The increasing global burden of cancer, together with the need for more sustainable resource management, has stimulated growing interest in the valorization of agro-industrial plant residues as sources of bioactive compounds with therapeutic potential. This review highlights the potential of plant by-products—including citrus peels, olive leaves, date palm residues, and tea and coffee processing wastes—as sustainable reservoirs of polyphenols and other phytochemicals with significant anticancer activity. Key compounds such as hesperidin and naringenin from citrus peels, oleuropein and hydroxytyrosol from olive leaves, quercetin and syringic acid from date palm residues, and chlorogenic acid and epigallocatechin gallate from tea and coffee by-products have demonstrated promising antitumor effects in both in vitro and in vivo studies. These molecules exert their activity through multiple mechanisms, including the inhibition of cancer cell proliferation, induction of apoptosis, regulation of the cell cycle, and modulation of major oncogenic signaling pathways such as PI3K/AKT, MAPK, NF-κB, and EGFR. For instance, hydroxytyrosol induces apoptosis and cell cycle arrest while inhibiting the PI3K/AKT and MAPK pathways. Quercetin limits metastasis and glycolysis and suppresses VEGF, PKM2, and AKT signaling. Ferulic acid suppresses tumor growth by inhibiting the PI3K/AKT and JAK2/STAT6 pathways, thereby promoting apoptosis (in vitro and in vivo). In addition to their pharmacological potential, the recovery of these compounds from plant waste supports circular economy strategies by reducing environmental impact and promoting the development of value-added products. Future research should focus on optimizing extraction methods, improving bioavailability and stability, and validating safety and efficacy through well-designed preclinical and clinical studies. The effects of different ultrasound parameters on some physicochemical properties and secoiridoid compositions of olive extracts were investigated. For this purpose, pH, acidity, photometric color index (PCI), total phenolic content, and secoiridoid phenolic compound composition analyses were carried out in olive extracts obtained by ultrasonic extraction at different operating parameters such as temperature, ultrasonic power, and extraction time. The data obtained were modeled and optimized by using the Box–Behnken design of RSM. Then, the comparison of experimental data versus mathematical estimations was performed by using both ANN and RSM. The results revealed that the pH values of the samples ranged between 4.94 and 5.23, and the average acidity value was 0.551 (% oleic acid). PCI values varied between 20.46 and 83.70. Total phenolic content ranged between 0.13 and 0.42 mg GAE (gallic acid equivalent)/g extract. Regarding secoiridoid phenolics, the ranges for oleuropein, oleacein, and oleocanthal were 5.33–34.39 ng/μL, 0.76–6.03 ng/μL, and 3.77–14.16 ng/μL, respectively. The optimized temperature, time, and ultrasonic power were 43.13 °C, 15 min, and 100% (of the maximum ultrasonic power of 90 W), respectively. The overall desirability of the process was obtained as 95.51%. RSM and ANN were both favorable in the estimation of experimental data with slight differences. Full article

This research focuses on the synthesis of a novel baking powder enriched with bioactive molecules recovered from olive pomace via ultrasound-assisted extraction using a hydro-ethanolic mixture. The functional ingredient was engineered by anchoring the extracted phytocompounds onto a starch backbone through a sustainable grafting technique. Biscuits formulated with the innovative ingredient showed an increased concentration of phenolic compounds (2.162 mg GAE/g), encompassing both phenolic acids (0.372 mg GAE/g) and flavonoids (0.360 mg CTE/g). Enhanced antioxidant efficacy was recorded, mostly in aqueous media (IC50 = 0.554 mg mL−1 against ABTS radical) compared to organic environments (IC50 = 0.132 mg mL−1 against DPPH radical). Furthermore, Oxitest and oxidation stability reactor analyses revealed exceptional antioxidant capacity (induction period = 37 ± 2 h). By an accelerated shelf-life test, a marked instrumental color difference was observed with the fortified sample showing a darker, redder/brown color (ΔE > 16), as also confirmed by trained panelists. On the contrary, similar scores were achieved for the olfactory, textural and tasting attributes of the two samples, as well as values of the friability index (<1 mm−1) evaluated by instrumental techniques. This approach represents a sustainable strategy, transforming a high-polluting agri-food by-product into a source of bioactive compounds for nutritional and technological improvement of baked foods. Landmark epidemiological studies and clinical trials, such as the Seven Countries Study, the Lyon Diet Heart Study, the PREDIMED Study and the CORDIOPREV Study, have shown significant reductions in cardiovascular events in those following the Mediterranean diet (MD). The aim of the present work is to summarize the most robust available evidence and the major biological pathways underlying the protective effects of the MD, with particular emphasis on the role of PAF inhibitors. Mechanistically, MD functions through a complex synergy of antioxidant, anti-inflammatory, and antithrombotic effects that collectively improve lipid profiles, enhance endothelial function, optimize postprandial metabolism and cell membrane signaling, making it a functional model for human longevity. The PAF-Implicated Atherosclerosis Theory has emerged as a key unifying framework, proposing that Platelet-Activating Factor (PAF)—a highly potent lipid inflammatory mediator—plays a central role in the initiation and progression of atherosclerosis. Oxidized LDL promotes the production of PAF and PAF-like lipids, leading to endothelial dysfunction, vascular inflammation, and atherosclerotic plaque formation. Traditional Mediterranean foods are rich in natural PAF inhibitors, particularly the polar lipid fractions of extra virgin olive oil, as well as wine, fish, vegetables, onions, and garlic. Animal studies demonstrate that these compounds can reduce or even regress atherosclerotic lesions, independently of serum cholesterol levels. Human dietary interventions have further shown that MD-based meals and functional foods enriched with PAF inhibitors reduce PAF activity and improve thrombosis-related biomarkers. This mechanistic framework helps explain phenomena such as the “French Paradox” and the cardio-protective effects associated with fish consumption. Moreover, the extraction of PAF inhibitors from Mediterranean food by-products, such as olive pomace, offers promising ecological and economic advantages. Collectively, targeting PAF and increasing dietary intake of PAF inhibitors represent promising strategies for the prevention and management of atherosclerosis and other inflammatory diseases, supporting the view that PAF may function as a major, modifiable risk factor in these conditions. The European Regulation (UE) 432/2012 includes a specific health claim for olive-oil-associated with its phenolic content, which is based on its protective role against lipid oxidation in the blood. To make use of the health claim, olive oil must have a minimum concentration in phenolic compounds of 250 mg/kg. Reviewing the health claim, the phenolic compounds referred to are the secoiridoid derivatives of hydroxytyrosol and tyrosol. A method based on absolute quantification of phenolic compounds in olive oil is proposed for the recognition of the health claim. The method involves liquid–liquid extraction with a 1:8 (v/v) oil:extract ratio to avoid saturation of the extract in oils with a higher phenolic content and its subsequent determination through LC–MS/MS in multiple reaction monitoring (MRM) mode, the gold standard technique in many application fields because of its analytical features. The optimized method was applied to a set of 100 extra virgin olive oils (EVOOs), and the results obtained were compared with the classic Folin–Ciocalteu method. The comparison between the two methods showed that the classic method is a non-selective method that can be affected by many interferences and that the Folin method underestimates the real phenolic content. The exponential growth of the fruit-processing industry generates significant quantities of organic by-products, such as peels, seeds, and pomace, which represent a rich but underutilized source of bioactive polyphenols. Valorizing these residues is critical for the transition toward a circular bioeconomy, yet conventional extraction methods remain solvent-intensive and kinetically inefficient. This review provides a comprehensive analysis of emerging green extraction technologies, specifically Ultrasound-Assisted (UAE), Microwave-Assisted (MAE), Enzyme-Assisted (EAE), Pressurized Liquid (PLE), and Supercritical Fluid Extraction (SFE), and Pulsed Electric Field (PEF), applied to key industrial matrices including apple, citrus, grape, olive, and coffee. Comparative data demonstrate that intensification technologies significantly outperform conventional maceration, with UAE and MAE reducing processing times by up to 90% while enhancing polyphenol yields by 20–55% through mechanisms such as acoustic cavitation and dipole rotation. Furthermore, high-pressure methods exhibit tunable selectivity, enabling the specific recovery of heat-sensitive anthocyanins and bound phenolics without the use of toxic organic solvents. The study concludes that the future of industrial valorization lies in the adoption of hybrid technologies and sequential biorefinery strategies to achieve high-purity isolates with minimal environmental impact. The extensive utilisation of chemical fertilisers and pesticides in agricultural contexts has precipitated substantial environmental degradation, thereby amplifying the repercussions of climate change. Furthermore, this overuse poses a threat to the sustainability and resilience of global food production systems. The utilisation of microalgae-based biostimulants is a novel and sustainable approach that has the potential to enhance crop productivity and resilience, while reducing dependence on chemical pesticides and their negative effects. The present study evaluated the effectiveness of two novel microalgae-based formulations on the performance of processing tomato (Solanum lycopersicum) crops under field conditions in Spain and Portugal. The formulation comprised enzymatically hydrolysed biomass from L. platensis, N. gaditana and A. obliquus, in combination with olive mill wastewater (alpechin) and aromatic plant extracts. The mixture was applied through drip irrigation and foliar spraying. The application of combined foliar and drip treatments resulted in a substantial enhancement in gross yield up to 51.9%. Concurrently, the acceptable raw material yield demonstrated a notable increase up to 44.9%. Furthermore, an increase in average fruit weight by 2–9 g was recorded. A subsequent foliar nutrient analysis revealed elevated concentrations of N, P, K, Ca, Mg, Fe, and Cu in the plants treated with biostimulants, achieving 3.61, 52.94, 5.96, 36.53, 22.28, 60.41 and 71.32% respectively in the plot L4 with foliar treatment. Although the efficacy of pest control measures was slightly lower than that of conventional pesticides, no significant increase in the incidence of diseased was observed. These findings indicated that microalgae-based biostimulants have the potential to function as sustainable agricultural inputs capable of enhancing crop yields and quality while reducing dependence on chemical fertilisers and pesticides. The outcomes of the study demonstrate the efficacy of microalgae-based formulations in enhancing the yield and quality of tomato crops. This is achieved while maintaining optimal plant health and reducing the reliance on synthetic fertilisers and pesticides. Full article

Mineral oil hydrocarbons (MOH), comprising saturated (MOSH) and aromatic (MOAH) compounds, are ubiquitous lipophilic contaminants. This review critically examines their occurrence, toxicology, analysis, contamination sources, and mitigation strategies in the olive oil sector. Emphasis is placed on analytical evolution, highlighting online LC-GC-FID and the EN ISO 20122:2024 standard, including advances in saponification and epoxidation to minimize biogenic interferences. Monitoring data reveal that virgin olive oils from the market can sometimes exceed the 2.0 mg/kg limit for the MOAH. Ten times higher levels are usually found in olive pomace oils (OPOs). In OPO, solvent extraction causes a significant reconcentration of hydrocarbons remaining on the solid matter after physical extraction and accumulating during the open-air storage of pomace. Conversely, for virgin oils, contamination can occur at multiple points along the supply chain, but harvesting emerged as the most important critical step, often due to accidental contact with lubricants, greases, or hydraulic fluids. Post-milling operations may also contribute to contamination. Mitigation strategies rely on Good Agricultural and Manufacturing Practices, focusing on the systematic replacement of technical-grade lubricants with food-grade alternatives. Additionally, olive washing can reduce initial MOSH content, while refining further lowers levels, particularly in lighter fractions. Full article

Improving nitrogen (N) use efficiency in olive cultivation is essential to address the environmental burden of N fertilizers, whose recovery efficiency rarely exceeds 55%. This study evaluates the agronomic and environmental performance of chabazite-rich zeolite as a soil amendment to enable 50% N-fertilizer reduction in olive growing. A seven-year field experiment (2017–2023) was conducted at two sites in Emilia-Romagna (Italy)—one irrigated (Brisighella) and one rainfed (Bertinoro)—comparing four autochthonous varieties under zeolite amendment (ZEO, 50% N) versus conventional fertilization (CNT, 100% N). Vegetative growth, productive parameters, oil quality and environmental impacts (Life Cycle Assessment, ISO 14040/44) were monitored. Under irrigation, ZEO maintained vegetative and productive equivalence with CNT, sustaining commercially viable yields (0.5–2.3 t ha−1). Under rainfed conditions, variety-specific responses emerged: Colombina exhibited 126.2% greater trunk diameter and near-universal fruiting competence (88.9% vs. 29–35% productive plants) under ZEO, while Capolga showed treatment convergence. LCA revealed higher per-unit environmental impacts for ZEO during early orchard phases due to front-loaded extraction burdens, progressively offset by annual N-input reductions. These findings demonstrate that zeolite amendment enables agronomically viable 50% N-fertilizer reduction, with efficacy modulated by water regime and genotype. Full article

Background: The Mediterranean diet (MD) represents a nutritionally balanced eating pattern characterized by high consumption of fruits, vegetables, legumes, nuts, whole grains, olive oil, fish, and extra-virgin olive oil as the principal fat source and limited intake of red meat and refined sugars. Emerging evidence indicates that the MD’s anti-inflammatory and antioxidant properties extend beyond systemic health, potentially reducing the risk and severity of periodontitis. This narrative review aimed to synthesize current evidence on the relationship between adherence to the MD and periodontal health outcomes. Methods: A comprehensive electronic literature search was conducted in PubMed without restrictions on publication date. Fourteen studies, ranging from 2019 to 2025, were included, encompassing human, clinical, experimental, and review designs that examined MD adherence and its effects on periodontal parameters. Eligible studies included cross-sectional, cohort, randomized controlled trials; systematic reviews; and animal models assessing clinical periodontal indices, inflammatory biomarkers, or microbial composition. Extracted data included study design, population characteristics, dietary assessment methods, and primary periodontal findings. Results: Most studies demonstrated that greater adherence to the MD was associated with improved periodontal parameters, including reduced probing pocket depth, clinical attachment loss, and bleeding on probing. Interventional trials showed significant reductions in systemic inflammatory markers such as IL-1β, TNF-α, and CRP, along with decreased counts of periodontopathogenic bacteria. Experimental studies further revealed the protective role of oleic acid and polyphenols in regulating macrophage activity, suppressing osteoclastogenesis, and enhancing IL-10 expression via epigenetic modulation. However, heterogeneity in dietary scoring systems, sample characteristics, and follow-up duration limited direct comparison, and not all associations reached statistical significance. Conclusions: Current evidence supports a beneficial association between MD adherence and periodontal health, mediated through anti-inflammatory, antioxidant, and microbiome-stabilizing mechanisms. Further standardized longitudinal and interventional studies are needed to confirm causality and refine nutritional strategies for periodontal disease prevention and management. Full article

In this study, an oleuropein-rich aqueous olive leaf extract was incorporated into a thermoresponsive sol–gel delivery system designed for localized application. The formulation was engineered to remain in a low-viscosity sol state at room temperature and to undergo a temperature-triggered sol-to-gel transition near physiological temperature (~33 °C), enabling in situ gel formation. Oleuropein content was quantified using a validated HPLC method, and the formulation was characterized with respect to physicochemical parameters, thermoreversible gelation behavior, particle size distribution, mechanical properties, and spreadability. Biological performance was evaluated through in vitro cytocompatibility (MTT assay), fibroblast migration (scratch assay), and collagen deposition (Sirius Red staining) in L929 fibroblasts, as well as antibiofilm activity against representative Gram-positive and Gram-negative bacterial strains. The developed sol–gel system demonstrated stable physicochemical characteristics, rapid and reversible thermogelation, suitable mechanical and spreading properties, concentration-dependent inhibition of biofilm formation, and acceptable cytocompatibility within the tested concentration range. Notably, the formulation supported fibroblast viability and collagen-associated responses at optimized concentrations. Overall, the results indicate that the proposed thermoresponsive sol–gel formulation represents a promising strategy for the localized delivery of oleuropein-rich olive leaf extract, combining physicochemical stability with dual wound-healing and antibiofilm functionality. This study evaluated olive leaves from three cultivars (Hojiblanca, Picual, and Arbequina) and olive pomace as complementary sources of bioactive compounds, comparing ultrasound-assisted extraction using organic solvents (UAE) with supercritical CO2 extraction (SFE). The aim was to determine how the plant matrix and extraction method influence phytochemical composition and functional properties, including antioxidant and antimicrobial activity. The results showed that both factors strongly affected extract composition and bioactivity. UAE favored the recovery of phenolic compounds associated with antioxidant activity, particularly in leaf extracts, while SFE promoted a distinct compositional profile enriched in flavonoids and lipophilic constituents, especially in olive pomace. Multivariate analysis confirmed a clear differentiation between matrices and extraction methods. Leaf extracts from Picual and Arbequina were mainly associated with phenolic compounds linked to antioxidant activity, including luteolin, ethyl vanillin, tyrosol, and isorhamnetin-3-O-glucoside. In contrast, olive pomace extracts were more strongly associated with flavonoids and lipophilic metabolites, such as triterpenes (oleanolic, maslinic, and ursolic acids) and lipid derivatives (oleic acid and lauric isopropanolamide). These compositional differences were reflected in biological activity: UAE extracts showed higher antioxidant activity, whereas SFE extracts, enriched in lipophilic and triterpenic compounds, exhibited stronger antimicrobial effects against Pseudomonas savastanoi and Hanseniaspora sp. Overall, these findings demonstrate that extraction-driven selectivity enables the production of olive-derived extracts with targeted functionalities, with UAE favoring antioxidant-oriented extracts and SFE promoting extracts enriched in lipophilic compounds with antimicrobial potential, particularly from olive pomace. This study aims to develop and characterize novel dermatocosmetic formulations designed to hydrate the skin, improve its appearance, reduce wrinkles, and provide antioxidant, anti-ageing, antimicrobial, and anti-inflammatory benefits, along with potential protection against UVA and UVB radiation. The formulations contain the following ingredients: xanthan gum (0.5%), Calendula officinalis oil (5%), Argania spinosa oil (5%), Helianthus annuus oil (5%), liposomes containing a hydroalcoholic extract of pomace from local red or white grapes (2%), an olive oil-based emulsifier (6%), vitamin E (0.5%), cetearyl alcohol (3%), propylene glycol (8%), and purified water (up to 100%). The natural ingredients used in these formulations, i.e., the red or white grape pomace extract from the aforementioned Romanian varieties, the oils of Calendula officinalis, Argania spinosa, and Helianthus annuus, xanthan gum, and the olive oil-based emulsifier (Olliva), promote the concept of ‘green cosmetics’. The use of liposomes to deliver bioactive substances from hydroalcoholic extracts allows the gradual release of active ingredients into the skin. An alternative for incorporating grape pomace extract into a cream-type matrix involves the use of liposomes. Liposomes loaded with red or white grape pomace extract were prepared using the thin-film hydration technique, followed by ultrasonication and extrusion. The obtained formulations were characterized using bio-physico-chemical analysis procedures in terms of consistency, colour, homogeneity, aroma, pH, stretch, texture, stability, and antioxidant activity/free radical scavenging capacity, as well as in vitro polyphenol release behaviour. These newly developed dermatocosmetic formulations were the subject of a patent application in Romania. Full article

Introduction: Olives have been used in traditional Mediterranean medicine for thousands of years to address the causes of inflammation, ageing and cognitive health. Traditional preparations of olive include olive oil and olive leaf extract, which are major components of diets that contribute to maintaining cognitive function and reducing neurodegenerative disease risk. Aims of the study: This systematic review aimed to synthesise experimental and limited human evidence on olive biophenols in neurodegenerative disease models, identify the most studied compounds, characterise their mechanisms of action, and evaluate key translational barriers. Materials and methods: Following PRISMA 2020 guidelines and registered with PROSPERO (CRD420251252252), primary studies investigating the effects of well-characterised olive biophenols in neurodegenerative relevant in vitro, in vivo, or human models were systematically reviewed. Each study was assessed for its design, experimental model, mechanistic outcomes and reported limitations. Risk of bias was evaluated using validated tools (SYRCLE/OHAT/ToxR) appropriate for preclinical and experimental study designs. Results: Among the 25 studies, 7 (28.0%) examined oleuropein or oleuropein aglycone, 10 (40.0%) focused on hydroxytyrosol or its derivatives, and 9 (36.0%) investigated oleocanthal. Most studies employed in vivo animal models (57.7%), predominantly transgenic mouse models of AD and toxin-induced PD models. Oleuropein-based studies reported inhibition of amyloid-β and α-synuclein aggregation with behavioural improvements. Hydroxytyrosol primarily exerted antioxidant and anti-inflammatory effects with modest cognitive benefits. Oleocanthal showed the most consistent anti-amyloid and anti-tau activity, including enhanced amyloid-β clearance across the blood–brain barrier. Most studies show a moderate risk of bias due to incomplete reporting, randomisation and blinding. Conclusions: Olive biophenols demonstrate consistent neuroprotective effects in preclinical models; however, translation to clinical application remains limited by pharmacokinetic constraints, methodological heterogeneity, and insufficient human evidence. The use of olive by-products in livestock farming is a valuable resource, given their high levels of bioactive compounds with antioxidant and health-promoting properties. This preliminary study adopted an integrated approach to evaluate the influence of dietary Olea europaea L. polyphenols on animal welfare, physiological stress response, intestinal morphofunctional traits, and meat quality in Neroametà finishing pigs, a novel Casertana × Large White genetic line (Neroametà). Thirty pigs reared under extensive farming conditions were randomly allocated to two groups (n = 15): a control group fed a standard diet (C) and a treatment group (OL) supplemented with 300 mg/head/day of olive polyphenolic extract for 90 days. The study focused on the systemic correlation between host health and product quality. Meat composition, rheological properties, meat antioxidant activity, stress parameters, and fatty acid profiles of the longissimus lumborum and psoas major muscles were analyzed. Results showed that the OL diet significantly modulated the HPA axis, as evidenced by a marked reduction in plasma ACTH and cortisol levels, alongside improved antioxidant status. These physiological changes were positively associated with a trophic effect on the intestinal mucosa, characterized by increased villus height and a more favorable villus/crypt ratio. Regarding meat quality, the OL group exhibited superior oxidative stability, optimized pH decline, and an improved intramuscular fatty acid profile (increased MUFA and n-3 PUFA, reduced SFA). Despite the pilot scale of 30 animals, these findings provide a solid foundation for characterizing the Neroametà breed. In conclusion, Olea europaea L. polyphenols act as a multi-level modulator, enhancing physiological resilience and meat quality, offering a sustainable strategy for high-quality pork production in line with circular economy and One Health principles.
