To enhance PharmD student leadership and advocacy skills, combat the paucity of trained pre-health advisors for pharmacy admissions, augment community relationships, and increase pharmacy admissions volume, we designed, implemented, and assessed PAALS, a Pre-health Academic Advising and Leadership System. PAALS was grounded in Astin’s Theory of Student Involvement and evaluated using the RE-AIM implementation science framework. RE-AIM measured outcomes across Reach, Effectiveness, Adoption, Implementation, and Maintenance as indicators of PAALS’s scale, fidelity, sustainability, and institutional embedding. Analysis of PAALS using the RE-AIM framework demonstrated the following outcomes: (1) Reach: 42 P1-P3 PharmD students participated as mentors; external partnerships expanded from 2 to 8 regional high schools and community programs; and more than 25 mentored learners successfully matriculated into the PharmD program. (2) Effectiveness: students enacted sustained leadership, advocacy, and mentoring roles. (3) Adoption: voluntary uptake of mentoring and governance roles by PharmD students occurred with repeated engagement by external partner institutions. (4) Implementation: Core program components were delivered consistently using existing institutional resources. (5) Maintenance: PAALS remained operational across five academic years despite student turnover, with leadership succession and institutional embedding sustained across cohorts. Our findings demonstrate that student-led advising and advocacy ecosystems address critical gaps in pharmacy-specific pre-health advising models. Full article

The flavivirus NS1 protein is a component of the viral replication complex and plays diverse, yet poorly understood, roles in the viral life cycle. To enable real-time visualization of the developing replication organelle and biochemical analysis of tagged NS1 and its interacting partners, we engineered a replication-competent yellow fever virus (YFV) replicon encoding a C-terminal fusion of NS1 with green fluorescent protein (NS1–GFP). The initial variant was non-viable in the absence of trans-complementation with wild-type NS1; however, viability was partially restored through the introduction of co-adaptive mutations in GFP (Q204R/A206V) and NS4A (M108L). Subsequent cell culture adaptation generated a 17-nucleotide frameshift within the NS1–GFP linker, resulting in a more flexible and less hydrophobic linker sequence. The optimized genome, in the form of a replicon, replicates in packaging cells that produce YFV structural proteins, as well as in naive BHK-21 cells. In the packaging cells, the adapted NS1–GFP replicon produces titers of infectious particles of approximately 10^6 FFU/mL and is genetically stable over five passages. The expressed NS1–GFP fusion protein localizes to the endoplasmic reticulum and co-fractionates with detergent-resistant heavy membranes, a hallmark of flavivirus replication organelles. This NS1–GFP replicon provides a novel platform for studying NS1 functions and can be further adapted for proximity-labeling strategies aimed at identifying the still-unknown protease responsible for NS1–NS2A cleavage. Our objective was to conduct a scoping review and narrative synthesis of qualitative studies that examined experiences of partners of cancer patients. We searched MEDLINE, Embase, CINAHL, PsycINFO, and Scopus for qualitative studies involving adult (≥18 years) partners (e.g., in a romantic relationship) of patients diagnosed with cancer and published in English. We extracted findings from included studies, along with key study and participant characteristics, and applied a narrative summary approach, a process that allowed us to identify synthesized themes across studies. Our search identified 15,729 records, of which 159 met the inclusion criteria. Included studies primarily collected data through interviews, with participants being predominantly female and middle-aged (55.2 ± 8.3 years). Partners were commonly in relationships with patients diagnosed with breast, genital–urinary, or gastrointestinal cancer. Our synthesis identified four conceptual themes—transformation of relationship dynamics and roles, distress and burden, coping strategies, and unmet needs and support gaps—which reflect the emotional, relational, and practical challenges partners navigate throughout the cancer trajectory. These findings highlight the need to better recognize and support the role of partners within the cancer care landscape as their wellbeing impacts care and experiences of patients with cancer. Interspecies communication is increasingly recognized as an affective–cognitive process co-created between humans and animals rather than a one-directional transmission of signals. This review integrates findings from ethology, neuroscience, welfare science, behavioral studies, and posthumanist ethics to examine how emotional expression, communicative intentionality, and relational engagement shape understanding across species. Research on primates, dogs, elephants, and marine mammals demonstrates that empathy, consolation, cooperative signaling, and multimodal perception rely on evolutionarily conserved mechanisms, including mirror systems, affective contagion, and oxytocin-mediated bonding. These biological insights intersect with ethical considerations concerning animal agency, methodological responsibility, and the interpretation of non-human communication. Emerging technological tools—bioacoustics, machine vision, and AI-assisted modeling—offer new opportunities to analyze complex vocal and behavioral patterns, yet they require careful contextualization to avoid anthropocentric misclassification. Synthesizing these perspectives, the review proposes a relational framework in which meaning arises through shared emotional engagement, embodied interaction, and ethically grounded interpretation. This approach highlights the importance of welfare-oriented, minimally invasive methodologies and supports a broader shift toward recognizing animals as communicative partners whose emotional lives contribute to scientific knowledge. This review primarily synthesizes empirical and theoretical research on primates and dogs, complemented by selected examples from elephants and marine mammals, which provide the most developed evidence base for the affective–cognitive and relational mechanisms discussed. Over the past five years, generative AI has carved out a major role in architecture, especially in education and visual idea generation. Most of the time, the literature talks about AI as a tool, an assistant, or sometimes a co-creator, always highlighting efficiency and the end product in architectural design. There is a steady rise in empirical studies, yet the real impact on how young architects learn still lacks a solid theory behind it. In this systematic review, we dig into peer-reviewed work from 2015 to 2025, looking at how generative AI fits into architectural design education. Using PRISMA guidelines, we pull together findings from 40 papers across architecture, design studies, human–computer interaction and educational research. What stands out is a clear tension: on one hand, students crank out more creative work; on the other, their reflective engagement drops, especially when AI steps in as a replacement during early ideation instead of working alongside them. To address this, we introduce the idea of “AI sparring”. Here, generative AI is not just a helper—it becomes a provocateur, pushing students to think critically and develop stronger architectural concepts. Our review offers new ways to interpret AI’s role, moving beyond seeing it just as a productivity booster. Instead, we argue for AI as an active, reflective partner in education, and we lay out practical recommendations for studio-based teaching and future research. This paper is a theoretical review and conceptual proposal, and we urge future studies to test these ideas in practice. Full article

Geothermal energy is a crucial component of climate adaptation and sustainability transitions, as it provides a dependable, low-carbon source of baseload power that can accelerate sustainable energy transitions and enhance climate resilience. Yet, in East Africa—one of the world’s most promising geothermal regions, with the East African Rift—a unique climate-energy opportunity zone—the harnessing of geothermal power remains slow and uneven. This study examines the contextual conditions that facilitate the successful and sustainable development of geothermal power in the region. Drawing on semi-structured interviews with 17 experienced professionals who have worked extensively on geothermal projects across East Africa, the analysis identifies how technical, institutional, managerial, and relational circumstances interact to shape outcomes. The findings indicate an interdependent configuration of success conditions, with structural, institutional, managerial, and meta-conditions jointly influencing project trajectories rather than operating in isolation. The most frequently emphasised enablers were resource confirmation and technical design, leadership and team competence, long-term stakeholder commitment, professional project management and control, and collaboration across institutions and communities. A co-occurrence analysis reinforces these insights by showing strong patterns of overlap between core domains—particularly between structural and managerial factors and between managerial and meta-conditions, highlighting the mediating role of managerial capability in translating contextual conditions into operational performance. Together, these interrelated circumstances form a system in which structural and institutional foundations create the enabling context, managerial capabilities operationalise this context under uncertainty, and meta-conditions sustain cooperation, learning, and adaptation over time. The study contributes to sustainability research by providing a context-sensitive interpretation of how project success conditions manifest in geothermal development under climate transition pressures, and it offers practical guidance for policymakers and partners working to advance SDG 7 (Affordable and Clean Energy), SDG 9 (Industry, Innovation and Infrastructure), and SDG 13 (Climate Action) in Africa. Background: Food insecurity among college students is a multidimensional challenge shaped by individual, interpersonal, institutional, community, and policy factors. Although many campuses require or provide meal plans, students may experience food insecurity when barriers related to agency (choice and autonomy), utilization (nutrition security), and availability persist. This study explored how undergraduate students at a private, urban U.S. university experience and navigate the multiple dimensions of food insecurity. Methods: We conducted in-depth, semi-structured interviews via Zoom between December 2024 and January 2025 with n = 22 undergraduate students recruited based on food security status, determined by a Fall 2024 longitudinal survey using the USDA Six-Item Short Form. Transcripts were double-coded by trained research assistants in ATLAS.ti using an inductive codebook. Thematic analyses followed a phronetic, iterative approach, organizing findings within a socio-ecological determinants framework and comparing themes by food security status. Results: We identified nine themes across four domains (individual, interpersonal, institutional and community, and political). At the individual level, constrained personal resources for groceries and cooking, time scarcity leading to skipped meals, and health impacts that detracted from academics emerged as key themes. Interpersonally, reliable family financial support was protective and informal support from peers/coaches filled gaps sporadically for some. At the institutional and community level, dining hall hours misaligned with student schedules, perceived limited variety and nutrition quality reduced food agency and utilization, and transportation impeded use of the sole grocery partner accepting university meal plan benefits. Notably, meal plans including unlimited meal swipes provided stable access but did not guarantee food security when food agency and utilization barriers persisted. Many students relied on campus events for free food; formal assistance (e.g., food pantry) was largely underused. At the policy level, Supplemental Nutrition Assistance Program (SNAP) awareness and enrollment was limited among our sample. Conclusions: Meal plan access alone is insufficient to ensure food security. Campus strategies should extend beyond access to prioritize flexibility, variety, and alignment with students’ schedules and preferences, while strengthening communication and eligibility support for external benefits. Future work should design and evaluate interventions that integrate all dimensions of food security and address institutional policies affecting students’ basic needs. Full article

Colorectal cancer (CRC) remains a major cause of cancer-related deaths in advanced disease, and activating KRAS/NRAS mutations limit the use of anti-EGFR antibodies to RAS–wild-type tumors. The cellular prion protein (PrPC) has been linked to aggressive and chemoresistant CRC, but its extracellular partners and functional relevance in KRAS-mutant disease are not fully defined. Here, we examined extracellular PrPC complexes and PrPC-associated signaling in CRC cell lines and xenografts using a neutralizing PrPC monoclonal antibody. Across a CRC panel that included SNU-C5/WT and its 5-fluorouracil- and oxaliplatin-resistant derivatives, HT-29 (KRAS–wild-type), and HCT-8 and LoVo (KRAS-mutant), co-immunoprecipitation showed that PrPC forms complexes with the 37/67 kDa laminin receptor (RPSA), with PrPC–RPSA association particularly increased in KRAS-mutant HCT-8 and LoVo cells. PrPC protein levels were higher in KRAS-mutant HCT-8, SW620, and SNU-407 cells than in HT-29, and PrPC neutralization reduced viability in all four lines. Accordingly, we assessed upstream RAS activity and found that active RAS (RAS-GTP) was higher in KRAS-mutant cells than in HT-29, and PrPC treatment was associated with reduced RAS-GTP levels. In the same KRAS-mutant setting, basal AKT phosphorylation exceeded that in HT-29, and PrPC treatment lowered AKT phosphorylation without changing total AKT. Moreover, PrPC treatment was associated with reduced ERK1/2 phosphorylation in KRAS-mutant cells, suggesting attenuation of downstream RAS pathway output. These signaling changes coincided with a decrease in the S-phase fraction and an increase in G1. In an HCT-8 (KRAS G13D) xenograft model, PrPC monotherapy inhibited tumor growth in a dose-dependent manner, and 5-fluorouracil (5-FU) monotherapy produced an intermediate effect. The combination of PrPC (10 mg/kg) and 5-FU (20 mg/kg) yielded the greatest tumor growth inhibition among the tested regimens. Consistent with this enhanced tumor control, immunofluorescence of xenograft tissues showed that PrPC, particularly with 5-FU, reduced intratumoral PrPC and PCNA and decreased CD31-positive microvessels and α-SMA–positive vessel structures. Taken together, these findings suggest that extracellular PrPC supports RAS–AKT signaling, proliferation, and tumor-associated angiogenesis in KRAS-mutant colorectal cancer, and that PrPC neutralization additively enhances 5-fluorouracil activity in KRAS-mutant models. The data provide a preclinical basis for evaluating PrPC antibodies in combination with fluoropyrimidine-based regimens in patients with KRAS-mutant CRC. Full article

Objectives: The diagnosis of infective endocarditis (IE) is clinically challenging. This study aimed to examine an endocarditis network and the effects of delayed diagnosis. Methods: We reviewed the patients who were admitted for infective endocarditis at our institution between January 2012 and December 2021. Infective endocarditis was diagnosed according to ESC/EACTS guidelines for the management of endocarditis. Details of admitting hospitals were obtained from the German Hospital Directory. Data are presented as medians (25th–75th quartiles) or absolute values (percentages) unless otherwise specified. Results: A total of 812 consecutive patients were admitted to our centre for IE. Exact records on the time to diagnosis were available for 707 patients (87.1%). The patients were divided into two groups based on the time to diagnosis, i.e., up to 7 days (n = 509; 72.0% group ED) and more than 7 days (n = 198; 28.0% group LD). The EuroSCORE II (p = 0.001) and the EndoSCORE (p = 0.019) were significantly higher in the LD group. The median time to diagnosis was shorter in university hospitals as compared to non-teaching hospitals (p = 0.008) and among patients admitted to cardiology and cardiac surgery departments (p < 0.001). Patients diagnosed later had higher rates of tracheostomy (p < 0.001), longer ICU (p = 0.004) and hospital stays (p < 0.001) and higher in-hospital mortality (p = 0.027). We found that a delayed diagnosis (p = 0.040), stroke (p = 0.004), age > 75 years (p = 0.044) and atrial fibrillation (p < 0.001) were independently associated with in-hospital mortality. Furthermore, survival at 1 and 5 years was significantly higher in the ED group (p < 0.001). Conclusions: The diagnosis of IE may be influenced by a multitude of factors. Our results indicate that a delayed diagnosis is independently associated with an increased rate of in-hospital mortality. According to our results, an early diagnosis of IE may be associated with improved outcomes. Full article

Clinical significance: Their morphological consistency and anatomical distribution offer valuable elements for differentiating between intentional and accidental trauma. Case Presentation: In this study, we report the case of a 40-year-old woman who presented with multiple cutaneous lesions attributed to repeated assaults by her intimate partner. The forensic medical examination revealed five discrete scars characterized by sharply demarcated borders, circular to oval shapes, and dimensions ranging from 0.7 to 1.5 cm. These lesions were anatomically located in regions not typically accessible for self-infliction. To reinforce the diagnostic interpretation and assess reproducibility, a controlled experimental protocol was conducted using porcine skin matrices. Cigarette burns were recreated under variable conditions of contact pressure and exposure duration. The lesions produced on the biological substrate exhibited morphological features consistent with those observed in the patient, suggesting compatibility with cigarette-induced thermal injury. Conclusions: These findings provide circumstantial support for the forensic interpretation but must be considered within the limitations of the experimental model. This integrated approach underscores the relevance of combining clinical forensic documentation with experimental validation to support medico-legal conclusions in cases of suspected interpersonal violence. The application of Generative Artificial Intelligence (GenAI)—defined as a class of AI systems capable of autonomously generating new content such as images, texts, and design solutions based on learned data patterns—has become increasingly widespread in creative design. By supporting ideation, rapid trial-and-error, and data-driven decision-making, GenAI enables designers to explore design alternatives more efficiently and enhances human–computer interaction experiences. In design practice, GenAI functions not only as a productivity-enhancing tool but also as a collaborative partner that assists users in visual exploration, concept refinement, and iterative development. However, users still face a certain learning curve before effectively adopting these technologies. Within the framework of human-centered artificial intelligence, contemporary design practices place greater emphasis on inclusivity across diverse user groups and on enabling intuitive “what-you-think-is-what-you-get” interaction experiences. From a sustainable design perspective, GenAI’s capabilities in digital simulation, rapid iteration, and automated feedback contribute to more efficient design workflows, reduced collaboration costs, and broader access to creative participation for users with varying levels of expertise. These characteristics play a crucial role in enhancing the accessibility of design resources and supporting the long-term sustainability of creative processes. Focusing on the context of China’s digital design industry, this study investigates the application of GenAI in design workflows through an empirical case study of Zhitu AI, a generative design tool developed by Beijing Didi Infinity Technology Development Co., Ltd. The study conducts a literature review to outline the role of GenAI in visual design processes and employs observation-based experiments and semi-structured interviews with users of varying levels of design expertise. The findings reveal key pain points across stages such as prompt formulation, secondary editing, and asset generation. Drawing on the Kano model, the study further identifies potential design opportunities and discusses their value in improving efficiency, supporting non-expert users, and promoting more sustainable and inclusive design practices. This study proposes a human-in-the-loop dynamic graph-based planning framework designed to elevate LLM-based Embodied Agents from simple tools to trustworthy collaborative partners. To achieve this, we address the trade-off between the structural rigidity of plan-centric approaches and the instability of reactive methods. The framework utilizes a Directed Acyclic Graph (DAG) with AND/OR nodes to ensure robustness while maintaining flexibility. Critically, the agent features an Automated Recovery Mechanism for self-correction and a Dynamic Modification Mechanism that employs Relevance Analysis to effectively translate human interventions (Switch, Add, Delete) into graph updates. Comparative experiments in Minecraft with 30 participants validated the method’s effectiveness. The proposed agent (Agent B) outperformed the reactive baseline (Agent A), reducing mission completion time by 9.3%. Notably, the agent demonstrated high instruction compliance and reduced user frustration by approximately 20%, leading to statistically higher satisfaction scores (PSSUQ). These results confirm that by ensuring planning robustness and responsiveness, the proposed framework successfully enables agents to function as trustworthy partners in complex environments. Oncolytic coxsackievirus B3 (oCVB3) strain PD-H has shown potent oncolytic efficacy and a remarkable safety profile in the treatment of colorectal cancer in vivo after intratumoral (i.t.) injection. In this study, we investigated the safety and efficiency of PD-H following intravenous (i.v.) virus administration. When injected i.v. into Balb/C mice bearing subcutaneous Colon-26 tumors, PD-H led to slightly reduced tumor progression and a significant increase in animal survival, but it also caused multi-organ infection and tissue damage. To improve the safety profile of PD-H, we inserted microRNA target sites (miR-TS) of the heart-specific miR-1, pancreas-specific miR-375, liver-specific miR-122, and brain-specific miR-124 or the tumor-suppressor miR-145 into the genome of PD-H and generated the viruses PD-622TS and PD-145TS. Both viruses replicated similarly and induced cytotoxicity comparable to that of PD-H in the colorectal carcinoma cell lines Colon-26 and CT-26Luc. Their replication was inhibited in HEK293T cells transiently transfected with the cognate microRNAs. In vivo, i.v. administration of PD-145TS and PD-622TS to healthy Balb/C mouse resulted in significantly lower viral titers in the organs of mice and led to significantly less-intense pathological alterations compared to PD-H. PD-622TS injected i.v. into Balb/C mice with CT-26Luc-induced peritoneal carcinomatosis did not induce off-target alterations in normal organs, but it failed to induce a therapeutic effect. These data indicate that PD-H or microRNA-regulated PD derivatives exhibit only limited therapeutic efficacy following i.v. injection in colorectal tumor-bearing mice. However, the newly engineered microRNA-regulated PD-H variants demonstrate improved safety profiles. Full article

In this analysis, the dynamic nexus between green governance, energy transition, and carbon emissions in the period spanning 1990 and 2022 for the twenty-one member economies of the Organization for Economic Cooperation and Development (OECD) and partner economies is examined. Employing Feasible Generalized Least Squares (FGLS), Driscoll–Kraay Standard Errors (DKSE), and Quantile-on-Quantile Regression (QQR), this analysis encompasses the effects of the use of renewable energy sources, economic growth, and changes in the population on carbon emissions. Results for the analysis show that the adoption of renewable energy sources, tough environmental regulations, and green innovation play a significant role in offsetting carbon emissions since the results are more pronounced at the tail ends of the distribution of carbon emissions. Conversely, changes in the level of population and economic growth are identified as potential exacerbators of environmental concerns. In offering implications for policymakers, this analysis argues that environmental laws and taxation and green innovation are potential means of improving environmental governance in achieving the United Nations’ Sustainable Development Goals and climate change commitments. By addressing the issue of differential environmental effects based on varying levels of carbon emissions, this analysis makes contributions to the expanding literature on sustainable environmental governance in the twenty-first-century energy economy.
