The main objective of this research consists in finding a rapid method for cheese lipidomics based on NMR data. This study plays an important role in differentiation and characterization of cheese samples in accordance with fat composition, especially in the case of fat substitution with exogenous animal or vegetal fat. Our findings play an important role in relation to religious requirements regarding non-allowed foods (pork fat, for example, in some cultures) and in the correct characterization of foods according to their lipidic profile. The approach consists in establishing a fingerprint region (0.86–0.93 ppm from 1H-NMR spectra) and then creating a database of the results obtained. The evaluation of the long-chain saturated fatty acids and the saturated short-chain fatty acids (C4 to C8) was established with a newly developed set of equations that make the computation possible even when mixtures of fats from different sources are present. This was accomplished by developing a new method for quantification of the fatty acid composition of different types of cheese, based on 1H-NMR spectroscopy. Principal component analysis (PCA) was applied to 40 cheese samples with varying degrees (0%, 5%, 12%, or 15%) of milk fat substitution (pork fat, vegetable fat, hydrogenated oils) and different clotting agents (calcium chloride or citric acid). The best sample discrimination was achieved using fatty acid profiles estimated from 1H-NMR data (using a total of six variables), explaining 89.7% of the total variance. Clear separation was observed between samples containing only milk fat and those with added fats. These results demonstrate that the integration of 1H-NMR spectroscopy with principal component analysis (PCA) provides a reliable approach for discriminating cheese samples according to their fat composition. The geochemistry of the fatty acids in the modern sediments in the Northern South Yellow Sea is still poorly studied, and studies on the geochemistry of the fatty acids in relatively long-core sediment samples are lacking. Thus, the fatty acids in the core sediments in the Northern South Yellow Sea were separated and identified to study their components and distribution characteristics, and the sources of organic matter and the early diagenetic evolution of the fatty acids in the sediments were discussed. The results show that saturated straight-chain fatty acids (methyl ester) have the highest content in the core sediments in the Northern South Yellow Sea, which account for 83.89% of the total fatty acids (methyl ester). nC16:0 is dominant, accounting for 30.48% of the n-saturated fatty acids (methyl ester). Unsaturated fatty acids (methyl ester) account for 7.59% of the total fatty acids (methyl ester). Binary unsaturated fatty acids (methyl ester) can only be detected in some samples, which are low in content and dominated by C18:2. Based on the components and distribution of the fatty acids (methyl ester) in the core sediments in the Northern South Yellow Sea, combined with the characteristics of other lipid biomarker compounds, the actual geological background, and previous research results, it is considered that the sources of organic matter in the core sediments are marine–terrestrial mixed materials, with terrestrial materials dominating. The fatty acids’ (methyl ester) CPI, the relative content of short-chain saturated fatty acids (methyl ester), and the unsaturated fatty acids (methyl ester) in the core sediments show non-obvious variation as the burial depth increases, reflecting that the fatty acids in the core sediments are strongly degraded at the early diagenetic stage, and this degradation is controlled by various complicated factors. Kidney transplant recipients face a substantial burden of premature mortality and morbidity, primarily due to persistent inflammation, cardiovascular risk, and nutritional deficiencies. Traditional nutritional interventions in this population have either focused on supplementing individual nutrients—often with limited efficacy—or required comprehensive dietary overhauls that compromise patient adherence. In this narrative review, we explore the rationale for dietary nut enrichment as a feasible, multi-nutrient strategy tailored to the needs of kidney transplant recipients. Nuts, including peanuts and tree nuts with no added salt, sugar, or oil, are rich in beneficial fats, proteins, vitamins, minerals, and bioactive compounds. We summarize the multiple post-transplant challenges—including obesity, sarcopenia, dyslipidemia, hypertension, immunological dysfunction, and chronic inflammation—and discuss how nut consumption may mitigate these issues through mechanisms involving improved micro-nutrient intake (e.g., magnesium, potassium, selenium), lipid profile modulation, endothelial function, immune support, and gut microbiota health. Additionally, we highlight the scarcity of randomized controlled trials in high-risk populations such as kidney transplant recipients and make the case for studying this group as a model for investigating the clinical efficacy of nuts as a nutritional intervention. We also consider practical aspects for future clinical trials, including the choice of study population, intervention design, duration, nut type, dosage, and primary outcome measures such as systemic inflammation. Finally, potential risks such as nut allergies and oxalate or mycotoxin exposure are addressed. Altogether, this review proposes dietary nut enrichment as a promising, simple, and sustainable multi-nutrient approach to support cardiometabolic and immune health in kidney transplant recipients, warranting formal investigation in clinical trials. Chronic inflammatory enteropathies (CIEs) in companion animals represent a group of idiopathic, immune-mediated gastrointestinal disorders in which the intestinal epithelium can be altered, affecting intestinal functionality, nutrient absorption, and microbiota composition. This review presents an overview of markers that could be used for the assessment of intestinal health, focusing extensively on functional biomarkers, with particular attention to fatty acids (including short-chain fatty acids, SCFAs) and amino acids. Studies have consistently shown reduced concentrations of SCFAs in companion animals with CIEs compared to healthy groups. These alterations occur with varying intensity depending on the type of enteropathy. Alterations in saturated, monounsaturated, and long-chain polyunsaturated fatty acids have also been reported in blood and feces, particularly in omega-3 and omega-6 derivatives, as well as in the elongase and desaturase indices responsible for endogenous synthesis. In addition, amino acids serve as precursors to key metabolites involved in mucosal immunity, oxidative stress regulation, and microbial homeostasis. In CIEs, alterations in systemic and fecal amino acid profiles have been observed, reflecting both host metabolic adaptation and microbial dysbiosis. Integrating fatty acid and amino acid profiles can help distinguish different types of enteropathies, providing additional discriminatory power for determining response to dietary treatment. Future research should aim to elucidate the causal relationships between metabolic alterations and disease pathogenesis, which could lead to novel dietary interventions targeting metabolic interactions between the microbiota and the host. A traditional dairy product from northern Colombia is suero costeño (SC), typically handmade through artisanal processes involving the natural fermentation of raw cow’s milk (RM); it is characterized by a creamy texture and a distinctive sensory profile, with a sour/salty taste and rancid odor. This study aimed to determine the chemical identity (using GC-FID/MSD) of SC and RM samples (from eight locations in the department of Córdoba-Colombia) by analyzing volatile components (trapped by HS-SPME and SDE) and fatty acid content. Consequently, the most notable results were as follows: (a) myristic (7–12%), stearic (12–17%), oleic (13–23%), and palmitic (21–29%) acids were the most abundant constituents [without significant differences among them (p > 0.05)] in both RM and SC fats; these were also expressed as polyunsaturated (2–5%), monounsaturated (26–36%), saturated (59–69%), omega-9 (19–30%), omega-6 (0.5–1.6%), and omega-3 (0.2–1.2%) fatty acids; (b) differences in the composition (p < 0.05) of the volatile fractions were distinguished between RM and SC samples; likewise, the SC samples differed (from each other) in their volatile composition due to the preparation processes applied (processes with raw milk and natural fermentation had less variability); nonetheless, it was possible to determine the volatilome for the artisanal product; and (c) the major components responsible for the chemical identity of SC were ethyl esters (of linear saturated and unsaturated acids, short/medium chains), aliphatic alcohols (linear/branched, short/long chains), aliphatic aldehydes (long chains, >C14), alkyl methyl ketones (long chains, >C11), sesquiterpenes (caryophyllane/humulane types), monoterpenes (mono/bi-cyclics), short-chain fatty acids, and aromatic alcohol/acid, among others. Obesity and cardiometabolic diseases (CMDs) have reached epidemic levels. Dysregulation of lipid metabolism is a risk factor for obesity and CMDs. Lipids are energy substrates, essential components of cell membranes, and signaling molecules. Fatty acids (FAs) are the major components of lipids and are classified based on carbon chain length and number, position, and stereochemistry of double bonds. They exert differential impacts on CMDs, such that saturated fat increases risks while very-long-chain n-3 FAs provide benefits. The functionalities of FAs, modulating membrane properties, acting as ligands for receptors, and serving as precursors for lipid mediators, are vital for insulin signaling, lipid metabolism, oxidative stress, and inflammatory response, collectively contributing to cardiometabolic health. This review examines recent advances in the characteristics and functional properties of different FAs in lipid structures, signaling pathways, and cellular metabolism to better understand the differential roles of different types of FAs in obesity and cardiometabolic health. In recent years, non-alcoholic beers have been gaining popularity. Among the various components that affect the flavor and sensory characteristics of beers, free fatty acids (FFAs) are minor components. However, due to their involvement in beer quality, fast and simple methods for analyzing FFAs in beers are of importance. In this work, we present a liquid chromatography–high-resolution mass spectrometry (LC-HRMS) method for the rapid determination of FFAs in beers, avoiding a tedious sample preparation and derivatization and allowing the simultaneous study of a large set of FAs, including medium-chain, long-chain, saturated, monounsaturated and polyunsaturated FAs. The method was applied in the analysis of twelve non-alcoholic and nine alcoholic beer samples from the local market, permitting the comparison of their FFA profiling. Among the 37 FAs studied, 29 were quantified, and palmitic, stearic, oleic and myristic acids were identified as the predominant FAs in both alcoholic and non-alcoholic beers. The majority of the predominant long-chain FAs, including palmitic, stearic and myristic acids, were found in decreased amounts in non-alcoholic beers, compared to the alcoholic ones, with the marked exception of oleic acid, which was increased in non-alcoholic beers. Among the medium-chain FAs, octanoic acid was found at lower concentrations in non-alcoholic beers, comparing to regular beers, while lauric acid was slightly increased. Principal component analysis (PCA) suggested the correlation of FFAs with the type of beer (alcoholic or non-alcoholic beer). Full article

The n-3 long-chain polyunsaturated fatty acids (n-3 LC-PUFAs) have a key role in maintaining fish growth and health. However, fish oil (FO), the main source of n-3 LC-PUFAs, is in relative shortage due to the rapid development of the aquaculture industry. In this study, we investigated the efficacy of replacing fish oil with mixtures of algal oil (AO) from Schizochytrium sp. and terrestrially sourced oils (animal oil poultry oil (PO) or vegetable oil rapeseed oil (RO)) in the diets of juvenile tiger puffer (average initial body weight 23.8 ± 1.51 g). An 8-week feeding trial was conducted using three experimental diets: a control diet containing 6% added FO (control FO-C) and two diets with 3% AO + 3% PO or RO (groups AO+PO and AO+RO, respectively), replacing FO. Each diet was fed to triplicate tanks with 25 fish in each tank. The weight gain, feed conversion ratio, body composition, and serum biochemical parameters were not significantly different among the three groups, except that the AO+PO group had a significantly lower muscle lipid content than the other two groups. The AO-added diets significantly increased the DHA content in whole fish, muscle, and liver samples but significantly reduced the EPA content. The oil mixture treatments significantly increased the contents of monounsaturated fatty acid (MUFA) but significantly decreased the contents of saturated fatty acids (SFAs) in the liver and whole fish samples. However, the MUFA and SFA contents in the muscle samples were not significantly different among the dietary groups. The diets with oil mixtures did not affect the hepatic histology but tended to result in the atrophy of intestinal villi. The treatment diets downregulated the hepatic gene expression of proinflammatory cytokines (il-1β and tnf-α) and the fibrosis marker gene, acta2. However, the AO+PO diet inhibited the intestinal gene expression of the tight junction protein, claudin 18. In the muscle, the treatment diets upregulated the expression of genes related to cell differentiation and apoptosis (myod, myog, myf6, myf5, bcl-2, and bax). In conclusion, Schizochytrium sp. oil in combination with terrestrial oils (poultry oil or rapeseed oil) can be an effective alternative to fish oil in the diets of tiger puffer, but the mixing strategy may be better modified in consideration of intestinal health. Endometrial cancer (EC) is a complex gynecologic malignancy that requires a deeper understanding of its molecular basis to improve therapeutic strategies. In this study, we investigated the role of fatty acid (FA) reprogramming in the progression of EC. We analyzed FA profiles to identify the stage-specific changes and gene expression profiles of key enzymes involved in FA synthesis, desaturation, elongation, transport, and oxidation at different stages of EC. Our results show that EC tissues have lower levels of saturated FA and branched-chain FA, higher levels of very long-chain FA, n-3 polyunsaturated FA (PUFA), and monounsaturated FA, with the exception of myristoleic acid. The differences in n-6 PUFA were inconsistent. Gene expression analysis revealed the upregulation of key enzymes controlling de novo FA synthesis, including ACACA, FASN, SCD1, and ELOVL1. In contrast, the expression of genes related to FA transport in the cell and β-oxidation was downregulated. The expression of some genes related to PUFA metabolism was upregulated, while others were downregulated. These results demonstrate a reprogramming of lipid metabolism in EC tissues and suggest potential targets for novel therapeutic interventions in EC. Full article

Background: Osteoporosis is frequently observed in patients with chronic pancreatitis, and both conditions are closely associated with systemic metabolic disorders. However, the underlying mechanisms linking chronic pancreatitis and osteoporosis remain unclear. Methods: In this study, we utilized high−performance liquid chromatography–mass spectrometry (HPLC−MS) to conduct metabolomics and lipidomics analyses on pancreatic, serum, and other tissues from a mouse model of chronic pancreatitis−induced osteoporosis (CP−OP), with the aim to elucidate the metabolism−related pathogenic mechanisms of CP−OP. Results: We identified over 405 metabolites and 445 lipids, and our findings revealed that several metabolites involving the tricarboxylic acid (TCA) cycle, as well as triacylglycerols and diacylglycerols with higher saturation, were significantly increased in the CP−OP model. In contrast, triglycerides with higher unsaturation were decreased. Differential pathways were enriched in n−3 long−chain polyunsaturated fatty acid metabolism in both pancreatic and bone tissues, and these pathways exhibited positive correlations with bone−related parameters. Furthermore, the modulation of these polyunsaturated fatty acids by Qingyi granules demonstrated significant therapeutic effects on CP−OP, as validated in mouse models. Conclusions: Through the metabolomics approach, we uncovered that disorders in polyunsaturated fatty acids play a critical role in the pathogenesis of CP−OP. This study not only enhances our understanding of the pathogenesis of CP−OP but also highlights the therapeutic potential of targeting polyunsaturated fatty acids as a future intervention strategy for osteoporosis treatment. Lipotoxicity, resulting from the buildup of excess lipids in non-adipose tissues, is increasingly recognized as a major contributor to the progression of kidney disease, highlighting the need for alternative models to assess its effects on renal cells. The main aim of this study was to investigate the usefulness of Caki-1, a human proximal tubule (PT) and renal cell carcinoma (RCC) representative cell line, as a 3D model system for studying free fatty acid-induced PT lipotoxicity. Caki-1 spheroids were generated and maintained on ultra-low attachment plates and characterized regarding time-dependent morphology changes. In optimal 3D culture conditions, Caki-1 cells formed well-defined large compact spheroids with uniform morphology, good circularity, and increased diameter from days 4–12. Chronic exposure to saturated palmitate resulted in dose- and time-dependent spheroid disintegration and cell death, including dispersed and flattened spheroid morphology, with increased dead cells in the peripheral layers and decreased spheroid core. Moreover, palmitate-treated spheroids showed a significant increase in cleaved poly(ADP-ribose) polymerase (PARP) and active caspase-3. Palmitate-induced PARP cleavage, as well as endoplasmic reticulum (ER) stress and autophagy dysfunction, were blunted by triacsin C, an inhibitor of long-chain acyl-CoA synthetases. In addition, co-incubation with unsaturated oleate prevented palmitate-induced spheroid disintegration and apoptotic cell death in Caki-1 3D culture. While fatty acid overload upregulated lipid droplet protein perilipin 2 in Caki-1 cells, knockdown of perilipin 2 by siRNAs resulted in an exacerbation of palmitate-induced cell death. Together, these results indicate that the 3D Caki-1 spheroid model is a simple and reproducible in vitro system for studying renal lipotoxicity and lipid metabolism that gives useful readouts at the molecular, cellular, and multicellular levels. Background/Objectives: This research aimed to investigate phospholipid fatty acid (PLFA) distribution in the brain, kidneys, and white adipose tissue (WAT) and lipid profiles in response to high-fat diets. Methods: Adult female C57BL/6 mice were fed high-fat diets containing 25% linseed, palm, or sunflower oil for 100 days. The fatty acid composition of dietary oils and tissue PL were analyzed using gas–liquid chromatography. Results: Linseed oil increased n-3 polyunsaturated fatty acids (PUFAs) with subsequent conversion into long-chain n-3 PUFAs in the brain and kidney PL, while only alpha-linolenic acid was elevated in WAT. Palm and sunflower oils resulted in unique PLFA distributions in the kidneys and WAT. Palm oil raised linoleic acid without conversion to pro-inflammatory n-6 PUFAs. Sunflower oil increased saturated palmitic acid, as opposed to the rise in monounsaturated oleic acid. Linseed oil also significantly improved lipid profiles, reducing LDL and increasing HDL levels while enhancing cardiovascular indices. Conclusions: This study demonstrates that dietary oils significantly impact organ-specific PLFA profiles, with linseed oil enriching brain and renal n-3 PUFAs, while palm and sunflower oils induce distinct modifications in the kidney and WAT. Moreover, linseed oil offers notable cardioprotective benefits due to the favorable lipid profile changes. These findings highlight the importance of dietary fat selection in achieving balanced lipid metabolism and suggest that diverse oil combinations may be essential for optimizing health outcomes. This study evaluated the effects of different lipid sources—fish oil (FO), soybean oil, rapeseed oil, and palm oil—on the growth and muscle quality of golden pompano (Trachinotus ovatus) cultured in offshore cages for 10 weeks. Three diets (D1–D3) were formulated: D1 used only fish oil, D2 blended fish, rapeseed oil, and palm oil, and D3 combined fish and soybean oils. Fish in the D1 group showed the highest weight gain, specific growth rate, and muscle protein content, significantly outperforming D3. No significant differences in muscle lipid content or edible quality were found between groups. D1 had the highest levels of long-chain and n-3 polyunsaturated fatty acids (PUFA), while D3 had higher n-6 PUFA. Saturated and monounsaturated fatty acids were higher in D1 and D2 than in D3. Muscle volatiles like aldehydes and amines were elevated in D1, with more pleasant flavors compared to D2 and D3. Muscle texture was superior in D2. These results suggest that rapeseed and palm oils can enhance growth, flavor, and texture in fish on low FO diets, offering a sustainable alternative to reduce reliance on marine-based feed in aquaculture. X-adrenoleukodystrophy (X-ALD) is a peroxisomal metabolic disorder caused by mutations in the ABCD1 gene encoding the peroxisomal ABC transporter adrenoleukodystrophy protein (ALDP). Similar mutations in ABCD1 may result in a spectrum of phenotypes in males with slow progressing adrenomyeloneuropathy (AMN) and fatal cerebral adrenoleukodystrophy (cALD) dominating most cases. Mouse models of X-ALD do not capture the phenotype differences and an appropriate model to investigate the mechanism of disease onset and progress remains a critical need. Here, we generated induced pluripotent stem cell (iPSC) lines from skin fibroblasts of two each of apparently healthy control, AMN, and cALD patients with non-integrating mRNA-based reprogramming. iPSC lines expanded normally and expressed pluripotency markers Oct4, SOX2, NANOG, SSEA, and TRA-1–60. Expression of markers SOX17, Brachyury, Desmin, OXT2, and beta tubulin III demonstrated the ability of the iPSCs to differentiate into all three germ layers. iPSC-derived lines from CTL, AMN, and cALD male patients were differentiated into astrocytes. Differentiated AMN and cALD astrocytes lacked ABCD1 expression and accumulated saturated very long chain fatty acids (VLCFAs), a hallmark of X-ALD, and demonstrated differential mitochondrial bioenergetics, cytokine gene expression, and differences in STAT3 and AMPK signaling between AMN and cALD astrocytes. These patient astrocytes provide disease-relevant tools to investigate the mechanism of differential neuroinflammatory response in X-ALD and will be valuable cell models for testing new therapeutics. Dietary lipid intake can influence the gut microbiota (GM) and their metabolites, such as short-chain fatty acids (SCFA) and bile acids, which are key mediators of health. The objective is to examine how dietary lipids’ quantity and quality influence the GM and metabolite profiles. A literature review of 33 studies in animals and humans was performed on the effects of saturated fatty acids (SFAs), monounsaturated fatty acids (MUFAs), polyunsaturated fatty acids (PUFAs), trans-fatty acids (TFAs), and sterols on GM composition and gut-derived metabolites. The results show that diets rich in MUFAs, n-3 PUFAs, and short-chain FAs have the potential to enhance beneficial bacteria and metabolites. In addition, trans-palmitoleic acid, conjugated linoleic acid, and phytosterols may also have potentially beneficial effects on GM, but more research is needed. Medium-chain FAs and n-6 PUFAs have variable effects on the GM. Conversely, intakes of high-fat diets, long-chain SFAs, industrial TFAs, and cholesterol disrupt GM balance. In conclusion, animal studies clearly demonstrate that dietary fats influence the GM and related metabolites. Yet, human studies are limited. Therefore, well-designed human studies that consider the whole diet and baseline health status are needed to better understand the effects of dietary lipids on GM. Full article
