Scorpions possess unique, ornate mid-ventral sensory organs called pectines. The pectines are used to process chemo- and mechanosensory information acquired from the ground as the animal walks, and they are implicated in a variety of behaviors including navigation and detection of mates and prey. Many previous researchers have investigated pecten function by cutting the organs from the animals (full ablation) and comparing their behaviors with those of intact scorpions. This drastic approach is likely to not only cause enormous stress to the ablated animals but also change their behavior. Here, we have developed a method for gently and reversibly impairing the pectines by partially covering them to prevent them from lowering to the ground. Specifically, we fabricated small rectangles of a commercially available lightly adhesive foil tape that we placed across the pectines and secured to the body wall with a thin strip of a more strongly adhesive lab tape. Using a repeated measures design, we monitored the animals’ locomotory activity overnight in small behavioral arenas under three conditions: unmodified (intact) control, pectines restrained, and sham control. We found that scorpions with their pectines restrained had a significant increase in both the distance and duration of movement when compared to unmodified and sham control animals. Our method allows for temporary, reversible compromise of pecten function and should be useful in fully understanding the role of pectines in behavior. Full article

Browning significantly impacts the commercial value of luffa (luffa cylindrica) and is primarily driven by the metabolic processes of phenolic acids. Investigating changes in phenolic acids during browning aids in understanding the physiological mechanisms underlying this process and provides a basis for improving storage, processing, variety breeding, and utilization of germplasm resources. This study compared browning-resistant (‘30’) and browning-sensitive (‘256’) luffa varieties using high-throughput sequencing and metabolomics techniques. The results revealed 55 genes involved in the phenylpropanoid biosynthesis pathway, including 8 phenylalanine ammonia-lyase (PAL) genes, 20 peroxidase (POD) genes, 2 polyphenol oxidase (PPO) genes associated with tyrosine metabolism, and 37 peroxisome-related genes. Real-time quantitative (qPCR) was employed to validate 15 browning-related genes, revealing that the expression levels of LcPOD21 and LcPOD6 were 12.5-fold and 25-fold higher in ‘30’ compared to ‘256’, while LcPAL5 and LcPAL4 were upregulated in ‘30’. Enzyme analysis showed that catalase (CAT) and phenylalanine ammonia-lyase (PAL) activities were higher in ‘30’ than in ‘256’. Conversely, superoxide dismutase (SOD) and polyphenol oxidase (PPO) activities were reduced in ‘30’, whereas CAT activity was upregulated. The concentrations of cinnamic acid, p-coumaric acid, trans-5-O-(4-coumaroyl)mangiferic acid, and caffealdehyde were lower in browning-resistant luffa ‘30’ than in browning-sensitive luffa ‘256’, suggesting that their levels influence browning in luffa. These findings elucidate the mechanisms underlying browning and inform strategies for the storage, processing, and genetic improvement of luffa. Full article

Background/Objectives: Cannabidiol (CBD) is a non-intoxicating cannabinoid touted for a variety of medical benefits, including alleviation of anxiety. While legalization of hemp-derived products in the United States (containing ≤0.3% delta-9-tetrahydrocannabinol [d9-THC] by weight) has led to a rapid increase in the commercialization of hemp-derived CBD products, most therapeutic claims have not been substantiated using clinical trials. This trial aimed to assess the impact of 6 weeks of treatment with a proprietary hemp-derived, full-spectrum, high-CBD sublingual solution similar to those available in the marketplace in patients with anxiety. Methods: An open-label pilot clinical trial (NCT04286594) was conducted in 12 patients with at least moderate levels of anxiety. Patients self-administered a hemp-derived, high-CBD sublingual solution twice daily during the 6-week trial (target daily dose: 30 mg/day CBD). Clinical change over time relative to baseline was assessed for anxiety, mood, sleep, and quality of life, as well as changes in cognitive performance on measures of executive function and memory. Safety and tolerability of the study product were also evaluated. Results: Patients reported significant reductions in anxiety symptoms over time. Concurrent improvements in mood, sleep, and relevant quality of life domains were also observed, along with stable or improved performance on all neurocognitive measures. Few side effects were reported, and no serious adverse events occurred. Conclusions: These pilot findings provide initial support for the efficacy and tolerability of the hemp-derived, high-CBD product in patients with moderate-to-severe levels of anxiety. Double-blind, placebo-controlled studies are indicated to obtain robust data regarding efficacy and tolerability of these types of products for anxiety. Full article

Background/Objectives: Dental practitioners widely use dental implants to treat traumatic cases. Titanium implants are currently the most popular choice among dental practitioners and surgeons. The discovery of newer polymeric materials is also influencing the interest of dental professionals in alternative options. A comparative study between existing titanium implants and newer polymeric materials can enhance professionals’ ability to select the most suitable implant for a patient’s treatment. This study aimed to investigate material property advantages of high-performance thermoplastic biopolymers such as PEEK and PEKK, as compared to the time-tested titanium implants, and to find the most suitable and economically fit implant material. Methods: Three distinct implant material properties were assigned—PEEK, PEKK, and commercially pure titanium (CP Ti-55)—to dental implants measuring 5.5 mm by 9 mm, along with two distinct titanium (TI6AL4V) abutments. Twelve three-dimensional (3D) models of bone blocks, representing the mandibular right molar area with Osseo-integrated implants were created. The implant, abutment, and screw were assumed to be linear; elastic, isotropic, and orthotropic properties were attributed to the cancellous and cortical bone. Twelve model sets underwent a three-dimensional finite element analysis to evaluate von Mises stress and total deformation under 250 N vertical and oblique (30 degree) loads on the top surface of each abutment. Results: The study revealed that the time-tested titanium implant outperforms PEEK and PEKK in terms of marginal bone preservation, while PEEK outperforms PEKK. Conclusions: This study will assist dental practitioners in selecting implants from a variety of available materials and will aid researchers in their future research. Full article

Urban greening initiatives, including the incorporation of street trees, have been widely recognized for a variety of environmental benefits. However, their economic impact on retail, in particular, the impact of street trees on the visibility of signs, has been underexplored. Street trees can obscure retail signs, potentially reducing customer engagement and discouraging retailers from paying higher rents for such locations. This paper investigates how the blocking of retail signage by street trees affects monthly rent in developed commercial districts in Seoul. It identifies, through Google Street View and state-of-the-art deep-learning-based semantic segmentation methods, environmental elements such as street trees, sidewalks, and buildings; quantifies their proportions; and analyzes their impact on rent using OLS regression, controlling for socio-economic variables. The results reveal that rents significantly diminish when street trees blocking views of retail signs increase. Our findings require more nuanced consideration by planners and policymakers in balancing both environmental and economic demands toward sustainable street design and planning. Full article

Huanglongbing (HLB), caused by Candidatus Liberibacter asiaticus (CLas), is the most devastating disease threatening global citrus production. Although no commercial citrus varieties exhibit complete HLB resistance, genotype-specific tolerance variations remain underexplored. This study conducted a comparative transcriptomic profiling of six commercially citrus cultivars in South China, four susceptible cultivars (C. reticulata cv. Tankan, Gongkan, Shatangju, and C. sinensis Osbeck cv. Newhall), and two tolerant cultivars (C. limon cv. Eureka; C. maxima cv Guanxi Yu) to dissect molecular mechanisms underlying HLB responses. Comparative transcriptomic analyses revealed extensive transcriptional reprogramming, with tolerant cultivars exhibiting fewer differentially expressed genes (DEGs) and targeted defense activation compared to susceptible genotypes. The key findings highlighted the genotype-specific regulation of starch metabolism, where β-amylase 3 (BAM3) was uniquely upregulated in tolerant varieties, potentially mitigating starch accumulation. Immune signaling diverged significantly: tolerant cultivars activated pattern-triggered immunity (PTI) via receptor-like kinases (FLS2) and suppressed ROS-associated RBOH genes, while susceptible genotypes showed the hyperactivation of ethylene signaling and oxidative stress pathways. Cell wall remodeling in susceptible cultivars involved upregulated xyloglucan endotransglucosylases (XTH), contrasting with pectin methylesterase induction in tolerant Eureka lemon for structural reinforcement. Phytohormonal dynamics revealed SA-mediated defense and NPR3/4 suppression in Eureka lemon, whereas susceptible cultivars prioritized ethylene/JA pathways. These findings delineate genotype-specific strategies in citrus–CLas interactions, identifying BAM3, FLS2, and cell wall modifiers as critical targets for breeding HLB-resistant cultivars through molecular-assisted selection. This study provides a foundational framework for understanding host–pathogen dynamics and advancing citrus immunity engineering. Full article

Petit Manseng is a variety of Vitis vinifera gaining popularity in Virginia, USA because it consistently produces high quality grapes under variable growing conditions. However, its high sugar and acid levels complicate dry wine production. The goal of this study was to characterize Petit Manseng ripening kinetics from veraison to harvest to identify optimal harvest timing for producing dry white wines, using Chardonnay as a comparator because of its popularity in Virginia, well-known ripening kinetics, and ability to produce high quality dry white wines. A total of 74 samples of Petit Manseng and Chardonnay grapes were collected from five commercial sites over 2 years and evaluated for berry weight, pH, titratable acidity (TA), malic acid, total soluble solids (TSS), glucose, and fructose, with ripening kinetics modeled using segmented regressions. Results indicated that harvest timing and grape variety were the primary factors influencing ripening kinetics. In contrast, growing location and vintage had limited impact. In Chardonnay grapes, TA declined from 21 to 7.1 g/L and TSS increased from 6.1 to 19.5 g/L. In Petit Manseng, TA declined from 25 to 10.8 g/L and TSS increased from 8.0 to 23.6 g/L. Acid depletion plateaued ~2 weeks after sugar accumulation plateaued in Petit Manseng grapes, though the plateaus were similar in Chardonnay grapes. Linear discriminant analysis (LDA) completely separated grapes based on pH or TA vs. sugars, but not malic acid vs. sugars, suggesting that tartaric acid is driving acidity differences between cultivars. These data indicate that regardless of when grapes are harvested, winemakers may need to employ targeted acid management strategies with Petit Manseng because of its ripening kinetics. Full article

Breeding programs often overlook the use of root traits. Therefore, we investigated the relevance of sorghum root traits in explaining its adaptation to combined drought and heat stress (CDHS). Six (i.e., three pre-release lines + three checks) sorghum genotypes were established at two low-altitude (i.e., <600 masl) locations with a long-term history of averagely very high temperatures in the beginning of the summer season, under two management (i.e., CDHS and well-watered (WW)) regimes. At each location, the genotypes were laid out in the field using a randomized complete block design (RCBD) replicated two times. Root trait data, namely root diameter (RD), number of roots (NR), number of root tips (NRT), total root length (TRL), root depth (RDP), root width (RW), width–depth ratio (WDR), root network area (RNA), root solidity (RS), lower root area (LRA), root perimeter (RP), root volume (RV), surface area (SA), root holes (RH) and root angle (RA) were gathered using the RhizoVision Explorer software during the pre- and post-flowering stage of growth. RSA traits differentially showed significant (p < 0.05) correlations with grain yield (GY) at pre- and post-flowering growth stages and under CDHS and WW conditions also revealing genotypic variation estimates exceeding 50% for all the traits. Regression models varied between pre-flowering (p = 0.013, R2 = 47.15%, R2 Predicted = 29.32%) and post-flowering (p = 0.000, R2 = 85.64%, R2 Predicted = 73.30%) growth stages, indicating post-flowering as the optimal stage to relate root traits to yield performance. RD contributed most to the regression model at post-flowering, explaining 51.79% of the 85.64% total variation. The Smith–Hazel index identified ICSV111IN and ASAREACA12-3-1 as superior pre-release lines, suitable for commercialization as new varieties. The study demonstrated that root traits (in particular, RD, RW, and RP) are linked to crop performance under CDHS conditions and should be incorporated in breeding programs. This approach may accelerate genetic gains not only in sorghum breeding programs, but for other crops, while offering a nature-based breeding strategy for stress adaptation in crops. Full article

Mango is highly sensitive to heat stress, which directly affects the yield and quality. The extreme heat waves of 2024, with temperatures reaching 41–47 °C over 25 days, caused significant impacts on sensitive cultivars. The impact of heat waves on ten commercial cultivars from subtropical regions viz.,‘Dashehari’, ‘Langra’, ‘Chausa’, ‘Bombay Green’, ‘Himsagar’, ‘Amrapali’, ‘Mallika’, ‘Sharda Bhog’, ‘Kesar’, and ‘Rataul’, and thirteen selected elite hybrids H-4208, H-3680, H-4505, H-3833, H-4504, H-1739, H-3623, H-1084, H-4264, HS-01, H-949, H-4065, and H-2805, is reported. The predominant effects that were observed include the following: burning symptoms or blackened tips, surrounded by a yellow halo, with premature ripening in affected parts and, in severe cases, tissue mummification. Among commercial cultivars, viz., ‘Amrapali’ (25%), ‘Mallika’ (30%), ‘Langra’ (30%), ‘Dashehari’ (50%), and ‘Himsagar’ and ‘Bombay Green’ had severe impacts, with ~80% of fruits being affected, followed by ‘Sharda Bhog’. In contrast, mid-maturing cultivars like ‘Kesar’, ‘Rataul’, and late-maturing elite hybrids, which were immature during the stress period, showed no symptoms, indicating they are tolerant. Biochemical analyses revealed significantly elevated total soluble solids (TSS > 25 °B) in affected areas of sensitive genotypes compared to non-affected tissues and tolerant genotypes. Aroma profiling indicated variations in compounds such as caryophyllene and humulene between affected and unaffected parts. The study envisages that the phenological maturity scales are indicators for the selection of climate-resilient mango varieties/hybrids and shows potential for future breeding programs. Full article

The agronomic effectiveness of biofertilizers is influenced by strain origin, genetic identity, crop genotype, soil type, and environmental conditions. For best results, both the plant and rhizobia strain must be adapted to the common harsh soil conditions in the tropics. While plant varieties have changed over the years, complementary research on new strains effectiveness under varying soil fertility conditions has lagged in southern Africa. Seven field experiments were established in the main soybean-producing areas of Zimbabwe in the north, central, and north–east regions to evaluate agronomic benefits of new rhizobia strains against the current exotic commercial strain (MAR1491). One site was irrigated (site 3), and the other six sites were rainfed (sites 1, 2, 4, 5, 6, and 7). While trends in inoculation response varied from site to site due to site conditions, inoculation with the strains NAZ15, NAZ25, and NAK128 consistently yielded high grain yields, which were similar to the current commercial strain MAR1491 and to application of mineral fertilizer (51.75 and 100 kg N ha−1). Grain yield levels were generally below 2 t ha−1 for sites 2, 3, and 5 and above 2 t ha−1 for sites 1, 4, and 6, while for the irrigated site 3, they ranged upwards of 3 t ha−1. When irrigated, all strains except NAK9 performed similarly in terms of grain yields and aboveground N uptake. Further testing on the inclusion of the indigenous strains NAZ15, NAZ25, and NAK128 in multi-strain commercial inoculant production targeting application in regions and soils where they excel beyond the current exotic strain MAR1491 is recommended. In this context, the present study aimed to explore the microbial diversity and functional characteristics of Tsounati variety olives from the Monemvasia region of Peloponnese, Greece, that were naturally fermented for three months. The bacterial and fungal microbiota of both olives and brines were fingerprinted throughout the fermentation through classical microbiological analysis combined with molecular techniques. Among the 148 isolated bacteria, 85 were lactic acid bacteria (LAB), and 63 belonged to the Enterobacteriaceae family, while the 178 fungal isolates comprised 136 yeasts and 42 non-yeast or yeast-like fungi. Metataxonomic analysis confirmed the dominance of the bacterial genera Lactiplantibacillus, Leuconostoc, along with the Enterobacteriaceae family, and it revealed the presence of Coleofasciculaceae cyanobacteria mostly in olives. The dominant fungal genera were yeasts, namely Saccharomyces, Nakazawaea, and Cyberlindnera. Using the Folin–Ciocalteu assay, the average total polyphenol content of Tsounati fermented olive samples was 761.80 ± 128.87 mg gallic acid equivalents kg−1 after 90 days of fermentation. The concentrations of the triterpenic, maslinic, and oleanolic acids, as determined by HPLC, remained stable throughout fermentation, with average values of 4764 and 1807 mg kg−1, respectively. Finally, sensory analysis revealed the rich aromatic character of Tsounati variety, highlighting its potential to be used for Greek-style table olive production. Full article

Opuntia ficus-indica L. (Mill) belongs to the Cactaceae family. The plant produces edible and juicy fruits called cactus pear, recognized for their pleasant flavor and functional properties. However, the fruits have a short shelf life, hard seeds, and the presence of glochidia in the pericarpel. Recently, by inducing parthenocarpy, seedless fruits of cactus pear have been obtained. They have attractive colors, soft and small seminal residues, with a similar flavor to their original seeded counterparts. Nevertheless, their postharvest physiological behavior has not yet been documented. The aim of this study was to compare the biochemical, anatomical, and physiological characteristics of pollinated fruits, CP30 red and CP40 yellow varieties, with their parthenocarpic counterparts (CP30-P and CP40-P), obtained by the application of growth regulators in preanthesis. Fruits of each type were harvested at horticultural maturity, and analyses were carried out on both pulp and pericarpel (peel), using a completely randomized design. Results showed that red fruits CP30 and CP30-P showed higher concentrations of betacyanins in pulp (13.4 and 18.4 mg 100 g−1 FW) and in pericarpel (25.9 and 24.1 mg 100 g−1 FW), respectively; flavonoid content was significantly higher in partenocarpic fruits compared with the pollinated ones. Parthenocarpy mainly affected the shelf life, in pollinated fruits, CP30 was 14 days but 32 days in CP30-P; for CP40, it was 16 days, and 30 days in CP40-P. Also, the partenocarpic fruits were smaller but with a thicker pericarpel, and lower stomatal frequency. Overall, parthenocarpic fruits represent a viable alternative for commercial production due to their extended shelf life, lower weight loss, and soft but edible pericarpel. Full article

Melanin is a complex natural pigment that imparts a variety of colors to the fruiting bodies of edible fungi, influencing both their nutritional quality and commercial value. Stropharia rugosoannulata is an emerging type of edible fungus that has been widely cultivated in recent years. It can be categorized into red and yellow varieties based on cap color, while its pigment characteristics remain unclear. In this study, the melanins from the two varieties were obtained using an alkaline extraction and acid precipitation method, followed by comprehensive characterization of their chemical properties and ultrastructural features. Both melanins displayed distinct absorption maxima at approximately 211 nm. The melanin extracted from the red variety consisted of 55.63% carbon (C), 7.40% hydrogen (H), 30.23% oxygen (O), 5.99% nitrogen (N), and 0.64% sulfur (S), whereas the yellow variety comprised 52.22% C, 6.74% H, 29.70% O, 5.91% N, and 0.99% S. Both types of melanin included eumelanin and phaeomelanin forms, with eumelanin being the predominant type. Variations in the quantities and relative proportions of eumelanin and phaeomelanin contributed to the observed color differences in the mushroom caps. Ultrastructural micrographs revealed the melanins were primarily localized in the cell wall, consistent with findings in other fungal species. These findings contribute valuable insights into fundamental knowledge and potential applications of mushroom pigments. Full article

Soybean meal (SBM) is extensively used as a predominant protein source in animal feed. However, raw soybean cannot be directly utilized in animal feed, due to the presence of the Kunitz trypsin inhibitor (KTi) and the Bowman–Birk protease inhibitor (BBi). These antinutritional factors inhibit the digestive enzymes in animals, trypsin and chymotrypsin, resulting in poor animal performance. To inactivate the activity of protease inhibitors, SBM is subjected to heat processing, a procedure that can negatively impact the soybean protein quality. Thus, it would be beneficial to develop soybean varieties with little or no trypsin inhibitors. In this study, we report on the creation of experimental soybean lines with significantly reduced levels of Bowman–Birk protease inhibitors. RNA interference (RNAi) technology was employed to generate several transgenic soybean lines. Some of these BBi knockdown soybean lines showed significantly lower amounts of both trypsin and chymotrypsin inhibitor activities. Western blot analysis revealed the complete absence of BBi in selected RNAi-derived lines. RNA sequencing (RNAseq) analysis demonstrated a drastic reduction in the seed-specific expression of BBi genes in the transgenic soybean lines during seed development. Confocal fluorescence immunolabeling studies showed that the accumulation of BBi was drastically diminished in BBi knockdown lines compared to wild-type soybeans. The absence of BBi in the transgenic soybean did not alter the overall protein, oil, and sulfur amino acid content of the seeds compared to wild-type soybeans. The seed protein from the BBi knockdown lines were more rapidly hydrolyzed by trypsin and chymotrypsin compared to the wild type, indicating that the absence of BBi enhances protein digestibility. Our study suggests that these BBi knockdown lines could be a valuable resource in order for plant breeders to incorporate this trait into commercial soybean cultivars, potentially enabling the use of raw soybeans in animal feed. Azospirillum is a well-studied genus of plant growth-promoting rhizobacteria (PGPR) and one of the most extensively researched diazotrophs. This genus can colonize rhizosphere soil and enhance plant growth and productivity by supplying essential nutrients to the host. Azospirillum–plant interactions involve multiple mechanisms, including nitrogen fixation, the production of phytohormones (auxins, cytokinins, indole acetic acid (IAA), and gibberellins), plant growth regulators, siderophore production, phosphate solubilization, and the synthesis of various bioactive molecules, such as flavonoids, hydrogen cyanide (HCN), and catalase. Thus, Azospirillum is involved in plant growth and development. The genus Azospirillum also enhances membrane activity by modifying the composition of membrane phospholipids and fatty acids, thereby ensuring membrane fluidity under water deficiency. It promotes the development of adventitious root systems, increases mineral and water uptake, mitigates environmental stressors (both biotic and abiotic), and exhibits antipathogenic activity. Biological nitrogen fixation (BNF) is the primary mechanism of Azospirillum, which is governed by structural nif genes present in all diazotrophic species. Globally, Azospirillum spp. are widely used as inoculants for commercial crop production. It is considered a non-pathogenic bacterium that can be utilized as a biofertilizer for a variety of crops, particularly cereals and grasses such as rice and wheat, which are economically significant for agriculture. Furthermore, Azospirillum spp. influence gene expression pathways in plants, enhancing their resistance to biotic and abiotic stressors. Advances in genomics and transcriptomics have provided new insights into plant-microbe interactions. This review explored the molecular mechanisms underlying the role of Azospirillum spp. in plant growth. Additionally, BNF phytohormone synthesis, root architecture modification for nutrient uptake and stress tolerance, and immobilization for enhanced crop production are also important. A deeper understanding of the molecular basis of Azospirillum in biofertilizer and biostimulant development, as well as genetically engineered and immobilized strains for improved phosphate solubilization and nitrogen fixation, will contribute to sustainable agricultural practices and help to meet global food security demands.
