Twinning in cattle is an increasingly common reproductive phenomenon that has significant implications for herd productivity and animal welfare. This retrospective study aimed to assess the occurrence rate of twinning (TW) in Italian Holstein multiparous cows and to identify the associated risk factors and consequences with respect to fertility, milk production, and cow survival. Data from 44,535 calvings across 61 Italian dairy farms from January 2019 to December 2020 were analyzed. The overall TW rate was found to be 3.79%, affected by factors such as parity (p < 0.0001), season of conception (p < 0.0001), previous lactation 305-day mature equivalent (p < 0.05), and prior days open (p < 0.0001). Cows experiencing TW had significantly shorter gestation lengths (p < 0.0001) compared to singletons and lower 305-day milk production estimates (11,094 kg for TW vs. 11,295 kg for singleton females and 11,318 kg for singleton males) (p < 0.0001). Moreover, TW was linked to a decreased pregnancy risk and an increased risk of culling (p < 0.0001) both in short- and long-term analysis. These findings underline the need for targeted management strategies to mitigate the adverse effects of twinning in dairy herds, especially where TW rates are notably high. Full article

Here, we evaluated the effect of dietary supplementation with an Olea europaea L. extract on the animal welfare and milk quality of dairy cows. Thirty Italian Holstein–Friesian dairy cows in the mid-lactation phase (90 to 210 days) were blocked into experimental groups based on parity class (namely, primiparous (P) (n = 10), secondiparous (S) (n = 10) and pluriparous (PL) (n = 10)) and received, for 60 days, Phenofeed Dry® at 500 mg/cow/day. Milk and blood samples were collected before the start of the treatment (T0), subsequently every 15 days (T1–T4) and at 45 days after the end of treatment (T5). In the serum, glucose and triglycerides, stress, the thyroid, lactation and sex hormones were measured; in the milk, lysozyme content as well as the fatty acid profile were assessed. In the whole animal, the enriched feed helped to maintain hormonal parameters in the physiological range while producing hypoglycemic (T4 vs. T0, for P and PL p < 0.001) and hypolipidemic effects (T4 vs. T0, for P p < 0.001 and for PL p < 0.01). At the milk level, it resulted in a reduction in total fat (T5 vs. T0, for P, S and PL p < 0.001) and in the saturated fatty acids (SFAs)/monounsaturated fatty acids (MUFAs) ratio paralleled by an increase in polyunsaturated fatty acids (PUFAs) (T5 vs. T0, for P, S and PL p < 0.001), protein content (lysozyme (T4 vs. T0, for P and PL p < 0.001)) and lactose (T5 vs. T0, for P, S and PL p < 0.001). Thus, the inclusion of natural bioactive molecules such as O. europaea L. polyphenols in the dairy cow diet may help to improve animal welfare and milk quality. Full article

The utilization of corn silage in animal diets is becoming a challenge, due to the crop’s reduced yield as a result of climate change. Alternative silage types, such as mixtures of Italian ryegrass and winter cereals, may be a good complement to corn silage in diet formulation. Therefore, it is important to investigate how these alternative sources influence milk fatty acid and odor profile, as well as how these quality parameters could be efficiently evaluated. In this study, a corn silage-based control (CTR) and four experimental (EXP) diets—which contained winter cereals (WC), as well as WC with Italian ryegrass (IRG) silages in different proportions—were fed to Holstein-Friesian cows (n = 32) in a single-blinded efficacy study during a series of 4-week periods, with 2 weeks of adaption to each feed before the main trial. Milk from each trial was subjected to fatty acid (FA) analysis and odor profiling through the utilization of gas chromatography and an electronic nose, respectively. The results show that milk FAs in the EXP-3 and EXP-4 groups (which contained mixed silages using WC) changed the most when compared with other groups. Moreover, with a 7 kg/day inclusion rate of WC + IRG and of the WC silages in the diets of the EXP-2 and EXP-3 groups, respectively, the milk from these groups had their n6:n3 ratio reduced, thus indicating possible health benefits to consumers. The odor variation between the milk of the WC + IRG and WC groups was greater than the variation between the milk of the CTR and EXP groups. The main volatile compound responsible for the odor of the CTR milk was ethyl-butyrate, whereas 2-propanol and butan-2-one dominated the WC milk; the milk samples of the WC + IRG groups were influenced largely by ethanol. The study proved that with a 7 kg/day inclusion of mixed silages including winter cereals plus Italian ryegrass, the FA and odor profile of bovine milk could be modified. Full article

Estrus detection is very important for the profitability of dairy herds. Different automatic systems for estrus detection have been developed over the last decades. Our study aimed to assess the ability of the electronic nose (EN) MENT-EGAS prototype to detect estrus, based on odor release from the perineal headspace in dairy cattle by direct sampling. The study was performed in an Italian dairy farm using 35 multiparous Holstein–Friesian cows. The cows were divided into three groups: group I included 10 lactating 5-month pregnant cows, group II included 19 lactating cycling cows, and group III included 6 cows that were artificially inseminated 18 days before the trial. Odors from the perineal headspace were collected using the MENT-EGAS prototype. In group I, odors were collected once a day for 5 consecutive days. In group II, odors were collected twice daily from day 18 until day 1 of the reproductive cycle. In group III, odors were also collected twice daily from the presumable day 18 of gestation until day 22. Principal component analyses (PCA) of the perineal headspace samples were performed. PCA in group I revealed no significant discrimination. PCA in group II revealed clear discrimination between proestrus and estrus, and between estrus and metestrus but no significant discrimination was obtained between proestrus and metestrus. PCA in group III revealed that in four cows the results were similar to group I and in two cows the results were similar to group II. On day 40 of the presumable pregnancy, the ultrasound examination revealed that only the four cows were pregnant and the other two cows were regularly cycling. On the basis of our findings, we conclude that it is possible to accurately detect estrus in dairy cattle from directly collected odor samples using the MENT-EGAS prototype. This represents the first study of estrus detection using an EN detection by direct sampling. EN technologies, such as MENT-EGAS, could be applied in the future in dairy cattle farms as a precise, non-invasive method for estrus detection. Full article

The objectives of this study were to investigate the main risk factors associated with calving ease (CE) in Italian Holstein cow herds, and to estimate the association between CE and subsequent survival, fertility and milk production. Data obtained from Holstein cows in 40 Italian herds were retrospectively investigated. Calvings were surveilled and classified into two categories of CE, unassisted calving or assisted calving, based on the need for intervention. The following factors were analyzed as possible risk factor affecting CE: herd, number of calvings/herd/year, age at first calving (AFC), cow parity, gestation length, twinning, calf sex, previous calving-to-conception interval, previous milk yield, dry period and close-up length, and season of calving. The association between CE and culling risk within the first 30 days-in-milk (DIM), cumulative 60-d milk yield and predicted 305-d milk yield, and pregnancy risk within 150 DIM were also investigated. Of the 47,672 calvings, 37,892 (79.5%) were unassisted, while 9780 (20.5%) required some type of assistance. Among the risk factors, only the AFC was not correlated with CE, while for all the other risk factors an association with CE was detected. Assisted calvings were associated with an increased culling risk at 30 DIM, decreased 60-d milk yield, decreased 305-d milk yield and reduced pregnancy risk at 150 DIM. In conclusion, dairy herd management should aim at correcting/reducing the risk factors in order to limit the incidence of assisted calving, and possibly improve the quality of calving assistance; controlling CE within the herd is crucial to reducing culling risk, and achieving higher lactation and reproductive performance. The adoption of the mixer wagon and total mixed ration aimed to decrease dysmetabolic diseases and improve feed efficiency in dairy cows. Differences between theoretical and eaten diets are imputable to errors in diet preparation or cow feed sorting. We proposed a method to measure the chemical composition and particle size distribution of the ration and determined its peNDF content through a portable Near Infra-Red spectrophotometer that allowed the calculation of two indexes: the homogeneity and the sorting indexes. In a cohort of 19 Italian Holstein breeding farms, we studied the correlation of these indexes with the mixer wagon settings. Determination coefficients in the validation (Rv2) for dry matter, crude protein, aNDF, and starch were 0.91, 0.54, 0.86, and 0.67, respectively. The ration fractions (%, w/w of wet weight) retained by the 3.8 and 1.8 mm sieves, and the bottom showed Rv2 of 0.46, 0.49, and 0.53, respectively. The homogeneity index regressed negatively with the mixer wagon load fullness (R2 = 0.374). The homogeneity-binary classification showed an odds ratio of 1.72 for dry matter and 0.39 for aNDF (p < 0.05). The sorting-binary classification showed an odds ratio of 2.54 for aNDF (p < 0.05). The studied farms showed low peNDF values (median = 17.9%). Full article

One critical point of dairy farm management is calving and neonatal first care. Timely calving assistance is associated with the reduction of calf mortality and postpartum uterine disease, and with improved fertility in dairy cattle. This study aimed to evaluate the performance and profitability of an intravaginal sensor for the prediction of stage II of labor in dairy farms, thus allowing proper calving assistance. Seventy-three late-gestating Italian Holstein cows were submitted to the insertion of an intravaginal device, equipped with light and temperature sensors, connected with a Central Unit for the commutation of a radio-signal into a cell phone alert. The remote calving alarm correctly identified the beginning of the expulsive phase of labor in 86.3% of the monitored cows. The mean interval from alarm to complete expulsion of the fetus was 71.56 ± 52.98 min, with a greater range in cows with dystocia (p = 0.012). The sensor worked correctly in both cold and warm weather conditions, and during day- or night-time. The intravaginal probe was well tolerated, as any cow showed lesions to the vaginal mucosa after calving. Using sex-sorted semen in heifers and beef bull semen in cows at their last lactation, the economic estimation performed through PrecisionTree™ software led to an income improvement of 119 € and 123 €/monitored delivery in primiparous and pluriparous cows, respectively. Remote calving alarm devices are key components of “precision farming” management and proven to improve animal welfare, to reduce calf losses and to increase farm incomes. Full article

Stress in livestock reduces productivity and is a welfare concern. At a physiological level, stress is associated with the activation of inflammatory responses and increased levels of harmful reactive oxygen species. Biomarkers that are indicative of stress could facilitate the identification of more stress-resilient animals. We examined twenty-one metabolic, immune response, and liver function biomarkers that have been associated with stress in 416 Italian Simmental and 436 Italian Holstein cows which were genotyped for 150K SNPs. Single-SNP and haplotype-based genome-wide association studies were carried out to assess whether the variation in the levels in these biomarkers is under genetic control and to identify the genomic loci involved. Significant associations were found for the plasma levels of ceruloplasmin (Bos taurus chromosome 1—BTA1), paraoxonase (BTA4) and γ-glutamyl transferase (BTA17) in the individual breed analysis that coincided with the position of the genes coding for these proteins, suggesting that their expression is under cis-regulation. A meta-analysis of both breeds identified additional significant associations with paraoxonase on BTA 16 and 26. Finding genetic associations with variations in the levels of these biomarkers suggests that the selection for high or low levels of expression could be achieved rapidly. Whether the level of expression of the biomarkers correlates with the response to stressful situations has yet to be determined. Full article

The growing interest of consumers for milk and dairy products of high nutritional value has pushed researchers to evaluate the feasibility of including fatty acids (FA) in selection programs to modify milk fat profile and improve its nutritional quality. Therefore, the aim of this study was to estimate genetic parameters of FA profile predicted by mid-infrared spectroscopy, milk yield, composition, and total and differential somatic cell count. Edited data included 35,331 test-day records of 25,407 Italian Holstein cows from 652 herds. Variance components and heritability were estimated using single-trait repeatability animal models, whereas bivariate repeatability animal models were used to estimate genetic and phenotypic correlations between traits, including the fixed effects of stage of lactation, parity, and herd-test-date, and the random effects of additive genetic animal, cow permanent environment and the residual. Heritabilities and genetic correlations obtained in the present study reflected both the origins of FA (extracted from the blood or synthesized de novo by the mammary gland) and their grouping according to saturation or chain length. In addition, correlations among FA groups were in line with correlation among individual FA. Moderate negative genetic correlations between FA and milk yield and moderate to strong positive correlations with fat, protein, and casein percentages suggest that actual selection programs are currently affecting all FA groups, not only the desired ones (e.g., polyunsaturated FA). The absence of association with differential somatic cell count and the weak association with somatic cell score indicate that selection on FA profile would not affect selection on resistance to mastitis and vice versa. In conclusion, our findings suggest that genetic selection on FA content is feasible, as FA are variable and moderately heritable. Nevertheless, in the light of correlations with other milk traits estimated in this study, a clear breeding goal should first be established. Full article

Food antioxidants enhance products shelf life and stability during technological treatments through the maintenance of their physical and chemical properties. Moreover, they are endowed with several positive effects on human health, including cell membranes preservation, enzyme functionality, and DNA integrity. Milk has been described in relation to a wide array of fat soluble and water-soluble antioxidant compounds, in particular vitamin A, C, and E, lactoferrin and peptides derived from casein and whey proteins. The total antioxidant activity (TAA) of milk is a novel and scarcely explored trait, defined as the sum of antioxidant contributions of the aforementioned compounds. On this background, the aims of the present study were to investigate the variability of milk TAA on a large scale exploiting predictions obtained through mid-infrared (MIR) spectroscopy and to estimate genetic parameters of this trait in Holstein cows. Individual milk samples were collected between January 2011 and December 2018 during the routine milk recording procedure. Samples were analysed for gross composition through MIR spectroscopy and MIR spectra were stored. Milk TAA was then predicted (pTAA) from the stored milk MIR spectra (111,653 test-day records of 9519 cows in 344 herds) using the previously developed prediction model; considering the prediction accuracy, pTAA might be considered a proxy of the TAA determined through the reference method. Overall, pTAA averaged 7.16 mmoL/L of Trolox equivalents, showed a nadir around 40 days after calving and increased thereafter, following a linear trend up to the end of lactation. The lowest pTAA was observed in milk sampled from June to September. Milk pTAA was heritable (0.401 ± 0.015) and genetically associated to fat yield (0.366 ± 0.049), crude protein (CP) yield (0.238 ± 0.052), fat percentage (0.616 ± 0.022) and CP percentage (0.754 ± 0.015). The official selection index of Italian Holstein put the 49% of the emphasis on fat and protein yield and percentage; therefore, it derives that an indirect favourable selection for milk pTAA should be already in progress in Italian Holstein population. In spite of the impressive advancements observed on both management and genetic factors, udder health still represents one of most demanding objectives to be attained in the dairy cattle industry. Udder morphology and especially teat condition might represent the first physical barrier to pathogens’ access. The objectives of this study were to investigate the genetic component of teat condition and to elucidate its relationship with both milk yield and somatic cell scores in dairy cattle. Moreover, the effect of selection for both milk yield and somatic cell scores on teat condition was also investigated. A multivariate analysis was conducted on 10,776 teat score records and 30,160 production records from 2469 Italian Holstein cows. Three teat scoring traits were defined and included in the analysis. Heritability estimates for the teat score traits were moderate to low, ranging from 0.084 to 0.238. When teat score was based on a four-classes ordinal scoring, its genetic correlation with milk yields and somatic cell score were 0.862 and 0.439, respectively. The scale used to classify teat-end score has an impact on the magnitude of the estimates. Genetic correlations suggest that selection for milk yield could deteriorate teat health, unless more emphasis is given to somatic cell scores. Considering that both at national and international level, the current selection objectives are giving more emphasis to health traits, a further genetic deterioration in teat condition is not expected. The aim of this study was to assess the effect of heat stress on dairy cow performance and on the expression of selected genes involved in milk protein metabolism. Eight Italian Holstein Friesian cows were kept under thermoneutral conditions (temperature–humidity index (THI) < 72, CON) for 8 days and under mild heat stress conditions (72 < THI < 78, HS) for an additional 8 days. The rectal temperature, feed intake, and milk yield were recorded during the last 3 days of the CON and HS periods. During the same time period, milk samples were collected to assess the composition and expression of selected genes involved in milk protein metabolism. Gene expression analyses were performed on somatic cells from milk, which are representative of mammary tissue. In terms of dairy cow performance, HS resulted in lower milk and protein yields and feed intake but higher rectal temperature than for CON (p < 0.05). Under HS, there were greater abundances of HSPA1A (p < 0.05) and BCL2 (p < 0.05), compared to CON, but similar levels of CSN2 (p > 0.05), CSN3 (p > 0.05), HSPA8 (p > 0.05), and STAT5B (p > 0.05) mRNA. Mild heat stress reduced the performance of dairy cows without affecting the expression of genes coding for caseins. Full article

Effects of Aloe arborescens Whole Plant Homogenate on Lipid Metabolism, Inflammatory Conditions and Liver Function of Dairy Cows during the Transition Period

The anti-hyperlipidemic and anti-inflammatory effects exerted by Aloe on monogastric mammals suggest it as a potential strategy to address the tremendous metabolic alterations that affect dairy cows during their transition to calving. A group of 20 multiparous Italian Holstein dairy cows were housed in freestalls and allocated into two homogeneous groups to receive either 200 g/d of water (CTR) or 200 g/day of Aloe arborescens Mill. whole plant homogenate through a rumen tube (AAM) between −14 and 14 days from calving (DFC). From −14 to 35 DFC, the BCS, and milk yield were measured, and blood samples were collected to assess the hematochemical profile. Data underwent ANOVA testing using a mixed model for repeated measurements, including the treatment and time and their interactions as fixed effects. Compared to CTR cows, AAM cows had a less pronounced BCS loss in early lactation (p < 0.01), indicating less mobilization of body reserves. Compared to CTR cows, AAM cows had a lower plasma concentration of nonesterified fatty acids and beta hydroxybutyrate (p < 0.01 and = 0.01 respectively) that, paired with the lower butterfat content and fat/protein ratio in their milk (p = 0.03 and < 0.01 respectively), indicates that Aloe reduced the mobilization of body fats. AAM cows had a reduced concentration of myeloperoxidase in plasma and a lower SCC in milk compared to CTR cows (p = 0.02 for both), indicating an anti-inflammatory effect of Aloe. Furthermore, AAM cows had a lower plasma concentration of ceruloplasmin (p < 0.05) and higher plasma concentration of cholesterol, retinol, and paraoxonase compared to CTR cows (p < 0.01, < 0.01 and < 0.05 respectively), indicating Aloe was effective in mitigating the acute phase response in early lactation. Finally, AAM cows had lower plasma creatinine concentrations around calving (p < 0.05), a lower concentration of plasma bilirubin, and a higher concentration of plasma tocopherol compared to CTR cows (p = 0.01 for both). These data suggest Aloe has anti-hyperlipidemic and anti-inflammatory effects on transition dairy cows that could have ameliorated liver and kidney function disruption and increased the availability of body antioxidants in early lactation. The majority of proteins in cow’s milk are caseins, which occur in four groups (α-s1, α-s2, β, and k) encoded by different genes (CSN1S1, CSN1S2, CSN2, and CSN3, respectively). In this study, we focused on the β-casein allele variants A1 and A2 due to their influence on milk’s technological characteristics and human health. Digestion of the β-casein variant A1 leads to the formation of β-casomorphin 7 (BCM-7), a bioactive peptide that has been suggested to be a possible cause of various human diseases and associated with low milk digestibility. The potential negative role of the β-casein variant A1 in human health has stimulated the planning of cattle breeding programs based on genetic selection to increase the frequency of the A2 variant, which is associated with increased milk digestibility. The aim of this work was to evaluate the frequencies of the different β-casein variants in Italian Holstein Friesian dairy cows from cattle farms located in central Italy to select a population of A2 homozygous animals. β-casein genotypes were identified by evaluating the presence of single nucleotide polymorphisms (SNPs) of the CSN2 gene using PCR and sequencing analysis. The frequency of the desirable β-casein variant A2 in the studied bovine population was 0.61. The frequency of the undesirable A1 variant in the studied bovine population was 0.30. The frequency of the A2 allele was higher than expected for the breed; therefore, genetic selection for the A2 variant in these animals could be achieved in a fairly short time using A2 homozygous bulls.
