Snail mucus is increasingly investigated as a biologically compatible source of multifunctional biomolecules for pharmaceutical and dermatological use. However, the chemical profile and biological activities of mucus from the Moroccan endemic terrestrial snail Otala tingitana remain poorly characterized. In addition, the influence of heliciculture diet on the composition and functional properties of the mucus remains unclear. Here, O. tingitana was reared for 140 days under controlled conditions and fed a basal flour diet or the same diet supplemented with 3% Rosmarinus officinalis, Origanum compactum, or Thymus zygis subsp. zygis. Mucus from wild snails was included for comparison. Mucus samples were chemically profiled by GC–MS and evaluated for antibacterial activity, antioxidant capacity, wound-healing efficacy in mice, and histological anti-inflammatory effects, and evaluated semi-quantitatively based on the degree of inflammatory cell infiltration. GC–MS identified 13 compounds and demonstrated clear diet-dependent shifts in dominant components. All mucus samples exhibited broad-spectrum antibacterial activity against Staphylococcus aureus, Listeria monocytogenes, Escherichia coli, and Salmonella Typhimurium (inhibition zones 10.31–14.30 mm; MIC 120–240 µg/mL), with predominantly bactericidal profiles (MBC/MIC < 4) and significantly enhanced activity in plant-supplemented groups (p < 0.05). Antioxidant performance improved markedly with medicinal-plant supplementation, reaching low IC50 values (best ≈ 1.18 mg/mL) compared with basal-diet mucus. In vivo, topical application accelerated wound closure, achieving complete healing in <21 days, versus 28 days in untreated controls. In addition, histological assessment showed faster resolution of inflammatory cell infiltration in treated groups. Collectively, these findings provide the first integrated evidence that O. tingitana mucus possesses antibacterial, antioxidant, wound-healing, and anti-inflammatory activities, and that heliciculture diet is a practical lever to optimize its bioactive profile. Further studies should prioritize standardized manufacturing, contaminant control, and safety/toxicology assessment before translational development. Full article

Background/Objectives: Fungal colonization and biofilm formation on intrauterine device (IUD) strings are known to contribute to recurrent infections and decreased contraceptive efficacy. This study aims to develop a novel approach to prevent Candida reservoir and biofilm formation on IUD strings, thereby lowering the risk of IUD-associated vulvovaginal candidiasis (VVC). Methods: Cervicovaginal samples were collected from human cervix using a sterile cytobrush, avoiding microbial contamination. Cytological examination using the Papanicolaou method was performed to detect the presence of Candida. The antifungal effect of the essential oils (EOs) was determined by broth dilution and disk diffusion methods. Antifungal and biofilm inhibitory effects of Thymus zygis (Tz) EO-coated IUD strings were determined by agar diffusion and crystal violet binding assays, while fungal growth on the coated strings was assessed using Scanning Electron Microscopy (SEM) and Energy-Dispersive X-ray (EDX) analysis. Results: Tz EO exhibited significantly lower minimum inhibitory concentration (MIC ≤ 0.06 µL/mL) and minimum fungicidal concentration (MFC = 0.24 µL/mL) values compared to Melaleuca alternifolia (Ma) EO (MIC > 0.24 µL/mL, MFC = 1.95 µL/mL), along with larger zones of inhibition (ZOI) against both Candida albicans (110.0 ± 6.0 mm vs. 91.3 ± 7.0 mm) and Candida glabrata (84.0 ± 13.1 mm vs. 50.0 ± 9.2 mm), indicating a stronger antifungal potential. On IUD strings coated with 4% (40 μL/g) Tz EO in hypromellose ointment, the biofilm formation of both C. albicans and C. glabrata strains was inhibited by 58.9% and 66.7%, respectively, as confirmed by SEM and EDX. Conclusions: Tz EO-coated IUD strings effectively inhibit Candida growth, suggesting a promising natural strategy to reduce recurrent IUD-associated fungal infections. However, before these results can be translated to clinical practice, additional research is needed. Future investigations may encompass an extended number of Candida isolates, stability and release studies of the EO in relation to the formulation, toxicity to vaginal mucosa, epithelial cells and sperm motility, and the effect on vaginal microbiotia. Six aromatic plants (Lavandula pedunculata subsp. sampaioana, Lavandula stoechas subsp. luisieri, Mentha × piperita, Origanum vulgare subsp. virens, Thymus mastichina, and Thymus zygis subsp. sylvestris) were analyzed to evaluate their essential oil (EO) yield, chemical composition, antioxidant activity, and antifungal capacity against two mold species, green mold (Penicillium digitatum) and blue mold (Penicillium italicum). The antioxidant activity was evaluated using the ABTS and DPPH methods, and the antifungal activity was determined using the disk diffusion method. The results of the antioxidant activity tests showed that the essential oil of Th. zygis subsp. sylvestris has the highest value for the ABTS method (161.70 ± 0.15 mM TROLOX eq. and 864.20 ± 0.81 g TROLOX eq/g EO) and the L. stoechas subsp. luisieri essential oil in the DPPH method (33.91 ± 1.21 mM TROLOX eq. and 184.99 ± 6.58 g TROLOX eq/g EO). Furthermore, the essential oils with lower antioxidant activity were L. pedunculata subsp. sampaioana for the ABTS method (3.84 ± 0.26 mM TROLOX eq. and 20.79 ± 1.41 g TROLOX eq/g EO) and Th. mastichina for DPPH method (0.96 ± 0.03 mM TROLOX eq. and 5.31 ± 0.16 g TROLOX eq/g EO). Th. zygis subsp. sylvestris exhibited the strongest antifungal activity, with medium inhibition halo values of 60.50 ± 5.77 mm and 54.33 ± 2.93 mm for P. digitatum and P. italicum, respectively. Cyperus esculentus L. var sativus is cultivated in Spain, only in the L’Horta Nord in the Valencia region. In this country, tubers are consumed fresh to make a popular beverage in the Valencia region called “horchata de chufa” (chufa milk). This drink is considered beneficial for human health thanks to its high nutritional value and medicinal importance in several treatments. This work evaluates the antifungal potential of the Thymus zygis essential oil against fungi found in tiger nut warehouses to preserve tubers under the best conditions. The analyzed commercial thyme essential oil belongs to the thymol/p-cymene/γ-terpinene chemotype. Thymol was found in larger quantities (51.34%), followed by the identified biogenetic precursors p-cymene (35.16%) and γ-terpinene (3.53%). Carvacrol also appeared, but in small quantities (3.53%). During in vitro tests, the T. zygis EO showed strong inhibition (98.85% to 91.81% MGI) against fungi Alternaria alternata, Fusarium andiyazi, Fusarium incarnatum, and Fusarium oxysporum at 300 µg/mL. It totally inhibited their growth (100% MGI) at 400 µg/mL, and did so strongly (75.94%, 72.02%, and 70.78%) with fungi Podospora australis, Penicillium commune, and Cladosporium subuliforme, respectively. Under in vivo conditions, formulated as a protective biofilm, and by forcing the environmental conditions of temperature and humidity to the maximum for fungus F. andiyazi growth on tiger nut tubers, the created film acted as a strong protector against fungal attacks. This study aimed to investigate the chemical composition and bioactivities of essential oils (EOs) from five Moroccan thyme species: Thymus broussonetii subsp. broussonetii, T. maroccanus, T. willdenowii, T. zygis subsp. gracilis, and T. satureioides, collected from various geographical regions. Gas chromatography–mass spectrometry (GC-MS) identified thymol, p-cymene, borneol, γ-terpinene, carvacrol, α-pinene, and camphene as major constituents, with variations across species. Inductively coupled plasma atomic emission spectroscopy (ICP-AES) revealed important levels of calcium (450.6–712.2 mg/kg), potassium (255.4–420.7 mg/kg), magnesium (97.3–150.7 mg/kg), and iron (1.95–15.1 mg/kg). The EOs demonstrated strong antioxidant activity in DPPH, FRAP, and β-carotene bleaching assays. Insecticidal activity against Aphis gossypii revealed the highest efficacy with T. willdenowii EO (LC50 = 6.2 µL/mL), followed by T. maroccanus and T. zygis subsp. gracilis. Additionally, the EOs exhibited potent enzyme inhibitory effects at 1 mg/mL on acetylcholinesterase (83.1–96.4%), tyrosinase (77.5–89.6%), and α-glucosidase (79.4–89.5%). These findings suggest that thyme EOs, particularly from T. willdenowii, T. zygis, and T. maroccanus, are promising candidates for integrated pest management and natural enzyme inhibitors. Their potential applications in medicinal and pharmaceutical fields underscore the need for further research to optimize their use under specific conditions and dosages. Full article

The use of combined essential oils (EOs) is a new technique that can improve their preservative effects while minimizing their sensory impact in foods. The aim of this study was to determine the chemical profile of three essential oils (EOs) extracted from Lavandula stoechas L. (Ls), Thymus zygis L. (Tz), and Eucalyptus camaldulensis Dehnh (Ec) and to evaluate their synergistic antibacterial activity for optimal inhibition against Bacillus subtilis, Escherichia coli, and Staphylococcus aureus using an augmented Simplex centroid mixing scheme. The essential oils were extracted by hydrodistillation and analyzed via gas chromatography–mass spectrometry. Anti-bacterial potency was evaluated by disk diffusion. Chemical analysis revealed the main compounds in Lavandula stoechas (Ls) essential oil: camphor (36.15%), followed by fenchone (16.57%) and Z-8-hydroxy linalool (8.28%). The Thymus zygis (Tz) essential oil is dominated by δ-terpineol (27.64%), δ-3-carene (15.7%), and thymol (14.17%). In contrast, the Eucalyptus camaldulensis (Ec) essential oil contains mainly 1,8-cineole (43.61%), γ-terpinene (11.71%), and α-terpineol (10.58%). The optimal mixture is the binary association of 40% E. camaldulensis EO and 60% T. zygis EO, which provides an effective inhibition diameter (ID) of 13.37 mm to inhibit S. aureus. Furthermore, the formulation of 27% and 73% EOs of E. camaldulensis and T. zygis, respectively, corresponds to the mixture required to achieve the optimum inhibition diameter (ID = 11.55 mm) against E. coli. In addition, the mixture of 29% EO of E. camaldulensis and 71% EO of T. zygis is the optimum mixture to inhibit B. subtilis, with an inhibition diameter of 12.31 mm. These findings highlight the potency of antibacterial formulations of these essential oils and suggest that they might be used as substitutes for conventional drugs to prevent the development of bacteria responsible for serious infections and food spoilage. Tetracyclines have a high resistance percentage in Salmonella spp. of both human and animal origin. Essential oils, such as cinnamon (Cinnamomum zeylanicum), clove (Eugenia caryophyllata), oregano (Origanum vulgare), and red thyme (Thymus zygis), have shown bactericidal activity against this bacterium. However, in many cases, the minimum inhibitory concentration (MIC) exceeds the cytotoxicity limits. The objective of this study was to assess the in vitro efficacy of combining oxytetracycline with essential these oils against field multidrug-resistant (MDR) Salmonella enterica strains. The MIC of each product was determined using the broth microdilution method. The interaction was evaluated using the checkerboard method, by means of the fractional inhibitory concentration index (FICindex) determination. The results showed a positive interaction (synergy and additivity) between oxytetracycline and the four oils tested, resulting in a reduction in both products’ MICs by 2 to 4 times their initial value, in the case of oils, and by 2 to 1024 times in the case of the antibiotic. The combination of oxytetracycline and cinnamon achieved the best results (FICindex 0.5), with a decrease in the antibiotic effective concentration to below the sensitivity threshold (MIC of the combined oxytetracycline 0.5 µg/mL). There was no antagonistic effect in any case, although differences in response were observed depending on the bacterial strain. The results of this study suggest that combining oxytetracycline with cinnamon oil could be an effective alternative for controlling tetracycline-resistant strains of Salmonella. However, its individual use should be further evaluated through in vitro susceptibility tests. Full article

Essential oils are of great interest due to their potent pharmaceutical and biological activities. In this study, essential oils extracted from Origanum compactum and Thymus zygis originating from the Middle Atlas of Morocco were investigated. Their chemical compositions were analyzed using gas chromatography and mass spectrometry, while the assessment of the trapping power of the radical (DPPH: 1,1-diphenyl-2-picrylhydrazyl) and the reducing antioxidant potential of ferric ions (FRAP: Ferric Reducing Antioxidant Power) were performed in order to evaluate the antioxidant activity. Their antibacterial potency was tested against six bacterial strains through the disk diffusion method. The chromatography analyses of the extracted essential oils highlighted the presence of two main components, namely carvacrol at 75.70% in O. compactum and thymol at 40.67% in T. zygis. The antioxidant activity tests showed that both essential oils demonstrated a significant antioxidant activity comparable to the positive control (e.g., ascorbic acid). The antibacterial activity results showed a strong antimicrobial effect for both essential oils, compared to synthetic antibiotics. This study affirms the presence of bioactive components with interesting antioxidant and antibacterial activities in the essential oils extracted from Origanum compactum and Thymus zygis, which could find several applications in the food and pharmaceutical industries through the substitution of synthetic antioxidants and antibiotics. With the growing issues of food spoilage, microbial resistance, and high mortality caused by cancer, the aim of this study was to evaluate T. zygis essential oil (TZEO) as a potential solution for these challenges. Here, we first performed GC/MS analysis which showed that the tested TZEO belongs to the linalool chemotype since the abundance of linalool was found to be 38.0%. Antioxidant activity assays showed the superiority of TZEO in neutralizing the ABTS radical cation compared to the DPPH radical. The TZEO was able to neutralize 50% of ABTS•+ at the concentration of 53.03 ± 1.34 μg/mL. Antimicrobial assessment performed by employing disc diffusion and minimal inhibitory concentration assays revealed TZEO as a potent antimicrobial agent with the highest inhibition activity towards tested gram-negative strains. The most sensitive on the treatment with TZEO was Enterobacter aerogenes showing an MIC 50 value of 0.147 ± 0.006 mg/mL and a MIC 90 value of 0.158 ± 0.024 mg/mL. Additionally, an in situ analysis showed great effects of TZEO in inhibiting gram-negative E. coli, P. putida, and E. aerogenes growing on bananas and cucumbers. Treatment with the TZEO vapor phase in the concentration of 500 μg/mL was able to reduce the growth of these bacteria on the food models to the extent > 90%, except for E. coli growth on the cucumber, which was reduced to the extent of 83.87 ± 4.76%. Furthermore, a test on the antibiofilm activity of the tested essential oil revealed its biofilm prevention effects against Salmonella enterica which forms biofilms on plastic and stainless-steel surfaces. Performed tests on the TZEO effects towards cell viability showed no effects on the normal MRC-5 cell line. However, the results of MTT assay of TZEO effects on three cancer cell lines (MDA-MB-231, HCT-116, and K562) suggest that TZEO exerted the strongest effects on the inhibition of the viability of MDA-MB-231 cells, especially after long-term treatment in the highest concentration applied with reducing the viability of the cells to 57%. Additionally, results of NBT and Griess assays suggest that TZEO could be a convenient candidate for future testing for developing novel antitumor therapies. Full article

Wall. ex Nees, Pelargonium sidoides DC., Echinacea species and a combination of ivy (Hedera helix L.), primrose (Primula veris L./Primula elatior L.) and thyme (Thymus vulgaris L./Thymus zygis L.) have promising clinical effects in uncomplicated, acute upper respiratory tract infections (URTI) treatment. However, mechanistic evidence of these herbal treatments is lacking. The objective of this Pstudy is to provide an overview of mechanistic evidence for these effects. Thirty-eight databases were searched. Included studies were mechanistic studies (in vitro, animal, and human studies and reviews) on these herbs; published before June 2021. Non-mechanistic studies or studies on combinations of herbs other than ivy/primrose/thyme were excluded. Furthermore, three experts in traditional, complementary and integrative healthcare (TCIH) research and pharmacognosy were interviewed to collect additional expert knowledge. The results show that A. paniculata acts through immunomodulation and antiviral activity, possibly supplemented by antibacterial and antipyretic effects. P. sidoides acts through antiviral, indirect antibacterial, immunomodulatory and expectorant effects. Echinacea species likely act through immunomodulation. The combination of ivy/primrose/thyme combines secretolytic and spasmolytic effects from ivy with antibacterial effects from thyme. Studies on primrose were lacking. This mechanistic evidence supports the difference-making evidence from clinical studies, contributes to evidence-based recommendations for their use in URTI treatment, and guides future mechanistic studies on URTI treatments. Plant essential oils (PEOs) are being studied as a potential alternative to synthetic pesticides in agriculture. PEOs have the potential to control pests both directly, by being toxic or repellent to pests, and indirectly, by activating plant’s defense mechanisms. In this study, the effectiveness of five PEOs (Achillea millefolium, Allium sativum, Rosmarinus officinallis, Tagetes minuta, and Thymus zygis) on controlling Tuta absoluta and their impact on the predator Nesidiocoris tenuis was examined. The study revelead that PEOs from A. millefolium and A. sativum-sprayed plants significantly reduced the number of T. absoluta-infested leaflets and did not affect the establishment and reproduction of N. tenuis. Additionally, the spraying of A. millefolium and A. sativum increased the expression of defense genes in the plants, triggering the release of herbivory-induced plant volatiles (HIPVs), such as C6 green leaf volatiles, monoterpenes, and aldehydes, which can be messengers in tritrophic interactions. The results suggest that PEOs from A. millefolium and A. sativum can provide a dual benefit for controlling arthropod pests, as they can directly exhibit toxicity against these pests while also activating plant defense mechanisms. Overall, this study provides new insights into using PEOs as a sustainable solution for controlling pests and diseases in agriculture, by reducing synthetic pesticides and promoting the use of natural predators. Medicinal plants represent an inexhaustible source of traditional and effective remedies thanks to the various active ingredients they contain. Secondary metabolites present in plant extracts, such as essential oils (EO), have remarkable pharmacological properties, including antimicrobial effects. Here, the chemical composition and antimicrobial effects of four Moroccan medicinal plants, Rosmarinus officinalis L. (R. officinalis), Mentha pulegium L. (M. pulegium), Salvia officinalis L. (S. officinalis), and Thymus zygis subsp. gracilis (Boiss.) R. Morales (T. zygis), traditionally used in Morocco to treat microbial infections, were addressed. EO were extracted using the hydrodistillation method, and analyzed by gas chromatography coupled with mass spectrometry (GC/MS). EO yields were of 3.64 ± 0.12, 3.53 ± 0.06, 2.48 ± 0.06, and 2.34 ± 0.08%, respectively, for M. pulegium, R. officinalis, S. officinalis, and T. zygis. The main bioactive components present in these EO were piperitenone (32.9%) and pulegone (32.8%) for M. pulegium, 1,8-cineol (43.8%) and camphor (18.7%) for R. officinalis, 1,8-cineole (16.8%) and trans-thujone (15.9%) for S. officinalis, and thymol (36.4%), carvacrol (24.1%) and cymene (23.5%) for T. zygis. These EO showed, according to the results of their antimicrobial activities, good effectiveness against bacteria and fungi. Moreover, the T. zygis EO showed the most potent activity against all bacteria studied, while that of R. officinalis, M. pulegium, and S. officinalis showed moderate activity against the Enterobacter cloacae of Streptococcus agalactiae and Escherichia coli. The antifungal activity tests revealed a strong antifungal activity for the T. zygis EO and a moderate activity for the S. officinalis EO. On the other hand, the EO of R. officinalis, and M. pulegium were found to be inactive at the doses used against the selected strains. In conclusion, our results show that the medicinal plants studied contain biologically active molecules with antimicrobial effects. They can replace synthesized molecules, especially in the formulation of additives, and for therapeutic, cosmetic, and food-processing purposes. Full article

The present work was aimed to study the toxicity of the essential oils of three aromatic and medicinal plants on the tomato leaf miner Tuta absoluta and the Medfly Ceratitis capitata as an alternative to conventional pesticides. We carried out a phytochemical and insecticide study of T. zygis L., S. officinalis L. and M. suaveolens Ehrh. essential oils (EOs) through the study of their chemical composition and their toxicity on C. capitata adults and T. absoluta larvae. The extraction of the EOs by hydrodistillation showed yields of 3.87 ± 0.03, 4.09 ± 0.23 and 4.35 ± 0.11 for T. zygis, S. officinalis L. and M. suaveolens Ehrh., respectively. The identification of the chemical composition of the EOs by GC/MS showed that oxygenated monoterpenes constituted the most abundant group for all the extracted EOs. The major compounds were rather diversified depending on plant species. In fact, the S. officinalis L. EO mainly contained trans-thujone (21.80 %), the M. suaveolens Ehrh. EO mainly contained piperitenone oxide (71.19%), and carvacrol (61.60%) was the main component of the T. zygis L. EO. An insecticidal effect was observed for the three studied EOs on C. capitata adults and T. absoluta larvae. The observed LD50 values were 0.80 µL/mL and 11.04 µL/mL for M. suaveolens and S. officinalis, respectively, on T. absoluta larvae. For C. capitata adults, the obtained LD50 values were 0.9 µL/mL and 11.78 µL/mL for M. suaveolens and T. zygis, respectively. The presented findings could contribute to the development of biopesticides for plants as a component of integrated pest management strategies in citrus and tomato crops. Full article

Innovative adhesive formulations have been developed in the laboratory based on urea-formaldehyde resin by adding medicinal plants to an industrial adhesive formulation containing raw materials: urea-formaldehyde resin, urea, ammonium sulphate and starch. Specifically, Thymus species (Thymus bleicherianus, Thymus capitates, Thymus satureioides, Thymus vulgaris and Thymus zygis) replaced part of the starch and were considered as the second filler in the formulations. The physico-chemical properties of the resulting adhesive formulations, such as: pH, viscosity, gel time, solids content, density, concentration of free formaldehyde and color were measured, and characterized using Scanning Electron Microscope (SEM), X-ray Diffraction (XRD), Differential Thermal Analysis (DTA), Thermogravimetric Analysis (TGA) and Fourier Transform Infrared spectroscopy (FTIR). In order to evaluate the mechanical performances of adhesive formulations based on plants, plywood panels were produced and their mechanical properties were studied. These mechanical properties included the shear strength, bending strength and the modulus of elasticity. The performance of these panels is comparable to that of plywood panels made using the standard adhesive formulation. From the results obtained, and following the statistical studies, the new adhesive formulations based on plants have the same physico-chemical properties, the same morphologies, and the same mechanical properties. Moreover, the novel adhesives are more viscous, and they have less free formaldehyde content than the commercial formulation. Full article

Medicinal and aromatic plants (MAPs) are potential sources of natural bioactive phytochemical compounds of an incredible worth for the food industry, such as polyphenols. Lamiaceae medicinal and aromatic plants from Granada’s high plateau, concretely Origanum bastetanum, Thymus zygis gracilis, Thymus longiflorus, Thymus membranaceus and Ziziphora hispanica, were evaluated under different conventional solid–liquid extraction conditions to obtain extracts enriched in bioactive compounds. Phenolic profile was detected by HPLC-QTOF-MS, identifying a high abundance of bioactive constituents. Furthermore, antioxidant and antiviral activities of the mentioned plants were studied as biological properties of interest for the improvement of food shelf-life. Thus, Origanum bastetanum showed the highest antioxidant potential for all assays. Antiviral activity was also tested against some important foodborne viruses, feline calicivirus (FCV), murine norovirus (MNV) and hepatitis A virus (HAV), with the highest activity obtained for Ziziphora hispanica, Thymus longiflorus and Origanum bastetanum. This research proposes the studied plants as rich sources of bioactive compounds with potential use as preservatives in the food industry. Full article
