Herbal medicines have significant industrial value in East Asia. Zizyphus jujuba Mill. var. spinosa, used in Korea for treating insomnia, is often confused with Zizyphus mauritiana Lam., which has unverified medicinal properties yet is sold at premium prices. This misclassification undermines consumer trust and poses health risks. This study proposes a deep learning-based classification system trained on RGB-GE data, combining grayscale and edge-detected images with RGB inputs to enhance feature extraction while reducing color-dependency. Our method achieves superior generalization while maintaining cost-effectiveness. The system incorporates Grad-CAM for model interpretation and reliability. By comparing accuracy and speed across basicCNN, DenseNet, and InceptionV3 models, we identified an optimal solution for on-site herbal medicine classification, achieving 98.36% accuracy with basicCNN, ensuring reliable quality control. Zizyphus jujuba Mill. cv. Goutou is an edible and medicinal fruit whose development significantly impacts the metabolism and accumulation of nutrients and is regulated by plant hormones. In this study, the metal element and triterpene acid content were investigated and transcriptomic analyses were conducted to evaluate changes in seven ripening stages (Stages I–VII) of Z. jujuba Mill. cv. Goutou. We first analyzed seven metal elements and found that the concentrations of Magnesium (Mg), Aluminum (Al), Calcium (Ca), Manganese (Mn), and Cuprum (Cu) were highest at Stage I; in comparison, the concentrations of Zinc (Zn) and Ferrum (Fe) were highest at Stage IV. Additionally, three triterpene acids were detected in the fruits, with the contents of betulinic acid and oleanolic acid being highest at Stage VII and that of ursolic acid being highest at Stage I. Subsequently, through Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) annotations, we identified 40, 18, 9, 6, and 11 differentially expressed genes involved in the auxin, abscisic acid (ABA), ethylene, gibberellic acid (GA), and jasmonic acid (JA) signaling pathways, respectively. Notably, genes associated with ABA, including ZjABA3, ZjABA4, ZjABA6, ZjABA7, ZjABA10, ZjABA11, ZjABA15-ZjABA19, ZjABA22-ZjABA25, and ZjABA27-ZjABA33, were downregulated from Stage I to Stage VII. Conversely, the expression of ZjACO in the ethylene signaling pathway was the highest at Stage VII. ZjMYC2-1, a JA signaling pathway gene, was significantly induced at Stage I compared to in the other stages. The genes ZjGID-1 and ZjTF-1, related to GA, exhibited the highest expression levels at Stage VI. Full article

Low-temperature storage is an effective method to extend the shelf life of harvested winter jujube fruit; however, the quality of winter jujube fruit decreases after refrigeration. To enhance the quality of winter jujube with minimum qualitative deterioration during cold storage, this study investigated the effects of different melatonin concentration (100, 150, and 200 µmol/L) treatments on the storage quality of winter jujube (during the white ripening stage) at 0 ± 1 °C (RH, 90 ± 5%) for 90 days. The relative indexes of the fruit quality and the antioxidant system were measured every 15 days. The results showed that different concentrations of melatonin treatments could maintain the fruit’s firmness, retard the fruit’s redness, and inhibit the decrease in weight, total soluble solid (TSS), titratable acidity (TA), and the contents of total phenols, flavonoids, glutathione, and ascorbic acid; they also inhibited the increase in relative conductivity, malondialdehyde (MDA), and hydrogen peroxide (H2O2) content of jujube fruits and enhanced antioxidant enzyme activities (superoxide dismutase (SOD), catalase (CAT), ascorbate peroxidase (APX), glutathione reductase (GR), and peroxidase (POD)). As a whole, the 200 µmol/L melatonin treatment had the best effect. Therefore, melatonin treatment can maintain the cold-stored quality of postharvest winter jujube by improving its antioxidant capacity. Full article

Grey jujube (Zizyphus jujuba Mill. cv. Huizao), a prominent cultivar from Xinjiang, China, is well known for its high nutritional value and medicinal benefits. This study investigates the effects of long-term storage on the quality attributes of grey jujube, focusing on color, texture, physicochemical properties, bioactive ingredients, amino acid profiles, sensory characteristics, and volatile compounds. Over a three-year storage period, significant changes were observed, including a decline in lightness and redness of the peel, accompanied by textural modifications such as increased hardness and chewiness, primarily attributed to moisture loss. Physicochemical analyses revealed significant reductions in moisture content, sugars (particularly reducing sugars), and bioactive compounds such as vitamin C, total flavonoids, and cyclic adenosine monophosphate (cAMP). In contrast, total acidity showed a significant increase over time. Sensory evaluation indicated a substantial loss of fresh aroma and flavor, with the development of off-flavors over time. Additionally, a comprehensive analysis of volatile compounds highlighted significant transformations in aroma profiles, with notable reductions in desirable esters and increases in acetic acid concentrations. This study investigates the quality changes of grey jujubes during storage from sensory and physicochemical perspectives, aiming to provide a novel approach for differentiating between newly harvested and aged grey jujubes. Furthermore, these findings provide theoretical support for improving and optimizing storage conditions. Future research should focus on elucidating the underlying mechanisms of these changes, identifying key markers for quality control during grey jujube storage, and providing a scientific basis for distinguishing between newly harvested and aged grey jujubes, while improving storage quality. Background: Sleep, a process physiologically vital for mental health, faces disruptions in various sleep disorders linked to metabolic and neurodegenerative risks. Zizyphus seed (Zizy) has long been recognized for its diverse pharmacological attributes, including analgesic, sedative, insomnia, and anxiety alleviation. Objectives: In this study, the sleep-prolonging effects of Zizy extract (100, 200 mg/kg), along with their characterizing compounds jujuboside A (JuA) (5, 10 mg/kg), were evaluated in a mouse model under a pentobarbital-induced sleep. Additionally, the efficacy of Zizy extract was examined on caffeine-induced insomnia in mice. Methods: To confirm the efficacy of Zizy extract on the structure and quality of sleep, an electroencephalogram (EEG) analysis of rats was performed using the MATLAB algorithm. Additionally, Western blot analysis and measurement of intracellular chloride influx were performed to confirm whether these effects acted through the gamma-aminobutyric acid (GABA)ergic system. Administration of Zizy extract showed no effect on the locomotor performance of mice, but the extract and their characteristic compounds significantly prolonged sleep duration in comparison to the pentobarbital alone group in the pentobarbital-induced sleep mouse model. Furthermore, this extract alleviated caffeine-induced insomnia in mice. Results: The administration of Zizy extract extended non-rapid eye movement sleep (NREMS) duration without inducing significant changes in the brain wave frequency. Zizy extract regulated the expression of GABAA receptor subunits and GAD65/67 in specific brain regions (frontal cortex, hippocampus, and hypothalamus). JuA increased intracellular chloride influx in human SH-SY5Y cells, and it was reduced by GABAA receptor antagonists. These results suggest that the sleep-maintaining effects of Zizy extract may entail GABAergic regulation. In summary, Zizy extract demonstrated sleep-prolonging properties, improved insomnia, and regulated sleep architecture through GABAergic system modulation. Conclusions: These findings suggest that Zizy extract has potential as a therapeutic agent for stress-related neuropsychiatric conditions such as insomnia. Full article

Alcohol-associated liver disease (ALD) is a major cause of chronic liver disease, with few effective treatments besides alcohol abstinence. Angelicae Gigantis Radix (AG), Glycyrrhizae Radix et Rhizoma (GR), Paeoniae Radix (PR), and Zizyphi Fructus (ZF) are traditional herbs used to treat various ailments, including liver diseases. While several studies have reported the beneficial effects of GR on ALD, the effects of AG, PR, and ZF remain underexplored. Therefore, their efficacy and mechanisms against ALD were investigated using an alcohol-related liver injury model. The model was induced by ethanol gavage in C57BL/6J mice for 14 days, followed by oral administration of AG, GR, PR, and ZF one hour post-induction. The administration of these herbs reduced liver weight, and improved serum biomarkers of liver injury (ALT, AST, albumin). The herbs enhanced hepatic antioxidant capacity (GSH, SOD, catalase) and suppressed the production of proinflammatory cytokines (TNF-α, IL-1β) and apoptotic changes (caspase-3). The mechanisms of action involved lipid-lowering gene modulation through regulation of the cytochrome P450 2E1/Sirtuin 1/Nrf2 pathways. Histopathological and immunohistochemical analyses revealed that these herbs attenuated hepatocyte damage and steatosis via antioxidant, anti-inflammatory, and antiapoptotic effects. These findings suggest that traditional herbs, particularly AG, could be promising alternative therapies for treating ALD. Full article

Zizyphus spinosus Hu (ZS), as a “medicinal and food-homologous” plant, has been used for a long history. The study was to assess the sedative and hypnotic effects among various parts of ZS. The model, diazepam (DZP), ZS kernel (ZSS), ZS flesh (ZSF), and ZS husk (ZSKS) group occurred subsequent to the successful establishment of the para-chlorophenylalanine induced insomnia model via intraperitoneal injection. The latency and duration of sleep in mice in each group were recorded. The substance basis of various parts of ZS was analyzed by the UPLC-QTOF-MS technique. The results showed that relative to the model group, DZP, ZSS, ZSF, and ZSKS groups demonstrated shortened sleep latency (p < 0.05) and extended sleep duration (p < 0.01). The GABA, 5-HT, and BDNF levels were significantly upregulated in the brain tissues of the mice in the DZP, ZSF, and ZSS groups (p < 0.01). However, the improvement in ZSKS was non-significant. Additionally, the mRNA and protein expression levels of 5-HT1AR, GABAARα1, and BDNF in mice in the DZP, ZSS, and ZSF groups were significantly enhanced (p < 0.01). However, the improvement in the ZSKS group was insignificant (p < 0.05). The examination of the substance composition across different parts revealed that the shared chemical basis contributing to the sedative and hypnotic potency of different parts of ZS may involve the presence of compounds such as (1) magnoflorine, (8) betulinic acid, (9) ceanothic acid, and (10) alphitolic acid. It provides a basis for further elucidation of the substance basis responsible for the functional and medicinal effects of ZS. Full article

The objective of this research was to investigate the effect of infrared-combined hot air intermittent drying (IIRHAD) on energy consumption, drying characteristics, and the quality of jujube slices. The water content of jujube slices decreased from 0.267 g/g to 0.05 g/g during the experiment, and the infrared heating plate’s temperature was fixed at 70 °C while the hot air temperature was fixed at 50 °C. Nine different intermittent ratios were used to dry jujube slices, and the results showed that intermittent treatment had varying effects on drying characteristics, energy consumption, and quality. In comparison to infrared-combined hot air drying (IRHAD), the effective drying time of red jujube slices was reduced by 40 to 100 min, the energy consumption decreased by 11.91% to 34.34%, and there were also varying degrees of improvement in the quality indicators. It was discovered that excessively long or short active drying and tempering periods had a negative impact on the drying process. Therefore, these factors should be further broken down and improved in the future. This research holds great importance for the future advancement and widespread use of IIRHAD in fruit and vegetable materials. Full article

Plant proteins are a good source of active peptides, which can exert physiological effects on the body. Predicting the possible activity of plant proteins and obtaining active peptides with oral potential are challenging. In this study, the potential activity of peptides from Zizyphus jujuba proteins after in silico simulated gastrointestinal digestion was predicted using the BIOPEP-UWM™ database. The ACE-inhibitory activity needs to be further investigated. The actual peptides in mouse intestines after the oral administration of Zizyphus jujuba protein were collected and analyzed, 113 Zizyphus jujuba peptides were identified, and 3D-QSAR models of the ACE-inhibitory activity were created and validated using a training set (34 peptides) and a test set (12 peptides). Three peptides, RLPHV, TVKPGL and KALVAP, were screened using the 3D-QSAR model and were found to bind to the active sites of the ACE enzyme, and their IC50 values were determined. Their values were 6.01, 3.81, and 17.06 μM, respectively. The in vitro digestion stabilities of the RLPHV, TVKPGL, and KALVAP peptides were 82%, 90%, and 78%. This article provides an integrated method for studying bioactive peptides derived from plant proteins. Full article

Although Ziziphus jujuba Mill (jujube) is used in folk medicine for hypnotic sedative, anxiolytic, and many other purposes, to date, only a few studies have revealed its sleep-promoting effects and related mechanisms. Currently, drugs used for the treatment of sleep disorders have various side effects, so it is essential to develop safe natural materials. Therefore, we evaluated the sleep-enhancing activity and mechanism of action of an aqueous extract of jujube seeds (ZW) fermented with Lactobacillus brevis L-32 in rodent models. The starch contained in ZW was removed by enzymatic degradation and fermented with L. brevis to obtain a fermented product (ZW-FM) with a high γ-aminobutyric acid (GABA) content. To evaluate the sleep-promoting effect of ZW-FM, pentobarbital-induced sleep tests were performed on ICR mice, and electroencephalography analysis was undertaken in Sprague Dawley rats. Additionally, the awakening relief effects of ZW-FM were confirmed in a caffeine-induced insomnia model. Finally, the mechanism of sleep enhancement by ZW-FM was analyzed using GABA receptor type A (GABAA) antagonists. The ZW-FM-treated groups (100 and 150 mg/kg) showed increased sleep time, especially the δ-wave time during non-rapid eye movement (NREM) sleep. In addition, the 150 mg/kg ZW-FM treatment group showed decreased sleep latency and increased sleep time in the insomnia model. In particular, NREM sleep time was increased and REM sleep time, which was increased by caffeine treatment, was decreased by ZW-FM treatment. ZW-FM-induced sleep increase was inhibited by the GABAA receptor antagonists picrotoxin, bicuculline, and flumazenil, confirming that the increase was the result of a GABAergic mechanism. These results strongly suggest that the increased GABA in water extract from jujube seeds fermented by L. brevis acts as a sleep-promoting compound and that the sleep-promoting activity is related to GABAA receptor binding. Accurate prediction of orchard evapotranspiration (ET) can optimize orchard water management. Based on the jujube (Zizyphus jujuba), ET was continuously measured from 2016 to 2019 using a large weighing lysimeter; the actual jujube ET was compared with the ET simulated with the Priestley–Taylor (P–T), Dual Crop Coefficient (Dual Kc), and Shuttleworth–Wallace (S–W) models, to verify the accuracy of the three models. The results showed that, from 2016 to 2019, the whole growth period of jujube ET was 532–592 mm and the crop coefficient was 0.85–0.93. The basal crop coefficients of the calibrated Dual Kc model were 0.4, 1.0, and 0.5 at the initial, middle, and ending growth stages, respectively. The overall simulation error of the Dual Kc model was low, and simulations were stable during the four years of the study. However, because of rough estimation the water stress coefficient (Ks) simulation accuracy will be reduced in the case of serious water shortage. The simulation error of the S–W model was greater than the simulation error of the Dual Kc model, and the simulations were unstable and vulnerable to interannual changes. The simulation error of the traditional P–T model was large. When the parameter “α” solution method was improved, the simulation accuracy was significantly improved, and the P–T model’s simulation accuracy was only slightly lower than that of the Dual Kc model. However, the model was easily affected by changes in net radiation and air temperature. Therefore, the Dual Kc model is recommended for estimating the ET of young jujube trees in arid areas. Fruit orchards are frequently irrigated with brackish water. Irrigation with poor quality water is also a major cause of salt accumulation in soil. An excess of salts results in stunted growth, poor yield, inferior quality and low nutritional properties. Melatonin is a low molecular weight protein that shows multifunctional, regulatory and pleiotropic behavior in the plant kingdom. Recently, its discovery brought a great revolution in sustainable fruit production under salinity-induced environments. Melatonin contributed to enhanced tolerance in Zizyphus fruit species by improving the plant defense system’s potential to cope with the adverse effects of salinity. The supplemental application of melatonin has improved the generation of antioxidant assays and osmolytes involved in the scavenging of toxic ROS. The tolerance level of the germplasm is chiefly based on the activation of the defense system against the adverse effects of salinity. The current study explored the contribution of melatonin against salinity stress and provides information regarding which biochemical mechanism can be effective and utilized for the development of salt-tolerant germplasm in Zizyphus. Currently, microencapsulation has become a viable method of nutrient delivery for the food industry. This work microencapsulated the bioactive compounds extracted from two neglected species (Balanites aegyptiaca and Ziziphus mauritiana) by freeze-drying. A combination of wall materials (whey protein and pectin; soy protein and maltodextrin) was chosen to prepare the microcapsules. The phytochemical and physicochemical characterization of the microcapsules was then carried out. The encapsulation yield ranged from 82.77% to 96.05% for Balanites and Ziziphus, respectively, whereas the efficiency was 76.18 ± 1.39% and 80.93 ± 1.71%. The stimulated in vitro gastrointestinal test showed that encapsulation increased the bioavailability of the bioactive compounds. The total carotenoids were the most bioavailable compounds with 85.89 ± 0.06% for Ziziphus and 70.46 ± 1.10% for Balanites, followed by total flavonoids for Zizyphus with 63.27 ± 1.56%. Furthermore, regardless of species or wavelengths, the HPLC analysis resulted in the identification of 17 bioactive metabolites. The predominant one was epicatechin, whose level ranged from 231.52 ± 5.06 to 250.99 ± 3.72 mg/100 g DW in Ziziphus and 91.80 ± 3.85 to 116.40 ± 4.09 mg/100 g DW in Balanites. In estimating the enzyme inhibition and antioxidant power, both studied fruits showed antidiabetic, inflammatory, and antioxidant effects. These findings suggest that natural bioactive compounds are abundant in the fruits of Z. mauritiana and B. aegyptiaca and could be a valuable source for the food and pharmaceutical industries. A homogalacturonan pectin (HG, designated as WJP-F80) was extracted from the peel of winter jujube (Zizyphus jujuba Mill. Cv. Dongzao) and separated via ethanol-graded precipitation. The structural and conformational features were elucidated through HPAEC-PAD, GC-MS, 2D NMR, and HPSEC-MALLS studies. In vivo assessments were carried out to evaluate the hepatoprotective effects of WJP-F80 against CCl4-induced injury of mice. Results showed that WJP-F80 was a linear 1,4-α-galacturonan with partially methyl-esterified at O-6 of GalpA and occasionally acetylation. The Mw of WJP-F80 was determined as 45.3 kDa, the polydispersity was calculated as 1.56, and the Rg was measured as 22.7 nm in 0.1 M NaNO3. The conformational analysis revealed that WJP-F80 exhibited as rigid stiff chain in low Mw range, while aggregation by self-assembly of HG chains lead to high Mw and random coil conformation. In vivo studies indicated that WJP-F80 can protect the livers of mice from acute injury induced via CCl4 by decreasing the serum biochemical markers of alanine aminotransferase (ALT) and aspartate aminotransferase (AST) to normal levels. This work provides a theoretical basis for the value-added deep processing of winter jujube. Full article

Salt injury, as an abiotic stress factor, seriously affects the quality and yield of crops. This study was conducted by analyzing alternative splicing in the control (CK), δ-aminolevulinic acid (ALA), NaCl, and ALA + NaCl treatments of wild jujube (Zizyphus spinosus (Bunge)Hu) using RNA-seq. It was found that the unique differential alternative splicing is closely related to the alleviation of salt stress and the analyzed intermediates of chlorophyll synthesis and chlorophyll content in the leaves. The results showed that the content and synthesis of chlorophyll were reduced and disrupted in wild jujube leaves under NaCl stress, and the exogenous spraying of ALA could alleviate the effect of NaCl stress on the content of chlorophyll. RNA-seq indicated that the alternative splicing of genes was dominated by exon skipping in all the experimental treatments. Gene Ontology and Kyoto Encyclopedia of Genes and Genomes analyses revealed that the CK and ALA + NaCl comparison groups were primarily enriched in porphyrin and chlorophyll metabolism, chloroplast, and energy metabolism pathways. It is hypothesized that ALA treatment can alleviate the effects of salt stress on chlorophyll by altering the alternative splicing of genes related to photosynthesis, chlorophyll metabolism, and energy metabolism in wild jujube. In addition, the verification of alternative splicing genes related to chlorophyll metabolism in wild jujube revealed that treatment with ALA significantly upregulated the expression of HEMH, UROIII, and ChlD genes in the leaves under salt stress and increased the content of the chlorophyll biosynthetic intermediates endogenous ALA, porphobilinogen, protoporphyrin IX, Mg-protoporphyrin IX, and protochlorophyllide, which served to alleviate the effects of NaCl stress on chlorophyll in wild jujube.
