Climate change has a significant negative impact on agriculture and food production. This trend requires technological development and the adaptation of new technologies in both the grapevine production and winemaking sectors. High temperatures and heat accumulation during the growing season result in faster ripening and a higher sugar content, leading to a higher alcohol content during fermentation. The negative consequences are an imbalanced wine character and consumer reluctance, as lower alcoholic beverages are now in high demand. Over the last decade, several methods have been developed to handle this impact and reduce the alcohol content of wines. In this study, we used the MASTERMIND® REMOVE membrane-based dealcoholization system to reduce the alcohol concentration in of Pinot gris wines from 12.02% v/v to 10.69% v/v and to investigate the effect on analytical parameters in three steps (0.5%, 1%, and 1.5% reductions) along the treatment. To evaluate the impact of the partial alcohol reduction and identify correlations between the wine chemical parameters, data were analyzed with ANOVA, PCA, multivariate linear regression and cluster analysis. The results showed that except for the extract, sugar content and proline content, the treatment had a significant effect on the chemical parameters. Both free and total SO2 levels were significantly reduced as well as volatile acid, glycerol and succinic acid levels. It must be highlighted that some parameters were not differing significantly between the untreated and the final wine, while the change was statistically verified in the intermediate steps of the partial alcohol reduction. This was the case for example for n-Propanol, i-Amylalcohol, Acetaldehyde, and Ethyl acetate. The multivariate linear regression model explained 18.84% of the total variance, indicating a modest but meaningful relationship between the alcohol content and the investigated analytical parameters. Our results showed that even if the applied instrument significantly modified some of the wine chemical parameters, those changes would not influence significantly the wine sensory attributes. The production of bionanocomposite films based on carboxymethyl derivatives of starch and cellulose with sodium montmorillonite (MMT-Na) as a filler was described. The developed films with high absorbency can be used in the preparation of adhesive dressings for wounds oozing as a result of abrasions or tattoos. Carboxymethyl cellulose (CMC), carboxymethyl starch (CMS), and potato starch were used as the raw materials for film manufacturing. Citric acid was used as a crosslinking agent and glycerol as a plasticizer. The following parameters were evaluated for the obtained films: solubility in water, swelling behavior, moisture absorption, and mechanical durability (tensile strength, elongation at break, and Young’s modulus). This study revealed that filler concentration has a significant influence on the stability, durability, and moisture absorption parameters of films. The best nanocomposite with a high absorption capacity was a two-component film CMS/CMC containing 5 pph of sodium montmorillonite and can be used as a base material for wound dressing, among other applications. Bilberry (Vaccinium myrtillus L.) fruits represent the recognized wellspring of bioactive compounds with various documented bioactivities. Although bilberry leaves are often treated as industrial by-products, they also represent a valuable source of phytochemicals with potential dermocosmetic applications. In this study, extracts of bilberry fruits and leaves were prepared using both conventional solvents (water and 50% ethanol) and natural deep eutectic solvents (NaDES) as green, biodegradable alternatives. The aim of this study was to examine the UV protective activity and inhibitory potential of those extracts against some enzymes (tyrosinase, hyaluronidase, collagenase) that are important in terms of skin conditioning and skin aging. The results of in vitro tests have shown the superiority of NaDES extracts compared to conventional extracts regarding all tested bioactivities. In addition, bilberry leaves extracts were more potent compared to fruit extracts in all cases. The most potent extract was bilberry leaf extract made with malic acid–glycerol, which exhibited strong anti-tyrosinase (IC50 = 3.52 ± 0.26 mg/mL), anti-hyaluronidase (IC50 = 3.23 ± 0.30 mg/mL), and anti-collagenase (IC50 = 1.84 ± 0.50 mg/mL) activities. The correlation analysis revealed correlation between UV protective and anti-tyrosinase, UV protective and anti-collagenase as well as between anti-hyaluronidase and anti-collagenase activity. UV protection and anti-tyrosinase activity correlated significantly with chlorogenic acid and hyperoside contents in extracts. The extracts with the best activities also demonstrated a good safety profile in a 24 h in vivo study on human volunteers. Full article

The batch, repeated batch and fed-batch cultivation strategies, in stirred tank bioreactors, were evaluated to maximize biomass production and the cells’ desulfurization activity (CDA) of Rhodococcus qingshengii IGTS8. The batch culture reached 2.62 g DCW/L biomass, with a productivity of 0.03 g DCW·L−1·h−1 and only 26% glycerol consumption. The repeated batch strategy reduced cultivation time during the first cycle, increasing biomass production by 15%, with 30% glycerol consumed and productivity 2.3 times higher than the batch process; however, subsequent cycles showed no further improvement. CDA peaked early in both modes but declined to 12–13 U/mg DCW by the end of the exponential growth phase. Fed-batch cultivation achieved 8.35 g DCW/L with 87% glycerol consumption, resulting in a threefold increase in volumetric productivity and a 1.7-fold higher specific growth rate compared with the batch mode. CDA remained stable during the fed-batch process and was approximately 40% higher compared with the batch and repeated batch processes. The fed-batch culture was used directly in a two-phase bubble column bioreactor for the desulfurization of dibenzothiophene (DBT), 4-methyl-dibenzothiophene (4-MDBT) and their mixture. The complete desulfurization of 1.4 mM DBT was achieved at a rate of 21.6 mmol DBT/kg DCW/h, while 0.9 mM 4-MDBT was fully converted but at a 2.5-fold lower rate. The simultaneous conversion of the DBT/4-MDBT mixture showed reduced efficiencies of 59.6% and 41.2%, respectively. Full article

The brown planthopper (BPH), Nilaparvata lugens, is the most destructive insect pest of rice. BPH infestations severely threaten rice yield worldwide. The gustatory receptor NlGr23 plays a critical role in mediating the repulsive reaction to oxalic acid of the BPH. We integrated transcriptomic and proteomic analyses to determine the metabolic and behavioral consequences of NlGr23 silencing. The RNAi-mediated knockdown of NlGr23 increased body weight and honeydew production, indicating enhanced feeding activity. The results of multiomics profiling revealed disrupted lipid homeostasis, identifying 187 differentially expressed genes and 150 differentially expressed proteins. These genes were enriched in pathways including glycerophospholipid metabolism, fatty acid biosynthesis, and AMPK signaling. The results of biochemical assays showed that NlGr23 silencing elevated triacylglycerol levels by 68.83%, and reduced glycerol and free fatty acid levels, suggesting impaired lipolysis. The NlGr23 loss-of-function mutation mechanistically activates the AMPK pathway, suppresses lipid breakdown, and promotes energy storage. This study established NlGr23 as a key regulator linking chemosensation to metabolic reprogramming, providing new insights into gustatory receptor-mediated energy homeostasis in the BPH. Full article

Glycerol fatty acid esters (GFAEs) are recognized for their potential to improve lipid metabolism, energy utilization, and gut health due to their excellent emulsifying and antimicrobial properties. The objective of this research was to investigate the effects of dietary GFAE supplementation on production performance, serum biochemical profiles, and rumen fermentation in beef cattle. Thirty crossbred Simmental bulls, averaging 507.42 ± 9.59 kg in body weight, were assigned to three distinct cohorts, with 10 animals in each cohort. The CON cohort was fed a basal diet devoid of GFAE, whereas the treatment cohorts (GFAE1 and GFAE2) received GFAE supplements at concentrations of 0.1% and 0.2% of the dietary dry matter, respectively. Compared with the control group, supplementation with 0.1% GFAE significantly increased the ADG of beef cattle by 12.14% (p < 0.05); compared with the GFAE2 group, ADG was 7.86% higher (p > 0.05). The digestibility of NDF and ADF was significantly enhanced in the GFAE1 group relative to the control group (p < 0.05). Dietary GFAE supplementation significantly elevated rumen acetate, propionate, and total volatile fatty acid concentrations in both the GFAE1 and GFAE2 groups compared to the control group (p < 0.05). In contrast to the control group, there was a notable rise in serum levels of T-AOC, UREA, and TG in both GFAE1 and GFAE2 groups (p < 0.05). Conversely, the concentration of HDL-C was significantly decreased in the GFAE2 group. Additionally, at the phylum level, the abundance of Fibrobacterota was significantly higher in the GFAE1 group than in the control group (p < 0.01). At the genus level, the proportions of Bacteroides and Fibrobacter were significantly higher in the GFAE1 group compared to the control group (p < 0.05). In conclusion, this study demonstrates that the addition of 0.1% GFAE to beef cattle diets significantly enhances the digestibility of ADF and NDF nutrients, increases serum total antioxidant capacity, urea, and triglycerides, optimizes rumen fermentation parameters and microbial community structure, and ultimately improves production performance. As a result of the findings from this research, it is suggested that 0.1% GFAE be incorporated into the diet for beef cattle. Full article

Tailoring culture media and supplementation strategies to the specific requirements of a target product is essential for enhancing microbial production efficiency. This work addresses an unexplored aspect of K. phaffii cultivation: optimizing culture media for metabolite production from xylose, diverging from the conventional focus on recombinant protein expression and the use of glycerol or methanol as primary substrates. Ethylene glycol biosynthesis in an engineered K. phaffii strain was improved by evaluating media and nutrient supplementation. Among the seven evaluated formulations, FM22 and d’Anjou were the most effective, with inositol and thiamine dichloride playing key roles in enhancing production. Salt concentrations in both media were optimized using Central Composite Design (CCD), reducing complexity while increasing yields. Ethylene glycol production increased by 54% in FM22 and 21% in d’Anjou, accompanied by a threefold and 26% reduction in the total salt content, respectively. The vitamin solution was streamlined from seven to two components, each at half the standard concentration. Trace element solutions were reduced to 25% of the original volume without compromising productivity. These findings underscore the dual benefit of culture medium optimization: improved ethylene glycol yields and simplified formulations, establishing a foundation for the development of more efficient and cost-effective bioprocesses using K. phaffii. The Mo:BiVO4/FeOOH photoelectrode was synthesized through the deposition of FeOOH onto the surface of the Mo:BiVO4 photoelectrode. The composite photoelectrode demonstrated a photocurrent of 1.8 mA·cm−2, which is three times greater than that observed for pure BiVO4. Furthermore, the glycerol conversion rate was recorded at 79 μmol·cm−2·h−1, approximately double that of pure BiVO4, while the selectivity for glyceraldehyde reached 49%, also about twice that of pure BiVO4. The incorporation of Mo has been shown to enhance the stability of the BiVO4. Additionally, Mo doping improves the efficiency of electron-hole transport and increases the carrier concentration within the BiVO4. This enhancement leads to a greater number of holes participating in the formation of iron oxyhydroxide (FeOOH), thereby stabilizing the FeOOH co-catalyst within the glycerol conversion system. The FeOOH co-catalyst facilitates the adsorption and oxidation of the primary hydroxyl group of glycerol, resulting in the cleavage of the C−H bond to generate a carbon radical (C). The interaction between the carbon radical and the hydroxyl group produces an intermediate, which subsequently dehydrates to form glyceraldehyde (GLAD). Full article

Various pretreatment methods for the valorization of sunflower husks (SHs) for H2 gas generation through fermentation by Escherichia coli were investigated. We analyzed thermal treatment (TT), acid hydrolysis (AH), and alkaline hydrolysis (AlkH) at different substrate concentrations (50 g L−1, 75 g L−1, 100 g L−1, and 150 g L−1) and dilution levels (undiluted, 2× diluted, and 5× diluted). A concentration of 75 g L−1 SH that was acid-hydrolyzed and dissolved twice in the medium yielded optimal microbial growth, reaching 0.3 ± 0.1 g cell dry weight (CDW) L−1 biomass. The highest substrate level enabling effective fermentation was 100 g L−1, producing 0.37 ± 0.13 (g CDW) × L−1 biomass after complete fermentation, while 150 g L−1 exhibited pronounced inhibitory effects. It is worth mentioning that the sole alkaline treatment was not optimal for growth and H2 production. Co-fermentation with glycerol significantly enhanced both biomass formation (up to 0.42 ± 0.15 (g CDW) × L−1)) and H2 production. The highest H2 yield was observed during batch growth at 50 g L−1 SH hydrolysate with 5× dilution, reaching up to 5.7 mmol H2 (g sugar)−1 with glycerol supplementation. This study introduces a dual-waste valorization strategy that combines agricultural and biodiesel industry residues to enhance clean energy generation. The novelty lies in optimizing pretreatment and co-substrate fermentation conditions to maximize both biohydrogen yield and microbial biomass using E. coli, a widely studied and scalable host. This study analyzes the aromatic profiles of kiwi-based fermented beverages, inoculated with varying proportions of milk kefir grains and incubated under different shaking rates. The experiments were designed using response surface methodology and three consecutive batch cultures were performed under each experimental condition. At the end of each fermentation, the grains were separated from the beverage and reused as the inoculum for fermenting fresh kiwi juice in the subsequent batch. Based on the results, together with the previously determined microbiological and chemical characteristics, two beverages were identified as having broader aromatic profiles, lower contents of sugars, ethanol, and acids, and high counts of lactic acid bacteria (LAB) and yeasts (>106 CFU/mL). These beverages were produced under relatively low agitation rates (38 and 86 rpm) and high inoculum proportions (4.33% and 4.68% w/v) during the second and third batch cultures, respectively. Over 28 days of refrigerated storage, the pH values of both beverages remained relatively stable, and the LAB counts consistently exceeded 106 CFU/mL. Yeast counts, along with the production of ethanol, glycerol, lactic acid, and acetic acid, increased slightly over time. In contrast, the concentrations of citric acid, quinic acid, total sugars, and acetic acid bacteria declined by day 28. Full article

The catalytic conversion of bio-based glycerol (Gly) into high-value glycerol carbonate (GC) has received great attention from both the academic and industrial fields. The development of highly efficient catalysts and economical industrial processes remains a challenging subject. In this mini-review, we summary the recent advances in catalyst design, characterization, mechanism, and catalytic process optimization, including the various synthetic strategies of GC, such as the coupling of CO2 and Gly or its derivatives like glycidol (GD), the transesterification of Gly with small carbonate-containing molecules, and the carbonylation of Gly with urea. The main difficulties and challenges faced by constructing high-performance catalysts and achieving scale production of GC have been put forward, and the future research directions and opportunities in catalyst innovation, reaction mechanism exploration, and continuous catalytic process improvement have also been suggested. Although these are established products, there is great interest in the development of new products, particularly new vinegars, and among these, the possibility of vinegars containing gluconic acid stands out. Gluconic acid in vinegar, mainly produced by acetic acid bacteria (AAB), is positively valued by consumers. Its content depends on the availability of glucose in the base wine; however, this hexose is preferentially consumed by the indigenous yeast population which conducts the previous alcoholic fermentation. For this reason, the use of non-conventional fructophilic yeasts, which consume fructose rather than glucose, is required. In this work, we isolated, screened, and identified osmophilic and fructophilic non-Saccharomyces yeasts from sun-dried grape must and tested them under different fermentation conditions in synthetic and natural grape musts, in order to obtain a base wine with ethanol and glucose content for the development of new vinegars containing gluconic acid. The isolate of the species Starmerella lactis-condensi was found to be an ideal candidate due to its fructophilic and osmophilic features, which allowed for the production of a base wine with high ethanol (11% v/v) and glucose (up to 200 g/L) content from a natural concentrated must. In fresh must, inoculation with Starmerella lactis-condensi resulted in faster and preferential fructose consumption over glucose compared to the control. However, both sugars were completely consumed at the end of the alcoholic fermentation; therefore, new fermentation strategies should be tested in this type of must. Furthermore, this strain could be of interest in oenology due to its high glycerol yield and low volatile acid production during alcoholic fermentation. The use of this strain could allow for the production of new wines with unique metabolic profiles suitable for further vinegar production. Full article

Floridoside (2-O-D-glycerol-α-D-galactopyranoside) is a natural product typically found in red algae. It serves as the algae’s carbon reserve and is produced as a protective response against osmotic and heat stress. Both floridoside and its acylated derivatives have been associated with modulating redox homeostasis and inflammatory responses. Therefore, we aimed to evaluate whether the newly synthesized floridoside phosphotriesters (1b1d, 1f1h) and acylated floridoside derivative (1e) can modulate the oxidative burst in stimulated human neutrophils. Synthetic strategies included the glycosylation of the thioglycoside donor with glycerol derivatives, having NIS/TfOH as the promoter. Phosphorylation was achieved with POCl3 in the presence of pyridine. The compounds were analysed for their cytotoxicity, with 1b and 1h being cytotoxic at 50 μM, while the others showed no cytotoxicity in the tested concentrations. The detection of the neutrophils’ oxidative burst was performed using multiple probes [luminol, aminophenyl fluorescein (APF), and Amplex Red (AR)] to evaluate reactive species levels. Compound 1e prevented the oxidative burst in activated human neutrophils (IC50 = 83 ± 7 μM). All the other tested compounds were ineffective in inhibiting APF and AR oxidation under the present experimental conditions. These findings highlight the potential of floridoside-based derivatives as candidates for targeting inflammatory pathways. Full article

Large-scale bioprocesses often suffer from spatial heterogeneities, which impact microbial performance and often lead to phenotypic population heterogeneity. To better understand these effects at the single-cell level, this study applied, for the first time, automated real-time flow cytometry (ART-FCM) to monitor L-phenylalanine production with an Escherichia coli triple reporter strain in a fed-batch process with glycerol as the carbon source. The strain was cultivated in both a well-mixed stirred-tank bioreactor (STR) and a scale-down two-compartment bioreactor (TCB), consisting of an STR and a coiled flow inverter (CFI) in bypass, to simulate spatial heterogeneities. ART-FCM enabled autonomous, high-frequency sampling every 20 min, allowing for real-time tracking of fluorescence signals linked to growth (rrnB-mEmerald), oxygen availability (narGHIJ-CyOFP1), and product formation (aroFBL-mCardinal2). The STR exhibited uniform reporter expression and higher biomass accumulation, while the TCB showed delayed product formation and pronounced phenotypic diversification depending on the set mean residence time in the CFI. Single-cell fluorescence distributions revealed that the shorter mean residence time in the CFI resulted in pronounced subpopulation formation, whereas longer exposure attenuated heterogeneity, indicating transcriptional adaptation. This finding highlights a critical aspect of scale-down studies: increased exposure duration to perturbations can enhance population robustness. Overall, this study demonstrates the relevance of ART-FCM, in combination with a multi-reporter strain, as a pioneering tool for capturing dynamic cellular behavior and correlating it to process performance, providing deeper insights into microbial heterogeneity under fluctuating bioprocess conditions. People have long been interested in plants rich in secondary metabolites and have tried to isolate them outside the plants. Purple coneflower is a crucial medicinal plant, known for its broad spectrum of bioactive substances. The type of solvent and the duration of maceration had an important impact on the phenolic level of purple coneflower leaves, flowers and roots. The flowers and leaves had a significantly higher phenolic concentration than the roots. The results of this study show the importance of stabilizing and regulating the polarity of the solvent and the duration of maceration to obtain the highest yield of phenolics from purple coneflower. The greatest yield of phenolics was gained with two natural extraction solvents, 5% acetic acid and glycerol, after 3 days of maceration, yielding (1696.05 mg 100 g−1 DW) and (2796.94 mg 100 g−1 DW) from the flowers of purple coneflower, respectively. For purple coneflower leaves and roots, the best extraction method was 40% ethanol after 3 days of maceration, and the total content analyzed was 1022.43 mg 100 g−1 DW and 1011.32 mg 100 g−1 DW, respectively. Different phenolics respond significantly to different maceration factors, but the best product from the flowers of purple coneflower is glycerol extract after 9 days of maceration. From the leaves of purple coneflower, we obtained the highest phenolic yield when producing extract in glycerol, a 40% ethanol tincture, or an acetic acid product after 3 days of maceration.
