Mercury (Hg) poses a significant threat to human health and ecosystems, garnering increased attention in environmental studies. This paper evaluates the dynamics of Hg accumulation in various common tree leaves, specifically white poplar, linden, and cherry plum, throughout their growing season. The findings offer valuable insights into air quality and the ability of urban vegetation to mitigate mercury pollution in urban areas. A case study was conducted in Turda, a town in northwestern Romania, where a former chlor-alkali plant operated throughout the last century. Although the plant ceased its electrolysis activities over 25 years ago, the surrounding soil remains contaminated with mercury (Hg) due to the significant amounts released during its operation. The results indicated that the Hg concentration varied between 2.4 and 7.3 mg kg−1 dry weight (dw), exceeding the intervention threshold for soil of 2.0 mg kg−1. Additionally, the Hg content in the leaf samples consistently increased over time, influenced by leaf age and tree species. The Hg content increased in the following order: cherry plum < white poplar < linden. On average, white poplar leaves accumulated 72 ng Hg g−1 dw, linden leaves 128 ng Hg g−1 dw, and cherry plum leaves 47 ng Hg g−1 dw during the six-month monitored period from April to September. The results obtained can be used to evaluate the potential of different tree species for mitigating atmospheric Hg contamination and to elaborate on the suitable management of fallen leaves in the autumn. This paper assesses the complexity and diversity of vegetation in grassy strips with spontaneous plants between tree rows in three fruit orchards (plum, cherry, apple) in Dolj County, Romania, using structural and biodiversity indices. It addresses the lack of data on spontaneous vegetation in Romanian orchards, supporting improved plantation management and native biodiversity conservation. The study found that grassy strips supported high wild herbaceous diversity and a complex, heterogeneous ecological structure, with the apple orchard showing the highest biodiversity. Species diversity, evaluated through species richness, evenness, and diversity indices (Shannon, Simpson, Menhinick, Gleason, etc.), showed species richness ranging from 30 species in the cherry orchard to 40 in the apple orchard. Several species, including Capsella bursa-pastoris, Geranium pusillum, Poa pratensis, Veronica hederifolia, Lolium perenne, and Convolvulus arvensis, were present in 100% of samples, making them constant species from a phytosociological perspective. Their presence indicates relatively stable plant communities in each orchard. From a phytocoenological view, an ecological plant community is defined not only by species composition but also by constancy and co-occurrence in sampling units. Dominance remained low in all orchards, indicating no single plant dominated, while evenness showed a uniform distribution of species. Full article

Biodegradable composite materials enhanced with food waste for tribological applications are proposed in this article. Polymer materials used as matrices included polypropylene and polylactic acid, which, according to the manufacturers’ claims, were made entirely or partially from biodegradable raw materials. Additionally, the matrices were enhanced with three types of waste materials: powders derived from cherry and plum stones, and pomace extracted from flax seeds. The composites differed in the percentage content of filler (15 or 25 wt.%) and particle size (d < 400 µm or d > 400 µm). Thirty-minute block-on-ring friction tests were performed to determine frictional behaviour (when pairing with steel), and the wear mechanisms were analysed using optical microscopy and scanning electron microscopy, supplemented with Raman spectroscopy. A notable effect of cherry and plum stone fillers was observed as a reduction in motion resistance, as measured by the friction coefficient. This reduction was evident across all material configurations in polypropylene-based composites and was significant at the lowest concentrations and granulation in polylactic acid composites. The effect of flaxseed pomace filler was ambiguous for both composite bases. Full article

Viral infections in stone fruit crops cause substantial economic losses across all sectors of production. Despite their significance, viruses affecting stone fruits remain under-investigated in Kazakhstan. Among these, plum pox virus (PPV, genus Potyvirus, family Potyviridae), commonly known as Sharka, is the most critical viral pathogen worldwide, severely threatening the sustainable cultivation of stone fruits and posing risks to food security. This study aimed to evaluate virus management strategies in stone fruit crops to facilitate the production of healthy planting material from valuable genotypes. Field surveys were conducted in plum and apricot orchards located in the Almaty region (Southeast Kazakhstan) and the Saryagash region (Southern Kazakhstan). Plant samples were tested for the presence of the following viruses: apple chlorotic leaf spot virus (ACLSV), apple mosaic virus (ApMV), PPV, prune dwarf virus (PDV), prunus necrotic ringspot virus (PNRSV), cherry green ring mottle virus (CGRMV), and myrobalan latent ringspot virus (MLRSV). Real-time RT-PCR diagnostics confirmed the presence of PPV in the ‘Stanley’ and ‘Ansar’ cultivars and Prunus armeniaca genotypes, while both PPV and ACLSV were detected in the ‘Ayana’ variety. Chemotherapy (Ribavirin), thermotherapy, cryotherapy, and shoot apical meristem (SAM) culture, both individually and in combination, were used to eliminate viruses and regenerate virus-free plants. Successful virus eradication was achieved for PPV and ACLSV. However, the ‘Stanley’ and ‘Ansar’ cultivars did not survive the treatment process, likely due to high thermo- or cryo-sensitivity. As a result of this research, an in vitro collection of virus-free plants was established, comprising eight rootstocks, six plum cultivars, and three apricot genotypes. Full article

The different impact and interaction of pulsed electric field (PEF) treatment and sulfite addition on the color, phenolic compounds, volatile profile, and sensory properties of a Primitivo wine were studied at bottling and after six months of storage. The results show that PEF treatment, promoting the electroporation of grape skin cells and the permeability of cell membranes, accelerated the extraction of anthocyanins and polyphenols only in the initial phase of maceration. After six months in bottles, wines treated only with PEF show lower levels of hydroxycinnamic acids and flavonols, but a significant increase in procyanidins B2, which are important for structure and color stability and a richer flavor profile, with higher concentrations of fruity esters and higher alcohols. The use of SO2 improves anthocyanin stabilization and facilitates the extraction of polyphenols. The wines from the PEF + SO2 combination maintain greater aromatic freshness, limiting the formation of oxidative compounds. Wines made from SO2 have a more balanced profile, with cherry, plum, and licorice aromas, although the combined PEF + SO2 treatment better preserves fresh fruit aromas, reducing the perception of dried fruits and herbaceous notes. Peaches (Prunus persica L. Batsch) and nectarines (Prunus persica L. Batsch var. nectarina [Ait.] Maxim) are economically important stone fruits consumed worldwide, both fresh and processed. Viruses and viroids significantly constrain the cultivation and productivity of peach orchards. Climate change may alter vector populations and lead to shifts in agricultural practices, influencing the spread of these viruses and viroids. Additionally, market globalization further intensifies the pressure on peach crops by facilitating the movement of pathogens, increasing the incidence of virus-induced diseases. In this study, we identified the viral and viroid communities in five peach cultivars from Bulgaria and assessed their impact on symptom development. RNA sequencing of symptomatic leaf samples revealed the presence of common peach viruses, such as plum pox virus and prunus necrotic ringspot virus. Notably, we identified peach latent mosaic viroid and cherry green ring mottle virus in Bulgarian peach orchards for the first time. Furthermore, bioassays of indicator plants, ELISA, and Sanger sequencing were performed for each peach tree to complement the RNA sequencing data. These findings provide valuable insights into the composition of viral and viroid pathogens affecting peaches in Bulgaria and will support the development of targeted strategies for monitoring and managing these pathogens, contributing to the sustainable production of peaches in the region. To learn about the yeast biodiversity of Hungarian honeys and to isolate osmotolerant yeasts, fifteen different honey varieties, beeswax, and bee bread were purchased, and samples of another, but highly osmotic material, tree sap (cherry, sour cherry, and plum), were collected from the northeastern region of the country. In total, 60 yeast strains were isolated and their taxonomic positions were determined by barcode sequences using ITS1-NL4 primers. The honey products contained mostly Zygosaccharomyces and Starmerella species. In addition, Hanseniaspora uvarum, Rhodotorula mucilaginosa and diobovata, Sporobolomyces roseus, Filobasidium magnum, Naganishia sp., and Aureobasidium pullulans were also present in smaller numbers. In contrast, tree saps contained Metschnikowia and Pichia fermentas cells. Further results suggest that some of the yeasts in honey can only “survive”, while others can propagate at high sugar levels, generally between 600 and 700 mg/g, with a predominance of fructose. Properties important for pathogenicity, such as invasive hyphae production, gelatin melting ability, and growth at 37 °C, were also examined. Hanseniaspora uvarum and Pichia fermentans representatives seemed to be negative for gelatin hydrolysis, while the other strains were able to melt gelatin. Although some of the strains could produce hyphae-like structures at 25 °C, none of them could grow at 37 °C. Full article

A novel primer (FaSt-R) targeting the Prunus-specific Falling Stone (FaSt) non-autonomous transposon was combined with start-codon-targeted (SCoT) primers to assess genetic diversity in 12 cultivars from six Prunus species and 28 cultivars of European plum. Compared to SCoT-only analyses, the SCoT–FaSt combination produced fewer total bands but a higher percentage of polymorphic bands, while maintaining comparable values for polymorphism information content, resolving power, gene diversity, and Shannon’s index. SCoT–FaSt markers generated bands across a broader size range, which made gel patterns less dense, enabling the more accurate detection of differentially amplified fragments. Neighbor-joining and principal component analyses confirmed that SCoT–FaSt markers provided sufficient phylogenetic resolution at both interspecific and intraspecific levels. The sequencing of 32 SCoT–FaSt amplicons revealed FaSt elements in 26 fragments, with SCoT primers preferentially annealing to GC-rich exonic and intergenic regions. Seventeen protein-coding and one RNA-coding gene were partially identified, with FaSt elements localized in UTRs and introns of genes with key physiological functions. Comparative analysis indicated a biased distribution of FaSt elements between the Cerasus and Prunus subgenera. In silico findings suggest that FaSt elements are more fragmented in cherry species, potentially contributing to subgeneric divergence. Overall, the SCoT–FaSt marker system is effective for evaluating Prunus genetic diversity, reconstructing phylogenetic relationships, and elucidating the genomic impact of an active Mutator-like transposon. This study is the first investigation of tree–insect–bacteria interactions in southern Romania, documenting the distribution of 19 insect species across various fruit trees and their insect-associated bacterial diversity. Insect species were identified through DNA barcoding, while bacterial communities in Anthomyia, Botanophila, Drosophila, and Scaptomyza insects were analyzed via 16S rRNA gene sequencing. Insect diversity varied across apple, cherry, plum, peach, and quince trees, with most species showing tree-specific distribution, except for Drosophila melanogaster, which was found on all tree species. Its presence was primarily influenced by fruit development stages rather than temperature changes. Insect bacterial communities comprised 51 genera across four phyla, predominantly Pseudomonadota and Bacillota, that varied by tree species rather than insect species, suggesting the potential role of these flies as bacterial vectors. Several potential pathogenic bacterial genera were identified as biomarkers within insect microbiomes, suggesting their involvement in disease transmission, particularly affecting apple and cherry trees. This study also provides the first report of seven insect species in Romania, being the first microbiome characterization of four dipteran species associated with regional fruit trees. Full article

Fish smoking is one of the oldest preservation methods and has traditional forms in most parts of the world, each with distinct variables. The presence of PAHs in smoked fish is of great concern for producers and consumers alike, as PAHs have great negative effects on human health. This study aimed to determine the physicochemical characteristics and polycyclic aromatic hydrocarbons (PAHs) content level in smoked rainbow trout Oncorhynchus mykiss (Walbaum, 1792) processed using a commercial portable grilling and smoking oven. Sawdust of hardwood (beech, cherry, sour cherry, walnut, and plum) and softwood (fir, willow) were used to produce aromatic essences. The principal component analysis (PCA) revealed that different tree species tend to accumulate different PAHs in different concentrations. In the case of the generalized additive model (GAM) analyzing fish meat, fir tree reduced the concentration of PAHs in fish meat. When GAM analyzed fish skin, cherry, sour cherry, and willow trees significantly reduced the concentrations of PAHs in fish skin compared to beech trees. Furthermore, the results regarding the skin of the fish suggest that it acts as a protective barrier, trapping smoke particulates and reducing the penetration of PAHs into the meat. The present method clearly shows that, at least in the case of Benzo[a]pyrene, it is safer than traditional methods of smoking. This highlights the need for further research into the physicochemical properties of fish tissue and their impact on PAH accumulation. Full article

The aim of this study was to investigate the antioxidant capacity of fruit wines and their protective effects against hydrogen peroxide-induced oxidative stress in rat synaptosomes in vitro. The wines were produced from strawberries and drupe fruits (i.e., plum, sweet cherry, peach, and apricot) through microvinification with a pure S. cerevisiae yeast culture. Fruit wines were produced with and without added sugar before the start of fermentation, whereas subvariants with and without pits were only applied to drupe fruit wines. First, synaptosomes were treated with the wines, while oxidative stress was induced with H2O2. Subsequently, the activities of antioxidant enzymes (superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GPx)) and the content of malondialdehyde (MDA), an indicator of membrane injury, were determined. In addition, the Briggs–Rauscher reaction (BR) was used to evaluate the inhibition capacity against free radicals. All investigated fruit wines increased the activity of the studied antioxidant enzymes and decreased MDA content compared to the corresponding controls (synaptosomes treated with H2O2). After synaptosomal treatment with plum wine, the highest activities were observed for SOD (5.57 U/mg protein) and GPx (0.015 U/mg protein). Strawberry wine induced the highest CAT activity (0.047 U/mg protein) and showed the best ability to reduce lipid peroxidation, yielding the lowest MDA level (2.68 nmol/mg). Strawberry, plum, and sweet cherry wines were identified as samples with higher antioxidant activity in both principal component analysis (PCA) and hierarchical cluster analysis (HCA). Finally, plum wine exhibited the highest inhibitory activity in the BR reaction (397 s). The results suggest that fruit wines could be considered potential functional food due to their protective effects against oxidative stress. Full article

In the past, the Silk Road was a vital trade route that spanned Eurasia, connecting East Asia to the Mediterranean Sea. The genus Prunus, belonging to the Rosaceae family and encompassing plums, peaches, apricots, cherries, and almonds, thrived as human travel along the Silk Road increased. The majority of fruits within this genus, whether wild or cultivated, are naturally sweet and easily preserved by drying for storage and transport. The interaction along the Silk Road between wild populations and diverse varieties of Prunus fruits led to the development of various hybrids. This article provides a summary of archaeological findings related to prominent Prunus fruits such as peaches, apricots, plums, cherries, and almonds, shedding light on their evolutionary history, genetic diversity, population structure, and historical dynamics crucial for species conservation. The origins of biodiversity may involve factors like migration of pre-adapted lineages, in situ variation, or the persistence of ancestral lineages. Furthermore, climate change is affecting spatial genetic patterns and potentially further threatening rare Prunus species. Evaluating the scope and composition of genetic diversity within germplasm collections is essential for enhancing plant breeding initiatives and preserving genetic resources in this changing context. From a molecular point of view, techniques such as genome-wide association studies (GWASs) and the identification of quantitative trait loci (QTLs) and genes responsible for phenotypic changes in cultivars and germplasm collections should be of great interest in these breeding programs, while genomic estimated breeding values (GEBVs) derived from genome-wide DNA polymorphism information can facilitate the selection of superior genotypes. Postharvest diseases cause considerable losses of fruits and vegetables during transportation and storage, and synthetic fungicides are the first option for their prevention. However, promising alternatives to chemical fungicides are currently available, and several post-harvest diseases can be controlled using microbial antagonists. This study utilised a comprehensive methodology to assess the antagonistic and synergistic interactions between four yeasts and two bacteria in conjunction with sodium bicarbonate (SB) during the treatment of sweet cherries and plums. The aim of this study was to evaluate the effects of microbial antagonists and sodium bicarbonate on fruits. The in situ treatments showed a protective effect exerted by the antagonists P. guillermondii and H. uvarum, and their combination with SB. However, in vivo studies did not indicate enhanced efficacy when combined with this compound. In conclusion, the use of microbial antagonists in conjunction with SB has been successful in preventing post-harvest rot of cherries and plums. Tests conducted on live organisms have shown that microbial antagonists are viable alternatives to synthetic fungicides for the control of stone fruit rot. Full article

Numerous health benefits have been attributed in the last decades to the regular consumption of fruits, vegetables, herbs and spices, along with honey, in a balanced diet. In this context, the aim of the present review was to provide the literature with the most relevant studies focusing on the determination protocols of these polyphenols and other reducing agents in selected fruits (orange, lemon, grapefruit, prunus, apricot, peach, plum, sweet cherry), spices (oregano, cinnamon, clove, saffron, turmeric) and honey of different botanical origin (nectar or honeydew). In addition, the content and the extraction methods of these compounds, along with their metabolic pathway, have been critically evaluated and discussed. Results showed that all fruits, spices and honey exhibit a considerably high antioxidant activity, which is mainly owed to their phytochemical content. Therefore, a balanced diet consisting of the combination of the foods studied herein may comprise a shield against chronic and other pathophysiological disorders and may be achieved through consecutive educational programs for consumers at an international level. Development of Loop-Mediated Isothermal Amplification (LAMP) Assay for In-Field Detection of American Plum Line Pattern Virus

American plum line pattern virus (APLPV) is the most infrequently reported Ilarvirus infecting stone fruit trees and is of sufficient severity to be classified as an EPPO quarantine A1 pathogen. In late spring, yellow line pattern symptoms were observed on leaves in a few flowering cherries (Prunus serrulata Lindl.) grown in a public garden in Northwest Italy. RNA extracts from twenty flowering cherries were submitted to Ilarvirus multiplex and APLPV-specific RT-PCR assays already reported or developed in this study. One flowering cherry (T22) with mixed prunus necrotic ringspot virus (PNRSV) and prune dwarf virus (PDV) infection also showed infection with APLPV. Blastn analysis of PCR products of the full coat protein (CP) and movement protein (MP) genes obtained from flowering cherry T22 showed 98.23% and 98.34% nucleotide identity with reference APLPV isolate NC_003453.1 from the USA. Then, a LAMP-specific assay was designed to facilitate the fast and low-cost identification of this virus either in the laboratory or directly in the field. The developed assay allowed not only the confirmation of APLPV (PSer22IT isolate) infection in the T22 flowering cherry but also the identification of APLPV in an asymptomatic flowering cherry tree (TL1). The LAMP assay successfully worked with crude flowering cherry extracts, obtained after manually shaking a single plant extract in the ELISA extraction buffer for 3–5 min. The developed rapid, specific and economic LAMP assay was able to detect APLPV using crude plant extracts rather that RNA preparation in less than 20 min, making it suitable for in-field detection. Moreover, the LAMP assay proved to be more sensitive in APLPV detection in flowering cherry compared to the specific one-step RT-PCR assay. The new LAMP assay will permit the estimation of APLPV geographic spread in the territory, paying particular attention to surrounding gardens and propagated flowering cherries in ornamental nurseries.
