence the growth performance, apparent nutrient metabolism, and intestinal development of Jingyuan chickens during the growing phase. Dietary ME and CP levels modulated the cecal microbiota composition, potentially inhibiting the abundance of harmful bacteria Desulfobacterota, while enriching the abundance of beneficial bacteria, thereby enhancing gut development and nutrient absorption. The combination of medium-level ME and CP (11.70 MJ/kg ME, 15.50% CP) demonstrated the most favorable outcomes in our study. Full article

Roan coat color is described as the dispersion of white hairs within an otherwise solid background-color coat. This phenotype is primarily expressed on the body of the horse, with the head and legs exhibiting few or no white hairs. Previous studies mapped the locus for roan to the KIT region and observed linked variants in a small number of breeds. Recently, we reported evidence for two independent haplotypes, RN1 and RN2, in the KIT region, which account for approximately 38% and 36% of roan horses, respectively. In the current report, using whole genome sequencing for unknown roan samples. We present a third novel haplotype, RN3, found in American Quarter Horses, that accounts for an additional 30% of American Quarter Horses negative for RN1 and RN2 that display the roan phenotype. Within this haplotype, we observe a variant chr3:79656505 (G > A), which we believe is a founder allele for the RN3 haplotype. In our sample set of horses, these three haplotypes account for more than 95% of the American Quarter Horse population studied and about 50–60% of roan horses in other breeds. Using whole genome sequencing of distantly related animals with a particular phenotype, together with a larger number of control horses, improves the odds of finding linked and/or causative variants. The objective of the study was to investigate the effects of low-protein amino acid-balanced (LPAB) diets supplemented with Astragalus polysaccharides (APSs) on the production performance, antioxidants, immunity, and biochemical index of laying hens in an elevated-temperature environment. Fifty-two-week-old Hy-Line Brown chickens (n = 768) were randomly divided into four groups, with eight replicates of 24 hens each. The control group was kept at 24 °C with a basal diet (CON), while the treatment groups were exposed to 32 °C and given the following diets: basal (HB), LPAB (HL), and LPAB with 0.5% APSs (HLA). Under heat stress, APSs increased the egg production rate and number of small white follicles, improved the yolk color, and lowered the feed conversion ratio. LPAB diets increased follicle-stimulating hormone, antioxidant enzyme activities, and anti-inflammatory cytokine activity and up-regulated related genes, whereas they reduced stress-related hormones, malondialdehyde concentrations, and triglyceride concentrations and down-regulated related genes. The addition of APSs enhanced immunoglobulin concentrations and cholesterol recovery and altered the expression of related genes. The study found that the adverse effects of high temperatures are directly related to oxidative stress. LAPB diets and APSs relatively alleviate these adverse effects. Therefore, the importance of feeding strategies such as LPAB diets and APSs for laying hens under heat stress conditions has been identified. Full article

Artificial insemination (AI) in rabbits depends largely on chilled semen storage, but the physiological responses to chilling and associated biochemical changes in seminal plasma (SP) remain poorly understood, particularly across breeds. This study aimed to compare the semen preservation capacity of Algerian local population (LAP) and New Zealand White (NZW) rabbits and to explore the relationship between SP oxidative stress biomarkers and sperm traits during 72 h of chilled storage at 5 °C. Semen pools (nine/breed) were evaluated at 0, 4, 24, 48, and 72 h for motility, viability, and acrosome status. Oxidative stress markers were also assessed in the SP, including malondialdehyde (MDA), reactive oxygen metabolites (ROMs), superoxide dismutase (SOD), glutathione peroxidase (GPX), and catalase (CAT). LAP sperm showed higher motility (p < 0.001) and viability (p < 0.05), particularly between 4 h and 48 h, and exhibited a lower rate of acrosome reaction (p < 0.001) from 48 h to 72 h. Lower SOD and higher CAT activity in LAP (p < 0.001), correlated with MDA and acrosome status, respectively, may reflect a more balanced antioxidant response. Lipid peroxidation did not appear to be the main factor driving sperm deterioration (p > 0.05). These results demonstrate that LAP rabbits exhibit better resilience to chilled storage compared to NZW and highlight the potential value of CAT and SOD activities as indicators of sperm resilience during chilled storage. Further studies are required to validate and extend these findings, with the aim of improving semen preservation strategies. Full article

Given the restrictions on animal growth promoters, alternative plant-based additives—particularly those rich in phenolic compounds, such as agro-industrial by-products—have been explored. These additives help to mitigate heat stress, which negatively affects productivity by impacting intestinal health and antioxidant status. This study evaluated the effects of individual and combined supplementation of ferulic acid (FA) and grape pomace (GP) on antioxidant enzyme activity, as well as intestinal histomorphometry, in finishing pigs under heat stress. Forty Yorkshire × Duroc pigs were randomly assigned to four treatments: control, 25 mg/kg FA, 2.5% GP, and MIX (FA + GP). FA supplementation increased intestinal villus height, while GP increased villus width in the duodenum and jejunum (p < 0.05). Antioxidant enzyme activity (SOD, CAT, and GPx) increased in pigs supplemented with GP (p < 0.05). These results suggest that GP and FA have potential as functional additives in monogastric diets, improving intestinal health and muscle antioxidant status and contributing to growth modulation. Condition factors are essential in fishery science. They provide insights into fish well-being according to the impact of environmental stressors. The Fulton condition factor (K) and hepatosomatic index (HSI), as well as two new indices (GILSI and GITI), were used to assess the well-being of herring and bycatch species (cod, flounder, sprat, long-spined bullhead, lumpfish) from the Baltic Sea. For Cyclopterus lumpus and Taurulus bubalis, K and HSI values were reported for the first time. Fish from different basins differed according to biometric measures (weight: 6.09–750.12 g; length: 8.6–41.8 mm) and condition indices (mean K: 1.19; HSI: 0.02; GITI: 0.04; GILSI: 0.03). Microplastics (MPs) were found in all species (21% (sprat)–79% (lumpfish)), averaging 3.1 items/fish. The organ-wise analysis revealed that 46% of the gills, 16% of the livers, and 38% of the gastrointestinal tracts were contaminated with MPs. The relationship between these indices and MPs abundance in the organs of herring was examined using PCA, revealing a negative impact of MPs on fish health. The morphometric features and the chemical composition of MPs were also detected. The dominant size ranges were 0.11–0.5 mm (30%) and 1.01–5 mm (28%). Blue fibers were the most common. Full article

This study investigated the reproductive and economic costs of four estrus synchronization protocols in multiparous Holstein dairy cows: Ovsynch with one or two artificial inseminations (AIs), and Double Ovsynch with one or two AIs. A total of 216 clinically healthy cows were randomly assigned to one of the four treatment groups (n = 54 each). Pregnancy diagnosis was performed by transrectal ultrasonography at 33 days post insemination. Economic calculations included the cost of hormonal treatments, insemination, and pregnancy diagnosis, reflecting market prices in large scale Romanian dairy operations. The highest pregnancy rate (64.8%) and lowest cost per confirmed pregnancy (EUR 89.51) were achieved in the group treated with Double Ovsynch followed by a single AI. The standard Ovsynch protocol with one AI yielded the lowest conception rate (42.6%), indicating limited effectiveness under fixed-time AI conditions. These findings suggest that Double Ovsynch with a single insemination is the most effective and economically viable protocol for reproductive management in intensively managed dairy herds. Lasia spinosa Thwaites (LST) has emerged as a potential supplement for enhancing male reproductive performance. This study evaluated the effects of long-term oral supplementation with LST on hematological parameters, semen characteristics, ultrasonographic measurements of the prostate gland and testes, and the cryopreservation potential of canine sperm. Six healthy male dogs received oral LST supplementation at a dosage of 10 mg/kg body weight once daily for 7 days (short-term). After a three-month washout period to ensure full physiological recovery, the same dogs underwent a long-term supplementation protocol (60 days). In the short-term trial, no clinically significant changes were observed in hematological or serum biochemical parameters, including complete blood count, alanine aminotransferase, creatinine, blood urea nitrogen, total protein, and albumin; all parameters were within normal reference ranges. Serum testosterone levels and semen characteristics were also unaffected (p > 0.05). During the long-term treatment, blood profiles and testosterone levels remained stable. Although prostatic and testicular volumes increased slightly, the changes were not statistically significant (p > 0.05). A significant increase in semen volume was observed (p < 0.05), while other semen parameters showed no significant differences. Notably, post-thaw sperm motility significantly improved at both 15 min and 4 h after thawing, and sperm viability was significantly enhanced at 4 h post-thaw (p < 0.05), suggesting a potential protective effect of LST during cryopreservation. These findings indicate that LST supplementation is physiologically safe and may improve canine sperm quality during freezing and thawing, supporting its potential application in reproductive health management. Continuous extradural infusions (CEIs) provide sustained analgesia, avoiding fluctuations seen with bolus administrations. An extradural catheter was placed in a cat with severe abdominal trauma following a vehicular accident. To provide sustained pain relief, a CEI of 0.5% ropivacaine at 0.18 mg/kg/h or 0.036 mL/kg/h was initiated, resulting in improved analgesia, as shown by lower scores on both the Feline Grimace Scale (FGS) and the Glasgow Composite Measure Pain Scale-Feline (CMPS-Feline). During treatment, the cat developed hypersalivation, which persisted after catheter removal at 72 h. Eventually, the cat’s condition deteriorated, with signs of sepsis leading to euthanasia due to a poor prognosis. Potential contributors to the cat’s decline included systemic infection, local anaesthetic systemic toxicity, or opioid-related effects. This report describes the placement of an extradural catheter in a cat, highlighting the potential of CEI in feline pain management while emphasising the need for further research on its pharmacokinetics and safety. Because of the relevance of Salmonella spp. in poultry, in 2016, the Brazilian Ministry of Agriculture, Livestock, and Supply updated its regulation for the control and monitoring of Salmonella in the poultry meat production chain. We herein present the results of the official monitoring of sampling cycles of carcasses performed by the Brazilian official veterinary inspection service (SIF) and the Center for Epidemiology, Statistics, and Public Health of the Veterinary School of the Federal University of Minas Gerais. Samples were collected in March–September 2017, during which 140 slaughterhouses slaughtered over three billion chickens. The establishments were classified according to the number of animals slaughtered per day, and cycles varied according to the size of the establishment. Sample weights were defined to increase the external validity of the data. Chicken carcasses were randomly sampled after chilling and dripping. A total of 1434 samples were analyzed in official laboratories using Vidas SLM and SPT systems, for positive samples were used the ISO 6579:2014. The estimated prevalence of Salmonella spp. in chicken carcasses was 17.88% (95% CI 14.34–22.05). Considering the representativeness of Brazilian chicken meat in the global market, constant evaluation and review of the program is necessary to reduce the prevalence of Salmonella. Full article

Differences in horseshoe materials may have effects on gait that could change perceived esthetic qualities. Objective information regarding effects of shoeing on gait characteristics of horses is scant. The aim of this study was to determine differences in gait characteristics for horses under various experimental shoeing conditions (barefoot, aluminum shoes, steel shoes) on two surfaces (asphalt and soft footing) using body- and hoof-mounted sensors. We hypothesized that shoeing would affect hoof arc height during early (arc height a) and late (arc height b) swing phases but would not affect other gait variables. Twelve healthy, adult, client-owned horses were evaluated at a trot on asphalt and soft footing under the three experimental shoeing conditions. No significant (p < 0.05) effects of shoeing were detected for gait symmetry (Q score), mediolateral hoof deviation, stride length, or midstance, breakover, swing, and landing stride phase times. Hoof arc height a was significantly (p < 0.001) lower for aluminum versus steel shoes for right and left forelimbs on asphalt and soft footing. Hoof arc height b was significantly higher for aluminum versus steel shoes on soft footing for left (p < 0.001) and right (p = 0.02) forelimbs. Findings indicate that shoe weights affect early and late swing phase hoof heights differently. Further investigation is warranted to determine whether measured hoof arc height changes affect subjective esthetics of gait. Full article

A Functional Regulatory Variant of FGF9 Gene Affected the Body Weight in Hu Sheep

Body weight (BW) is a critical economic trait closely linked to livestock meat production performance and producer profitability. In the present study, we measured individual BW of 1070 male Hu sheep at six growth stages (80, 100, 120, 140, 160, and 180 days of age) and conducted descriptive statistical analyses. Results showed that the coefficient of variation (CV) for BW at each stage was higher than 13%. Additionally, we investigated the expression patterns of the FGF9 gene and its associations with single nucleotide polymorphisms (SNPs) and BW. Quantitative real-time PCR (qRT-PCR) revealed FGF9 is significantly higher expressed in the hypophysis tissues that in the other tested tissues. Association analyses indicated that the SNP FGF9:c.382-1264C>T was significantly associated with BW across different measurement periods. Finally, we performed assay for transposase-accessible chromatin using sequencing (ATAC-seq) and cleavage under targets and tagmentation (CUT&Tag) techniques to identify the OCRs (open chromatin regions) and regulatory regions in the hypophysis tissues of Hu sheep, and we obtained an average of 221243, 52692, and 20957.5 peaks in the ATAC-seq, H3K27ac, and H3K4me3 data per sample. Simultaneously, by integrating the SNPs of FGF9 and hypophysis tissue’s epigenomic data, we found that the SNP FGF9:c.382-1264C>T was located in the adjacent OCR- and H3K27ac-modified peaks. Therefore, we propose that the SNP FGF9:c.382-1264C>T plays an important role in regulating body weight in sheep. Overall, this study generated a valuable epigenetic dataset (ATAC-seq, H3K27ac, and H3K4me3) in sheep hypophysis tissue and identified a novel functional regulatory variant for improving body weight in sheep. Activation of Focal Adhesion Pathway by CIDEA as Key Regulatory Axis in Lipid Deposition in Goat Intramuscular Precursor Adipocytes

Intramuscular fat (IMF) content determines the quality of goat meat and is regulated by the comprehensive effect of the proliferation and adipogenesis of intramuscular preadipocytes. Our previous RNA-seq data revealed that cell death-inducing DNA fragmentation factor alpha (DFFA)-like effector (CIDE) A was upregulated during the development of intramuscular fat in the longissimus dorsi muscle tissue, implying an important role in lipid homeostasis. However, the mechanism by which CIDEA, a member of the CIDE family, regulates intramuscular fat deposition in goat muscle is unknown, so we explored the function and underlying mechanism of CIDEA in goat intramuscular preadipocytes. To address this, we altered CIDEA in intramuscular preadipocytes and resolved the effect and mechanism of CIDEA in adipogenesis through RT-PCR, Western blot, triglyceride and LD determinations, CCK-8, and RNA-seq. It was found that CIDEA increased lipid droplets (LDs) and triglyceride contents and inhibited cell proliferation. Meanwhile, the lipid metabolism-related genes PPARγ, C/EBPα, SREBP1c, PLIN1, TIP47, ADFP, DGAT1, ACC, FASN, ACSL1, and FABP3 were upregulated, while the lipolysis and β-oxidation genes HSL, ACOX1, and CPT1B, as well as the proliferation marker gene CDK1, were all downregulated upon CIDEA overexpression. Differentially expressed genes in CIDEA dysregulation groups through RNA-seq were selected and were enriched in the apelin and focal adhesion signaling pathways. Specifically, the Western blot and rescue assays found that focal adhesion, but not apelin, was the key signaling pathway in CIDEA regulating lipid deposition in goat intramuscular preadipocytes. In summary, this study reveals that CIDEA promotes lipid deposition in intramuscular preadipocytes through the focal adhesion pathway and inhibits cell proliferation. This work clarifies the functional role and downstream signaling pathway of CIDEA in intramuscular fat deposition and provides theoretical support for improving meat quality by targeting key phenotype-related genes.
