Objective: This study examined reading behavior and screen time in children and adolescents and their associations with sleep behavior and behavioral difficulties. Methods: This study was conducted as part of the LIFE Child study (Germany). Participants were 579 6- to 10.5-year-old children (younger age group) and 972 10.5- to 18-year-old children and adolescents (older age group). Information on reading (duration of reading, on paper or electronically), screen time, sleep difficulties, and behavioral difficulties was assessed via parental (younger sample) or self-reported (older sample) questionnaires. Associations of reading behavior and screen time with child age, sex, maternal education, behavioral difficulties, and sleep difficulties were assessed using linear regression analyses. Results: Girls reported longer reading times than boys, whereas boys reported longer daily screen time. In the younger age group, reading time increased with age. Daily screen time increased with age in both age groups. Higher screen time was significantly associated with more sleep difficulties in both age groups and with more behavioral difficulties in the older age group. In the younger age group, reading time was not associated with screen time, sleep difficulties, or behavioral difficulties. In the older age group, however, longer reading times were significantly associated with longer screen time, more behavioral difficulties, and more problematic sleep. Conclusions: These results indicate that both the use of electronic media and reading behavior are relevant for understanding child health and development. The findings suggest that guidance for families of older children should address not only screen use but also balance and context of reading activities. Full article

Neonatal screening using Dried Blood Spot (DBS) cards is an important and successful public health facility in Europe, enabling early detection of congenital disorders for early treatment and prevention of overt disease. Biobanks of stored DBS cards offer significant potential for biomedical research. Biobanking and secondary use of DBS cards also raise ethical and legal issues, particularly concerning the rights of parents and children. This study provides a comprehensive overview of legislation and legal practices governing DBS biobanking across 29 European countries, based on a survey conducted in collaboration with the International Society for Neonatal Screening. Findings reveal a highly heterogeneous landscape: 15 countries have national legislation, five have regional guidelines, and nine lack formal regulations. Only four countries require explicit parental consent for DBS storage, with considerable variation in approval processes for research use. A harmonization of practices, with the European General Data Protection Regulation (GDPR) as a basis for future regulation, supplemented by clearer guidance on ‘public interest’ and robust safeguards for individual rights may lead to more transparent and consistent governance, which is essential to balance scientific progress with the protection of parental and children’s rights in DBS-based research across Europe. 2,3-Butanediol (2,3-BD) is a versatile bulk chemical, serving as a high-boiling green solvent, a key monomer for polyesters and polyurethanes, and a crucial intermediate in the production of fine chemicals and cosmetics. Additionally, it is a key intermediate for pharmaceuticals and agrochemicals, supporting green chemistry. Microbial production of 2,3-BD offers a sustainable bioprocess but faces the challenge of high-concentration product inhibition. In an effort to improve 2,3-BD tolerance and production in Corynebacterium glutamicum, the engineered strain CGK4 was subjected to atmospheric and room temperature plasma (ARTP) mutagenesis, from which three superior mutant strains were isolated. Genomic and reverse engineering analyses revealed two mutations beneficial for tolerance and seven mutations beneficial for production. Among these, mutations atpGL250P, cobTV192V, and ctaEL79P increased 2,3-BD titer by 18.0%, 14.5%, and 15.0%, respectively. Furthermore, we found that while certain mutations markedly enhanced stress tolerance, the highest-producing strains achieved a balanced metabolic state rather than maximal tolerance, indicating a non-linear relationship between the two traits. Finally, through mutation combination optimization, the optimal strain K4-cobT-ctaE was constructed. In shake-flask fed-batch fermentation, this strain produced 84.35 ± 3.04 g/L (2R,3R)-2,3-BD, which was 38.3% higher than that of the parental strain. Our findings provide a promising approach to addressing 2,3-BD tolerance challenges and promote the development of microbial production platforms for industrial application. Full article

Pornography use during adolescence is a relevant issue from social, educational, and public health perspectives. As with other behaviours, its correlates may involve complex and nonlinear relationships. This study uses data from the 2023 Spanish Survey on Drug Use in Secondary Education (ESTUDES), a major source for analysing potentially addictive behaviours among adolescents in Spain because of its large sample size (original sample: N = 42,208; complete-case analytical sample: N = 33,543). Pornography use was modelled as a binary outcome using logistic regression, XGBoost, LightGBM, and CatBoost. The models included sociodemographic characteristics, family-related factors, parental control, substance use, selected sexual behaviours, indicators of mental well-being, and addictive Internet use. The four methods showed similar predictive performance. CatBoost achieved the highest AUC and the lowest Brier score and log loss, whereas LightGBM obtained the highest accuracy, specificity, and precision. Logistic regression yielded the highest sensitivity, negative predictive value, balanced accuracy, and F1-score. SHAP analysis identified sex as the most influential predictor, followed by addictive Internet use, cannabis and alcohol use, family conflict, and selected risky sexual behaviours. The findings suggest that pornography use among adolescents forms part of a broader behavioural and psychosocial profile. Whereas sex operated mainly as a strong direct predictor, problematic Internet use showed substantial interactions with age and cannabis and alcohol use. This study demonstrates how explainable machine learning can complement conventional regression by identifying the main correlates, nonlinear patterns, and interactions associated with adolescent pornography use. Background/Objectives: Clariid catfishes are a cornerstone of Southeast Asian aquaculture, where intensive breeding and interspecific hybridization have significantly decreased genetic diversity. To address the limited understanding of growth-governing molecular determinants, we characterized the genomic architecture and evolutionary dynamics of the myogenic factor 5 (Myf5) gene, a master regulator of myogenesis, in 511 individuals from 17 catfish populations in Thailand. Methods: A total of 511 individuals representing 17 catfish populations were analyzed. A 334-bp fragment of exon 1 of the Myf5 gene was sequenced using Illumina short-read technology. Genetic diversity indices were calculated to evaluate sequence variation, population structure, and selective constraints acting on the locus. Results: Sequence analysis of a 334-bp segment of exon 1 using Illumina short-read technology identified 17 haplotypes and 92 variable sites, which revealed moderate allelic richness despite low overall nucleotide diversity (π ≈ 0.008). Evolutionary analyses indicated that the Myf5 locus is governed by stringent purifying selection (ω = 0.063), wherein site-specific constraints preserve the stability of the α-helix folding architecture, which is essential for myogenic regulation. Population genetic analyses revealed substantial differentiation among taxa (56% among-population variation), while Bayesian clustering successfully resolved a hierarchical structure (K = 2) that distinguished Clarias gariepinus, Clarias macrocephalus, and Clarias batrachus. These findings demonstrate that species-specific Myf5 haplotypes serve as diagnostic markers for identifying parental species and assisting in the identification of F1 hybrid stocks. Conclusions: Overall, this study provides the molecular markers for species identification, broodstock authentication, hybrid detection, and sustainable genetic management of Thai Clariid resources, which are critical for balancing production efficiency with the conservation of native biodiversity in Southeast Asia. The study evaluated the effects of dietary compound enzyme preparations (CEPs) supplementation on antioxidant capacity, immune response, and intestinal health of pigeons. A total of 288 White King parent pigeons were randomly assigned to four treatments, with six replications of 12 pigeons each. The control (CK) group received a basal diet, whereas the CEP groups were fed a basal diet with 0.25 g/kg (low compound enzyme preparation group, LCEP), 0.5 g/kg (middle compound enzyme preparation group, MCEP), and 1.0 g/kg (high compound enzyme preparation group, HCEP) CEPs. Results showed that supplementation with 0.25 g/kg of CEPs significantly increased duodenal crypt depth (CD), as well as jejunal villus height (VH) and the villus height-to-crypt depth ratio (VCR) (p < 0.05). It also increased the immunoglobulins of serum, such as IgA, IgG, and IgM, as well as enhanced antioxidant capacity by increasing superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GSH-Px) activities of serum (p < 0.05). Furthermore, we found that CEPs altered ileum content microbiota composition, increasing the relative abundance of p_Bacteroidota and g_Bifidobacterium (p < 0.05). In addition, untargeted metabolomics analysis identified 92 upregulated and 95 downregulated metabolites. Pathway enrichment analysis of these differential metabolites revealed that CEP supplementation altered the metabolomic profile of the ileum contents, with differential metabolites mainly enriched in alanine, aspartate and glutamate metabolism, fatty acid biosynthesis, and unsaturated fatty acid biosynthesis pathways. These results demonstrated that dietary supplementation with CEPs at different doses improved antioxidant capacity to varying degrees and maintained intestinal immune balance by modulating microbiota, which provides a theoretical basis for the rational application of compound enzyme preparations in breeding pigeon feed. Moreover, this study provides mechanistic evidence that CEPs can improve intestinal health, immunity, and antioxidant capacity. It also fills a key knowledge gap in pigeon nutrition by linking enzyme use with gut microbiota and metabolic regulation. Full article

X-linked adrenoleukodystrophy (X-ALD) is the most common peroxisomal disorder and is associated with serious clinical consequences. While newborn screening (NBS) could facilitate early identification and timely intervention, its implementation for X-ALD remains controversial due to ethical concerns arising from its X-linked recessive inheritance pattern, particularly regarding female newborns. This study aims to explore the perspectives of healthcare professionals and the general public on NBS for X-ALD in Hong Kong. An online survey with 20 quantitative questions on ethical considerations was conducted from May to August 2024. Among a total of 259 responses, most respondents (99.2%) supported NBS for their male newborns, primarily because it facilitates early diagnosis and effective management. The majority of the respondents were in favor of offering NBS to female newborns, citing potential benefits for the management of adult-onset disease, enhanced family planning and support, and opportunities for extended family screening. However, some respondents expressed concerns regarding (1) psychological stress and anxiety from uncertain disease onset and frequent monitoring; (2) potential genetic discrimination and adverse impact on insurance premiums/coverage; (3) affordability and accessibility of expensive treatments, such as gene therapy; and (4) ethical issues regarding children’s “right to an open future”, particularly for late-onset female X-ALD. To address these concerns while respecting family autonomy, we propose an opt-in system with clear, balanced information for parents, combined with a three-tier screening algorithm. In summary, while inclusion of X-ALD in Hong Kong’s NBS program receives strong community support, targeted measures are needed to mitigate the identified ethical and practical barriers. Full article

Climate change is altering grape composition and ripening patterns, thereby influencing the selection criteria for wine yeast strains adapted to warmer conditions and fluctuating sugar–acid balances. Previously, we sequenced and annotated the Champagne–Epernay–Geisenheim white wine yeast CEG. CEG is a hybrid tetraploid strain with more than three genome equivalents of Saccharomyces cerevisiae and only 1/3 of a S. kudriavzevii genome. To address climate change issues, we used the limited sporulation capacity of CEG to isolate meiotic progeny. The obtained spore clones were subjected to targeted selection procedures, identifying progeny with enhanced fermentation performance under higher temperature conditions. Spore clones were also selected to carry the FOT4 allele encoding a Fungal Oligopeptide Transporter gene. We identified suitable strains, particularly GYBC 1467, that surpassed the parental strain at 25 °C with enhanced volatile aroma production. Genome sequencing of three spore clones indicated differential loss of S. kudriavzevii DNA. This approach demonstrates that, via a single round meiosis, spore clone isolates with substantially improved fermentation performance compared with the parental strain can be obtained. This provides a fast route for the generation of resilient wine yeasts capable of maintaining wine quality in increasingly difficult fermentation environments. Full article

Urologic malignancies (UMs), including prostate cancer (PCa), bladder cancer (BCa), renal cell carcinoma (RCC), and testicular germ cell tumors (TGCT), are governed by interconnected molecular pathways that regulate proliferation, angiogenesis, metabolism, invasion, immune escape, and treatment resistance. Key pathways include PI3K/AKT/mTOR, VEGF/VEGFR, EGFR, FGFR, c-MET, androgen receptor signaling, DNA damage response, inflammatory transcriptional programs, and regulation of the tumor microenvironment. This structured expert review evaluates mechanistic, translational, epidemiological, and clinical evidence on food- and botanical-derived nutraceuticals that may influence cancer-related signaling, redox balance, inflammation, metabolic adaptation, and host–tumor interactions. Nutraceuticals are considered investigational adjunctive exposures rather than anticancer treatments or alternatives to standard care. Relevant literature was identified through a structured, non-systematic search of PubMed/MEDLINE, Scopus, Web of Science Core Collection, and Embase from database inception to 17 June 2026, supplemented by backward and forward citation searches. Eligible evidence comprised preclinical, observational, interventional, pharmacokinetic, formulation, safety, and drug–nutraceutical interaction studies. Evidence was evaluated by cancer type, compound class, molecular context, formulation, exposure, translational maturity, and safety, and was classified into five stages: prevention signal, mechanistic plausibility, bioavailability and exposure feasibility, exposure-linked biomarker activity, and clinical benefit. An exposure–species–compartment framework was used to assess whether parent compounds and relevant metabolites reached systemic, urinary, or target-tissue concentrations compatible with the proposed effects. Findings based solely on supraphysiological concentrations of unconjugated parent compounds were considered hypothesis-generating unless supported by human exposure or tissue-distribution data. Curcumin, green tea catechins, isoflavones, carotenoids, flavonols, stilbenes, and triterpenoids affect several cancer-related pathways, primarily in experimental models. Translation to clinical practice is constrained by inconsistent formulations, limited bioavailability, inadequate target-tissue exposure data, few biomarker-linked studies, and possible interactions with anticancer therapies. PCa and BCa provide the most suitable settings for exposure-verified mechanistic studies. In RCC, safety and treatment interactions should be prioritized, whereas in TGCTs, non-interference with curative cisplatin-based therapy must be demonstrated. Future studies should use chemically defined formulations, verify clinically relevant exposure, incorporate mechanism-matched biological-response endpoints, and confirm compatibility with established treatment. Current evidence does not support nutraceuticals as treatments for urologic malignancies. Full article

Background/Objectives: The middle cerebral artery (MCA) bifurcation is a common site of intracranial aneurysms. We evaluated associations between MCA bifurcation geometry and aneurysm presence and explored relationships between rupture and aneurysm size using a standardized 3D morphometry pipeline. Methods: This single-center, retrospective, propensity score-matched case–control study included 133 MCA bifurcation aneurysms and 266 matched controls, with bifurcation and sac-level geometry quantified using 3D rotational DSA. Associations with aneurysm presence were evaluated using generalized estimating equation (GEE) logistic regression clustered by patient ID. The discriminatory ability of individual geometric markers was assessed using ROC-AUC. Results: After matching, baseline covariates were balanced. In univariate comparisons, aneurysm-bearing bifurcations had a wider angle between the two M2 branches (opening angle), smaller dominant and non-dominant M1–M2 branch angles, smaller parent diameters and cross-sectional areas, a higher opening angle-to-parent diameter ratio (opening-to-caliber index), and lower parent-to-dominant-branch and daughter-branch diameter ratios. In multivariable GEE analysis, a larger opening angle, smaller dominant-branch angle, and lower D1/D2 ratio were independently associated with aneurysm presence; each 5° increase in opening angle corresponded to approximately 40% higher odds. The opening-to-caliber index showed the numerically highest single-marker ROC-AUC but did not remain independently associated with aneurysm presence after adjustment. Among the aneurysms, rupture was not associated with bifurcation geometry; ruptured aneurysms were larger. Across size strata, the opening angle and opening-to-caliber index increased, and the dominant-branch angle decreased. Conclusions: MCA bifurcation aneurysm presence was associated with a wider, more asymmetric bifurcation geometry, whereas rupture appeared more closely related to sac-level morphology. Full article

Proactive mental health consultation–liaison (C-L) services in neonatal intensive care units (NICUs) are increasingly recognized as essential to infant health and family functioning, yet there is limited guidance on how to develop sustainable and effective programs in real-world clinical settings. This paper describes the development, implementation, and iterative refinement of a NICU mental health C-L program using a learning health system (LHS) framework. Grounded in a theory-driven model that conceptualizes the infant as the identified patient, the program was designed to target proximal mechanisms influencing infant regulation, parent–infant interaction, and medical course. Consistent with LHS principles, program elements were introduced incrementally and refined through embedded data collection, rapid feedback loops, and multidisciplinary team learning. Data sources emphasized feasibility, reach, implementation processes, and acceptability rather than hypothesis-driven outcome testing. Over six years, this approach supported the expansion of multiple integrated service components, including dyadic intervention services, parent mental health screening and referral pathways, unit-wide family engagement initiatives, and neurobehavioral care practices. Rather than presenting a fixed protocol, this paper offers a replicable framework for developing NICU mental health C-L services that balances developmental theory, implementation realities, and sustainability. The LHS approach provides a pragmatic roadmap for NICUs seeking to build or refine mental health services in contexts characterized by clinical complexity, evolving evidence, and system-level constraints. 2-phenylbenzimidazole-5-sulfonic acid (PBSA) is a commonly used organic ultraviolet (UV) filter frequently found in aquatic environments, raising substantial ecological health concerns. While some toxic effects of PBSA on aquatic organisms have been reported, its intergenerational developmental and neurotoxic risks remain poorly understood. In this study, we established a zebrafish life-cycle exposure model to explore the intergenerational toxicity of environmentally relevant concentrations of PBSA (0.2, 2, and 20 μg/L). Offspring were categorized into three exposure groups: parental exposure only (F0+/F1−), parental exposure with continuous F1 exposure (F0+/F1+), and only F1 exposure without parental treatment (F0−/F1+). Our findings demonstrate the transfer of PBSA from parental gonads to F1 embryos. Parental lifetime exposure significantly inhibited somitogenesis and increased mortality and malformation rates in the F1 generation, with the most pronounced developmental damage observed in the F0+/F1+ group. Whole-mount immunohistochemistry revealed that PBSA notably reduced motor neuron axon length in F1 larvae, accompanied by downregulation of developmental-related genes including gap43, mbp, and shha. Mechanistically, the F0+/F1− group exhibited a marked increase in MDA levels. Excessive ROS accumulation and reduced CAT activity were specifically observed in the F0+/F1+ group, whereas the F0+/F1− and F0−/F1+ groups showed significantly elevated CAT activity, jointly driving developmental and neurotoxic changes. In silico predictions indicated low acute aquatic toxicity of PBSA, contradicting its observed intergenerational risks. Our findings demonstrate that parental lifetime PBSA exposure can induce significant intergenerational neurodevelopmental toxicity in zebrafish offspring by disrupting oxidative balance and neural gene expression, with continuous offspring exposure further aggravating the adverse effects. This research underscores that traditional single-generation toxicity assessments underestimate the ecological dangers of UV filters. It also offers new insights into the environmental risk evaluation of PBSA and similar emerging contaminants. Full article

Parental education, as institutionalized family cultural capital, is significantly associated with children’s academic performance. Using provincial-level academic quality monitoring data, this study employs hierarchical linear models and path analysis to examine how parental education levels and matching patterns relate to urban primary school students’ academic performance. Key findings: Higher overall parental or paternal education is positively associated with performance; however, maternal education exhibits an inverted U-shaped relationship. Compared to both parents non-highly-educated families, students perform significantly better when both parents are highly educated or when fathers are highly educated and mothers are not, but significantly worse in families where the mother is highly educated while the father is not. Path analysis indicates that the negative association in mother-hypergamy families is partly statistically associated with reduced paternal learning involvement, which in turn is related to heightened academic pressure and poorer outcomes. The study suggests that the lower academic performance observed in mother-hypergamy families may be partly associated with reduced paternal learning involvement, highlighting the importance of family educational configuration and balanced co-parenting dynamics. These findings suggest that the association between parental education and children’s academic performance is not straightforward. While consistent with cultural reproduction and family investment perspectives, the detrimental effect of the mother-hypergamy pattern highlights the importance of family configuration and points to the potential roles of role conflict, co-parenting dynamics, and expectation-pressure mechanisms that warrant direct empirical investigation in future research. This study examined how attachment to parents influences attachment to God across Christianity, Islam, and Hinduism. Grounded in attachment theory and extending prior work on parental correspondence, participants from the United States and India completed measures of attachment to mothers, fathers, and God. Consistent with the correspondence hypothesis, parental attachment patterns mirrored attachment to God. For Christian participants, both avoidance and anxiety toward fathers predicted attachment to God, reflecting Christianity’s paternal conception of the divine. For Muslim participants, both avoidance and anxiety toward mothers predicted attachment to God, consistent with the maternal role in transmitting Islamic faith. Among Hindu participants, avoidance toward mothers predicted avoidance toward God, and anxiety toward both parents predicted anxiety toward God, reflecting the balance of masculine and feminine divine representations in Hinduism. These findings underscore cultural differences in spiritual attachment and highlight the importance of parental influences in understanding cross-religious variations in faith development. Screen Time and Myopia-Related Outcomes in European Children: A Systematic Review

Myopia is an increasingly common refractive disorder in children, with growing concern regarding the potential role of screen time and digital device use in its development and progression. This systematic review assessed the association between screen-related behaviours and myopia in European children and adolescents. A systematic search of Ovid Embase, Ovid MEDLINE, Scopus, Web of Science and Cochrane/CENTRAL was conducted in accordance with PRISMA guidance. Original English-language studies published within the last 10 years were included if they involved participants aged <18 years, were conducted in Europe, and investigated screen time, digital device use, near-work behaviour or related lifestyle exposures in relation to myopia-related outcomes. A total of 1196 records were identified across the databases. After duplicate removal, title and abstract screening, and full-text assessment, 14 studies were included. Overall, the evidence suggested an association between screen-related behaviours, prolonged near work, reduced outdoor exposure and myopia-related outcomes in European children. However, findings were heterogeneous, and screen exposure was often measured using broad or proxy variables, including total screen time, smartphone use, internet data consumption, computer use, handheld near-screen use and COVID-19-related lifestyle change. Outdoor activity appeared to be a protective factor, while parental myopia was an important risk factor and confounder. Most included studies were observational, many were cross-sectional, and several relied on self- or parent-reported exposure measures, limiting causal inference and introducing potential recall bias. Overall, the certainty of evidence was low to very low. Practical advice should therefore be framed around balanced digital device use, regular breaks from prolonged near work, appropriate viewing distances and increased outdoor activity, while further longitudinal studies using objective screen-use measures, cycloplegic refraction and axial length outcomes are needed to clarify causality.
