The Anti-Müllerian hormone (AMH) is widely recognized for promoting Müllerian duct regression in higher vertebrates and regulating key reproductive functions like steroidogenesis, folliculogenesis, and Leydig cell development. In teleost fish, which lack Müllerian ducts, Amh primarily influences male reproductive functions, including sex determination, testis differentiation, and germ cell proliferation. In adult fish, Amh supports gonad development and spermatogenesis, but its role in teleost gonadal physiology remains largely underexplored. This study reveals a novel steroidogenic function in the European sea bass (Dicentrarchus labrax) using in vitro testis culture, in vivo plasmid injection, and cell-based transactivation assays. The Amh-induced significant increase in androgen levels was also confirmed in Japanese medaka (Oryzias latipes) treated with recombinant sea bass Amh. Beyond activating the canonical Smad pathway, Amh also triggered the cAMP/PKA signalling pathway via its cognate type II receptor, Amhr2. Inhibitors of these pathways independently and synergistically counteracted Amh-induced CRE-Luc activity, indicating pathway crosstalk. Moreover, inhibition of the cAMP pathway suppressed Amh-induced androgen production in testis cultures, emphasizing the crucial role of protein kinase A in mediating Amh steroidogenic action. These findings uncover a novel steroidogenic function of Amh in teleosts and highlight its broader role in male reproductive physiology. Full article

Background: Fish escape events from aquaculture facilities are increasing and pose significant ecological, economic, and traceability concerns. Accurate methods to differentiate between wild, cultured, and escaped fish are essential for fishery management and seafood authentication. Methods: This study analyzed muscle tissue from Sparus aurata, Dicentrarchus labrax, and Argyrosomus regius using a multiomics approach. Heavy metals were quantified by ICP-MS, fatty acid profiles were assessed via GC-MS, and metabolomic and lipidomic signatures were identified using 1H NMR spectroscopy. Multivariate statistical models (MDS and PLS-LDA) were applied to classify fish origins. Results: Wild seabream showed significantly higher levels of arsenic (9.5-fold), selenium (3.5-fold), and DHA and ARA fatty acids (3.2-fold), while cultured fish exhibited increased linoleic and linolenic acids (6.5-fold). TMAO concentrations were up to 5.3-fold higher in wild fish, serving as a robust metabolic biomarker. Escaped fish displayed intermediate biochemical profiles. Multivariate models achieved a 100% classification accuracy across species and analytical techniques. Conclusions: The integration of heavy metal analysis, fatty acid profiling, and NMR-based metabolomics enables the accurate differentiation of fish origin. While muscle tissue provides reliable biomarkers relevant to human exposure, future studies should explore additional tissues such as liver and gills to improve the resolution of traceability. These methods support seafood authentication, enhance aquaculture traceability, and aid in managing the ecological impacts of escape events. Full article

Synthetic flavors from standardized processes have recently emerged as a promising and sustainable alternative to traditional feed attractants. In this study, two attractive (F25, cheese; F35, caramel) and one repulsive (F32-, coconut) synthetic flavors were individually added (1% w/w) to a commercial diet for European seabass (Dicentrarchus labrax) and tested over a 90-day feeding trial (30 fish per tank, in triplicate; initial weight 72.48 ± 8.04 g) to assess their impact on fish growth performance, welfare, and the modulation of brain appetite and monoaminergic pathways. None of the tested flavors negatively affected overall fish health. The F35 flavor enhanced feed intake (90.1 ± 5.6%) and growth (SGR 2.2 ± 0.2%) and positively influenced appetite-related and monoaminergic signals, thus being more effective than the F25 one (80.4 ± 3.2 and 1.6 ± 0.1%, respectively). A weekly feeding rotation between F35 and F25 (ROT group) resulted in suboptimal outcomes compared to F35 administration alone. The F32- flavor was not clearly perceived as strongly aversive by seabass and did not impair zootechnical performance. These findings highlight the potential of attractive synthetic flavors to improve diet palatability in a carnivorous species of commercial value, offering novel insights for more sustainable and cost-effective aquaculture feeding strategies. The anti-Müllerian hormone (amh) and its receptor, amhr2, along with the downstream bone morphogenetic protein receptors (bmprs), have been recognized as the central regulators in teleost sex determination (SD) and differentiation. However, their evolution and function in reproduction among diverse teleost lineages may represent species-specific patterns and still need more explanation. In this study, systematic investigations of amh signaling genes, including amh, amhy (Y-linked paralog of amh), amhr2, bmpr1, and bmpr2, were conducted among teleost species. The results revealed generally conserved gene copy number, phylogeny, structure, and synteny, among teleost amh signaling genes. Notably, significantly accelerated evolutionary rates (dN/dS) were found in teleost amhy compared to amh, and amh exhibited faster molecular evolution in amhy-SD teleosts than in non-amhy-SD teleosts, suggesting their enhanced evolutionary plasticity in teleosts. Expression profiling identified testis-biased expression of the most amh signaling genes in fish species with different SD genes and mechanisms, including Lateolabrax maculatus and Dicentrarchus labrax from Order Perciformes, Cynoglossus semilaevis and Paralichthys olivaceus from Order Pleuronectiformes, and Salmo salar and Oncorhynchus mykiss from Order Salmoniformes, with ovary-biased expression also found in Salmoniformes. A weighted gene co-expression network analysis further uncovered strong species-specific functional interactions between amh signaling components and genes of germ-cell development, the meiotic process, etc. Collectively, the integrated evidence from this study supports the hypothesis that amh signaling provides the key molecules governing sex differentiation in a species-specific manner in diverse teleost lineages, independent of its SD role, and interacts with functions of both testis and ovary development. This article documents the first outbreak of aeromoniasis caused by Aeromonas veronii in farmed European seabass (Dicentrarchus labrax) along the Greek Ionian Sea coast. In late spring 2024, commercially sized fish exhibited anorexia, hemorrhages, and ulcers on the skin, accompanied by elevated morbidity and mortality rates. The outbreak spread rapidly across local farms in Sagiada Bay, reaching its peak in late summer, and extending into the Astakos Gulf, southern in the Ionian Sea. The postmortem examination revealed hemorrhages, organomegaly, abscess formation, and granulomatous inflammation. Aeromonas veronii was isolated from all examined individuals in nutrient media and confirmed by biochemical and molecular methods. Whole genome sequencing and phylogenetic analysis demonstrated genetic homogeneity among two strains from two different areas along the Ionian Sea and a close evolutionary relationship with other Aeromonas veronii strains from the Aegean Sea. Although genetically similar, the isolates exhibited differences in phenotypic and biochemical characteristics, indicating regional variability. The present study provides an overview of the pathology, clinical characteristics and progression of aeromoniasis in Ionian Sea aquaculture, highlighting the need for continued monitoring, in-depth genomic and phenotypic assessment, and the design of region-specific preventive strategies, including autogenous vaccines, for effective disease management. Argonauta argo (Linnaeus, 1758), commonly known as the greater argonaut, is a pelagic octopod with a circumglobal distribution in warm and temperate seas. Although the species is occasionally reported in the Mediterranean Sea, confirmed in situ observations of live individuals remain scarce, particularly in the eastern basin. Here, we document the occurrence of a live female A. argo entangled in aquaculture nets at a sea bass (Dicentrarchus labrax) facility off the coast of Lesvos Island in the northeastern Aegean Sea, Greece. This represents the first confirmed observation in this part of the Mediterranean and only the second confirmed observation of a live individual in Greek waters. The individual was photographed in situ and identified based on diagnostic morphological features. The encounter occurred under specific hydrodynamic and environmental conditions potentially favorable to the species, including enhanced prey availability and structural refuge within the aquaculture infrastructure. This record contributes to the limited dataset on Mediterranean argonauts and underscores the need for further research into their ecology, distribution patterns, and potential interactions with anthropogenic marine structures. Growing health and environmental concerns have intensified the search for natural antioxidants to replace synthetic alternatives to prevent spoilage of seafood. Long-term intake of synthetic antioxidants has been linked to liver toxicity, reproductive issues, and cancer risks in animals. This study therefore evaluates the efficacy of Aloe vera gel (AVG) as a natural antioxidant in preserving the quality of sea bass (Dicentrarchus labrax, Linnaeus 1758) slices during cold storage at 4 ± 1 °C for 13 days. Sea bass slices were coated with 100% and 75% AVG and analyzed for physical (color and texture), chemical (pH, TVB-N, TBARS, and PV), and sensory changes. Results showed that AVG significantly reduced lipid oxidation, as indicated by lower peroxide (PV) and thiobarbituric acid reactive substances (TBARS) values in treated samples compared to controls. While sensory, color, and texture parameters remained consistent across all groups, 75% AVG-coated slices extended the shelf life by four days, and the 100% AVG treatment achieved a two-day extension, based on TVB-N values. These findings highlight AVG’s potential as a natural, eco-friendly alternative to synthetic antioxidants for aquatic product preservation. Full article

This review examines the current state of the supply chain management for Dicentrarchus labrax (sea bass) and Sparus aurata (sea bream), two key commercial fish species in the Mediterranean. It provides a comprehensive analysis of sustainable innovations in aquaculture, processing, and packaging, with particular attention to circular economy-based biopreservation techniques. A major focus is on the Integrated Multi-Trophic Aquaculture (IMTA) system, an advanced farming approach that enhances sustainability, promotes circular resource utilization, and improves fish welfare. By fostering ecological balance through the co-cultivation of multiple species, IMTA contributes to the overall quality of fish products for human consumption. Beyond aquaculture, the review addresses the critical challenge of food loss, which stems from the high perishability of fish during storage and processing. In this regard, it highlights recent advancements in biopreservation strategies, including the application of antagonistic microorganisms, their metabolites, and plant-derived extracts. Particular attention is given to the development of edible antimicrobial films, with a focus on the valorization of citrus processing by-products for their production. By centering on innovations specific to the Mediterranean context, this review underscores that a holistic, integrative approach to supply chain management is essential for transitioning the aquaculture sector toward greater efficiency and sustainability. This study investigates the effects of pineapple by-products on feed preservation during storage at two different temperatures (25 °C and 4 °C) and on European sea bass (Dicentrarchus labrax) stress resistance. Four isoproteic, isolipidic, and isoenergetic diets were manufactured: CTRL—negative control, commercial diet without added antioxidants; VITE—positive control, CTRL diet with 100 mg kg−1 of vitamin E; and P2 and S2—VITE diet with 2% pineapple peel or stem flour, respectively. The fish (13.5 ± 0.8 g) were split into four replicate groups per diet and fed ad libitum for 12 weeks, after which they were subjected to a stress challenge of air exposure (1 min) followed by confinement (5 min, 100 kg m−3). Despite storage time lowering the antioxidant properties of all diets, P2 and S2 showed increased antioxidant capacity (DPPH, ABTS•+, and ORAC) before and after storage. The diets were well accepted by the fish, and the VITE-fed fish showed significantly lower lipid peroxidation values in the liver and muscle compared to all remaining diets. However, pineapple by-product inclusion did not result in increased fish stress resistance. Further optimization is required for the successful use of pineapple by-products as natural antioxidants in aquafeeds. Full article

The six6 and vgll3 genes play evolutionarily conserved roles in developmental processes and life history traits across species, including teleosts. Notable differences in genotype and allele frequencies of these genes have been observed between farmed and wild populations of European seabass and gilthead seabream, suggesting potential roles in traits associated with domestication. Here, we hypothesized that genetic variations in the six6 and vgll3 are associated with distinct expression profiles that underlie domestication-related traits in the two species. Using quantitative PCR (qPCR), we examined the expression profiles of these genes in early developmental stages across genotypes linked to domestication. Our results confirmed that vgll3 and six6 genotypes significantly influenced their expression in gilthead seabream, with statistically significant differences between genotypes. In European seabass, six6 expression did not significantly differ among genotypes, although heterozygous larvae showed higher variability that decreased at the juvenile stage. Meanwhile, no genetic variation was observed in vgll3, precluding genotype-specific expression analysis. Altogether, our findings provide the first evidence confirming that prior DNA-based associations of these genes with domestication traits are reflected at the mRNA level, particularly in vgll3 in gilthead seabream. This highlights the potential functional relevance of these genetic variations in shaping expression profiles linked to domestication in the species. Full article

Fish viral infections have great environmental and economic implications in aquaculture. Nervous necrosis virus (NNV) is a pathogen affecting more than 120 different species, causing high mortality and morbidity. Herein, we study how NNV re-infection affects the European sea bass (Dicentrarchus labrax, L.) head kidney transcriptome in disease-resistant and -susceptible sea bass families. To determine how each family responds to re-infection, we performed the RNA-sequencing analysis of experimentally NNV-infected D. labrax. Fish were experimentally infected in a long-term study, and one month after the last recorded death, all surviving fish were re-infected by the same NNV strain. Fish tissues were sampled 7 days upon re-infection. The transcriptome profiles of infected vs. non-infected fish revealed 103 differentially expressed genes (DEGs) for the resistant family and 336 DEGs for the susceptible family. Only a few pathways were commonly enriched in the two families, further indicating that the resistant and susceptible families utilize completely different mechanisms to fight the NNV re-infection. Protein–protein interaction analysis identified a variety of hub genes for the resistant and the susceptible families, quite distinct in their function on NNV resistance. In conclusion, NNV-resistant and -sensitive sea bass transcriptomes were analyzed following NNV survivors’ viral re-infection, offering a glimpse into how host attempts to control the infection depending on its genetic background in relation with virus resistance. Brewer’s spent grain (BSG) is one such by-product. Solid-state fermentation (SSF) of BSG with Aspergillus ibericus enhances its nutritional value and digestibility for European seabass. The present study further evaluates the potential of dietary inclusion of BSG-SSF on growth performance, feed utilization, plasma metabolite profile, intermediary metabolism, and oxidative status of European seabass juveniles compared to the unfermented product. A practical diet (45% protein; 18% lipids) was tested against diets incorporating 10% or 20% of BSG or BSG-SSF, replacing plant-protein feedstuffs. Triplicate groups of European seabass juveniles (49 g initial weight) were fed for 10 weeks. Unfermented BSG (10% and 20%) reduced growth and feed efficiency. In comparison, the 20% BSG-SSF diet promoted growth and feed efficiency similar to the control group, while the 10% BSG-SSF diet surpassed the control diet. Whole-body protein content was unaffected, but lipid and energy content decreased with increasing BSG levels, regardless of fermentation. Plasma glucose and phospholipid levels and hepatic activities of glucokinase and malic enzymes decreased with increasing BSG, irrespective of fermentation. BSG-SSF incorporation increased plasma triglyceride levels and decreased hepatic transaminase activities but did not affect hepatic key enzyme activity of β-oxidation or lipogenesis. It also reduced antioxidant enzyme activity and lipid peroxidation. In conclusion, BSG negatively impacted growth performance, while BSG-SSF supported inclusion levels up to 20% without performance loss. Further, the 10% BSG -SSF diet outperformed the control diet. This study investigates the effects on hard-bottom macro-zoobenthic communities of converting a mariculture plant into an Integrated Multi-Trophic Aquaculture (IMTA) system. This study was conducted from 2018 to 2021 in the semi-enclosed Mar Grande basin of Taranto (northern Ionian Sea), on a facility located 600 m off the coastline, with a production capacity of 100 tons per year of seabass (Dicentrarchus labrax) and seabream (Sparus aurata). The results from seasonal sampling performed in a treatment site, where bioremediators (filter-feeding invertebrates such as sponges, polychaetes, mussels, and macroalgae) were deployed, and a control site without bioremediators were compared. Before the IMTA installation, the hard substrates under the cages were sparsely inhabited, with significant sediment coverage. By 2021, the treatment site exhibited revitalized and more diverse macro-zoobenthic communities, with species richness increasing from 83 taxa in 2018 to 104 taxa, including notable growth in sponges, annelids, mollusks, and bryozoans. In contrast, the control site showed no substantial changes in biodiversity over the same period. Biodiversity indices, including Shannon and Margalef indices, improved significantly at the treatment site, particularly during summer months, highlighting a more resilient and balanced benthic environment. Taxonomic distinctness (delta+) and multivariate analyses (PERMANOVA, PCO) confirmed significant spatial and temporal shifts in community structure at the treatment site, driven by IMTA implementation. SIMPER analysis identified key taxa contributing to these changes, which played a pivotal role in structuring the community. The emergence of filter feeders, predators, and omnivores at the treatment site suggests enhanced nutrient cycling and trophic complexity, while the decline in opportunistic species further supports improved environmental conditions. These findings demonstrate the potential of IMTA to promote recovery and sustainable mariculture practices, also offering a comprehensive understanding of its positive effects on hard-bottom benthic community dynamics. Tenebrio molitor (TM) meal is a potential alternative ingredient to plant and fishmeal in aquafeeds due to its high protein content, digestibility, and low environmental impact. However, its low n-3 PUFA (EPA and DHA) content represents one of its main limitations. This study evaluated the effects of replacing fishmeal and plant-derived ingredients (plant proteins and soybean oil) with full-fat TM meal on the growth performance and lipid composition of tissues in juvenile European seabass (Dicentrarchus labrax). Four experimental diets, including 5–10% TM for replacing fishmeal and soybean oil (FM5 and FM10) and 10–20% TM for substituting plant protein ingredients and soybean oil (PI10 and PI20), were tested over 49 days against a control diet (CT). Fish fed on PI20 exhibited the highest growth performance (final weight: 49.73 g and SGR: 1.78% day−1) compared to fish fed on FM (final weight: 47.0 g and SGR: 1.62% day−1) and CT (final weight: 48.45 g and SGR: 1.71% day−1). In general, the diets containing TM modulated the lipid profile in fish tissues by reducing the liver lipid content and muscle triglycerides and increasing the n-3/n-6 ratio compared to the CT diet. However, the most effective approach for modulating those parameters seems to be using the TM meal for replacing the plant ingredients instead of substituting fishmeal. Moreover, this feeding strategy, demonstrated to be useful for promoting the growth of fish owing to the use of the TM meal for replacing fishmeal, resulted in a slight reduction in the final weight of the fish. Host–pathogen interactions and the design of vaccines for aquaculture fish viruses are challenging and call for innovative approaches. This study explores the potential of adenoviral (Ad) vectors Ad5 and chimeric Ad5/40 as gene delivery tools for fish brain cells susceptible to neurotropic viruses. For this purpose, European sea bass (Dicentrarchus labrax) DLB-1 and gilthead seabream (Sparus aurata) SaB-1 brain cell lines were infected with Ad5 or Ad5/40 vectors expressing GFP, and we evaluated their capacity for infection by fluorescence microscopy and flow cytometry, as well as their antiviral innate immune response by the transcription of gene markers (irf3 and mx). We found that both vectors are able to infect DLB-1 and SaB-1 brain cell lines to similar levels, as demonstrated by fluorescence microscopy and flow cytometry, though the infection efficiency was low. In addition, infection with Ad vectors regulated the transcription of genes related to the interferon-mediated antiviral immune response. Our results indicate that the Ad5/40 vector achieves better infection and consistent cellular distribution. These findings suggest that these vectors may offer targeted gene delivery and local immune responses. Full article
