The color of flowers in Osmanthus fragrans is regulated by carotenoid metabolism. The orange-red variety, Dangui, is believed to have evolved from the yellow variety, Jingui, through a natural bud mutation. This study uses the Jingui cultivar ‘Jinqiu Gui’ (JQG) and its bud mutation cultivar ‘Huolian Jindan’ (HLJD) as materials, combining genome resequencing, ultrastructural observation, targeted metabolomics, and transcriptomic analysis to elucidate the molecular and cellular mechanisms underlying flower color variation. Phylogenetic analysis confirms that HLJD is a natural bud mutation of JQG. Ultrastructural observations reveal that during petal development, chromoplasts are transformed from proplastids. In HLJD petals, starch granules degrade more slowly and exhibit abnormal morphology, resulting in chromoplasts displaying crystalline, tubular, and fibrous composite structures, in contrast to the typical spherical plastoglobuli found in JQG. Targeted metabolomics identified 34 carotenoids, showing significant increases in the levels of ε-carotene, γ-carotene, α-carotene, and β-carotene in HLJD petals compared to JQG, with these levels continuing to accumulate throughout the flowering process, while the levels of the cleavage products α-ionone and β-ionone decrease. Transcriptomic analysis indicates that carotenoid metabolic pathway genes do not correlate directly with the phenotype; however, 49 candidate genes significantly associated with pigment accumulation were identified. Among these, the expression of genes such as glycoside hydrolases (LYG036752, etc.), sucrose synthase (LYG010191), and glucose-1-phosphate adenylyltransferase (LYG003610) are downregulated in HLJD. This study proposes for the first time the pathway of “starch degradation delay → chromoplast structural abnormalities → carotenoid cleavage inhibition” for deepening flower color, providing a new theoretical model for the metabolic regulation of carotenoids in non-photosynthetic tissues of plants. This research not only identifies key target genes (such as glycoside hydrolases) for the color breeding of O. fragrans but also establishes a theoretical foundation for the color enhancement of other ornamental plants. Hippeastrum, a perennial herbaceous plant belonging to the Amaryllidaceae family, is widely cultivated for its large, vibrant flowers with diverse petal colors, which have significant ornamental and economic value. However, the mechanisms underlying anthocyanin accumulation in Hippeastrum petals remain poorly understood. To fully explore the involved regulation mechanism was significant for the breeding of Hippeastrum and other Amaryllidaceae family plants. In this study, we selected six Hippeastrum cultivars with distinctly different petal colors. We used metabolomic profiling and high-throughput transcriptomic sequencing to assess varied anthocyanin profiles and associated expression of genes in their biosynthetic pathways. Four key anthocyanins were identified: cyanidin, cyanidin-3-O-rutinoside, delphinidin-3-glucoside, and delphinidin-3-rutinoside. Weighted gene co-expression network analysis (WGCNA) correlated the abundance of these four anthocyanins with transcriptomic data, to suggest three regulatory modules. Nine transcription factors families in these modules were identified and some of them were validated using qRT-PCR. Y2H assay isolated some transcription factors interacted with TTG1 (WD40 protein), including MYB3/39/44/306 and bHLH13/34/110, illustrating the possibility of forming MBW complexes. Our study provides a comprehensive characterization of anthocyanin composition. These findings laid a theoretical foundation for future research on the regulatory mechanisms of pigment accumulation and the breeding of Hippeastrum cultivars with novel petal colors. Full article

The mismatch between the rapid expansion of breeding scale and outdated techniques has hindered the development of the sea cucumber (A. japonicus) industry. Our previous work revealed that ecological seedling breeding can produce red-colored A. japonicus, a phenotype not observed in traditional artificial breeding, where individuals are typically green. To investigate the molecular and genetic basis of this novel red coloration, we compared the growth conditions of red sea cucumbers and green sea cucumbers, as well as the differences in the pigment composition, gene expression and metabolites of their body walls. Red individuals showed higher body length and weight, and elevated levels of astaxanthin, lutein, canthaxanthin, and β-carotene in the body wall. Transcriptomic and metabolomic analyses identified differentially expressed genes and metabolites associated with pigmentation. In particular, FMO2 and WDR18, involved in the cytochrome P450 drug metabolism pathway, were significantly upregulated in red individuals and are known to play roles in pigment biosynthesis and light signal perception. Key metabolites such as astaxanthin and fucoxanthin were implicated in body color formation. Moreover, genes in the arachidonic acid metabolism pathway were highly expressed, suggesting that dietary factors may contribute to pigment synthesis and accumulation. These findings provide novel insights into the mechanisms underlying body color variation in A. japonicus and offer potential for improved breeding strategies. Full article

Insects’ body coloration may be indirectly influenced by their host plants. Perina nuda (Lepidoptera: Lymantriidae), commonly known as the Banyan Tussock Moth and a serious pest of banyan trees (Ficus spp.) in southern China, exhibits light body coloration during its first- to third-instar stages, with its coloration progressively darkening as it matures, but little is known of the relationship between larval body coloration and host plants. To address this gap, we examined the R (red), G (green), B (blue), and L (lightness) values of the head, dorsal thorax and abdomen, stripe, dorsal mid-line, and tail of larvae fed on different hosts and host endogenous substance by using quantitative image analysis and chemical determination. Our results revealed that larval body coloration exhibited conserved ontogenetic patterns but varied significantly with host species, developmental age, and anatomical region. Redundancy analysis identified chlorophyll-b as the dominant driver, strongly associating with dorsal thorax–abdomen pigmentation. Flavonoids exhibited subthreshold significance, correlating with darker dorsal mid-line coloration, while nutrients (sugars, proteins) showed negligible effects. Linear regression revealed weak but significant links between leaf and larval body coloration in specific body regions. These findings demonstrate that host plant endogenous substances play a critical role in shaping larval body coloration. This study provides a foundation for understanding the ecological and biochemical mechanisms underlying insect pigmentation, with implications for adaptive evolution and pest management strategies. Introduction: This study aims to elucidate the impact of AQP1 on cardiac function and the intestinal microbiota in mice with chronic heart failure and to further investigate the broad effects of AQP1 on the gut microbiota composition in these mice. Methods: AQP1 knockout mice were used as the experimental group, with wild-type mice serving as the control group. The study evaluated the effects of AQP1 on various physiological parameters, including blood pressure, heart rate, cardiac function, cardiac color Doppler ultrasound, and 24 h urine collection. Additionally, the high-throughput sequencing of gut microbiota was performed to identify key microbial communities. Results: The deletion of the AQP1 gene did not significantly alter key cardiovascular metrics such as systolic blood pressure (SBP), mean blood pressure (MBP), or left ventricular mass (LV mass). However, we found that AQP1 knockout affected 24 h urine output in mice. Echocardiography results showed that AQP1 expression influenced LV mass, LVAW; d, and LVPW; s. Moreover, substantial differences were observed in the intestinal microbiota profiles between AQP1 knockout mice with heart failure and their wild-type counterparts. These findings suggest that AQP1 may contribute to cardiac dysfunction in mice with chronic heart failure through the regulation of gut microbiota. Conclusion: Our investigation provides initial insights into the role of AQP1 in modulating the intestinal microbiota in a murine model of heart failure. However, the precise mechanisms underlying this association require further exploration and detailed analysis. Full article

Chinese lacks explicit word boundary markers, creating frequent temporary segmental ambiguities where character sequences permit multiple plausible lexical analyses. Skilled native (L1) Chinese readers resolve these ambiguities efficiently. However, mechanisms underlying word segmentation in second language (L2) Chinese reading remain poorly understood. Our study investigated: (1) whether L2 readers experience greater difficulty processing temporary segmental ambiguities compared to L1 readers, and (2) whether visual boundary cues can facilitate ambiguity resolution in L2 reading. We measured the eye movements of 102 skilled L1 and 60 high-proficiency L2 readers for sentences containing temporarily ambiguous three-character incremental words (e.g., “音乐剧” [musical]), where the initial two characters (“音乐” [music]) also form a valid word. Sentences were presented using either neutral mono-color displays providing no segmentation cues, or color-coded displays marking word boundaries. The color-coded displays employed either uniform coloring to promote resolution of the segmental ambiguity or contrasting colors for the two-character embedded word versus the final character to induce a segmental misanalysis. The L2 group read more slowly than the L1 group, employing a cautious character-by-character reading strategy. Both groups nevertheless appeared to process the segmental ambiguity effectively, suggesting shared segmentation strategies. The L1 readers showed little sensitivity to visual boundary cues, with little evidence that this influenced their ambiguity processing. By comparison, L2 readers showed greater sensitivity to these cues, with some indication that they affected ambiguity processing. The overall sentence-level effects of color coding word boundaries were nevertheless modest for both groups, suggesting little influence of visual boundary cues on overall reading fluency for either L1 or L2 readers. Pleurotus citrinopileatus, a valuable edible fungus characterized by its distinctive light yellow coloration and saprophytic growth on elm wood, has garnered increasing scientific interest due to its diverse bioactive constituents. Among these, polysaccharides derived from P. citrinopileatus (PCPs) have received the most extensive research attention. This review summarizes recent advances in the chemical structure and biological activities of PCPs. Structurally, PCPs are primarily composed of repeating units such as →3)-α-D-Glcp-(1→ and →6)-α-D-Galp-(1→. Functionally, PCPs exhibit a range of bioactivities, including immunomodulatory, hypoglycemic, and antitumor effects. Furthermore, the underlying mechanisms associated with these biological activities are also explored. This review aims to provide a comprehensive reference for future studies and facilitate the development and application of PCPs as potential functional food ingredients or therapeutic agents. The objective of this study was to formulate a compound suspension comprising paeoniflorin and curcumin, assess its quality characteristics, and investigate its protective efficacy against acute liver injury in mice. The prescriptions were screened using a single-factor test, and nine groups of suspensions were prepared using the dispersion method. Fifty KM mice (four weeks old) were selected and randomly divided into five groups: the CON, LD, PF, CUR, and PC groups. The doses of both paeoniflorin and curcumin were 100 mg/kg BW, and different suspensions were given to different groups by gavage for 14 days. All the groups except the CON group were injected intraperitoneally with 20 μg/kg LPS and 700 mg/kg D-GalN on the last day. According to the results, the suspension prepared using the optimal prescriptions was orange-yellow in color, with homogeneous turbidity and good re-dispersibility. The combination treatment could reduce the severity of pathological injuries of liver, improve the ultrastructure of hepatocytes, increase the activities of T-SOD, GSH-Px, and CAT, decrease the levels of IFN-γ, TNF-α, and IL-1, and down-regulate the expression of genes such as TLR4, MyD88, IκBα, and NLRP3. The underlying mechanism might be associated with the enhancement of antioxidant enzyme activities, inhibition of the TLR4/NF-κB/NLRP3 signaling pathway, and suppression of inflammasome assembly and release in hepatic tissues. Rationally designing and synthesizing chemosensors capable of simultaneously detecting both anions and cations via water content modulation is challenging. In this study, we synthesized and characterized a novel triazine calixarene derivative-based iodide and copper ion-selective fluorescent “turn-off” sensor. This dual-channeled fluorescent probe is able to recognize Cu2+ and I ions simultaneously in aqueous systems. The fluorescent sensor s4 was synthesized by displacement reaction of acridine with 1, 3-bis (dichloro-mono-triazinoxy) benzene in acetonitrile. Mass spectrometry (MS), UV-vis, and fluorescence spectra were acquired to characterize the fluorescence response of s4 to different cations and anions, while infrared (IR) spectroscopy and isothermal titration calorimetry (ITC) were employed to study the underlying selectivity mechanism of s4 to Cu2+ and I. In detail, s4 displayed extremely high sensitivity to Cu2+ with over 80% fluorescence decrement caused by the paramagnetic nature of Cu2+ in the aqueous media. The reversible fluorescence response to Cu2+ and the responses to Cu2+ in the solution of other potential interferent cations, such as Li+, Na+, K+, Ca2+, Cd2+, Zn2+, Sr2+, Ni2+, Co2+ were also investigated. Probe s4 also exhibited very good fluorescence selectivity to iodide ions under various anion (F, Cl, Br, NO3, HSO4, ClO4, PF6, AcO, H2PO4) interferences. In addition to the fluorescent response to I, s4 showed a highly selective naked-eye-detectable color change from colorless to yellow with the other tested anions. Full article

Mechanisms of Exogenous L-Lysine in Influencing the Quality of Low-Sodium Marinated Braised Beef

During the processing of marinated braised beef, excessive sodium intake is likely to occur, which can lead to various health issues. Exogenous L-lysine (L-Lys), as an essential amino acid for the human body, has the capability to enhance the quality of low-sodium meat products. This study aimed to investigate the effects of exogenous L-Lys on the quality of low-sodium plain boiled beef and marinated braised beef, as well as its underlying mechanisms of action. Among them, the substitution rate of KCl was 60%. This study was conducted with three batches of experiments, each batch serving as an independent parallel. For low-sodium plain boiled beef, the optimal addition level of L-Lys was screened out through the research on the effects on meat quality indicators, water distribution, microstructure, and sensory evaluation. For the quality of low-sodium plain boiled beef, in terms of microstructure, the addition of L-Lys reduced muscle fiber breakage and voids, thereby improving its microstructural characteristics. Combined with quantitative descriptive analysis (QDA), the optimal level of additional L-Lys was subsequently determined to be 0.6%. It was further processed into marinated braised beef in soy sauce, and a comparative analysis was conducted with low-sodium marinated braised beef in soy sauce without L-Lys addition for shear force, meat color, thiobarbituric acid reactive substances (TBARS), and total viable count (TVC) during the storage periods of 0, 3, 6, 9, and 12 d. The results show that the redness (a*) value significantly increased within 0–12 d (p < 0.05), leading to a more stable meat color. Moreover, the addition of L-Lys significantly reduced the shear force and thiobarbituric acid reactive species (TBARS) values in the marinated braised beef (p < 0.05), thereby optimizing the tenderness of the marinated braised beef and inhibiting lipid oxidation. Although the total viable count (TVC) of the L-Lys group was higher than that of conventional low-sodium marinated braised beef in soy sauce from 9 to 12 d, both groups of products had undergone spoilage by day 12; therefore, the addition of L-Lys had no effect on the shelf life of the products. Comprehensive analysis suggested that the addition of exogenous L-Lys could optimize beef quality by enhancing hydration, improving muscle structural properties, and exerting antioxidant synergistic effects. The large yellow croaker, or Larimichthys crocea, is highly prized for its golden color and nutritional content. The purpose of this study was to investigate the differences in body composition, mucus biochemical indices and body color in five strains of large yellow croakers (body weight: 347.01 ± 5.86 g). To conduct genetic diversity analyses of the populations, a total of 50 tailfin samples were randomly chosen from the following populations of large yellow croakers: wild (LYC1), Dai-qu population (LYC2), Yongdai 1 (LYC3), Min-yuedong population (LYC4), and Fufa 1 (LYC5). The findings demonstrated that the LYC3 group’s pigment contents, crude protein, crude lipid, and chromatic values were comparable to those of the LYC1 group (p > 0.05). There was no significant difference between the LYC1 and LYC5 groups’ mucus superoxide dismutase (SOD) and catalase (CAT) activities (p > 0.05). The alkaline phosphatases (ALP), acid phosphatases (ACP), and lysozyme (LYS) activities of the mucus in the LYC1 group were not significantly different from the LYC3 group (p > 0.05). The back skin mRNA expressions of tyrosinase (tyr), tyrosinase-related protein 1 (tyrp1), dopachrome tautomerase (dct), microphtalmia-associated transcription factor (mitf), and melanocortin 1 receptor (mc1r) were significantly up-regulated in the LYC2 and LYC4 groups compared to the LYC1, LYC3, and LYC5 groups (p < 0.05). Forkhead box d3 (foxd3), paired box 3 (pax3), purine nucleoside phosphorylase 4a (pnp4a), aristaless-like homeobox 4a (alx4a), cAMP dependent protein kinase (pka), anaplastic lymphoma kinase (alk), leukocyte receptor tyrosine kinase (ltk), and colony stimulating factor (fms) were among the mRNA expressions of the abdominal skin in the LYC1, LYC3, and LYC5 groups significantly higher than those in the LYC2 and LYC4 groups (p < 0.05). In conclusion, the LYC3 group’s crude protein, crude lipid, carotenoid, and lutein contents were most similar to those of the large yellow croaker found in the wild. Furthermore, the molecular mechanism underlying the variations in body color among the various strains of large yellow croakers was supplied for additional research. Full article

Rice-field eel (Monopterus albus) slices, an important aquatic product in Southeast Asia, are prone to spoilage and deterioration during cold chain storage. In this study, the effects of a composite preservative (ε-polylysine, Vitamin C (Vc), epigallocatechin gallate (EGCG), and phloretin) on the muscle quality (color, texture, water holding capacity (WHC)) of rice-field eel slices during refrigeration storage at 4 °C for up to 7 days was investigated, and the underlying mechanism was elucidated by the integrated microbiome and metabolomics, in addition to Elisa and Low-Field Nuclear Magnetic Resonance (LF-NMR). After 7 days of storage, the WHC, shear force, and a* decreased by 11.39%, 34.37%, and 49.20% in treated samples, and by 19.18%, 38.38%, and 54.87% in control samples, respectively. The addition of the composite preservative significantly increased Hexokinase, Pyruvate kinase, and Creatine kinase, while it decreased the total viable count (TVC), total volatile basic nitrogen (TVB-N), thiobarbituric acid reactive substance (TBARS), and Lactic acid. Preservative treatment maintained the moisture content of the eel slices during storage and prevented bright red oxymyoglobin from transforming into brown metmyoglobin. Microbiota composition (especially Pseudomonas) and metabolic pathways (including amino acid and its metabolites, nucleotide and its metabolite, and organic acid and its derivatives, etc.) were obviously altered by the preservative treatment. Pseudomonas, tryptophan-aspartic acid (Trp-Asp), D-Glucose 6-phosphate, Succinic Acid, Biliverdin 1, 5-Diaminopentane, and Tyramine, etc., are potential biomarkers for the quality changes of eel slices during refrigeration. These findings provide an in-depth understanding of the improvement of the eel slice quality during refrigeration storage by the composite preservative. Full article

During processing and storage, vegetables often experience undesirable color changes, including fading, lightening, or yellowing and softening, due to browning (enzymatic and non-enzymatic) and chlorophyll degradation. These changes diminish commercial and nutritional value. Therefore, it is necessary to maintain vegetable color and improve the quality of vegetable-based dishes. To address these issues, it is a scientific and practical necessity to summarize and discuss existing strategies and innovative techniques. This review first highlights the mechanisms of vegetable browning. This review then provides a comprehensive overview of recent advances in methods for color preservation, focusing on underlying mechanisms and techniques for inhibiting color changes from physical, chemical, and biological perspectives. A review of innovative technologies suggests that effective color preservation in vegetables is achieved by inhibiting the conditions that lead to three unfavorable color change reactions: enzymatic browning, non-enzymatic browning, and chlorophyll degradation. Current research frequently employs combined approaches that integrate two or more techniques to mitigate these adverse color changes. Moreover, most of these methods could simultaneously inhibit the three reaction processes. Future research directions are proposed for in-depth investigations into the molecular mechanisms of color changes in vegetables and the impact of treatments on the nutritional value. Full article

The local goat breeds of Chongqing represent the typical goat populations of Southwestern China and play a significant role in global goat research. However, studies on these goats remain limited. Chongqing goats exhibit diverse coat colors, a crucial economic trait essential for individual and breed identification. In this study, we performed genome-wide association analysis (GWAS) of Chongqing goats to identify candidate genes associated with coat color. The results of the GWAS revealed that the ASIP, AHCY, and ITCH genes on chromosome 13 are significantly associated with coat color variation in Chongqing goats. Furthermore, haplotype analysis, gene function annotation, and pathway analysis enrichment further confirmed that a significant region on chromosome 13 is associated with coat color in local Chongqing goats, and that ASIP and AHCY are related to black coat color, whereas ITCH is potentially associated with white coat expression. Additionally, we also identified TPK1 as being associated with coat color in goats, which has hardly been reported. The newly identified genes expand our understanding of the complex genetic architecture underlying pigmentation traits in livestock. Our study identifies the genes related to coat color in local goats, which enriches the genetic resource pool of goats and contributes to a deeper understanding of the genetic mechanisms underlying coat color in these breeds. Phalaenopsis pulcherrima are known for their captivating floral morphology and diverse colors, demonstrate exceptional resilience to adverse environmental conditions, and exhibit significant potential for hybrid breeding. However, current research on flower coloration is still limited. The data from this study indicates that variations in anthocyanin levels are the primary determinants of the difference between white and purple colors. Through RNA-seq, we identified 469 genes that were differentially expressed. Furthermore, our bioinformatics exploration uncovered two potential transcription factors, PpMYB1 and PpbHLH1, which play regulatory roles in anthocyanin accumulation. Y2H assays demonstrated that these two TFs could form heterodimers and interact with each other. Afterwards, transient expression assays were conducted for the first time in P. pulcherrima flowers, revealing that overexpression of PpMYB1 alone or in combination with PpbHLH1 resulted in purple petal pigmentation. Overexpressing PpMYB1 in tobacco resulted in more purple-colored corollas, stamens, pistils, and pods compared to control plants. Y1H and dual-luciferase assays provided further evidence that PpMYB1 and PpbHLH1 interact with the promoters of the structural genes PpF3H, PpDFR, and PpANS in the anthocyanin biosynthesis pathway, thereby driving their robust expression. This study not only enhances our understanding of the molecular mechanisms underlying anthocyanin synthesis but also holds significant practical implications for advancing plant hybrid breeding and genetic engineering applications in flower color regulation.
